Institute for Multiscale Modeling of Biological Interactions

The Institute for Multiscale Modeling of Biological Interactions (IMMBI) has two primary goals: Foster interdisciplinary collaborations among faculty and their research laboratories that will lead to novel applications of multiscale simulation and modeling methods in the biological sciences and engineering; and Building on the unique biophysical/biology-based engineering foundations of the participating faculty, train scientists and engineers to apply computational methods that collectively span multiple time and length scales of biological organization. The success of IMMBI will be defined by the following: Size and quality of the applicant pool for pre-doctoral and post-doctoral fellows; Academic performance; Quality of the pre-doctoral and post-doctoral research; Impact of the research broadly and to the DOE (ASCR program) mission; Distinction of the next career step for pre-doctoral and post-doctoral fellows; and Faculty collaborations that result from IMMBI activities. Specific details about accomplishments during the three years of DOE support for IMMBI have been documented in Annual Progress Reports (April 2005, June 2006, and March 2007) and a Report for a National Academy of Sciences Review (October 2005) that were submitted to DOE on the dates indicated. An overview of these accomplishments is provided.

Optical People Counting for Demand Controlled Ventilation: A Pilot Study of Counter Performance

This pilot scale study evaluated the counting accuracy of two people counting systems that could be used in demand controlled ventilation systems to provide control signals for modulating outdoor air ventilation rates. The evaluations included controlled challenges of the people counting systems using pre-planned movements of occupants through doorways and evaluations of counting accuracies when naive occupants (i.e., occupants unaware of the counting systems) passed through the entrance doors of the building or room. The two people counting systems had high counting accuracy accuracies, with errors typically less than 10percent, for typical non-demanding counting events. However, counting errors were high in some highly challenging situations, such as multiple people passing simultaneously through a door. Counting errors, for at least one system, can be very high if people stand in the field of view of the sensor. Both counting system have limitations and would need to be used only at appropriate sites and where the demanding situations that led to counting errors were rare.

The Advanced Helical Generator (open access)

The Advanced Helical Generator

A high explosive pulsed power (HEPP) generator called the Advanced Helical Generator (AHG) has been designed, built, and successfully tested. The AHG incorporates design principles of voltage and current management to obtain a high current and energy gain. Its design was facilitated by the use of modern modeling tools as well as high precision manufacture. The result was a first-shot success. The AHG delivered 16 Mega-Amperes of current and 11 Mega-Joules of energy to a quasi-static 80 nH inductive load. A current gain of 154 times was obtained with a peak exponential rise time of 20 {micro}s. We will describe in detail the design and testing of the AHG.

The Case for the Younger Dryas Extraterrestrial Impact Event: Mammoth, Megafauna and Clovis Extinction

The onset of>1000 years of Younger Dryas cooling, broad-scale extinctions, and the disappearance of the Clovis culture in North America simultaneously occurred 12,900 years ago followed immediately by the appearance of a carbon-rich black layer at many locations. In situ bones of extinct megafauna and Clovis tools occur only beneath this black layer and not within or above it. At the base of the black mat at 9 Clovis-age sites in North America and a site in Belgium numerous extraterrestrial impact markers were found including magnetic grains highly enriched in iridium, magnetic microspherules, vesicular carbon spherules enriched in cubic, hexagonal, and n-type nanodiamonds, glass-like carbon containing Fullerenes and nanodiamonds, charcoal, soot, and polycyclic aromatic hydrocarbons. The same impact markers were found mixed throughout the sediments of 15 Carolina Bays, elliptical depressions along the Atlantic coast, whose parallel major axes point towards either the Great Lakes or Hudson Bay. The magnetic grains and spherules have an unusual Fe/Ti composition similar to lunar Procellarum KREEP Terrane and the organic constituents are enriched in 14C leading to radiocarbon dates often well into the future. These characteristics are inconsistent with known meteorites and suggest that the impact was by a previous unobserved, possibly extrasolar …

A letter report issued by the Government Accountability Office with an abstract that begins "In order to participate in federal student financial aid programs, law schools must be accredited by an agency recognized by the Department of Education (Education). Accreditation is intended to ensure that schools provide basic levels of quality in their educational programs, and Education recognizes those accrediting agencies that it concludes can reliably determine the quality of education provided by the schools and programs they accredit. The American Bar Association's (ABA) Council of the Section of Legal Education and Admissions to the Bar serves as the main accrediting agency for law schools, and students who attend one of the 200 ABA-accredited law schools can take the bar examination in any jurisdiction in the country. There are also several law schools that are accredited by other Education-recognized accrediting agencies such as the Western Association of Schools and Colleges. Graduates of some of these non-ABA-accredited law schools are eligible to take the bar examination in their own state, but may not do so nationwide. Identification of Novel Cell Wall Components

Our DOE Biosciences-funded work focused on the fungal cell wall and morphogenesis. We are especially interested in how new cell wall material is targeted to appropriate areas for polar (asymmetric) growth. Polar growth is the only way that filamentous fungi explore the environment to find suitable substrates to degrade. Work funded by this grant has resulted in a total of twenty peer-reviewed publications. In work funded by this grant, we identified nine Aspergillus nidulans temperature-sensitive (ts) mutants that fail to send out a germ tube and show a swollen cell phenotype at restrictive temperature, the swo mutants. In other organisms, a swollen cell phenotype is often associated with misdirected growth or weakened cell walls. Our work shows that several of the A. nidulans swo mutants have defects in the establishment and maintenance of polarity. Cloning of several swo genes by complementation also showed that secondary modification of proteins seems is important in polarity. We also investigated cell wall biosynthesis and branching based on leads in literature from other organisms and found that branching and nuclear division are tied and that the cell wall reorganizes during development. A letter report issued by the Government Accountability Office with an abstract that begins "Concerns exist that a more severe pandemic outbreak than 2009's could cause large numbers of people staying home to increase their Internet use and overwhelm Internet providers' network capacities. Such network congestion could prevent staff from broker-dealers and other securities market participants from teleworking during a pandemic. The Department of Homeland Security (DHS) is responsible for ensuring that critical telecommunications infrastructure is protected. GAO was asked to examine a pandemic's impact on Internet congestion and what actions can be and are being taken to address it, the adequacy of securities market organizations' pandemic plans, and the Securities and Exchange Commission's (SEC) oversight of these efforts. A letter report issued by the Government Accountability Office with an abstract that begins "Since 2005, the Department of Veterans Affairs (VA) has been undertaking an initiative to develop an integrated financial and asset management system known as the Financial and Logistics Integrated Technology Enterprise (FLITE). FLITE is the successor to an earlier initiative known as the Core Financial and Logistics System (CoreFLS) that the department undertook in 1998 and discontinued in 2004 because it failed to support VA's operations. In light of the past performance of CoreFLS and the Office of Management and Budget's designation of FLITE as high risk, GAO was asked to (1) determine the status of pilot system development and (2) evaluate key program management processes, including VA's efforts to institute effective human capital management, develop a reliable program cost estimate, use earned value management (a recognized means for measuring program progress), establish a realistic program schedule, employ effective requirements development and management, and perform independent verification and validation. STEADY STATE FLAMMABLE GAS RELEASE RATE CALCULATION AND LOWER FLAMMABILITY LEVEL EVALUATION FOR HANFORD TANK WASTE

This report assesses the steady state flammability level under off normal ventilation conditions in the tank headspace for 28 double-shell tanks (DST) and 149 single shell-tanks (SST) at the Hanford Site. Flammability was calculated using estimated gas release rates, Le Chatelier's rule, and lower flammability limits of fuels in an air mixture. This revision updates the hydrogen generation rate input data for all 177 tanks using waste composition information from the Best Basis Inventory Detail Report (data effective as of August 4,2008). Assuming only barometric breathing, the shortest time to reach 25% of the lower flammability limit is 11 days for DSTs (i.e., tank 241-AZ-10l) and 36 days for SSTs (i.e., tank 241-B-203). Assuming zero ventilation, the shortest time to reach 25% of the lower flammability limit is 10 days for DSTs (i.e., tank 241-AZ-101) and 34 days for SSTs (i.e., tank 241-B-203).

SYNTHETIC SLING FAILURE - EVALUATIONS & RECOMMENDATIONS

The information and evaluations provided in this report were compiled to address the recurring problem of synthetic sling failure. As safety is the number one priority in all work aspects, a solution must be devised to prevent accidents from occurring. A total of thirteen cases regarding synthetic sling failure were evaluated in order to determine their causes, effects, and preventative measures. From the collected data, it was found that all cases in which the synthetic sling contacted the edge of its load resulted in sling failure. It is required that adequate synthetic sling protection devices be used to protect slings in any lift where the sling comes in direct contact with the edge or corner of its load. However, there are no consensus codes or standards stating the type, material, or purpose of the type of protective device used to protect the sling from being cut. Numerous industry standards and codes provide vague descriptions on how to protect synthetic slings. Without a clear, concise statement of how to protect synthetic slings, it is common for inadequate materials and sling protection devices to be used in an attempt to meet the intent of these requirements. Update and Improve Subsection NH –– Alternative Simplified Creep-Fatigue Design Methods

This report described the results of investigation on Task 10 of DOE/ASME Materials NGNP/Generation IV Project based on a contract between ASME Standards Technology, LLC (ASME ST-LLC) and Japan Atomic Energy Agency (JAEA). Task 10 is to Update and Improve Subsection NH -- Alternative Simplified Creep-Fatigue Design Methods. Five newly proposed promising creep-fatigue evaluation methods were investigated. Those are (1) modified ductility exhaustion method, (2) strain range separation method, (3) approach for pressure vessel application, (4) hybrid method of time fraction and ductility exhaustion, and (5) simplified model test approach. The outlines of those methods are presented first, and predictability of experimental results of these methods is demonstrated using the creep-fatigue data collected in previous Tasks 3 and 5. All the methods (except the simplified model test approach which is not ready for application) predicted experimental results fairly accurately. On the other hand, predicted creep-fatigue life in long-term regions showed considerable differences among the methodologies. These differences come from the concepts each method is based on. All the new methods investigated in this report have advantages over the currently employed time fraction rule and offer technical insights that should be thought much of in the improvement of creep-fatigue evaluation procedures. …

The Utility of Social Modeling for Proliferation Assessment project (PL09-UtilSocial) investigates the use of social and cultural information to improve nuclear proliferation assessments, including nonproliferation assessments, Proliferation Resistance (PR) assessments, safeguards assessments, and other related studies. These assessments often use and create technical information about a host State and its posture towards proliferation, the vulnerability of a nuclear energy system (NES) to an undesired event, and the effectiveness of safeguards. This objective of this project is to find and integrate social and technical information by explicitly considering the role of cultural, social, and behavioral factors relevant to proliferation; and to describe and demonstrate if and how social science modeling has utility in proliferation assessment. This report describes a modeling approach and how it might be used to support a location-specific assessment of the PR assessment of a particular NES. The report demonstrates the use of social modeling to enhance an existing assessment process that relies on primarily technical factors.
