Optimized Volumetric Scanning for X-Ray Array Sources

Non-destructive evaluation (NDE) is the science and technology of determining non-invasively the internal structure of manufactured parts, objects, and materials. NDE application areas include medicine, industrial manufacturing, military, homeland security, and airport luggage screening. X-ray measurement systems are most widely used because of their ability to image through a wide range of material densities (from human tissue in medical applications to the dense materials of weapon components). Traditional x-ray systems involve a single source and detector system that rotate and/or translate about the object under evaluation. At each angular location, the source projects x-rays through the object. The rays undergo attenuation proportional to the density of the object's constitutive material. The detector records a measure of the attenuation. Mathematical algorithms are used to invert the forward attenuated ray projection process to form images of the object. This is known as computed tomography (CT). In recent years, the single-source x-ray NDE systems have been generalized to arrays of x-ray sources. Array sources permit multiple views of the object with fewer rotations and translations of the source/detector system. The use of microarrays in microbial ecology

Microarrays have proven to be a useful and high-throughput method to provide targeted DNA sequence information for up to many thousands of specific genetic regions in a single test. A microarray consists of multiple DNA oligonucleotide probes that, under high stringency conditions, hybridize only to specific complementary nucleic acid sequences (targets). A fluorescent signal indicates the presence and, in many cases, the abundance of genetic regions of interest. In this chapter we will look at how microarrays are used in microbial ecology, especially with the recent increase in microbial community DNA sequence data. Of particular interest to microbial ecologists, phylogenetic microarrays are used for the analysis of phylotypes in a community and functional gene arrays are used for the analysis of functional genes, and, by inference, phylotypes in environmental samples. A phylogenetic microarray that has been developed by the Andersen laboratory, the PhyloChip, will be discussed as an example of a microarray that targets the known diversity within the 16S rRNA gene to determine microbial community composition. Monodisperse metal nanoparticle catalysts on silica mesoporous supports: synthesis, characterizations, and catalytic reactions

The design of high performance catalyst achieving near 100% product selectivity at maximum activity is one of the most important goals in the modern catalytic science research. To this end, the preparation of model catalysts whose catalytic performances can be predicted in a systematic and rational manner is of significant importance, which thereby allows understanding of the molecular ingredients affecting the catalytic performances. We have designed novel 3-dimensional (3D) high surface area model catalysts by the integration of colloidal metal nanoparticles and mesoporous silica supports. Monodisperse colloidal metal NPs with controllable size and shape were synthesized using dendrimers, polymers, or surfactants as the surface stabilizers. The size of Pt, and Rh nanoparticles can be varied from sub 1 nm to 15 nm, while the shape of Pt can be controlled to cube, cuboctahedron, and octahedron. The 3D model catalysts were generated by the incorporation of metal nanoparticles into the pores of mesoporous silica supports via two methods: capillary inclusion (CI) and nanoparticle encapsulation (NE). GeoChips for Analysis of Microbial Functional Communities

Functional gene arrays (FGA) are microarrays that contain probes for genes encoding proteins or enzymes involved in functions of interest and allow for the study of thousands of genes at one time. The most comprehensive FGA to date is the GeoChip, which contains ~;;24,000 probes for ~;;10,000 genes involved in the geochemical cycling of C, N, P, and S, as well as genes involved in metal resistance and reduction and contaminant degradation. This chapter details the methods necessary for GeoChip analysis. Methods covered include preparation of DNA (whole community genome amplification and labeling), array setup (prehybridization steps), hybridization (sample and hybridization buffers), and post hybridization steps (slide washing and array scanning).

Associations between iron oxyhydroxide nanoparticle growth and metal adsorption/structural incorporation

The interaction of metal ions and oxyanions with nanoscale mineral phases has not yet been extensively studied despite the increased recognition of their prevalence in natural systems as a significant component of geomedia. A combination of macroscopic uptake studies to investigate the adsorption behavior of As(V), Cu(II), Hg(II), and Zn(II) onto nanoparticulate goethite ({alpha}-FeOOH) as a function of aging time at elevated temperature (75 C) and synchrotron-based X-ray studies to track changes in both the sorption mode and the rate of nanoparticle growth reveal the effects that uptake has on particle growth. Metal(loid) species which sorb quickly to the iron oxyhydroxide particles (As(V), Cu(II)) appear to passivate the particle surface, impeding the growth of the nanoparticles with progressive aging; in contrast, species that sorb more slowly (Hg(II), Zn(II)) have considerably less impact on particle growth. Progressive changes in the speciation of these particular metals with time suggest shifts in the mode of metal uptake with time, possibly indicating structural incorporation of the metal(loid) into the nanoparticle; this is supported by the continued increase in uptake concomitant with particle growth, implying that metal species may transform from surface-sorbed species to more structurally incorporated forms. Sulfate, one of the inorganic constituents that could be present in the nuclear waste repository, forms complexes with U(VI) and affects its migration in the environment. Results show that the complexation of U(VI) with sulfate is enhanced by the increase in temperature. The effect of temperature on the complexation and speciation of U(VI) in sulfate solutions is discussed.

The Value of Renewable Energy as a Hedge Against Fuel Price Risk: Analytic Contributions from Economic and Finance Theory

For better or worse, natural gas has become the fuel of choice for new power plants being built across the United States. According to the Energy Information Administration (EIA), natural gas-fired units account for nearly 90% of the total generating capacity added in the U.S. between 1999 and 2005 (EIA 2006b), bringing the nationwide market share of gas-fired generation to 19%. Looking ahead over the next decade, the EIA expects this trend to continue, increasing the market share of gas-fired generation to 22% by 2015 (EIA 2007a). Though these numbers are specific to the US, natural gas-fired generation is making similar advances in many other countries as well. A large percentage of the total cost of gas-fired generation is attributable to fuel costs--i.e., natural gas prices. For example, at current spot prices of around $7/MMBtu, fuel costs account for more than 75% of the levelized cost of energy from a new combined cycle gas turbine, and more than 90% of its operating costs (EIA 2007a). Furthermore, given that gas-fired plants are often the marginal supply units that set the market-clearing price for all generators in a competitive wholesale market, there is a direct link between natural gas prices and wholesale …

Rapid Data Assimilation in the Indoor Environment: theory and examples from real-time interpretation of indoor plumes of airborne chemicals

Releases of acutely toxic airborne contaminants in or near a building can lead to significant human exposures unless prompt response measures are identified and implemented. Commonly, possible responses include conflicting strategies, such as shutting the ventilation system off versus running it in a purge (100percent outside air) mode, or having occupants evacuate versus sheltering in place. The right choice depends in part on quickly identifying the source locations, the amounts released, and the likely future dispersion routes of the pollutants. This paper summarizes recent developments to provide such estimates in real time using an approach called Bayesian Monte Carlo updating. This approach rapidly interprets measurements of airborne pollutant concentrations from multiple sensors placed in the building and computes best estimates and uncertainties of the release conditions. The algorithm is fast, capable of continuously updating the estimates as measurements stream in from sensors. The approach is employed, as illustration, to conduct two specific investigations under different situations.

Nature uses enzymes to activate otherwise unreactive compounds in remarkable ways. For example, DNases are capable of hydrolyzing phosphate diester bonds in DNA within seconds,[1-3]--a reaction with an estimated half-life of 200 million years without an enzyme.[4] The fundamental features of enzyme catalysis have been much discussed over the last sixty years in an effort to explain the dramatic rate increases and high selectivities of enzymes. As early as 1946, Linus Pauling suggested that enzymes must preferentially recognize and stabilize the transition state over the ground state of a substrate.[5] Despite the intense study of enzymatic selectivity and ability to catalyze chemical reactions, the entire nature of enzyme-based catalysis is still poorly understood. For example, Houk and co-workers recently reported a survey of binding affinities in a wide variety of enzyme-ligand, enzyme-transition-state, and synthetic host-guest complexes and found that the average binding affinities were insufficient to generate many of the rate accelerations observed in biological systems.[6] Therefore, transition-state stabilization cannot be the sole contributor to the high reactivity and selectivity of enzymes, but rather, other forces must contribute to the activation of substrate molecules. Structural Genomics of Minimal Organisms: Pipeline and Results

The initial objective of the Berkeley Structural Genomics Center was to obtain a near complete three-dimensional (3D) structural information of all soluble proteins of two minimal organisms, closely related pathogens Mycoplasma genitalium and M. pneumoniae. The former has fewer than 500 genes and the latter has fewer than 700 genes. A semiautomated structural genomics pipeline was set up from target selection, cloning, expression, purification, and ultimately structural determination. At the time of this writing, structural information of more than 93percent of all soluble proteins of M. genitalium is avail able. This chapter summarizes the approaches taken by the authors' center.

Modeling Water Management in Polymer-Electrolyte Fuel Cells

Fuel cells may become the energy-delivery devices of the 21st century with realization of a carbon-neutral energy economy. Although there are many types of fuel cells, polymerelectrolyte fuel cells (PEFCs) are receiving the most attention for automotive and small stationary applications. In a PEFC, hydrogen and oxygen are combined electrochemically to produce water, electricity, and waste heat. During the operation of a PEFC, many interrelated and complex phenomena occur. These processes include mass and heat transfer, electrochemical reactions, and ionic and electronic transport. Most of these processes occur in the through-plane direction in what we term the PEFC sandwich as shown in Figure 1. This sandwich comprises multiple layers including diffusion media that can be composite structures containing a macroporous gas-diffusion layer (GDL) and microporous layer (MPL), catalyst layers (CLs), flow fields or bipolar plates, and a membrane. During operation fuel is fed into the anode flow field, moves through the diffusion medium, and reacts electrochemically at the anode CL to form hydrogen ions and electrons. The oxidant, usually oxygen in air, is fed into the cathode flow field, moves through the diffusion medium, and is electrochemically reduced at the cathode CL by combination with the generated protons and electrons. …

Chapter 3: Evaluating the impacts of carbonaceous aerosols on clouds and climate

Any attempt to reconcile observed surface temperature changes within the last 150 years to changes simulated by climate models that include various atmospheric forcings is sensitive to the changes attributed to aerosols and aerosol-cloud-climate interactions, which are the main contributors that may well balance the positive forcings associated with greenhouse gases, absorbing aerosols, ozone related changes, etc. These aerosol effects on climate, from various modeling studies discussed in Menon (2004), range from +0.8 to -2.4 W m{sup -2}, with an implied value of -1.0 W m{sup -2} (range from -0.5 to -4.5 W m{sup -2}) for the aerosol indirect effects. Quantifying the contribution of aerosols and aerosol-cloud interactions remain complicated for several reasons some of which are related to aerosol distributions and some to the processes used to represent their effects on clouds. Aerosol effects on low lying marine stratocumulus clouds that cover much of the Earth's surface (about 70%) have been the focus of most of prior aerosol-cloud interaction effect simulations. Since cumulus clouds (shallow and deep convective) are short lived and cover about 15 to 20% of the Earth's surface, they are not usually considered as radiatively important.
