PROTEIN QUALITY CONTROL IN BACTERIAL CELLS: INTEGRATED NETWORKS OF CHAPERONES AND ATP-DEPENDENT PROTEASES.

It is generally accepted that the information necessary to specify the native, functional, three-dimensional structure of a protein is encoded entirely within its amino acid sequence; however, efficient reversible folding and unfolding is observed only with a subset of small single-domain proteins. Refolding experiments often lead to the formation of kinetically-trapped, misfolded species that aggregate, even in dilute solution. In the cellular environment, the barriers to efficient protein folding and maintenance of native structure are even larger due to the nature of this process. First, nascent polypeptides must fold in an extremely crowded environment where the concentration of macromolecules approaches 300-400 mg/mL and on average, each ribosome is within its own diameter of another ribosome (1-3). These conditions of severe molecular crowding, coupled with high concentrations of nascent polypeptide chains, favor nonspecific aggregation over productive folding (3). Second, folding of newly-translated polypeptides occurs in the context of their vehtorial synthesis process. Amino acids are added to a growing nascent chain at the rate of {approx}5 residues per set, which means that for a 300 residue protein its N-terminus will be exposed to the cytosol {approx}1 min before its C-terminus and be free to begin the folding process. Signaling to the P53 Tumor Suppressor Through Pathways Activated by Genotoxic and Non-Genotoxic Stresses.

The p53 tumor suppressor is a tetrameric transcription factor that is post-translational modified at {approx}18 different sites by phosphorylation, acetylation, or sumoylation in response to various cellular stress conditions. Specific posttranslational modifications, or groups of modifications, that result from the activation of different stress-induced signaling pathways are thought to modulate p53 activity to regulate cell fate by inducing cell cycle arrest, apoptosis, or cellular senescence. Here we review the posttranslational modifications to p53 and the pathways that produce them in response to both genotoxic and non-genotoxic stresses.

The 14th Workshop will provide a forum for an informal exchange of technical and scientific information between international researchers in the photovoltaic and relevant non-photovoltaic fields. It will offer an excellent opportunity for researchers in private industry and at universities to prioritize mutual needs for future collaborative research. The workshop is intended to address the fundamental properties of PV silicon, new solar cell designs, advanced solar cell processing techniques, and cell-related module issues. A combination of oral presentations by invited speakers, poster sessions, and discussion sessions will review recent advances in crystal growth, new cell designs, new processes and process characterization techniques, cell fabrication approaches suitable for future manufacturing demands, and solar cell encapsulation. This year's theme, ''Crystalline Si Solar Cells: Leapfrogging the Barriers,'' reflects the continued success of crystalline Si PV in overcoming technological barriers to improve solar cell performance and lower the cost of Si PV. Laboratories for the 21st Century: Energy Analysis

This study, done for the joint DOE-EPA Laboratories for the 21st Century program ("Labs 21"), used a simplified computer model to analyze the effects of energy efficiency measures in laboratory buildings in four different climates: those of Minneapolis, Denver, Seattle, and Atlanta. Results show that using variable-air-volume fume hoods can reduce lab energy costs as much as$1 per square foot in any climate. Energy-recovery systems such as enthalpy wheels also save varying amounts of energy and money in all climates. Savings for other measures, such as heat pipes and evaporated cooling, are also included.

To economize on energy costs and improve the safety, comfort, and health of building occupants, building design teams can incorporate daylighting, energy efficiency, renewable energy, and passive solar design into all projects in which these elements are technically and economically feasible. The information presented here will help project leaders begin the process and manage the inclusion of sustainable design in the procurement process. The section on establishing selection criteria contains key elements to consider before selecting an architectural and engineering (A/E) firm. Evaluation of the Compressed Air Challenge(R) Training Program (Executive Summary)

The final report of the evaluation of the Compressed Air Challenge (CAC) Training Program. The training program is designed to provide plant personnel and compressed air system vendors with knowledge and tools required to effect improvements to the energy efficiency and overall performance of plant compressed air systems. As of May 2001, 3,029 individuals had attended the CAC Fundamentals of Compressed Air Training Systems and 925 individuals had attended ''Advanced Management of Compressed Air Systems''. These individuals represented 1,400-1,500 separate business establishments. The evaluation is based on three main research tasks: analysis of the CAC registration database, interviews with 100 end-user personnel who attended the CAC training, and interviews with 100 compressed air system vendors and consulting engineers who attended the training sessions.

Handbook for Planning and Conducting Charrettes for High-Performance Projects

The purpose of this handbook is to furnish guidance for planning and conducting a"high-performance building" charrette, sometimes called a"greening charrette." The handbook answers typical questions that will arise, such as"What is a charrette?""Why conduct a charrette?""What topics should we cover during the charrette?" and"Whom should we invite?" It also contains samples of agendas, invitation letters, and other commonly used charrette materials. This handbook also outlines the characteristics of a good charrette facilitator. It gives suggestions for the types of experts to invite to the event to motivate participants and answer their questions. The handbook includes sample presentations that can be used by these experts to ensure they address the required technical content. It suggests the types of participants, including technical, political, and community representatives, to invite to the charrette. It offers advice for forming effective breakout groups to ensure that a broad range of complementary expertise is represented in each group. We have also included guidance on how best to include key decision makers and stakeholders who are able to attend only portions of the event.

Assessing the Potential for Renewable Energy on National Forest System Lands

This technical report and CD for the U.S. Department of Agriculture, Forest Service (USFS), evaluates the potential for renewable energy resource development on National Forest System (NFS) lands. USFS can use the report findings to consider potential for development of solar and wind energy resources on NFS lands, in land management decisions. The Geographical Information System (GIS) based analysis resulted in the following findings: (1) Ninety-nine National Forest Units have high potential for power production from one or more of these solar and wind energy sources; and (2) Twenty National Forest Units in nine states have high potential for power production from two or more of these solar and wind energy sources.
