Advanced Recombinant Manganese Peroxidase for Biosynthesis of Lignin Bioproducts, Phase I Final Report, STTR Grant #: DE-SC0007503.

The core purpose of this Phase I STTR was to evaluate the feasibility of a new method of producing a recombinant version of manganese peroxidase (MnP) enzyme. MnP is a potentially valuable enzyme for producing high value lignin products and also for industrial de-coloring operations such as biobleaching of pulp and color removal from textile dye effluents. This lignin-modifying enzyme is produced in small amounts by the native host, a white rot fungus. Previous work by Oregon State University developed a secreted recombinant version of the enzyme in the yeast Pichia pastoris. Unfortunately, the expression is barely moderate and the enzyme is heavily glycosylated, which inhibits purification. In this work, the gene for the enzyme is given a tag which targets production of the enzyme to the peroxisome. This is a promising approach since this location is also where heme and hydrogen peroxide are sequestered, which are both necessary cofactors for MnP. More than ten recombinant strains were constructed, verified, and expressed in the Pichia system. Constitutive (GAP) and methanol-induced promoters (AOX) were tried for peroxisomal targeted, cytosolic, and secreted versions of MnP. Only the secreted strains showed activity. Novel Low Cost, High Reliability Wind Turbine Drivetrain

Clipper Windpower, in collaboration with United Technologies Research Center, the National Renewable Energy Laboratory, and Hamilton Sundstrand Corporation, developed a low-cost, deflection-compliant, reliable, and serviceable chain drive speed increaser. This chain and sprocket drivetrain design offers significant breakthroughs in the areas of cost and serviceability and addresses the key challenges of current geared and direct-drive systems. The use of gearboxes has proven to be challenging; the large torques and bending loads associated with use in large multi-MW wind applications have generally limited demonstrated lifetime to 8-10 years [1]. The large cost of gearbox replacement and the required use of large, expensive cranes can result in gearbox replacement costs on the order of $1M, representing a significant impact to overall cost of energy (COE). Direct-drive machines eliminate the gearbox, thereby targeting increased reliability and reduced life-cycle cost. However, the slow rotational speeds require very large and costly generators, which also typically have an undesirable dependence on expensive rare-earth magnet materials and large structural penalties for precise air gap control. Electroweak Baryogenesis and Higgs Properties

We explore the connection between the strength of the electroweak phase transition and the properties of the Higgs boson. Our interest is in regions of parameter space that can realize electroweak baryogenesis. We do so in a simplified framework in which a single Higgs field couples to new scalar fields charged under SU(3){sub c} by way of the Higgs portal. Such new scalars can make the electroweak phase transition more strongly first-order, while contributing to the effective Higgs boson couplings to gluons and photons through loop effects. For Higgs boson masses in the range 115 {approx}< m{sub h} {approx}< 130 GeV, whenever the phase transition becomes strong enough for successful electroweak baryogenesis, we find that Higgs boson properties are modified by an amount observable by the LHC. We also discuss the baryogenesis window of the minimal supersymmetric standard model (MSSM), which appears to be under tension. Furthermore, we argue that the discovery of a Higgs boson with standard model-like couplings to gluons and photons will rule out electroweak baryogenesis in the MSSM.

ALTERNATIVES ANALYSIS FOR SELECTING ET #3 SITE

Engineered trenches (ETs) are considered to be a cost-effective method for disposing Low Level Waste (LLW). Based on waste forecasts from waste generators, the last engineered trench in operation (ET No.2) is anticipated to close in FY14, requiring development of a new ET. Solid Waste requested that SRNL develop an assessment report that reviews four disposal options for this new ET (ET No.3) and determine which option would provide the 'best' Performance Assessment (PA) disposal limits for LLW (Appendix A). Those four options (see option footprint locations in Figure 1-1) are: (1) Disposal at grade on TRU Pads 7-13 where soil would be mounded over waste packages; (2) Excavation at a slightly modified SLIT No.13 location - near the Used Equipment Storage Area; (3) Excavation at a modified SLIT No.12 location - near the 643-26E Naval Reactor Component Disposal Area; and (4) Excavation east of TRU Pad No.26 that replaces northeast portions of four slit trench (ST) disposal units in the eastern set of STs. The Jackson-Vanik Amendment and Candidate Countries for WTO Accession: Issues for Congress

This report gives an analysis of the unconditional most-favored-nation (MFN) status, or in U.S. statutory parlance, normal trade relations (NTR) status, which is a fundamental principle of the World Trade Organization (WTO). This conflicts with the U.S. laws under Title IV of the Trade Act of 1974 that limits trade status with several nations undergoing accession into the WTO. On June 12, 2012, Sen. Max Baucus introduced a bill with bipartisan co-sponsorship to authorize PNTR for Russia. The report includes information about MFN status and the WTO, the Jackson-Vanik Amendment restricting trade, the case of China, and prospective WTO accessions.

BUNCOMBE COUNTY WASTEWATER PRE-TREATMENT AND LANDFILL GAS TO ENERGY PROJECT

The objective of this project was to construct a landfill gas-to-energy (LFGTE) facility that generates a renewable energy source utilizing landfill gas to power a 1.4MW generator, while at the same time reducing the amount of leachate hauled offsite for treatment. The project included an enhanced gas collection and control system, gas conditioning equipment, and a 1.4 MW generator set. The production of cleaner renewable energy will help offset the carbon footprint of other energy sources that are currently utilized.

China's Building Energy Use: A Long-Term Perspective based on a Detailed Assessment

We present here a detailed, service-based model of China's building energy use, nested in the GCAM (Global Change Assessment Model) integrated assessment framework. Using the model, we explore long-term pathways of China's building energy use and identify opportunities of reducing greenhouse gas emissions. The inclusion of a structural model of building energy demands within an integrated assessment framework represents a major methodological advance. It allows for a structural understanding of the drivers of building energy consumption while simultaneously considering the other human and natural system interactions that influence changes in the global energy system and climate. We also explore a range of different scenarios to gain insights into how China's building sector might evolve and what the implications might be for improved building energy technology and carbon policies. The analysis suggests that China's building energy growth will not wane anytime soon, although technology improvement will put downward pressure on this growth. Also, regardless of the scenarios represented, the growth will involve the continued, rapid electrification of the buildings sector throughout the century, and this transition will be accelerated by the implementation of carbon policy.

This report describes progress made during the final phase of our DOE-funded program on Nanostructured Magnetic Materials. This period was quite productive, resulting in the submission of three papers and presentation of three talks at international conferences and three seminars at research institutions. Our DOE-funded research efforts were directed toward studies of magnetism at surfaces and interfaces in high-quality, well-characterized materials prepared by Molecular Beam Epitaxy (MBE) and sputtering. We have an exceptionally well-equipped laboratory for these studies, with: Thin film preparation equipment; Characterization equipment; Equipment to study magnetic properties of surfaces and ultra-thin magnetic films and interfaces in multi-layers and superlattices.

The goal of the project was to demonstrate the commercial feasibility of geopressured-geothermal power development by exploiting the extraordinarily high pressured hot brines know to exist at depth near the Sweet Lake oil and gas field in Cameron Parish, Louisiana. The existence of a geopressured-geothermal system at Sweet Lake was confirmed in the 1970's and 1980's as part of DOE's Geopressured-Geothermal Program. That program showed that the energy prices at the time could not support commercial production of the resource. Increased electricity prices and technological advancements over the last two decades, combined with the current national support for developing clean, renewable energy and the job creation it would entail, provided the justification necessary to reevaluate the commercial feasibility of power generation from this vast resource.
