Since October 2001, the United States has provided Pakistan with over $10 billion for military, economic, and development activities in support of the critical U.S. national security goals of destroying terrorist threats and closing terrorist safe havens. A major component of this effort has been U.S. Coalition Support Funds (CSF) reimbursed to Pakistan. Employment Verification: Challenges Exist in Implementing a Mandatory Electronic Employment Verification System

Testimony issued by the Government Accountability Office with an abstract that begins "In 1996, the former U.S. Immigration and Naturalization Service, now within the Department of Homeland Security (DHS), and the Social Security Administration (SSA) began operating a voluntary pilot program, recently named the E-Verify program, to provide participating employers with a means for electronically verifying employees' work eligibility. Legislation has been introduced in Congress to require all employers to electronically verify the work authorization status of their employees. In this testimony GAO provides observations on the E-Verify system's capacity and costs, options for reducing delays and improving efficiency in the verification process, ability to detect fraudulent documents and identity theft, and vulnerability to employer fraud and misuse. This testimony is based on GAO's products issued from August 2005 through June 2007 and updated information obtained from DHS and SSA in April 2008. Testimony issued by the Government Accountability Office with an abstract that begins "For more than a decade, GAO has reported that Medicare has paid higher than market rates for medical equipment and supplies provided to beneficiaries under Medicare Part B. Since 1989, Medicare has used fee schedules primarily based on historical charges to set payment amounts. But this approach lacks flexibility to keep pace with market changes and increases costs to the federal government and Medicare's 44 million elderly and disabled beneficiaries. The Balanced Budget Act of 1997 required the Centers for Medicare & Medicaid Services (CMS)--the agency that administers Medicare--to test competitive bidding as a new way to set payments. CMS did this through a demonstration in two locations in which suppliers could compete on the basis of price and other factors for the right to provide their products. The Medicare Prescription Drug, Improvement, and Modernization Act of 2003 (MMA) required CMS to conduct competitive bidding on a large scale and suppliers to obtain accreditation. Photoconductive Detectors with Fast Temporal Response for Laser Produced Plasma Experiments.

Processes during laser plasma experiments typically have time scales that are less than 100 ps. The measurement of these processes requires X-ray detectors with fast temporal resolution. We have measured the temporal responses and linearity of several different X-ray sensitive Photoconductive Detectors (PCDs). The active elements of the detectors investigated include both diamond (natural and synthetic) and GaAs crystals. The typical time responses of the GaAs PCDs are approximately 60 ps, respectively. Some characterizations using X-ray light from a synchrotron light source are presented.

Diagnostics for Fast Ignition Science

The concept for Electron Fast Ignition Inertial Confinement Fusion demands sufficient laser energy be transferred from the ignitor pulse to the assembled fuel core via {approx}MeV electrons. We have assembled a suite of diagnostics to characterize such transfer. Recent experiments have simultaneously fielded absolutely calibrated extreme ultraviolet multilayer imagers at 68 and 256eV; spherically bent crystal imagers at 4 and 8keV; multi-keV crystal spectrometers; MeV x-ray bremmstrahlung and electron and proton spectrometers (along the same line of sight); nuclear activation samples and a picosecond optical probe based interferometer. These diagnostics allow careful measurement of energy transport and deposition during and following laser-plasma interactions at extremely high intensities in both planar and conical targets. Augmented with accurate on-shot laser focal spot and pre-pulse characterization, these measurements are yielding new insight into energy coupling and are providing critical data for validating numerical PIC and hybrid PIC simulation codes in an area that is crucial for many applications, particularly fast ignition. Novel aspects of these diagnostics and how they are combined to extract quantitative data on ultra high intensity laser plasma interactions are discussed, together with implications for full-scale fast ignition experiments.

THERMODYNAMIC AND MASS BALANCE ANALYSIS OF EXPANSIVE PHASE PRECIPITATION IN SALTSTONE

This report assesses the potential for future precipitation of expansive phases that could cause fracturing in saltstone. It examines the equilibrium case using The Geochemist's Workbench{reg_sign} reaction path model. The scenarios simulated examine the effects of different possible infiltrating fluids, different saltstone formulations, and different amounts of minerals available for reaction. Mineralogy of the vault cement and saltstone were estimated using reported chemical compositions of each. The infiltrating fluid was assumed to be either rainwater equilibrated with vault cement or rainwater itself. The simulations assumed that minerals were homogeneously distributed in saltstone and that each pore volume of infiltration reached equilibrium with the mineral assemblage. Fracturing that initiates in pores by expansive phase precipitation is unlikely to occur in saltstone because the maximum amount of porosity filled is 34%. If less than 100% of the saltstone minerals are available for reaction, less porosity will be lost to expansive phases. Likewise, the formulation of saltstone used will affect the amount of porosity filled by expansive phases.

Using X-Rays to Test CVD Diamond Detectors for Areal Density Measurement at the National Ignition Facility

At the National Ignition Facility (NIF), 192 laser beams will compress a target containing a mixture of deuterium and tritium (DT) that will release fusion neutrons, photons, and other radiation. Diagnostics are being designed to measure this emitted radiation to infer crucial parameters of an ignition shot. Chemical Vapor Deposited (CVD) diamond is one of the ignition diagnostics that will be used as a neutron time-of-flight detector for measuring primary (14.1 MeV) neutron yield, ion temperature, and plasma areal density. This last quantity is the subject of this study and is inferred from the number of downscattered neutrons arriving late in time, divided by the number of primary neutrons. We determine in this study the accuracy with which this detector can measure areal density, when the limiting factor is detector and electronics saturation. We used laser-produced x-rays to reproduce NIF signals in terms of charge carriers density, time between pulses, and amplitude contrast and found that the effect of the large pulse on the small pulse is at most 8.4%, which is less than the NIF accuracy requirement of {+-} 10%.

ASGRAD FY07 Annual Report

This is the annual project report for the ASGRAD project - Amorphous Semiconductors for Gamma Radiation Detection. We describe progress in the development of new materials for portable, room temperature, gammaradiation detection at Pacific Northwest National Laboratory. High Z, high resistivity, amorphous semiconductors are being designed for use as solid-state detectors at near ambient temperatures; their principles of operation are analogous to single-crystal semiconducting detectors. Compared to single crystals, amorphous semiconductors have the advantages of rapid, cost-effective, bulk-fabrication; nearnet-shape fabrication of complicated geometries; compositional flexibility; and greater electronic property control. The main disadvantage is reduced-charge carrier mobility. The focus of this project is to develop optimized amorphous semiconductor materials for gamma detection applications that leverage their material advantages while mitigating their limitations. During the second year of this project, several important milestones were accomplished. Major accomplishments were: (1) Significant processing - property and composition - property correlations were determined for Cd-Ge-As glasses; (2) Radiation response testing was successfully demonstrated on three different amorphous semiconductor materials (Cd-Ge-As, As-Se, and As-Se-Te systems) at ambient and near ambient temperatures; (3) Advanced, enabling Schottky contacts were developed for Cd-Ge-As compounds, this will allow these materials to perform at ambient temperatures; and (4) The collaborative …

LARG at chromosome 11q23 has functional characteristics of a tumor suppressor in human breast cancer

Deletion of 11q23-q24 is frequent in a diverse variety of malignancies, including breast and colorectal carcinoma, implicating the presence of a tumor suppressor gene at that chromosomal region. We show here that LARG, from 11q23, has functional characteristics of a tumor suppressor. We examined a 6-Mb region on 11q23 by high-resolution deletion mapping, utilizing both loss of heterozygosity (LOH) analysis and microarray comparative genomic hybridization (CGH). LARG (also called ARHGEF12), identified from the analyzed region, was underexpressed in 34% of primary breast carcinomas and 80% of breast cancer cell lines including the MCF-7 line. Multiplex ligation-dependent probe amplification on 30 primary breast cancers and six breast cancer cell lines showed that LARG had the highest frequency of deletion compared to the BCSC-1 and TSLC1 genes, two known candidate tumor suppressor genes from 11q. In vitro analysis of breast cancer cell lines that underexpress LARG showed that LARG could be reactivated by trichostatin A, a histone deacetylase inhibitor, but not by 5-Aza-2{prime}-deoxycytidine, a demethylating agent. Bisulfite sequencing and quantitative high-throughput analysis of DNA methylation confirmed the lack of CpG island methylation in LARG in breast cancer. An integrative genomic and proteomic analysis of PIK3CA, PTEN and AKT mutations in breast cancer

Phosphatidylinositol-3-kinase (PI3K)/AKT pathway aberrations are common in cancer. By applying mass spectroscopy-based sequencing and reverse phase protein arrays to 547 human breast cancers and 41 cell lines, we determined the subtype specificity and signaling effects of PIK3CA, AKT and PTEN mutations, and the effects of PIK3CA mutations on responsiveness to PI3K inhibition in-vitro and on outcome after adjuvant tamoxifen. PIK3CA mutations were more common in hormone receptor positive (33.8%) and HER2-positive (24.6%) than in basal-like tumors (8.3%). AKT1 (1.4%) and PTEN (2.3%) mutations were restricted to hormone receptor-positive cancers with PTEN protein levels also being significantly lower in hormone receptor-positive cancers. Unlike AKT1 mutations, PIK3CA (39%) and PTEN (20%) mutations were more common in cell lines than tumors, suggesting a selection for these but not AKT1 mutations during adaptation to culture. PIK3CA mutations did not have a significant impact on outcome in 166 hormone receptor-positive breast cancer patients after adjuvant tamoxifen. PIK3CA mutations, in comparison with PTEN loss and AKT1 mutations, were associated with significantly less and indeed inconsistent activation of AKT and of downstream PI3K/AKT signaling in tumors and cell lines, and PTEN loss and PIK3CA mutation were frequently concordant, suggesting different contributions to pathophysiology. System Design and the Safety Basis

The objective of this paper is to present the Bechtel Jacobs Company, LLC (BJC) Lessons Learned for system design as it relates to safety basis documentation. BJC has had to reconcile incomplete or outdated system description information with current facility safety basis for a number of situations in recent months. This paper has relevance in multiple topical areas including documented safety analysis, decontamination & decommissioning (D&D), safety basis (SB) implementation, safety and design integration, potential inadequacy of the safety analysis (PISA), technical safety requirements (TSR), and unreviewed safety questions. BJC learned that nuclear safety compliance relies on adequate and well documented system design information. A number of PIS As and TSR violations occurred due to inadequate or erroneous system design information. As a corrective action, BJC assessed the occurrences caused by systems design-safety basis interface problems. Safety systems reviewed included the Molten Salt Reactor Experiment (MSRE) Fluorination System, K-1065 fire alarm system, and the K-25 Radiation Criticality Accident Alarm System. The conclusion was that an inadequate knowledge of system design could result in continuous non-compliance issues relating to nuclear safety. Decreased expression of RNA interference machinery, Dicer and Drosha, is associated with poor outcome in ovarian cancer patients

The clinical and functional significance of RNA interference (RNAi) machinery, Dicer and Drosha, in ovarian cancer is not known and was examined. Dicer and Drosha expression was measured in ovarian cancer cell lines (n=8) and invasive epithelial ovarian cancer specimens (n=111) and correlated with clinical outcome. Validation was performed with previously published cohorts of ovarian, breast, and lung cancer patients. Anti-Galectin-3 siRNA and shRNA transfections were used for in vitro functional studies. Dicer and Drosha mRNA and protein levels were decreased in 37% to 63% of ovarian cancer cell lines and in 60% and 51% of human ovarian cancer specimens, respectively. Low Dicer was significantly associated with advanced tumor stage (p=0.007), and low Drosha with suboptimal surgical cytoreduction (p=0.02). Tumors with both high Dicer and Drosha were associated with increased median patient survival (>11 years vs. 2.66 years for other groups; p<0.001). In multivariate analysis, high Dicer (HR=0.48; p=0.02), high-grade histology (HR=2.46; p=0.03), and poor chemoresponse (HR=3.95; p<0.001) were identified as independent predictors of disease-specific survival. Findings of poor clinical outcome with low Dicer expression were validated in separate cohorts of cancer patients. A spatial symmetry of the field is not always represented by a symmetry in the Fourier spectrum when magnetic coordinates are used because of the distortion of the toroidal angle. The practical importance of spectral distortion is illustrated with a tokamak example.

Testimony issued by the Government Accountability Office with an abstract that begins "Collaboration between the federal government and the aviation industry has led to reductions in aviation emissions, but growing air traffic has partially offset these reductions. The Federal Aviation Administration (FAA), together with the National Aeronautics and Space Administration (NASA), the Environmental Protection Agency (EPA), and others, is working to increase the efficiency, safety, and capacity of the national airspace system and at the same time reduce aviation emissions, in part, by transforming the current air traffic control system to the Next Generation Air Transportation System (NextGen). This effort involves new technologies and air traffic procedures that can reduce aviation emissions and incorporates research and development (R&D) on emissions-reduction technologies. Reducing aviation emissions is important both to minimize their adverse health and environmental effects and to alleviate public concerns about them that could constrain the expansion of airport infrastructure and aviation operations needed to meet demand. This testimony addresses (1) the scope and nature of aviation emissions, (2) the status of selected key federal efforts to reduce aviation emissions, and (3) next steps and challenges in reducing aviation emissions.
