Molecular analysis of the metabolic rates of discrete subsurface populations of sulfate reducers

Elucidating the in situ metabolic activity of phylogenetically diverse populations of sulfate-reducing microorganisms that populate anoxic sedimentary environments is key to understanding subsurface ecology. Previous pure culture studies have demonstrated that transcript abundance of dissimilatory (bi)sulfite reductase genes is correlated with the sulfate reducing activity of individual cells. To evaluate whether expression of these genes was diagnostic for subsurface communities, dissimilatory (bi)sulfite reductase gene transcript abundance in phylogenetically distinct sulfate-reducing populations was quantified during a field experiment in which acetate was added to uranium-contaminated groundwater. Analysis of dsrAB sequences prior to the addition of acetate indicated that Desulfobacteraceae, Desulfobulbaceae, and Syntrophaceae-related sulfate reducers were the most abundant. Quantifying dsrB transcripts of the individual populations suggested that Desulfobacteraceae initially had higher dsrB transcripts per cell than Desulfobulbaceae or Syntrophaceae populations, and that the activity of Desulfobacteraceae increased further when the metabolism of dissimilatory metal reducers competing for the added acetate declined. In contrast, dsrB transcript abundance in Desulfobulbaceae and Syntrophaceae remained relatively constant, suggesting a lack of stimulation by added acetate. The indication of higher sulfate-reducing activity in the Desulfobacteraceae was consistent with the finding that Desulfobacteraceae became the predominant component of the sulfate-reducing community. Surface structure of protonated R-plane-sapphire (1-102) studied by sum frequency vibrational spectroscopy

Sum frequency vibrational spectroscopy was used to study the protonated R-plane (1{bar 1}02 ) sapphire surface. The OH stretch vibrational spectra show that the surface is terminated with three hydroxyl moieties, two from AlOH{sub 2} and one from Al{sub 2}OH functional groups. The observed polarization dependence allows determination of the orientations of the three OH species. The results suggest that the protonated sapphire (1{bar 1}02 ) surface differs from an ideal stoichimetric termination in a manner consistent with previous X-ray surface diffraction (crystal truncation rod) studies. However, in order to best explain the observed hydrogenbonding arrangement, surface oxygen spacing determined from the X-ray diffraction study requires modification.

Fossil fuels are abundant, inexpensive to produce, and are easily converted to usable energy by combustion as demonstrated by mankind's dependence on fossil fuels for over 80% of its primary energy supply (13). This reliance on fossil fuels comes with the cost of carbon dioxide (CO{sub 2}) emissions that exceed the rate at which CO{sub 2} can be absorbed by terrestrial and oceanic systems worldwide resulting in increases in atmospheric CO{sub 2} concentration as recorded by direct measurements over more than five decades (14). Carbon dioxide is the main greenhouse gas linked to global warming and associated climate change, the impacts of which are currently being observed around the world, and projections of which include alarming consequences such as water and food shortages, sea level rise, and social disruptions associated with resource scarcity (15). The current situation of a world that derives the bulk of its energy from fossil fuel in a manner that directly causes climate change equates to an energy-climate crisis. Although governments around the world have only recently begun to consider policies to avoid the direst projections of climate change and its impacts, sustainable approaches to addressing the crisis are available. An Accumulator/Pre-Booster for the Medium-Energy Electron Ion Collider at Jlab

Future nuclear physics facilities such as the proposed electron ion collider (MEIC) will need to achieve record high luminosities in order to maximize discovery potential. Among the necessary ingredients is the ability to generate, accumulate , accelerate, and store high current ion beams from protons to lead ions. One of the main components of this ion accelerator complex for MEIC chain is the accumulator that also doubles as a pre-booster, which takes 200 MeV protons from a superconducting linear accelerator, accumulates on the order of 1A beam, and boosts its energy to 3GeV, before extraction to the next accelerator in the chain, the large booster. This paper describes its design concepts, and summarizes some preliminary results, including linear optics, space charge dynamics, and spin polarization resonance analysis.

Differential growth responses of soil bacterial taxa to carbon substrates of varying chemical recalcitrance

Soils are immensely diverse microbial habitats with thousands of co-existing bacterial, archaeal, and fungal species. Across broad spatial scales, factors such as pH and soil moisture appear to determine the diversity and structure of soil bacterial communities. Within any one site however, bacterial taxon diversity is high and factors maintaining this diversity are poorly resolved. Candidate factors include organic substrate availability and chemical recalcitrance, and given that they appear to structure bacterial communities at the phylum level, we examine whether these factors might structure bacterial communities at finer levels of taxonomic resolution. Analyzing 16S rRNA gene composition of nucleotide analog-labeled DNA by PhyloChip microarrays, we compare relative growth rates on organic substrates of increasing chemical recalcitrance of >2,200 bacterial taxa across 43 divisions/phyla. Taxa that increase in relative abundance with labile organic substrates (i.e., glycine, sucrose) are numerous (>500), phylogenetically clustered, and occur predominantly in two phyla (Proteobacteria and Actinobacteria) including orders Actinomycetales, Enterobacteriales, Burkholderiales, Rhodocyclales, Alteromonadales, and Pseudomonadales. Taxa increasing in relative abundance with more chemically recalcitrant substrates (i.e., cellulose, lignin, or tannin-protein) are fewer (168) but more phylogenetically dispersed, occurring across eight phyla and including Clostridiales, Sphingomonadalaes, Desulfovibrionales. An Analysis of the Effects of Photovoltaic Energy Systems on Residential Selling Prices in California.

An increasing number of homes with existing photovoltaic (PV) energy systems have sold in the U.S., yet relatively little research exists that estimates the marginal impacts of those PV systems on the sales price. A clearer understanding of these effects might influence the decisions of homeowners, home buyers and PV home builders. This research analyzes a large dataset of California homes that sold from 2000 through mid-2009 with PV installed. Across a large number of hedonic and repeat sales model specifications and robustness tests, the analysis finds strong evidence that homes with PV systems sold for a premium over comparable homes without. The effects range, on average, from approximately $3.9 to $6.4 per installed watt (DC), with most models coalescing near $5.5/watt, which corresponds to a premium of approximately $17,000 for a 3,100 watt system. The research also shows that, as PV systems age, the premium enjoyed at the time of home sale decreases. Additionally, existing homes with PV systems are found to have commanded a larger sales price premium than new homes with similarly sized PV systems. Constitutive relationships for elastic deformation of clay rock: Data Analysis

Geological repositories have been considered a feasible option worldwide for storing high-level nuclear waste. Clay rock is one of the rock types under consideration for such purposes, because of its favorable features to prevent radionuclide transport from the repository. Coupled hydromechanical processes have an important impact on the performance of a clay repository, and establishing constitutive relationships for modeling such processes are essential. In this study, we propose several constitutive relationships for elastic deformation in indurated clay rocks based on three recently developed concepts. First, when applying Hooke's law in clay rocks, true strain (rock volume change divided by the current rock volume), rather than engineering strain (rock volume change divided by unstressed rock volume), should be used, except when the degree of deformation is very small. In the latter case, the two strains will be practically identical. Second, because of its inherent heterogeneity, clay rock can be divided into two parts, a hard part and a soft part, with the hard part subject to a relatively small degree of deformation compared with the soft part. Real space soft x-ray imaging at 10 nm spatial resolution

Using Fresnel zone plates made with our robust nanofabrication processes, we have successfully achieved 10 nm spatial resolution with soft x-ray microscopy. The result, obtained with both a conventional full-field and scanning soft x-ray microscope, marks a significant step forward in extending the microscopy to truly nanoscale studies.

Determination of the Fe magnetic anisotropies and the CoO frozen spins in epitaxial CoO/Fe/Ag(001)

CoO/Fe/Ag(001) films were grown epitaxially and studied by X-ray Magnetic Circular Dichroism (XMCD) and X-ray Magnetic Linear Dichroism (XMLD). After field cooling along the Fe[100] axis to 80 K, exchange bias, uniaxial anisotropy, and 4-fold anisotropy of the films were determined by hysteresis loop and XMCD measurements by rotating the Fe magnetization within the film plane. The CoO frozen spins were determined by XMLD measurement as a function of CoO thickness.We find that among the exchange bias, uniaxial anisotropy, and 4-fold anisotropy, only the uniaxial magnetic anisotropy follows thickness dependence of the CoO frozen spins.

BETR Global - A geographically explicit global-scale multimedia contaminant fate model

We present two new software implementations of the BETR Global multimedia contaminant fate model. The model uses steady-state or non-steady-state mass-balance calculations to describe the fate and transport of persistent organic pollutants using a desktop computer. The global environment is described using a database of long-term average monthly conditions on a 15{sup o} x 15{sup o} grid. We demonstrate BETR Global by modeling the global sources, transport, and removal of decamethylcyclopentasiloxane (D5).

Calculating the probability of injected carbon dioxide plumes encountering faults

One of the main concerns of storage in saline aquifers is leakage via faults. In the early stages of site selection, site-specific fault coverages are often not available for these aquifers. This necessitates a method using available fault data to estimate the probability of injected carbon dioxide encountering and migrating up a fault. The probability of encounter can be calculated from areal fault density statistics from available data, and carbon dioxide plume dimensions from numerical simulation. Given a number of assumptions, the dimension of the plume perpendicular to a fault times the areal density of faults with offsets greater than some threshold of interest provides probability of the plume encountering such a fault. Application of this result to a previously planned large-scale pilot injection in the southern portion of the San Joaquin Basin yielded a 3% and 7% chance of the plume encountering a fully and half seal offsetting fault, respectively. Subsequently available data indicated a half seal-offsetting fault at a distance from the injection well that implied a 20% probability of encounter for a plume sufficiently large to reach it.

Optical modeling of Fresnel zoneplate microscopes

Defect free masks remain one of the most significant challenges facing the commercialization of extreme ultraviolet (EUV) lithography. Progress on this front requires high-performance wavelength-specific metrology of EUV masks, including high-resolution and aerial-image microscopy performed near the 13.5 nm wavelength. Arguably the most cost-effective and rapid path to proliferating this capability is through the development of Fresnel zoneplate-based microscopes. Given the relative obscurity of such systems, however, modeling tools are not necessarily optimized to deal with them and their imaging properties are poorly understood. Here we present a modeling methodology to analyze zoneplate microscopes based on commercially available optical modeling software and use the technique to investigate the imaging performance of an off-axis EUV microscope design. The modeling predicts that superior performance can be achieved by tilting the zoneplate, making it perpendicular to the chief ray at the center of the field, while designing the zoneplate to explicitly work in that tilted plane. Although the examples presented here are in the realm of EUV mask inspection, the methods described and analysis results are broadly applicable to zoneplate microscopes in general, including full-field soft-x-ray microscopes rou tinely used in the synchrotron community.

In-Tank Elutriation Test Report And Independent Assessment

The Department of Energy (DOE) Office of Environmental Management (EM) funded Technology Development and Deployment (TDD) to solve technical problems associated with waste tank closure for sites such as Hanford Site and Savannah River Site (SRS). One of the tasks supported by this funding at Savannah River National Laboratory (SRNL) and Pacific Northwest Laboratory (PNNL) was In-Tank Elutriation. Elutriation is the process whereby physical separation occurs based on particle size and density. This report satisfies the first phase of Task WP_1.3.1.1 In-Tank Elutriation, which is to assess the feasibility of this method of separation in waste tanks at Hanford Site and SRS. This report includes an analysis of scoping tests performed in the Engineering Development Laboratory of SRNL, analysis of Hanford's inadvertent elutriation, the viability of separation methods such as elutriation and hydrocyclones and recommendations for a path forward. This report will demonstrate that the retrieval of Hanford salt waste tank S-112 very successfully decreased the tank's inventories of radionuclides. Analyses of samples collected from the tank showed that concentrations of the major radionuclides Cs-136 and Sr-90 were decreased by factors of 250 and 6 and their total curie tank inventories decreased by factors of 60,000 and 2000. A coupled THC model of the FEBEX in situ test with bentonite swelling and chemical and thermal osmosis

The performance assessment of a geological repository for radioactive waste requires quantifying the geochemical evolution of the bentonite engineered barrier. This barrier will be exposed to coupled thermal (T), hydrodynamic (H), mechanical (M) and chemical (C) processes. This paper presents a coupled THC model of the FEBEX (Full-scale Engineered Barrier EXperiment) in situ test which accounts for bentonite swelling and chemical and thermal osmosis. Model results attest the relevance of thermal osmosis and bentonite swelling for the geochemical evolution of the bentonite barrier while chemical osmosis is found to be almost irrelevant. The model has been tested with data collected after the dismantling of heater 1 of the in situ test. The model reproduces reasonably well the measured temperature, relative humidity, water content and inferred geochemical data. However, it fails to mimic the solute concentrations at the heater-bentonite and bentonite-granite interfaces because the model does not account for the volume change of bentonite, the CO{sub 2}(g) degassing and the transport of vapor from the bentonite into the granite. The inferred HCO{sub 3}{sup -} and pH data cannot be explained solely by solute transport, calcite dissolution and protonation/deprotonation by surface complexation, suggesting that such data may be affected also by other …

Effects of nitrogen additions on above- and belowground carbon dynamics in two tropical forests

Anthropogenic nitrogen (N) deposition is increasing rapidly in tropical regions, adding N to ecosystems that often have high background N availability. Tropical forests play an important role in the global carbon (C) cycle, yet the effects of N deposition on C cycling in these ecosystems are poorly understood. We used a field N-fertilization experiment in lower and upper elevation tropical rain forests in Puerto Rico to explore the responses of above- and belowground C pools to N addition. As expected, tree stem growth and litterfall productivity did not respond to N fertilization in either of these Nrich forests, indicating a lack of N limitation to net primary productivity (NPP). In contrast, soil C concentrations increased significantly with N fertilization in both forests, leading to larger C stocks in fertilized plots. However, different soil C pools responded to N fertilization differently. Labile (low density) soil C fractions and live fine roots declined with fertilization, while mineral-associated soil C increased in both forests. Decreased soil CO2 fluxes in fertilized plots were correlated with smaller labile soil C pools in the lower elevation forest (R2 = 0.65, p\0.05), and with lower live fine root biomass in the upper elevation forest (R2 = 0.90, …

The Division of Nuclear Chemistry and Technology (NUCL) of the American Chemistry Society (ACS) is sponsoring a symposium entitled "New Directions in Isotope Production, Nuclear Forensics and Radiochemistry Supported by the DOE" at the 240th ACS National Meeting in Boston, MA 22-26 August 2010. Radiochemistry and nuclear science is a critical area of research and funding for which the DOE has provided support over the years. Radiochemistry is undergoing a renaissance in interdisciplinary areas including medicine, materials, nanotechnology, nuclear forensics and energy. For example, interest in nuclear energy is growing in response to global warming. The field of nuclear forensics has grown significantly since 9/11 in response to potential terror threats and homeland security. Radioactive molecular imaging agents and targeted radiotherapy are revolutionizing molecular medicine. The need for radiochemists is growing, critical, and global. The NUCL Division of the ACS has been involved in various areas of radiochemistry and nuclear chemistry for many years, and is the host of the DOE supported Nuclear Chemistry Summer Schools. Do Photovoltaic Energy Systems Effect Residential Selling Prices? Results from a California Statewide Investigation.

An increasing number of homes in the U.S. have sold with photovoltaic (PV) energy systems installed at the time of sale, yet relatively little research exists that provides estimates of the marginal impacts of those PV systems on home sale prices. This research analyzes a large dataset of California homes that sold from 2000 through mid-2009 with PV installed. We find strong evidence that homes with PV systems sold for a premium over comparable homes without PV systems during this time frame. Estimates for this premium expressed in dollars per watt of installed PV range, from roughly $4 to $6.4/watt across the full dataset, to approximately $2.3/watt for new homes, to more than $6/watt for existing homes. A number of ideas for further research are suggested.

The focus of task WP 2.3.6 is to improve the U.S. Department of Energy’s (DOE’s) understanding of filtration operations for high-level waste (HLW) to improve filtration and cleaning efficiencies, thereby increasing process throughput and reducing the Na demand (through acid neutralization). Developing the cleaning/backpulsing requirements will produce much more efficient operations for both the Hanford Tank Waste Treatment and Immobilization Plant (WTP) and the Savannah River Site (SRS), thereby significantly increasing throughput by limiting cleaning cycles. The scope of this work is to develop the understanding of filter fouling to allow developing this cleaning/backpulsing strategy.
