Polymerase Chain Reaction-based Suppression of Repetitive Sequences in Whole Chromosome Painting Probes for FISH

We have developed a method to suppress the PCR amplification of repetitive sequences in whole chromosome painting probes by adding Cot-1 DNA to the amplification mixture. The repetitive sequences in the Cot-1 DNA bind to their homologous sequences in the probe library, prevent the binding of primers, and interfere with extension of the probe sequences, greatly decreasing PCR efficiency selectively across these blocked regions. A second labeling reaction is then done and this product is resuspended in FISH hybridization mixture without further addition of blocking DNA. The hybridization produces little if any non-specific binding on any other chromosomes. We have been able to successfully use this procedure with both human and rat chromosome probes. This technique should be applicable in producing probes for CGH, M-FISH and SKY, as well as reducing the presence of repetitive DNA in genomic libraries.

When I accepted the assignment of Associate Director for Chemistry and Materials Science (CMS), I talked to you about the need for strategic balance and excellence in all our endeavors. We also discussed how to take the directorate to the next level. The long-range CMS strategic plan presented here was developed with this purpose in mind. It also aligns with the Lab's institutional long-range science and technology plan and its 10-year facilities and infrastructure site plan. X-ray detector calibrations in the 280-eV to 100-keV energy range

The absolute sensitivity for several different types of radiation detectors has been measured using x-rays in the energy range of 280 eV to 100 keV. The photons in this energy range are produced using three separate x-ray-generating facilities. The detectors include a silicon semiconductor, two photoelectric diode detectors employing aluminum and gold photocathodes, and three detectors incorporating plastic scintillators and photodiodes. The plastic scintillators were MEL-150C, Pilot B, and NE102.

Cross-field injection of a charged, polarized, ion-electron beam

An early idea for fueling a controlled fusion device had been the injection of a polarized mixture of ions and electrons across a magnetic field and into the device. Now, the beam intensity (several kA/cm/sup 2/) required for this technique is available from pulsed ion diodes. Remaining feasibility questions involve beam optics and trapping. The most obvious advantage over neutral-beam injection is avoidance of the need to produce high-energy atoms. Therefore, the technique will compete best at ion energies above 100 keV. The method appears feasible for pulsed startup of mirror machines, but not for steady-state injection into a plasma.

Glass transition temperatures of epoxy resins by pulsed nuclear magnetic resonance spectroscopy

Pulsed nuclear magnetic resonance spectroscopy has been used to measure the glass transition temperatures of cured epoxy resins. These measurements make it possible to monitor the cure and determine the glass transition temperature as a function of the curing conditions and the concentration of the components. Knowledge of the glass transition temperature of the cured epoxies allows screening of them for a number of uses, including adhesives and coatings operations.

Hydrogen--deuterium exchange in water vapor: the mass spectrometric sensitivities and the equilibrium constant

The equilibrium constant, K/sub HDO/, for the reaction H/sub 2/O + D/sub 2/O = 2HDO can be expressed as an intensity ratio, I, measured mass spectrometrically, times a sensitivity ratio, S, measured in mass spectrometric calibration experiments. The latter is difficult to measure and previously was assumed to be unity. The 2.4 percent discrepancy between K's from theoretical calculations and direct mass spectrometric measurements might be explained by another value of S. An indirect measurement of S using a pulsed-molecular beam quadrupole mass filter that has a unique three-chamber, three-leak gas inlet system is reported. The results show the sensitivities are probably equal and therefore S = 1. Systematic errors were found in the procedure, however, which precluded an unambiguous test of the theory.

Determination of Free Nitric Acid in UNH Solutions, Problem Assignment No. 208-X2A, Progress Report

Technical report abstract: Since UNH is a highly dissociated compound which ionizes to NO3 and (UO2), the latter ion being very weakly basic, a solution of pure UNH is quite acid (pH about 3). It is possible to titrate to a phenolphthalein and point with standard caustic solution and obtain a figure which represents the free acid plus the NO3 from the the UNH. However, titration of the free nitric acid alone in such a solution presents quite a problem. By precipitating and removing uranium from solution by means of potassium ferrocyanide, the residual free acid may be titrated directly by means of standard caustic using either methyl red or phenolphthalein indicators, with an error of less than 4%. A method is given to determine formic acid, should that acid be present in the mixture.

The Influence of Operation on Tuballoy

An attempt is made to estimate the influence of operation upon the mechanical properties of tubealloy. It is to be emphasized that the work is highly speculative, being base upon a set of judicious assumptions regarding the transport material within the metal and should not be used as a substitute for a set of well-planned experiments. The two principal effects considered are the hardening which results from the atomic displacement produced by the fission fragments and the changes in mechanical properties arising from the presence of the fission products. The principal conclusion to be drawn is that the disruption of the metal which accompanies operation takes place at at sufficiently high rate that it could readily cause serious changes in the mechanical properties of the slups. the reversal of the disruption resulting from thermal effects probably is sufficiently high to prevent serious embrittlement as a consequence of displacement through most of the volume of the slug if not at the surface. On the other hand, the migration of atoms also appears to be sufficiently high that the rare gas products may have time to diffuse to cracks and produce embrittlement.

Remote Area Scintillation Monitoring System

This report was written to describe briefly several possible variations of such systems along with probable cost estimates. Previous work has been shown that the application of scintillation detectors is the simplest and most reliable means for such monitoring. By proper detector selection and measuring methods, the system can be of an approximate gamma dose-rate with gamma energy independence above about 100 Kev. The same system using detector change only is directly applicable to beta, gamma, slow neutron and fast neutron detection. Any type of alarming device desired can be incorporated.

Uranium Cold Extrusion (open access)

Uranium Cold Extrusion

Several hollow uranium cores of "C" size I & E diameters were fabricated by cold extrusion (550 to 750 F) at Hunter Douglas Aluminum Corporation. Results show diameter control and reproducibility are excellent. Preferred orientation induced by this process is completely removed by a single standard beta heat treatment.
