Advanced Modeling and Materials in Kraft Pulp Mills

This CRADA provided technical support to the Weyerhaeuser Company on a number of issues related to the performance and/or selection of materials at a number of locations in a pulp and paper mill. The studies related primarily to components for black liquor recovery boilers, but some effort was directed toward black liquor gasifiers and rolls for paper machines. The purpose of this CRADA was to assist Weyerhaeuser in the evaluation of materials exposed in various paper mill environments and to provide direction in the selection of alternate materials, when appropriate.

Web-interfaced Nonlinear Optical Waveguide and Photonic Crystal Simulator

We report on the development of new methods for the computation of spectral bandpass properties of photonic crystals and for the electromagnetic wave propagation in second order nonlinear optical waveguides. The former is based on a generalization of characteristic matrices while the latter is based on path integrals. Accurate and efficient propagation methods and algorithms form the basis for the construction of design tools for integrated optics.

Direct to Digital Holography

In this CRADA, Oak Ridge National Laboratory (ORNL) assisted nLine Corporation of Austin, TX in the development of prototype semiconductor wafer inspection tools based on the direct-to-digital holographic (DDH) techniques invented at ORNL. Key components of this work included, testing of DDH for detection of defects in High Aspect Ratio (HAR) structures, development of image processing techniques to enhance detection capabilities through the use of both phase and intensity, and development of methods for autofocus on the DDH tools.

During the project period, we have interacted with between 15 and 20 companies in the optical fiber telecommunications equipment industry, and our students have participated in a number of highly visible projects with companies such as Ciena, Science Applications International Corporation, KDD, ATT, Virtual Photonics, Inc., Phaethon Telecommunications, PhotonEx, and others. The project led to many successful interactions and numerous job offers for our students.

Disruption of NBS1 gene leads to early embryonic lethality in homozygous null mice and induces specific cancer in heterozygous mice

Nijmegen breakage syndrome (NBS) is a rare autosomal recessive chromosome instability syndrome characterized by microcephaly, growth retardation, immunodeficiency, and cancer predisposition, with cellular features similar to that of ataxia telangiectasia (AT). NBS results from mutations in the mammalian gene Nbs1 that codes for a 95-kDa protein called nibrin, NBS1, or p95. To establish an animal model for NBS, we attempted to generate NBS1 knockout mice. However, NBS1 gene knockouts were lethal at an early embryonic stage. NBS1 homozygous(-/-) blastocyst cells cultured in vitro showed retarded growth and subsequently underwent growth arrest within 5 days of culture. Apoptosis, assayed by TUNEL staining, was observed in NBSI homozygous(-/-) blastocyst cells cultured for four days. NBSI heterozygous(+/-) mice were normal, and exhibited no specific phenotype for at least one year. However, fibroblast cells from NBSI heterozygous(+/-) mice displayed an enhanced frequency of spontaneous transformation to anchorage-independent growth as compared to NBS1 wild-type(+/+) cells. Furthermore, heterozygous(+/-) mice exhibited a high incidence of hepatocellular carcinoma after one year compared to wild-type mice, even though no significant differences in the incidence of other tumors such as lung adenocarcinoma and lymphoma were observed. Global Expression Studies of Yersinia Pestis Pathogenicity

The aim of these studies continues to be the investigation into the molecular mechanisms that underlie the virulence process in Yersinia pestis. In particular, the focus of this work centers on the identification of novel genes and pathways responsible for the pathogenic properties of this organism. In spite of more than four decades of intense investigation in this field, the dilemma as to what makes Y. pestis such a virulent and lethal pathogen remains unanswered. The method being employed makes use microarray technology (DNA chip) that enables the examination of the global activities of the whole complement of genes in this pathogen. Two primary resources available to the investigators (one directly obtained from a separate CBNP-funded project) make these studies possible: (1) Whole genome comparisons of the genes in Y. pestis and its near neighbors with attenuated or non pathogenic characteristics, and (2) the ability to duplicate in vitro, conditions that mimic the infection process of this pathogen. Advances in Low-Defect Multilayers for EUVL Mask Blanks

Low-defect multilayer coatings are required to fabricate mask blanks for Extreme Ultraviolet Lithography (EUVL). The mask blanks consist of high reflectance E W multilayers on low thermal expansion substrates. A defect density of 0.0025 printable defects/cm{sup 2} for both the mask substrate and the multilayer is required to provide a mask blank yield of 60%. Current low defect multilayer coating technology allows repeated coating-added defect levels of 0.05/cm{sup 2} for defects greater than 90 nm polystyrene latex sphere (PSL) equivalent size for lots of 20 substrates. Extended clean operation of the coating system at levels below 0.08/cm{sup 2} for 3 months of operation has also been achieved. Two substrates with zero added defects in the quality area have been fabricated, providing an existence proof that ultra low defect coatings are possible. Increasing the ion source-to-target distance from 410 to 560 mm to reduce undesired coating of the ion source caused the defect density to increase to 0.2/cm{sup 2}. Deposition and etching diagnostic witness substrates and deposition pinhole cameras showed a much higher level of ion beam spillover (ions missing the sputter target) than expected. Optimal Elevation and Configuration of Hanford's Double-Shell Tank Waste Mixer Pumps

The objective of this study was to compare the mixing performance of the Lawrence pump, which has injection nozzles at the top, with an alternative pump that has injection nozzles at the bottom, and to determine the optimal elevation for the alternative pump. Sixteen cases were evaluated: two sludge thicknesses at eight levels. A two-step evaluation approach was used: Step 1 to evaluate all 16 cases with the non-rotating mixer pump model and Step 2 to further evaluate four of those cases with the more realistic rotating mixer pump model. The TEMPEST code was used.

The Computational Complexity of the Minimum Degree Algorithm

The Minimum Degree algorithm, one of the classical algorithms of sparse matrix computations, is widely used to order graphs to reduce the work and storage needed to solve sparse systems of linear equations. There has been extensive research involving practical implementations of this algorithm over the past two decades. However, little has been done to establish theoretical bounds on the computational complexity of these implementations. We study the Minimum Degree algorithm, and prove time complexity bounds for its widely used variants.

Lawrence Livermore National Laboratory East Avenue/Emergency Response Planning Traffic Study

The Lawrence Livermore National Laboratory (LLNL) and the Sandia National Laboratory (SNL) are located at the eastern end of the City of Livermore. Recently the research facilities have been placed on heightened security alerts due to the events of September 11. To respond to the security concerns, LLNL and SNL have proposed to place East Avenue between South Vasco Road and Greenville Road under administrative control. This type of control would require security check points at both ends of this segment of East Avenue, including a truck inspection facility west of the Greenville Road intersection. In this configuration, East Avenue would be closed to general public traffic. The purpose of this traffic study is to determine the potential traffic impact of placing East Avenue under administrative control. The primary focus of the traffic study is to evaluate the proposed modifications to the geometry and operation of East Avenue between South Vasco Road and Greenville Road. The study also includes a review of a traffic study prepared for the closure of East Avenue in 1989 to determine if the assumptions and conclusions of that study remain valid.

Supersymmetry and the Cosmic Ray Positron Excess

We explore several supersymmetric alternatives to explain predictions for the cosmic ray positron excess. Light sneutrino or neutralino LSP's, and a fine-tuned model designed to provide a delta-function input, can give adequate statistical descriptions of the reported HEAT data if non-thermal production of the relic cold dark matter density dominates and/or if"boost factors" (that could originate in uncertainties from propagation or local density fluctuations) to increase the size of the signal are included. All the descriptions can be tested at the Tevatron or LHC, and some in other WIMP detecting experiments.

Combined hyperlipidemia (CHL) is a common disorder of lipidmetabolism that leads to an increased risk of cardiovascular disease. Thelipid profile of CHL is characterised by high levels of atherogeniclipoproteins and low levels of high-density-lipoprotein-cholesterol.Apolipoprotein (APO) A5 is a newly discovered gene involved in lipidmetabolism located within 30kbp of the APOA1/C3/A4 gene cluster. Previousstudies have indicated that sequence variants in this cluster areassociated with increased plasma lipid levels. To establish whethervariation at the APOA5 gene contributes to the transmission of CHL, weperformed linkage and linkage disequilibrium (LD) tests on a large cohortof families (n=128) with familial CHL (FCHL). The linkage data producedevidence for linkage of the APOA1/C3/A4/A5 genomic interval to FCHL (NPL= 1.7, P = 0.042). The LD studies substantiated these data. Twoindependent rare alleles, APOA5c.56G and APOC3c.386G of this gene clusterwere over-transmitted in FCHL (P = 0.004 and 0.007, respectively), andthis was associated with a reduced transmission of the most commonAPOA1/C3/A4/A5 haplotype (frequency 0.4425) to affected subjects (P =0.013). The APOA5c.56G allele was associated with increased plasmatriglyceride levels in FCHL probands, whereas the second, andindependent, APOC3c.386G allele was associated with increased plasmatriglyceride levels in FCHL pedigree founders. CO2 Sequestration in Coalbed Methane Reservoirs: Experimental Studies and Computer Simulations

One of the approaches suggested for sequestering CO{sub 2} is by injecting it in coalbed methane (CBM) reservoirs. Despite its potential importance for CO{sub 2} sequestration, to our knowledge, CO{sub 2} injection in CBM reservoirs for the purpose of sequestration has not been widely studied. Furthermore, a key element missing in most of the existing studies is the comprehensive characterization of the CBM reservoir structure. CBM reservoirs are complex porous media, since in addition to their primary pore structure, generated during coal formation, they also contain a variety of fractures, which may potentially play a key role in CO{sub 2} sequestration, as they generally provide high permeability flow paths for both CO{sub 2} and CH{sub 4}. In this report we present an overview of our ongoing experimental and modeling efforts, which aim to investigate the injection, adsorption and sequestration of CO{sub 2} in CBM reservoirs, the enhanced CH{sub 4} production that results, as well as the main factors that affect the overall operation. We describe the various experimental techniques that we utilize, and discuss their range of application and the value of the data generated. Several DOE sites have been contaminated by transuranicradionuclide (TRU) discharges including neptunium and plutonium. Theirinteraction with the surrounding geological media can affect thetransport and remediation of these radionuclides in the environment.Manganese based minerals, present as minor phases in the vadose zone, canpreferentially sequester TRU over other minerals present in largerquantities. The objective of this project is to understand theinteractions between plutonium and neptunium and manganese oxyhydroxideminerals to predict potential hazards they represent to the environment,as well as to provide important scientific information for the design ofeffective remediation strategies for contaminated DOE sites.

Apolipoprotein A5: A newly identified gene impacting plasmatriglyceride levels in humans and mice

Apolipoprotein A5 (APOA5) is a newly described member of theapolipoprotein gene family whose initial discovery arose from comparativesequence analysis of the mammalian APOA1/C3/A4 gene cluster. Functionalstudies in mice indicated that alteration in the level of APOA5significantly impacted plasma triglyceride concentrations. Miceover-expressing human APOA5 displayed significantly reducedtriglycerides, while mice lacking apoA5 had a large increase in thislipid parameter. Studies in humans have also suggested an important rolefor APOA5 in determining plasma triglyceride concentrations. In theseexperiments, polymorphisms in the human gene were found to define severalcommon haplotypes that were associated with significant changes intriglyceride concentrations in multiple populations. Several separateclinical studies have provided consistent and strong support for theeffect with 24 percent of Caucasians, 35 percent of African-Americans and53 percent of Hispanics carrying APOA5 haplotypes associated withincreased plasma triglyceride levels. In summary, APOA5 represents anewly discovered gene involved in triglyceride metabolism in both humansand mice whose mechanism of action remains to be deciphered.

20th Edition of TOP500 List of World's Fastest Supercomputers Released MANNHEIM, Germany; KNOXVILLE, Tenn.;&BERKELEY, Calif. In what has become a much-anticipated event in the world of high-performance computing, the 20th edition of the TOP500 list of the world's fastest supercomputers was released today (November 15, 2002). The Earth Simulator supercomputer installed earlier this year at the Earth Simulator Center in Yokohama, Japan, is with its Linpack benchmark performance of 35.86 Tflop/s (trillions of calculations per second) retains the number one position. The No.2 and No.3 positions are held by two new, identical ASCI Q systems at Los Alamos National Laboratory (7.73Tflop/s each). These systems are built by Hewlett-Packard and based on the Alpha Server SC computer system.

CALCIUM CARBONATE PRODUCTION BY COCCOLITHOPHORID ALGAE IN LONG TERM, CARBON DIOXIDE SEQUESTRATION

Predictions of increasing levels of anthropogenic carbon dioxide (CO{sub 2}) and the specter of global warming have intensified research efforts to identify ways to sequester carbon. A number of novel avenues of research are being considered, including bioprocessing methods to promote and accelerate biosequestration of CO{sub 2} from the environment through the growth of organisms such as coccolithophorids, which are capable of sequestering CO{sub 2} relatively permanently. Calcium and magnesium carbonates are currently the only proven, long-term storage reservoirs for carbon. Whereas organic carbon is readily oxidized and releases CO{sub 2} through microbial decomposition on land and in the sea, carbonates can sequester carbon over geologic time scales. This proposal investigates the use of coccolithophorids--single-celled, marine algae that are the major global producers of calcium carbonate--to sequester CO{sub 2} emissions from power plants. Cultivation of coccolithophorids for calcium carbonate (CaCO{sub 3}) precipitation is environmentally benign and results in a stable product with potential commercial value. Because this method of carbon sequestration does not impact natural ecosystem dynamics, it avoids controversial issues of public acceptability and legality associated with other options such as direct injection of CO{sub 2} into the sea and ocean fertilization. A Development of On-Line Temperature Measurement Instrumentation for Gasification Process Control Progress Report

This progress report covers continuing work to develop a temperature probe for a coal gasifier. A workable probe design requires finding answers to crucial questions involving the probe materials. We report on attempts to answer those questions. We designed, assembled, and tested a portable test fixture that can give relative quantitative data on the condition of phosphors. It needs a more-sensitive detector for optimum performance. We ordered an appropriate detector. An experimental test of the survivability of thermographic phosphor in an ambient environment similar to that in a slagging gasifier showed no substantial deterioration of the phosphor. We consider this result so important that we delayed the date of publication of this report by one month to accommodate it. We assembled the first version of a prototype probe and were preparing to test it at the time of this report.
