Disregard restriction header and footer - We audited the procedures, data, and controls that the Reserve Component Process Action Team used to develop Army Reserve Component proposals for inclusion in the Base Realignment and Closure 2005 process. Although we identified some areas where the Process Action Team needed to strengthen procedures, the team took corrective actions during the audit, and we concluded that the team had appropriate procedures for identifying candidate facilities and functions to study for possible closure or realignment. Our review of the support for the critical data elements for 59 Reserve Component proposals identified differences in amounts reported and what was supported by documentation or audit results. However, the team took actions to mitigate the risk of using incorrect data and established procedures to correct and recertify the data. Specifically, the team:

Significant time and effort are often required to solve and complete a macromolecular crystal structure. The development of automated computational methods for the analysis, solution and completion of crystallographic structures has the potential to produce minimally biased models in a short time without the need for manual intervention. The PHENIX software suite is a highly automated system for macromolecular structure determination that can rapidly arrive at an initial partial model of a structure without significant human intervention, given moderate resolution and good quality data. This achievement has been made possible by the development of new algorithms for structure determination, maximum-likelihood molecular replacement (PHASER), heavy-atom search (HySS), template and pattern-based automated model-building (RESOLVE, TEXTAL), automated macromolecular refinement (phenix.refine), and iterative model-building, density modification and refinement that can operate at moderate resolution (RESOLVE, AutoBuild). These algorithms are based on a highly integrated and comprehensive set of crystallographic libraries that have been built and made available to the community. The algorithms are tightly linked and made easily accessible to users through the PHENIX Wizards and the PHENIX GUI.

Nuclear fission, nuclear fusion, and renewable energy (including biofuels) are the only energy sources capable of satisfying the Earth's need for power for the next century and beyond without the negative environmental impacts of fossil fuels. This chapter will address some of the many issues associated with the identification of signatures based on genomic DNA/RNA, which can be used to identify and characterize pathogens for biodefense and microbial forensic goals. For the purposes of this chapter, we define a signature as one or more strings of contiguous genomic DNA or RNA bases that are sufficient to identify a pathogenic target of interest at the desired resolution and which could be instantiated with particular detection chemistry on a particular platform. The target may be a whole organism, an individual functional mechanism (e.g., a toxin gene), or simply a nucleic acid indicative of the organism. The desired resolution will vary with each program's goals but could easily range from family to genus to species to strain to isolate. The resolution may not be taxonomically based but rather pan-mechanistic in nature: detecting virulence or antibiotic-resistance genes shared by multiple microbes. Entire industries exist around different detection chemistries and instrument platforms for identification of pathogens, and we will only briefly mention a few of the techniques that we have used at Lawrence Livermore National Laboratory (LLNL) to support our biosecurity-related work since 2000.
