Comparison of Heteranthera Dubia (Jacq.) MacM.-associated Macroinvertebrates Between Georgraphical Regions in the United States

Macroinvertebrates associated with the aquatic plant, water stargrass (Heteranthera dubia), were sampled from 12 waterbodies in four regions of the United States from June to August 2005. Taxa richness, evenness, and diversity were lowest in the Lower Midwest (LMW) region, and higher in Northern sites, especially the Upper Midwest (UMW), and Northeast (NE). While relative abundance varied from site to site and region to region, utilization of the plant by functional groups remained fairly constant. Collector-gatherers consistently comprised the largest portion of invertebrates sampled. The shredder/ herbivore functional group comprised an average of 17 % of total groups. Through an exhaustive literature review, it was found that shredder/ herbivores of water stargrass have not been reported in the literature. Because of this, the herbivore group was analyzed separately and consisted of 2,383 specimens representing 23 species. The most common groups were Rhopalosiphum sp., Nectopsyche spp. and chironomids. No differences were found in herbivore diversity or evenness between sampling regions, but species richness was significantly different.

Modulation of the Coelomic Fluid Protein Profile in the Earthworm, Lumbricus Terrestris, After Exposure to Copper as Copper Sulfate

Proteomic techniques were used to analyze the protein profile of earthworm, Lumbricus terrestris, coelomic fluid collected by either whole body dissection method or the coelomic cavity puncture method. Data demonstrated that collection of coelomic fluid using the coelomic cavity puncture method protocol resulted in a 32% reduction, 377 +/- 4.5 vs 253+/- 19.9 (p=0.0007), in the number of individual proteins. It was determined that the coelomic cavity puncture method yielded a "cleaner" preparation, one less contaminated with extraneous proteins from intestinal tissue, gut contents, and body wall materials. This protocol was used in all later studies. The same proteomic techniques were used to evaluate the effects that exposure to Cu (1.0 μg/cm2) as CuSO4 had on the earthworm coelomic fluid profile. Comparison of protein profile from exposed earthworms demonstrated a significant reduction in the number of proteins expressed (184 ± 2.64 vs 253 ±19.9 p=0.0192) when compared to control organisms. Cu exposure also resulted in a modulation of the protein profile with treated earthworms expressing 47 new proteins that were not identified in unexposed worm coelomic fluid. Adaptive Advantages of Carotenoid Pigments in Alpine and Subalpine Copepod Responses to Polycyclic Aromatic Hydrocarbon Induced Phototoxicity

Alpine zooplankton are exposed to a variety of stressors in their natural environment including ultraviolet radiation. Physiological coping mechanisms such as the accumulation of photoprotective compounds provide these zooplankton protection from many of these stressors. Elevated levels of carotenoid compounds such as astaxanthin have been shown to help zooplankton survive longer when exposed to ultraviolet radiation presumably due to the strong antioxidant properties of carotenoid compounds. This antioxidant capacity is important because it may ameliorate natural and anthropogenic stressor-induced oxidative stress. While previous researchers have shown carotenoid compounds impart increased resistance to ultraviolet radiation in populations of zooplankton, little work has focused on the toxicological implications of PAH induced phototoxicity on zooplankton containing high levels of carotenoid compounds. This thesis discusses research studying the role that carotenoid compounds play in reducing PAH induced phototoxicity. By sampling different lakes at elevations ranging from 9,500' to 12,700' in the front range of the Colorado Rocky Mountains, copepod populations containing different levels of carotenoid compounds were obtained. These populations were then challenged with fluoranthene and ultraviolet radiation. Effects of Suspended Multi-Walled Carbon Nanotubes on Daphnid Growth and Reproduction

Multi-walled carbon nanotube aggregates can be suspended in the aqueous phase by natural organic matter. These aggregates are ingested by filter feeding zooplankton. Ingested aggregates result in decreased growth and decreased reproduction. These effects may be caused by reduction in energy input from normal feeding behavior. pH alters natural organic matter structure through changes in electrostatic repulsion. Altered natural organic matter structure changes multi-walled carbon nanotube aggregate size. This size variation with variation in pH is significant, but not large enough a change in size to alter toxicity, as the aggregate size range remains well within the particle size selection of the organisms.

Familial Hypertrophic cardiomyopathy (HCM) causes ventricle walls to thicken and often leads to sudden death especially in adults. Mutations in the subfragment 2 (S2) of β-cardiac myosin are implicated in the genetic disorder. This S2 region is a coiled-coil rod region resulting from the dimeric form of myosin II. It has been proposed that an elastic quality allows normal S2 to absorb force during the powerstroke according to the sliding filament model. To test the flexibility of single molecules of S2 against levels of physiological force, the Gravitational Force Spectrometer (GFS) is being developed. This novel system employs a standard microscope on an equatorial mount that allows the spectrometer to be rotated freely in space. Stationary glass beads are attached to a microscope slide where the molecule is tethered between the stationary bead and a smaller mobile bead. The GFS is oriented so that the force of gravity can act on the mobile bead and so impart a small force to the tethered subfragment. Additionally, a video system in conjunction with ImageJ software makes a distance measurement of the molecule possible with a resolution of around 11 nm. Triclosan (TCS) is an antimicrobial compound found in several types of common household products. After being washed down the drain, TCS will then end up in the local watershed. Although numerous studies have been conducted to evaluate the fate and effects of TCS in aquatic environments, there have been no studies evaluating the role arbuscular mycorrhizal fungi (AM) play in a plants response to TCS exposure. Three species of wetland plants native North Texas were inoculated with AM spores and exposed to 0, 0.4 g/L and 4.0 g/L TCS concentrations. Root morphology of E. prostrata and S. herbacea showed AM and exposure responses. S. herbacea produced the greatest amounts biomass and TCS bioaccumulation, in all but one treatment. It also displayed opposing results to E. prostrata in measures of root length, root surface area, relative root mass, relative shoot mass and shoot:root ratio. TCS root tissue concentrations increased with increased exposures for both E. prostrata and S. herbacea. Even though E. prostrata had the lowest levels in each measure of biomass production, it had the highest amount of root TCS bioaccumulation in the AM inoculated 4.0 g/L treatment. Exploring the Evolutionary History of North American Prairie Grouse (Genus: Tympanuchus) Using Multi-locus Coalescent Analyses

Conservation biologists are increasingly using phylogenetics as a tool to understand evolutionary relationships and taxonomic classification. The taxonomy of North American prairie grouse (sharp-tailed grouse, T. phasianellus; lesser prairie-chicken, T. pallidicinctus; greater prairie-chicken, T. cupido; including multiple subspecies) has been designated based on physical characteristics, geography, and behavior. However, previous studies have been inconclusive in determining the evolutionary history of prairie grouse based on genetic data. Therefore, additional research investigating the evolutionary history of prairie grouse is warranted. In this study, ten loci (including mitochondrial, autosomal, and Z-linked markers) were sequenced across multiple populations of prairie grouse, and both traditional and coalescent-based phylogenetic analyses were used to address the evolutionary history of this genus. Results from this study indicate that North American prairie grouse diverged in the last 200,000 years, with species-level taxa forming well-supported monophyletic clades in species tree analyses. With these results, managers of the critically endangered Attwater's prairie-chicken (T. c. attwateri) can better evaluate whether outcrossing Attwater's with greater prairie-chickens would be a viable management tool for Attwater's conservation.

Oxygen availability is critical for survival for most organisms. The nematode, C. elegans, has been useful for studying genetic regulation of anoxia tolerance due to the oxygen deprivation response mechanisms shared with other metazoans. Studies examining long-term anoxia (72h, LTA) tolerance have only been conducted at adult day 1. To investigate the effect of aging on anoxia tolerance wild-type and mutant strains were exposed to LTA between adult day 1 and day 9. Wild-type isolates and daf-16(mu86) (FOXO transcription factor regulated by insulin-signaling) and aak-2(gt33) (catalytic subunit of AMP-activated protein kinase) strains were anoxia sensitive at day 1 and displayed increased LTA tolerance with aging correlated with reproductive senescence followed by a decline in survivorhsip through day 9. The daf-2(e1370) (insulin receptor homologue of C. elegans), glp-1(e2141) (a lin-12/Notch receptor) and fog-2(q71) (required for spermatogenesis) strains were LTA-tolerant through day 5. I conclude that aging influences LTA-tolerance in a strain- and age-dependent manner. In addition to being LTA-tolerant the daf-2(e1370) and glp-1(e2141) strains have a longevity phenotype that is suppressed by loss of kri-1 or daf-12. Many freshwater mussels are imperiled, due to a number of interrelated factors such as habitat alteration, degradation of water quality, and impoundments. The Golden Orb mussel (Quadrula aurea, I. Lea, 1859) is endemic to the state of Texas and is currently a candidate for the endangered species list, as the number of known populations has been declining in recent years. Little is currently known about Q. aurea aside from basic distribution data. This study is focused on evaluating a combination of macro-habitat and micro-habitat variables to determine their influence on the distribution and density of this species. Macro-habitat variables, including dominant land cover, surface geology, and soil erodibility factor, did not have a significant relationship with mussel distributions. The best model of micro-habitat variables that impacts the Q. aurea distributions is comprised of relative substrate stability (RSS) at moderate flows and current velocity at low flows. For all mussel species in this study, current velocity at low flows is the primary variable that influences distribution. Q. aurea are associated with habitats where larger sediment particles (large gravel and cobble) help to stabilize the substrate in areas with higher current velocities. oligodendrocytes migrate from the corpus callosum into the overlying cortex. The incidence of cilia did not change markedly across age groups, and did not vary consistently with the number of processes per cell, which was used as an indication of the maturation stage of OPCs and young OLs. The mean percent of Olig1 immunopositive (Olig1+) cells having cilia across ages was 33.1% + 16.5%, with all ages combined. In O4+ cells of these mice, 56.7 + 3.6% had primary cilia. If it is the case that adult OLs do not have cilia, the point in the lineage when primary cilia are lost is still unknown. Adult mice that had been injected with cyclopamine to block cilia-dependent Shh signaling were examined to determine whether the rate of generating new OPCs was influenced. Examination of the Relationship Between Glucuronic Acid and Vascular Damage in Rats

The goal of this experiment was to examine the role of glucuronic acid in the development of vascular damage in the kidneys and retinas of diabetic individuals. Glucuronic acid was provided to rats in their water at various concentrations in order to increase plasma levels of the compound. Kidneys and retinas were excised and compared to control specimens using microscopy to determine the effect of elevated blood glucuronic acid levels on the occurrence of microaneurysms in renal capillary networks. No differences were seen between the treatment and control groups. Further study needs to be conducted to determine a more suitable time frame for this experiment.

Expression of G-protein Coupled Receptors in Young and Mature Thrombocytes and Knockdown of Gpr18 in Zebrafish

In this study, a novel method based on biotinylated antibodies and streptavidin coated magnetic beads was used to separate the thrombocyte subpopulations from zebrafish whole blood. DiI-C18, a lipophilic dye, labels only young thrombocytes when used at low concentrations. Commercially available biotinylated anti-Cy3 antibody was used to label the chromophore of DiI-C18 on the young thrombocytes and streptavidin coated magnetic beads were added subsequently, to separate young thrombocytes. The remaining blood cells were probed with custom-made biotinylated anti-GPIIb antibody and streptavidin magnetic beads to separate them from other cells. Further, thrombocytes are equivalents of mammalian platelets. Platelets play a crucial role in thrombus formation. The G-protein coupled receptors (GPCRs) present on the platelet surface are involved during platelet activation and aggregation processes. So, thrombocytes were studied for the presence of GPCRs. The GPCR mRNA transcripts expressed in the young and mature thrombocytes were subjected to densitometry analysis and pixel intensities of the bands were compared using one way ANOVA. This analysis did not show significant differences between the young and mature GPCR mRNA transcripts but identified a novel GPCR, GPR18 that was not reported in platelets earlier. Novel Role of Trypsin in Zebrafish

It has been shown previously in our laboratory that zebrafish produce trypsin from their gills when they are under stress, and this trypsin is involved in thrombocyte activation via PAR2 during gill bleeding. In this study, I investigated another role of the trypsin that is secreted from zebrafish. This investigation has demonstrated a novel role of trypsin in zebrafish. Not only did this investigation demonstrate the role of trypsin in zebrafish behavior, but also it showed that PAR2 might be the receptor that is involved in trypsin-mediated behavioral response. In addition, we have shown that Gq and ERK inhibitors are able to block the trypsin pathway and prevent the escaping behavior. Finally, the results of this investigation suggest that the cells that respond to trypsin are surface cells, which have an appearance similar to that of neuromast cells.

The Influence of Urban Green Spaces on Declining Bumble Bees (Hymenoptera: Apidae)

Bumble bees (Bombus spp.) are adept pollinators of countless cultivated and wild flowering plants, but many species have experienced declines in recent decades. Though urban sprawl has been implicated as a driving force of such losses, urban green spaces hold the potential to serve as habitat islands for bumble bees. As human populations continue to grow and metropolitan areas become larger, the survival of many bumble bee species will hinge on the identification and implementation of appropriate conservation measures at regional and finer scales. North Texas is home to some the fastest-growing urban areas in the country, including Denton County, as well as at least two declining bumble bee species (B. pensylvanicus and B. fraternus). Using a combination of field , molevular DNA and GIS methods I evaluated the persistence of historic bumble bee species in Denton County, and investigated the genetic structure and connectivity of the populations in these spaces. Field sampling resulted in the discovery of both B. pensylvanicus and B. fraternus in Denton County's urban green spaces. While the relative abundance of B. fraternus in these spaces was significantly lower than historic levels gleaned from museum recors, that of B. pensylvanicus was significantly higher. Biodiversity and Genetic Structure of Benthic Macroinvertebrates Along an Altitudinal Gradient: A Comparison of the Windhond and Róbalo River Communities on Navarino Island, Chile

Altitudinal gradients in Sub-Antarctic freshwater systems present unique opportunities to study the effect of distinct environmental gradients on benthic macroinvertebrate community composition and dispersal. This study investigates patterns in biodiversity, dispersal and population genetic structure of benthic macroinvertebrate fauna across an altitudinal gradient between two watersheds on Navarino Island in southern Chile. Patterns in diversity, density, evenness and functional feeding groups were not significantly different across the altitudinal gradient in both the Windhond and Róbalo Rivers. Taxa richness in both rivers generally increased from the headwaters of the river to the mouth, and functional feeding group patterns were consistent with the predictions of the River Continuum Concept. Population genetic structure and gene flow was investigated by sampling the mitochondrial cytochrome oxidase I gene in two invertebrate species with different dispersal strategies. Hyalella simplex (Amphipoda) is an obligate aquatic species, and Meridialaris chiloeense (Ephemeroptera) is an aquatic larvae and a terrestrial winged adult. Contrasting patterns of population genetic structure were observed. Results for Hyalella simplex indicate significant differentiation in genetic structure in the Amphipod populations between watersheds and lower genetic diversity in the Róbalo River samples, which may be a result of instream dispersal barriers. Informing Conservation Management Using Genetic Approaches: Greater Sage-Grouse and Galápagos Short-Eared Owls as Case Studies

Small isolated populations are of particular conservation interest due to their increased extinction risk. This dissertation investigates two small wild bird populations using genetic approaches to inform their conservation. Specifically, one case study investigated a Greater Sage-grouse (Centrocercus urophasianus) population located in northwest Wyoming near Jackson Hole and Grand Teton National Park. Microsatellite data showed that the Jackson sage-grouse population possessed significantly reduced levels of neutral genetic diversity and was isolated from other Wyoming populations. Analysis with single nucleotide polymorphisms (SNPs) and microsatellite data provided further evidence that the population's timing of isolation was relatively recent and most likely due to recent anthropogenic habitat changes. Conservation recommendations include maintaining or increasing the population's current size and reestablishing gene flow with the nearest large population. The second case study investigated the genetic distinctiveness of the Floreana island population of the Galápagos Short-eared Owl (Asio flammeus galapagoensis). Mitochondrial DNA sequence data did not detect differences across nine island populations, yet microsatellite and morphometric data indicated that limited gene flow existed with the population and surrounding island populations, which appeared asymmetric in direction from Floreana to Santa Cruz with no indication of gene flow into Floreana. Since their emergence in the early 1990s, neonicotinoid use has increased exponentially to make them the world's most prevalent insecticides. Although there is considerable research concerning the lethality of neonicotinoids, their sub-lethal and developmental effects are still being explored, especially with regards to non-mammalian species. The goal of this research was to investigate the effects of the neonicotinoid imidacloprid on the morphological and physiological development of northern bobwhite quail (Colinus virginianus). Bobwhite eggs (n = 650) were injected with imidacloprid concentrations of 0 (sham), 10, 50, 100 and 150 grams per kilogram of egg mass, which was administered at day 0 (pre-incubation), 3, 6, 9, or 12 of growth. Embryos were dissected on day 19 when they were weighed, staged, and examined for any overt structural deformities. Embryonic heart, liver, lungs and kidneys were also weighed and preserved for future use. Treated embryos exhibited increased frequency of severely deformed beaks and legs, as well as larger hearts and smaller lungs at the higher dosing concentrations. Some impacts are more pronounced in specific dosing periods, implying that there may be critical windows of development when embryos are highly susceptible to neonicotinoid exposure. Since phages use the host resources to replicate themselves after infection, the different sizes of the phage genome should influence the replication rate. We, therefore, hypothesized that the smaller genomes should burst the cell faster than the larger ones. As well, the shorter genomes would have greater burst sizes because they should replicate faster. Here, we obtained 16 phages of various genome length. All phages were isolated on Streptomyces griseus and available in our phage bank at the University of North Texas. We performed one-step growth studies for the 16 phages, as well as determined the host doubling time from its growth curve. The results show that S. griseus grown in nutrient broth has a doubling time of 5 hours and 22 minutes. This doubling time is used as a guideline for the phage growth studies. Because the filamentous nature of the host caused several difficulties during the experiment, we isolated single cells by sonication and centrifugation. After the cell number was determined by viable cell count, the cells were infected with each type of phage using a multiplicity of infection (MOI) of 0.5. Superbursts: Investigation of Abnormal Paroxysmal Bursting Activity in Nerve Cell Networks In Vitro

Superbursts (SBs) are large, seemingly spontaneous activity fluctuations often encountered in high density neural networks in vitro. Little effort has been put forth to define and analyze SBs which are paroxysmal bursting discharges. Through qualitative and quantitative means, I have described specific occurrences of superbursting activity. A complex of paroxysmal bursting has been termed a "superburst episode," and each individual SB is a "superburst event" which is comprises a fine burst structure. Quantitative calculations (employing overall spike summations and coefficient of variation (CV) calculations), reveal three distinct phases. Phase 1 is a "build up" phase of increasingly strong, coordinated bursting with an average of a 17.6% ± 13.7 increase in activity from reference. Phase 2, the "paroxysmal" phase, is comprised of massive coordinated bursting with high frequency spike content. Individual spike activity increases by 52.9% ± 14.6. Virulence Factor Production in PyrE Mutants of Pseudomonas Aeruginosa

It has been shown previously in our lab that mutations in the pyrimidine pathway reduced the ability of Pseudomonas aeruginosa to produce virulence factors. Knockout mutations in pyrB, pyrC and pyrD genes of the pyrimidine pathway showed that virulence factor production was decreased. Pyoverdin, pyocyanin, hemolysin, iron chelation, motility, and adherence are all considered virulence factors. Here I further investigate the effects of mutations in the pyrimidine pathway by studying a pyrE mutant. I studied the effect of the pyrE mutation on the production of the above virulence factors. Just like the effect of pyrB, pyrC and pyrD mutations,the pyrE mutation also showed that the bacteria were deficient in producing virulence factors when compared to the wild type. The broader impact of this research would be the possibility of finding drugs that could treat patients infected with P. aeruginosa and possibly extend the lives of chronically infected patients with cystic fibrosis.

The Effect of Natural Gas Well Setback Distance on Drillable Land in the City of Denton, Texas

Municipalities protect human health and environmental resources from impacts of urban natural gas drilling through setback distances; the regulation of distances between well sites and residences, freshwater wells, and other protected uses. Setback distances have increased over time, having the potential to alter the amount and geographical distribution of drillable land within a municipality, thereby having implications for future land use planning and increasing the potential for future incompatible land uses. This study geographically applies a range of setback distances to protected uses and freshwater wells in the city limits of Denton, Texas to investigate the effect on the amount of land remaining for future gas well development and production. Denton lies on the edge of a productive region of the Barnett Shale geological formation, coinciding with a large concentration of drillable land in the southwestern region of the study area. This region will have the greatest potential for impacts to future municipal development and land use planning as a result of future gas well development and higher setback standards. Defining Components Linked to Bacterial Nutritional Utilization of Cyanide as a Sole Nitrogen Source

One of the challenges in biology is placing a function on the myriad of gene sequences having become available from rapid advances in genome sequencing. One such example is a gene cluster (Nit1C) found in bacteria that is tied to the unusual ability of certain bacteria to grow when supplied cyanide as the sole nitrogen source. The term cyanotrophs has been applied to such bacteria, for which a genetic linkage between cyanotrophy and Nit1C was demonstrated for 10 separate bacteria. In addition to growth, cyanide induced the expression of Nit1C genes in all organisms tested, and in one case, deletion of one of the Nit1C genes (nitC) caused a loss of growth. Of the ten bacteria able to grow cyanotrophically, all gave evidence of harboring Nit1C on their genome except for two (Pseudomonas fluorescens Pf11764 and P. monteilii BCN3), which were sequenced and the presence of Nit1C was also confirmed. A broader search of bacteria identified 270 separate strains with the cluster, all limited to bacteria spanning the phyla Firmicutes, Actinobacteria, Proteobacteria and Cyanobacteria. The Bobwhite Population Decline: Its History, Genetic Consequences, and Studies on Techniques for Locating and Assessing Current Populations

The northern bobwhite (Colinus virginianus) population decline is a severe, rangewide phenomenon beginning >150 years ago and continuing today. In this investigation, I: 1. document the timeline of bobwhite population decline and unintended genetic consequences of attempted remedies, 2) develop a model useful for predicting possible locations of potentially sustainable bobwhite populations in semiarid rangeland in Texas and Oklahoma, and 3) examine the relationship between population monitoring data and meteorological factors. While breeding season call counts of male bobwhite have been used for >70 years to provide estimates of fall populations for hunting, most studies of call counts have focused on mathematics and statistical accuracy of the count, largely overlooking the influence of meteorological factors on call counts. Here, I present the results of >4,400 individual point counts and examine their relationship with meteorological variables recorded at each stop. Humidity was positively correlated with the number of birds recorded (ρ = 0.275, p < 0.001) and temperature was negatively correlated (ρ = -0.252, p < 0.001). Comparison and Genetic Analysis of Host Specificity in Cluster BD1 Bacteriophages infecting Streptomyces

Bacteriophages are viruses that specifically infect bacteria. When a phage infects a bacterium, it attaches itself to the surface of the bacteria and injects its DNA into the intracellular space. The phage DNA hijacks the cellular machinery of the bacteria and forces it to produce phage proteins. Eventually, the bacteria cell bursts or lyses, releasing new phage. The bacteria act as a host for phage reproduction. The ability for a phage to infect multiple bacterial species is known as host range. In siphoviridae bacteriophages, host range is thought to primarily be determined by proteins at the tip of their tail fibers. These proteins act as anti-receptors to specific receptors on the surface of bacteria. In siphoviridae Gram-positive infecting phages, the genes that code these proteins are typically located between the tape measure protein gene and the endolysin gene. It is hypothesized that phages that have similar anti-receptor proteins will have similar host range. In this study, the host ranges of 12 BD1 bacteriophages were tested on 9 different Streptomyces species.
