The Effects of Electrochemical Therapy on Colon-25 Tumors in Balb-C Mice

The purpose of the research was to treat immunodeficient mice, implanted with colon-25 tumors, with continuous and interrupted electrochemical therapy (ECT). ECT involves the placement of two electrodes, an anode near the center of the tumor and a cathode into the tumor periphery. A constant voltage is applied across the electrodes for a given period of time. The data showed that the interrupted and continuous ECT resulted in a decrease in mean tumor growth as compared to that of the sham controls. The histology of both ECT groups showed an increase presence of large vacuoles, randomly distributed tumor cells as well as the presence of "crevicing" in the medullary tissue. The differential leukocyte counts showed a distinct neutrophilia and lymphopenia in all groups at day 20 post tumor implantation. The results from the experimental groups appeared to support the findings of previous investigators.

Establishment and Competitive Ability of Nelumbo Lutea in Relation to Myriophyllum Spicatum

Limitations from reduced light and increasing water depth on Nelumbo lutea seedlings were determined in tank experiments. Survival was high in all tested light levels. Total biomass increased significantly with increasing light. Biomass allocation shifted significantly to root production between 3 and 6 weeks in the 10 and 24% levels. Survival decreased with increasing planting depth, and biomass of survivors reduced significantly between 0.5, 1.0, and 1.5 m depths. Nelumbo lutea and Myriophyllum spicatum populations were monitored for one season in a 0.7 ha pond to track changes in species dominance. Myriophyllum spicatum dominated early, and N. lutea dominated from July through October, suppressing M. spicatum at all depths. Competitive interactions between N. lutea and M. spicatum were investigated for two seasons in a container experiment situated within a pond. Where established, N. lutea dominated in the presence of M. spicatum. However, N. lutea could not be established in depths greater than 1 meter.

Development of a Pre-Impact Environmental Site Characterization for the Bryan Mound, Texas Strategic Petroleum Reserve

This thesis presents a model for developing site-specific contingency plans to be used during spill response, remediation, and post-spill monitoring using the Strategic Petroleum Reserve's Bryan Mound site as a case study. Bird and vegetation observations provide baseline data for biological conditions, and sediment sampling for total petroleum hydrocarbons serves as a chemical component of the model. Results demonstrate previously unknown conditions that would hinder remediation and affect the persistence of petroleum contaminants. Results also established previously unmapped dominant bird and vegetation types likely to be impacted by a spill at the site. This model points to a reconsideration of individual facilities' responsibilities when planning for large-scale disasters and protecting the sensitive ecosystems surrounding their sites.

This manual has been designed for a class of twenty- four students concurrently enrolled in the lecture course. The laboratory aids in the learning process and fosters an interest in the science of genetics. This manual and the experiments contained within are both informative and fun. The manual correlates with and expands upon the genetics course. Each investigation, with the exception of the Drosophila melanogaster project, can be completed in a 3-4 hour timeframe. This manual provides a “hands on” experience of theories simply discussed in the lecture course. This manual is intended to be a one-source manual where each investigation is designed to include an adequate introduction. Special attention has been given for each investigation with both the student and instructor in mind.

Landscape forest modeling of the Luquillo Experimental Forest, Puerto Rico.

This thesis contributes to modeling the dynamics of forest community response to environmental gradients and disturbances over a mountain landscape. A gap model (FACET) was parameterized for species of various forest types (Tabonuco, Colorado, Dwarf and Palm), for many terrain conditions and was modified and extended to include species response to excess soil moisture and hurricanes. Landscape cover types were defined by dominance of species of each forest type and canopy height. Parameters of the landscape model (MOSAIC) were calculated from multiple runs of FACET. These runs were determined by combining terrain variables (elevation and soil) and hurricane risk. MOSAIC runs were analyzed for distribution patterns. Geographic Information Systems software was used to process terrain variables, hurricane risk and MOSAIC model output.

Tobacco Phospholipase D β1: Molecular Cloning and Biochemical Characterization

Transgenic tobacco plants were developed containing a partial PLD clone in antisense orientation. The PLD isoform targeted by the insertion was identified. A PLD clone was isolated from a cDNA library using the partial PLD as a probe: Nt10B1 shares 92% identity with PLDβ1 from tomato but lacks the C2 domain. PCR analysis confirmed insertion of the antisense fragment into the plants: three introns distinguished the endogenous gene from the transgene. PLD activity was assayed in leaf homogenates in PLDβ/g conditions. When phosphatidylcholine was utilized as a substrate, no significant difference in transphosphatidylation activity was observed. However, there was a reduction in NAPE hydrolysis in extracts of two transgenic plants. In one of these, a reduction in elicitor- induced PAL expression was also observed.

Water Quality Mapping on Lake Texoma USA

The primary objective of this study was to develop and evaluate a system capable of rapid, continuous collection of water quality and locational data on Lake Texoma. Secondary objectives included developing monthly distribution maps for chlorophyll-a, turbidity, and specific conductivity in Lake Texoma and investigating the spatial and temporal relationships between these common water quality indicators. A modified YSI multiprobe was used to develop a system capable of surveying the lake within 4 days with samples at 330 to 400 meter intervals. Data generated with this system compared favorably with previous studies of Lake Texoma. Two sets of raster format maps were developed for the monthly distributions of chlorophyll-a, turbidity, and specific conductivity across the lake. Spatial and temporal relationships generally took the form of decreasing gradients running from the lake arms towards the Main Lake Zone in the case of chlorophyll-a and turbidity. Or, in the case of specific conductivity, a decreasing gradient from the Red River arm to the Washita River arm. All three water quality indicators were strongly influenced by river discharge levels.

Integrating life cycle analysis and the ecological footprint calculator to foster sustainable behaviors

Many tools have been developed to assess global, national or regional sustainable development policies. However, as governments develop sustainable policies, individuals must also feel empowered to affect their personal impact on the planet. This thesis integrates three sustainability concepts that lend themselves to individual sustainability: The natural step, life cycle assessment, and the ecological footprint. TNS serves to provide the meaning and substance toward sustainable development. LCA helps provide the framework for assessing sustainability. The EF calculator determines the driving components and measures the qualitative decisions made through TNS and LCA. From the analysis of the household footprint calculator a simplified footprint calculator was developed to assist individuals and communities in setting benchmarks and goals as they move away from over-consumption and towards a sustainable lifestyle.

Management Tools for Prescribed Burning for Tallgrass Prairie Restoration at the Lewisville Lake Environmental Learning Area

The Lewisville Lake Environmental Learning Area (LLELA) is a wildlife management area with tallgrass prairie, an endangered ecosystem. Essential ecosystem processes, especially fire, are part of restoration. To support fire management efforts at LLELA and surrounding areas, this project evaluated and developed tools for fire restoration. The four primary prairie grasses respond favorably to burning. Fuel loads and fuel models vary by scale and survey method. One- and 10-hour fuel moisture can be predicted using a statistical model; 100- and 1,000-hour fuel moisture cannot. Historic weather data suggests that burning can occur when it is most effective. The production of ozone precursors produced by burning is comparable to those emitted every six minutes by regional automobiles.

Pyrimidine Genes in Pseudomonas Species

This thesis is a comparative study of gene arrangements in Pseudomonas species, and is organized into three major sections. The first section compares gene arrangements for different pathways in Pseudomonas aeruginosa PAO1 to determine if the gene arrangements are similar to previous studies. It also serves as a reference for pyrimidine gene arrangements in P. aeruginosa. The second part compares the physical, and genetic maps of P. aeruginosa PAO1 with the genome sequence. The final section compares pyrimidine gene arrangements in three species of Pseudomonas. Pyrimidine biosynthesis and salvage genes will be aligned for P. aeruginosa PAO1, P. putida KT2440, and P. syringae DC3000. The whole study will gives insight into gene patterns in Pseudomonas, with a focus on pyrimidine genes.

The aquatic insect communities of Holbrook Creek and Cochetopa Creek in Colorado.

The first objective for this problem in lieu of thesis project was to gather, identify to the lowest practical taxonomic level and organize all available aquatic insects collected from high altitude Colorado aquatic systems during the summers of 1994, 1996, 1998, and 2002 for the University of North Texas Environmental Science Field Course (BIOL 5650). The curated collection will be housed in the Elm Fork Natural History Museum, located at the University of North Texas. The second objective was to provide a summary and discussion of the occurrence and distribution of the aquatic insects collected from Mt. Blanca in 1994, 1996, and 1998 and to create a taxa list of aquatic insects collected from Cochetopa Creek during the summer of 2002.

Comparative Bioavailability of Dietary and Dissolved Cadmium to Freshwater Aquatic Snails

Heavy metal bioaccumulation in aquatic organisms may occur through direct or indirect uptake routes. Research indicates that the significance of uptake route varies with contaminant and organism exposed. The relative importance of different metal sources in aquatic systems was investigated by exposing freshwater snails to dietary or dissolved sources of cadmium. Snails were exposed to control, contaminated food only, contaminated water only, and contaminated food and water treatments. During the 15-day exposure, samples were taken to determine Cd concentration in snail soft tissue, snail shell, algal food, and overlying water. Analyses of snail soft tissue and shells indicate that exposure route significantly affects Cd concentrations in the tissues. In both cases, dissolved Cd is the primary contributor to metal body burden.

Heart rate and oxygen consumption during the critical prenatal period in chicken embryos (Gallus gallus): Influence of light cues and the onset of pulmonary ventilation.

To examine if a rhythm can be entrained in either heart rate or oxygen consumption in late stage embryos (days 17-19.5) with light as a zeitgeber, chicken embryos were incubated in complete darkness (D:D) and 12:12 light:dark cycle (L:D). Light had no impact on oxygen consumption (390 µL O2∙min-1∙egg-1) but increased heart rate for non-internally pipped embryos (260 to 270 beats∙min-1 during light cycle). Oxygen consumption increased independent of pipping while heart rate increased (255 to 265 beats∙min-1) in D:D embryos due to pipping. A light-induced rhythm or effect occurred in heart rate but not oxygen consumption, suggesting heart rate and oxygen consumption may be uncoupled.

Isolation of a Pseudomonas aeruginosa Aspartate Transcarbamoylase Mutant and the Investigation of Its Growth Characteristics, Pyrimidine Biosynthetic Enzyme Activities, and Virulence Factor Production

The pyrimidine biosynthetic pathway is an essential pathway for most organisms. Previous research on the pyrimidine pathway in Pseudomonas aeruginosa (PAO1) has shown that a block in the third step of the pathway resulted in both a requirement for exogenous pyrimidines and decreased ability to produce virulence factors. In this work an organism with a mutation in the second step of the pathway, aspartate transcarbamoylase (ATCase), was created. Assays for pyrimidine intermediates, and virulence factors were performed. Results showed that the production of pigments, haemolysin, and rhamnolipids were significantly decreased from PAO1. Elastase and casein protease production were also moderately decreased. In the Caenorhabditis elegans infection model the nematodes fed the ATCase mutant had increased mortality, as compared to nematodes fed wild type bacteria. These findings lend support to the hypothesis that changes in the pyrimidine biosynthetic pathway contribute to the organism's ability to effect pathogenicity.

Concentration-dependent Effects of D-Methylphenidate on Frontal Cortex and Spinal Cord Networks in vitro

Spontaneously active frontal cortex and spinal cord networks grown on microelectrode arrays were used to study effects of D-methylphenidate. These central nervous system tissues have relatively low concentrations of dopaminergic and noradrenergic neurons compared to the richly populated loci, yet exhibit similar neurophysiological responses to methylphenidate. The spontaneous spike activity of both tissues was inhibited in a concentration-dependent manner by serial additions of 1-500 µM methylphenidate. Methylphenidate is non-toxic as spike inhibition was recovered following washes. The average concentrations for 50% spike rate inhibition (IC50 ± SD) were 118 ± 52 (n= 6) and 57 ± 43 (n = 11) for frontal cortex and spinal cord networks, respectively. A 3 hour exposure of a network to 1 mM methylphenidate was nontoxic. The effective concentrations described in this study are within the therapeutic dosage range. Therefore, the platform may be used for further investigations of drug mechanisms.

Solid phase microextraction of amino-dinitrotoluenes in tissue.

TNT (2,4,6-trinitrotoluene) readily and predominantly transforms to 2ADNT (2-amino-4,6-dinitrotoluene) and 4ADNT (4-amino-2,6-dinitrotoluene) in environmental matrixes and tissues. Solid phase microextraction (SPME) was used to extract ADNTs (amino-dinitrotoluenes) from tissue as a potential method to investigate the recalcitrance of metabolically-generated ADNTs versus absorbed ADNTs. Tubifex tubifex was allowed to metabolize TNT into ADNTs in 24-hr static non-renewal exposure test followed by 24-hr depuration in clean reconstituted hard water. Polyacrylate-coated (PA) SPME fibers were then deployed and agitated in tissue homogenates containing metabolically-generated ADNTs for 48 hr to provide a measure of available ADNTs. Extractability of ADNTs from T. tubifex tissue containing metabolically-generated ADNTs was significantly less than extractability of ADNTs from T. tubifex tissue containing absorbed ADNTs: 50-60% and 81-90% of expected extractability based on fiber-water partition ratio. The lower SPME extractability of metabolically-generated ADNTs may stem from the unavailability of metabolically-generated ADNTs sequestered in tissue or bound to tissue macromolecules during metabolism of TNT to ADNT. Tissue extractions using SPMEs may be able to estimate such bound organic residues in tissue and serve as potential indicators of toxicological bioavailability and biomagnification potential of tissue-associated organic compounds.

The Life History and Contributions to the Ecology of Camelobaetidius variabilis Wiersema 1998 (Ephemeroptera: Baetidae) in Honey Creek, Oklahoma

A study of the life history and ecology of Camelobaetidius variabilis was conducted in Honey Creek, OK from February 2003-April 2004. Nymph development was assessed using changes in external morphology. Laboratory reared nymphs were used to calculate number of degree days to complete development (772 degree days at 20.8° C ±.38° C), which was used to determine voltinism. Field collected nymph microhabitat distribution was used in assessing microhabitat distribution. Nymphal thermoregulation was assessed during the winter and spring by comparing nymphal numbers present in shaded and un-shaded habitats. Camelobaetidius variabilis nymphs showed preference for algal microhabitats during the spring and leaf packs in the winter. Nymphs inhabited leaf packs to increase metabolic rate during the winter. Increased temperatures aid in development of nymphs. Camelobaetidius variabilis exhibited a multivoltine life cycle with six overlapping generations.

Rainfall-runoff changes due to urbanization: a comparison of different spatial resolutions for lumped surface water hydrology models using HEC-HMS.

Hydrologic models were used to examine the effects of land cover change on the flow regime of a watershed located in North-Central Texas. Additionally, the effect of spatial resolution was examined by conducting the simulations using sub-watersheds of different sizes to account for the watershed. Using the Army Corps of Engineers, Hydrologic Engineering Center Hydrologic Modeling System (HEC-HMS), two different modeling methods were evaluated at the different sub-watershed resolutions for four rainfall events. Calibration results indicate using the smaller spatial resolutions improves the model results. Different scenarios for land cover change were evaluated for all resolutions using both models. As land cover change increased, the amount of flow from the watershed increased.

Analysis of the One-Horned Rhinoceros (Rhinoceros Unicornis) Habitat in the Royal Chitwan National Park, Nepal.

This study analyzes the remaining suitable habitat of the one-horned rhinoceros, Rhinoceros unicornis, in Royal Chitwan National Park of Nepal. An April 2003 Landsat image was classified into eight land cover types: wetland, sand, water, mixed forest, sal forest, agriculture, settlement, and grassland. This image was converted into habitat suitability maps using cover, food, and water. The rhinoceros prefers grassland habitat with oxbow lakes and closed canopy during the monsoon season. Nominal values of five parameters were used to create a map of habitat suitability index. The map was categorized into four habitat classes: highly unsuitable, unsuitable, moderately suitable habitat, and suitable. Landscape metrics, patch metrics and class metrics associated with habitat were determined through the use of FRAGSTATS.

Determination of Dissociation Constants for GABAA Receptor Antagonists using Spontaneously Active Neuronal Networks in vitro

Changes in spontaneous spike activities recorded from murine frontal cortex networks grown on substrate-integrated microelectrodes were used to determine the dissociation constant (KB) of three GABAA antagonists. Neuronal networks were treated with fixed concentrations of GABAA antagonists and titrated with muscimol, a GABAA receptor agonist. Muscimol decreased spike activity in a concentration dependent manner with full efficacy (100% spike inhibition) and a 50% inhibitory concentration (IC50) of 0.14 ± 0.05 µM (mean ± SD, n=6). At 10, 20, 40 and 80 µM bicuculline, the muscimol IC50 values were shifted to 4.3 ± 1.8 µM (n=6), 6.8 ± 1.7 µM (n=6), 19.3 ± 3.54 µM (n=10) and 43.5 µM (n=2), respectively (mean ± SD). Muscimol titration in the presence of 10, 20, 40 µM of gabazine resulted in IC50s values of 20.1 (n=2), 37.17 (n=4), and 120.45 (n=2), respectively. In the presence of 20, 80, and 160 µM of TMPP (trimethylolpropane phosphate) the IC50s were 0.86 (n=2), 3.07 (n=3), 6.67 (n=2) µM, respectively. Increasing concentrations of GABAA antagonists shifted agonist log concentration-response curves to the right with identical efficacies, indicating direct competition for the GABAA receptor. FLP-mediated conditional loss of an essential gene to facilitate complementation assays

Commonly, when it is desirable to replace an essential gene with an allelic series of mutated genes, or genes with altered expression patterns, the complementing constructs are introduced into heterozygous plants, followed by the selection of homozygous null segregants. To overcome this laborious and time-consuming step, the newly developed two-component system utilizes a site-specific recombinase to excise a wild-type copy of the gene of interest from transformed tissues. In the first component (the first vector), a wild-type version of the gene is placed between target sequences recognized by FLP recombinase from the yeast 2 μm plasmid. This construct is transformed into a plant heterozygous for a null mutation at the endogenous locus, and progeny plants carrying the excisable complementing gene and segregating homozygous knockout at the endogenous locus are selected. The second component (the second vector) carries the experimental gene along with the FLP gene. When this construct is introduced, FLP recombinase excises the complementing gene, leaving the experimental gene as the only functional copy. The role of prostaglandins, nitric oxide and oxygen in the ductus arteriosi of the pre-term chicken embryo (Gallus domesticus).

The chicken ductus arteriosi (DA) are two embryonic blood vessels that shunt blood away from the non-ventilated lungs and towards the body and chorioallantoic membrane. I show that prostaglandins have a diminished role in maintaining chicken DA patency and nitric oxide inhibits oxygen induced contraction of the day 19 proximal DA in a time dependent manner. The pathways governing oxygen induced contraction in the chicken DA are similar to those found in mammals and include contributions from ROS, Kv channels, L-type Ca2+ channels, and the Rho kinase pathway. Longer exposure to high oxygen generates increased oxygen induced constriction of the day 19 DA that may be mediated through the Rho kinase pathway.

Home range analysis of rehabilitated and released great horned owls (Bubo virginianus) in Denton County, Texas, through radio telemetry.

Raptor rehabilitation has become commonplace globally, yet studies on the survival and adaptation of great horned owls (Bubo virginianus) after release has been neglected to an appreciable extent. The primary objective of this study is to provide quantitative data on the success of rehabilitated and released great horned owls in the North Texas region. Owls (N=12) were rehabilitated and released onto the Ray Roberts Greenbelt Corridor in Denton County, Texas, and monitored using radio telemetry to evaluate home range (November 2002 - February 2005). With approximately 75% of the birds released for this study surviving until transmitter battery failure, it is believed that the rehabilitation process was successful for these birds.

The Medicago truncatula NIP gene is located on M. truncatula Linkage Group 1. Informative recombinants showed crossovers that localize the NIP gene between markers 146O17 and 23C16D. Marker 164N9 co-segregates with the NIP gene, and the location of marker 164N9 is between markers 146O17 and 23C16D. Based upon data from the Medicago genome sequencing project, a subset of the model legume Medicago truncatula bacterial artificial chromosomes (BACs) were used to create a physical map on the DNA in this genetic internal. BACs near the potential NIP gene location near marker 164N9 were identified, and used in experiments to predict the physical map by a BAC-by-BAC strategy. Using marker 164N9 as a center point, and chromosome walking outward, the physical map toward markers 146O17 and 23C16D was built. The chromosome walk consisted of a virtual walk, made with existing sequence of BACs from the Medicago genome project, hybridizations to filters containing BAC DNA, and PCR reactions to confirm that predicted overlapping BACs contained DNA that yielded similar PCR products.
