Synthesis and Screening of a Combinatorial Peptide Library for Ligands to Target Transferrin: Miniaturizing the Library
Combinatorial libraries are used in the search for ligands that bind to target proteins. Fmoc solid-phase peptide synthesis is routinely used to generate such libraries. Microwave-assisted peptide synthesis was employed here to decrease reaction times by 80-90%. Two One-Bead-One-Compound combinatorial libraries were synthesized on 130μm beads (one containing 750 members and the other 16, 807). The use of smaller solid supports would have many important practical advantages including; increased library diversity per unit mass, smaller quantities of library needed to generate hits, and screening could be conducted by using a standard flow cytometer. To this end, a miniaturized peptide library was synthesized on 20 μm beads to demonstrate proof of principle. A small sample from the 16,807-member library was screened against transferrin-AlexaFluro 647, a protein responsible for iron transport in vivo. A number of hits were identified and sequenced using techniques coupling nanomanipulation with nanoelectrospray mass spectrometry.

An Exploration of the Relationship Between Principal Leadership Efficacy, Principal Computer Self-Efficacy, and Student Achievement
The purpose of this study was to examine whether or not relationships exist between principals' technology proficiency and student achievement as indicated by 2008 Texas Assessment of Knowledge and Skills (TAKS) ninth grade reading scaled scores. Secondly, the study examined whether or not relationships exist between principals' leadership self efficacy and student achievement as indicated in the 2008 Texas Assessment of Knowledge and Skills (TAKS) ninth grade reading scaled scores. Lastly, the select principal's personal and school demographic variables (principal gender, total years of experience as a professional, total years as principal at current school, total years of principal experience, highest degree earned, school economic status, school size) were considered within the study. The survey instruments used in this study were the Technology Proficiency Self Assessment Scale (TPSA) developed by Ropp in 2000 and the Principal's Self-Efficacy Scale (PSES) developed by Tschannen-Moran and Garies in 2004. A total of 129 Texas principal's participated in the study. Multiple regressions were utilized and effect size was considered to determine the strength of the relationship between variables. Easter Seals North Texas Autism Treatment Program Evaluation: Child Progress
This study reports and evaluates child outcome measures at a non-profit autism treatment program providing applied behavior analysis (ABA) based services to children age 3 to 8. To accomplish this, a review was conducted of available outcome data for a 1 year period. Several categories of outcome measures have been reported in the autism treatment literature (post-intervention educational placement, cognitive status, developmental and achievement status and/or progress, autism symptom reduction, and diagnostic reclassification). This study found that the program relied on 2 sources of data to evaluate child outcome: Hawaii Early Learning Profile® and program goal mastery. Children are making progress as indicated by these measures. The findings are discussed in relation to broader outcome recommendations.

A Performance Guide for Pearls I and Pearls II by Roland Szentpali
This dissertation is a performance guide for the euphonium solos Pearls I and Pearls II, written by Roland Szentpali. This performance guide allows performers to better understand the jazz styles within each movement and provides them with a resource for performing these particular pieces as well as other jazz influenced pieces. This performance guide is specific to euphonium repertoire and written for euphonium performers and educators. This is also a resource for a solo work in the repertoire that is performed regularly as well as a new work that will soon be published. A brief history of the development of euphonium repertoire and the influence of jazz is provided. The performance guide analyzes each movement and provides insight to extended techniques, common performance problems, errata, and jazz styles that each movement is based on. The guide also provides several suggestions for interpretation and for performance preparation. Illustrations from the scores have been provided for each example.

Rediscovering Fernande Decruck's Sonate en ut# pour saxophone alto (ou alto) et orchestre: A Performance Analysis
French composer Fernande Decruck (1896-1954) composed over forty works for the saxophone, but her music fell into obscurity soon after her death. In recent years, the Sonate en ut# pour saxophone alto (ou alto) et orchestre (1943) has been rediscovered, performed, and recorded by prominent concert saxophonists. This document takes a historical approach by examining Decruck's biography, as well as a theoretical approach to provide a deeper understanding and appreciation of her work through analysis. The first four chapters of this document provide biographical background on Decruck, her career, professional associations, and her husband, Maurice Decruck, saxophonist and music publisher. Additionally, an examination of her saxophone output includes a brief discussion of her compositional development. Fernande Decruck dedicated her sonata to French saxophone virtuoso Marcel Mule, but a version for solo viola also exists. From the discrepancies between the versions, one might infer that portions of the work were composed originally for the viola. There are also two versions of the accompaniment: one for full orchestra and the other for piano. Analysis comprises the bulk of this study. Metabolic Engineering of Raffinose-Family Oligosaccharides in the Phloem Reveals Alterations in Patterns of Carbon Partitioning and Enhances Resistance to Green Peach Aphid
Phloem transport is along hydrostatic pressure gradients generated by differences in solute concentration between source and sink tissues. Numerous species accumulate raffinose-family oligosaccharides (RFOs) in the phloem of mature leaves to accentuate the pressure gradient between source and sinks. In this study, metabolic engineering was used to generate RFOs at the inception of the translocation stream of Arabidopsis thaliana, which transports predominantly sucrose. To do this, three genes, GALACTINOL SYNTHASE, RAFFINOSE SYNTHASE and STACHYOSE SYNTHASE, were expressed from promoters specific to the companion cells of minor veins. Two transgenic lines homozygous for all three genes (GRS63 and GRS47) were selected for further analysis. Sugars were extracted and quantified by high performance anion exchange chromatography with pulsed amperometric detection (HPAEC-PAD), and 21-day old plants of both lines had levels of galactinol, raffinose, and stachyose approaching 50% of total soluble sugar. All three exotic sugars were also identified in phloem exudates from excised leaves of transgenic plants whereas levels were negligible in exudates from wild type leaves. Differences in starch accumulation or degradation between wild type and GRS63 and GRS47 lines were not observed. Understanding Quality of Life in Older Adults
I analyzed the 2004 and 2006 Health and Retirement Study data to test structural equation models of the quality of life (QOL) construct. The participants (N = 1352) were non-institutionalized individuals aged 42 and older (M = 65.70, SD = 10.88), with an average education of 12.73 years (SD = 2.96) and of varied ethnicities. The results indicated that physical functioning, affective experience, life satisfaction and social support could serve as indicators for a second order QOL factor. Furthermore, the developed QOL model explained 96% of the variance of the CASP-19 (Control, Autonomy, Self-realization and Pleasure), a QOL measure that reflects fulfillment of psychological needs. The results also indicated that Depression and Life Satisfaction are related through reciprocal causation and that Physical Functioning is more likely to cause a change in Depression than the reverse. The results suggest that QOL is a complex, multidimensional concept that should be studied at different levels of analysis.

My Texas History Notebook: Interactive Lesson Plans for 4th and 7th Graders Grant Materials
These grant materials were prepared for a project funded by Humanities Texas to develop and present online lesson plans that incorporate the interactive notebook style of learning. This method of teaching combines three pivotal concepts in educational research: spiral curriculum theory, multiple intelligences, and cooperative interaction. This problem-based learning system incorporates preview assignments, multiple intelligences teaching strategies, graphically organized notes, and processing assignments that require students to demonstrate their knowledge. The documents explore the research and methodology behind this style of pedagogy, and the elements of an interactive notebook. A work plan details how the project would work with master teachers to develop this curriculum. The project was funded for $5,000.

Child-Parent Relationship Therapy (CPRT) with Adoptive Families: Effects on Child Behavior, Parent-Child Relationship Stress, and Parental Empathy
This randomized controlled study is a preliminary investigation on the effects of Child-Parent Relationship Therapy (CPRT) with 61 adoptive parents. The participants in this study identified themselves as the following: 54 European American, 3 Black American, 3 Hispanic/Latino, and 1 individual who chose not to indicate ethnicity. The study included 23 couples and 15 individual mothers. The CPRT is a structured, time limited approach that trains caregivers to be an active participant as a therapeutic change agent in their child's life. Results from a two (group) by two (measures) split plot ANOVA indicated that adoptive parents who participated in 10 weeks of CPRT reported statistically significant decreases in child behavior problems and parent child-relationship stress. Statistically significant increases in parent empathy were also reported by raters blinded to the study. CPRT demonstrated a medium to large treatment effect on reducing children's behavior problems and parent-child relationship stress. In addition, CPRT demonstrated a large treatment effect on increasing parental empathy. The results of the study provide preliminary support for CPRT as a responsive intervention for adoptive parents and their children.

Using Web-Based Instruction to Teach Music Theory in the Piano Studio: Defining, Designing, and Implementing an Integrative Approach
This dissertation rationalizes the best use of Web-based instruction (WBI) for teaching music theory to private piano students in the later primary grades. It uses an integrative research methodology for defining, designing, and implementing a curriculum that includes WBI. Research from the fields of music education, educational technology, educational psychology, and interaction design and children receive primary consideration. A synthesis of these sources outlines several research-based principles that instructional designers can use to design a complete blended learning environment for use within the piano studio. In addition to the research-based principles, the precise methods of determining instructional tasks and implementing the program online are described in detail. A full implementation is then deployed, and piano teachers evaluate the extent to which the online program fulfills the research-based principles. This dissertation does not argue for the complete migration of theory instruction from traditional workbook approaches to an entirely Web-based medium but rather outlines the best use of face-to-face instruction, collaboration amongst students, teachers, and parents, and interaction with a Web-based program. This formative research provides a complete model of integrating WBI within the piano studio that can guide instructional designers and music educators.

The Effect of High-Probability Request Sequences on Latency to Comply with Instructions to Transition in a Child With Severe Mental Retardation
This study investigated the effect of implementing high-probability request sequences prior to the delivery of instructions to transition in a child with severe mental retardation. Data were collected on latency to comply with a low-probability request to transition and a modified version of the low-probability request. Implementation of high-probability request sequences resulted in shortened latencies to comply with the modified low-probability request instructing the child to engage in a preferred activity located at the endpoint of the transition.

Using Financial Rankings to Identify Characteristics of Libraries Serving Highly Profitable Private Law Firms
This purpose of this study was to develop evidence of a relationship between law libraries and private law firm profitability for law library administrators to use when making strategic decisions that influence the value of their libraries. The highest ranked administrator at each private law firm listed on the 2008 Am Law 200 was invited to complete an online benchmarking survey. The adjusted sample population totaled 179 firms. Fifty-one valid surveys were completed for a 28.5% response rate. Descriptive and statistical analyses were conducted using 26 independent variables (law library characteristics) and a single dependent variable, Revenue per Equity Partner, developed from data published for the Am Law 200. The most significant contributions of this study are: development of important law library financial and return on investment benchmarks; a listing of characteristics that have been empirically shown to impact law firm productivity; identification of optimum reporting structure for the law library administrator. Six characteristics positively impact Revenue per Equity Partner: to whom the library Administrator reports, number of library staff per library, number of Library staff per library, range in hourly bill rate for library staff time, practice areas most often supported. Culture and Self-Representation in the Este Court: Ercole Strozzi's Funeral Elegy of Eleonora of Aragon, a Text, Translation, and Commentary.
This dissertation presents a previously unedited text by one of the most distinguished- yet neglected-Latin writers of the Italian Renaissance, Ercole Strozzi (1471-1508), a poet and administrator in the court of Ferrara. Under the Este Dukes, Ferrara became a major center of literary and artistic patronage. The Latin literary output of the court, however, has received insufficient scholarly scrutiny. The text is a verse funeral elegy of Eleonora of Aragon (1450-1493), the first Duchess of Ferrara. Eleonora was a remarkable woman whose talents and indefatigable efforts on behalf of her husband, her children, and her state, won her accolades both at home and abroad. She also served as a prototype for the remarkable careers of her two daughters, Isabella d'Este, and Beatrice d'Este, who are celebrated for their erudition and patronage of arts and letters. The text is a mirror of the Estense court and reveals to us how its members no doubt saw themselves, at the very peak of its temporal power and the height of its prestige as a center of cultural creativity. A Geoarchaeological Investigation of Site Formation in the Animas River Valley at Aztec Ruins National Monument, NM
This paper presents an investigation of sedimentary deposition, soil formation, and pedoturbation in the Animas River Valley to determine the provenience of archaeological deposits in an open field at Aztec Ruins National Monument, NM outside of the Greathouse complex. Four stratigraphic pedounits correlated with active fan deposition have been proposed for the lower terrace in the project area with only one of these units retaining strong potential for buried archaeological deposits from the Anasazi late Pueblo II/Pueblo III period. The distal fan on the lower terrace and the Animas River floodplain appear to show poor potential for archaeological deposits either due to shallow sediment overburden with historic disturbance or alluvial activity during or after occupation. Based on these findings, four other zones of similar fan development have been identified throughout the Animas Valley and are recommended for subsurface testing during future cultural resource investigations.

Incorporating Flow for a Comic [Book] Corrective of Rhetcon
In this essay, I examined the significance of graphic novels as polyvalent texts that hold the potential for creating an aesthetic sense of flow for readers and consumers. In building a justification for the rhetorical examination of comic book culture, I looked at Kenneth Burke's critique of art under capitalism in order to explore the dimensions between comic book creation, distribution, consumption, and reaction from fandom. I also examined Victor Turner's theoretical scope of flow, as an aesthetic related to ritual, communitas, and the liminoid. I analyzed the graphic novels Green Lantern: Rebirth and Y: The Last Man as case studies toward the rhetorical significance of retroactive continuity and the somatic potential of comic books to serve as equipment for living. These conclusions lay groundwork for multiple directions of future research.

Impacts of Comprehensive Climate and Energy Policy Options on the U.S. Economy
This study compiles and updates the findings of 16 comprehensive state climate action plans and extrapolates the results to the nation. The study then takes those results and using a widely accepted econometric model projects the national impact of these policies on employment, incomes, gross domestic product (GDP) and consumer energy prices. Finally, using the bottom-up data developed by the states and aggregated here, the study models the national impact of major features of the Kerry-Lieberman climate bill under consideration in Congress in 2010. These state action plans and supporting assessments were proposed by over 1,500 stakeholders and technical work group experts appointed by 16 governors and state legislatures to address climate, energy and economic needs through comprehensive, fact-based, consensus-driven, climate action planning processes conducted over the past five years with facilitative and technical assistance by the Center for Climate Strategies. Findings show potential national improvements from implementation of a top set of 23 major sector-based policies and measures drawn from state plans.
