Activity of the kinesin spindle protein inhibitor ispinesib (SB-715992) in models of breast cancer

Ispinesib (SB-715992) is a potent inhibitor of kinesin spindle protein (KSP), a kinesin motor protein essential for the formation of a bipolar mitotic spindle and cell cycle progression through mitosis. Clinical studies of ispinesib have demonstrated a 9% response rate in patients with locally advanced or metastatic breast cancer, and a favorable safety profile without significant neurotoxicities, gastrointestinal toxicities or hair loss. To better understand the potential of ispinesib in the treatment of breast cancer we explored the activity of ispinesib alone and in combination several therapies approved for the treatment of breast cancer. We measured the ispinesib sensitivity and pharmacodynamic response of breast cancer cell lines representative of various subtypes in vitro and as xenografts in vivo, and tested the ability of ispinesib to enhance the anti-tumor activity of approved therapies. In vitro, ispinesib displayed broad anti-proliferative activity against a panel of 53 breast cell-lines. In vivo, ispinesib produced regressions in each of five breast cancer models, and tumor free survivors in three of these models. The effects of ispinesib treatment on pharmacodynamic markers of mitosis and apoptosis were examined in vitro and in vivo, revealing a greater increase in both mitotic and apoptotic markers in the MDA-MB-468 model …

Analysis of major trends in U.S. commercial trucking, 1977-2002.

This report focuses on various major long-range (1977-2002) and intermediate-range (1982-2002) U.S. commercial trucking trends. The primary sources of data for this period were the U.S. Bureau of the Census Vehicle Inventory and Use Survey and Truck Inventory and Use Survey. In addition, selected 1977-2002 data from the U.S. Department of Energy/Energy Information Administration and from the U.S. Department of Transportation/Federal Highway Administration's Highway Statistics were used. The report analyzes (1) overall gasoline and diesel fuel consumption patterns by passenger vehicles and trucks and (2) the population changes and fuels used by all commercial truck classes by selected truck type (single unit or combination), during specified time periods, with cargo-hauling commercial trucks given special emphasis. It also assesses trends in selected vehicle miles traveled, gallons per vehicle miles traveled, and gallons per cargo ton-mile traveled, as well as the effect of cargo tons per truck on fuel consumption. In addition, the report examines long-range trends for related factors (e.g., long-haul mileages driven by heavy trucks) and their impacts on reducing fuel consumption per cargo-ton-mile and the relative shares of total commercial fuel use among truck classes. ANOMALY STRUCTURE OF SUPERGRAVITY AND ANOMALY CANCELLATION

We display the full anomaly structure of supergravity, including new D-term contributions to the conformal anomaly. This expression has the super-Weyl and chiral U(1){sub K} transformation properties that are required for implementation of the Green-Schwarz mechanism for anomaly cancellation. We outline the procedure for full anomaly cancellation. Our results have implications for effective supergravity theories from the weakly coupled heterotic string theory.

Army Working Capital Fund: Actions Needed to Improve Budgeting for Carryover at Army Ordnance

A letter report issued by the Government Accountability Office with an abstract that begins "From fiscal years 2004 through 2008, new orders received by the eight Army ordnance activities increased from $788 million to $1.5 billion. To the extent that the ordnance activities do not complete work at year end, the ordered and funded work is carried over into the next fiscal year. While past congressional defense committees recognized the need for carryover, the committees have on occasion raised concerns that carryover may be more than needed. GAO was asked to determine (1) whether the reported actual total carryover increased or decreased from fiscal years 2004 through 2008 and, if the carryover increased, the actions the Army is taking to reduce it; (2) the primary reasons for carryover at the eight ordnance activities; and (3) whether carryover amounts exceeded ceilings for fiscal years 2006, 2007, and 2008, and whether the methodology used to calculate the ceiling for the ordnance activities was reasonable. GAO analyzed reported carryover and related data at the eight activities."

Complexation of Plutonium (IV) with Fluoride at Variable Tempeartures

Complexation of Pu(IV) with fluoride was studied by solvent extraction at 25, 40 and 55 C in 2.2 mol {center_dot} kg{sup -1} HClO{sub 4}. The distribution ratio of Pu(IV) between the organic and aqueous phases decreased as the concentration of fluoride was increased due to the formation of Pu(IV)-F complexes in the aqueous phase. Two complexes, PuF{sup 3+} and PuF{sub 2}{sup 2+}, were identified under the conditions in this work and their stability constants at 25, 40 and 55 C and I = 2.2 mol {center_dot} kg{sup -1} HClO{sub 4} were determined from the distribution data. The Specific Ion Interaction approach (SIT) was used to extrapolate the constants to the state of infinite dilution. Data from this work indicate that the complexation of Pu(IV) with fluoride is endothermic and entropy-driven. The complexation becomes stronger at higher temperatures.

Device fabrication and transport measurements of FinFETs built with 28Si SOI wafers towards donor qubits in silicon

We report fabrication of transistors in a FinFET geometry using isotopically purified silicon-28 -on-insulator (28-SOI) substrates. Donor electron spin coherence in natural silicon is limited by spectral diffusion due to the residual 29Si nuclear spin bath, making isotopically enriched nuclear spin-free 28Si substrates a promising candidate for forming spin quantum bit devices. The FinFET architecture is fully compatible with single-ion implant detection for donor-based qubits, and the donor spin-state readout through electrical detection of spin resonance. We describe device processing steps and discuss results on electrical transport measurements at 0.3 K.

DOES CRITICAL MASS DECREASE AS TEMPERATURE INCREASES: A REVIEW OF FIVE BENCHMARK EXPERIMENTS THAT SPAN A RANGE OF ELEVATED TEMPERATURES AND CRITICAL CONFIGURATIONS

Five sets of benchmark experiments are reviewed herein that cover a diverse set of fissile system configurations. The review specifically focused on the change in critical mass of these systems at elevated temperatures and the temperature reactivity coefficient ({alpha}{sub T}) on the system. Because plutonium-based critical benchmark experiments at varying temperatures were not found at the time this review was prepared, only uranium-based systems are included, as follows: (1) HEU-SOL-THERM-010 - UO{sub 2}F{sub 2} solutions with high U{sup 235} enrichment; (2) HEU-COMP-THERM-016 - uranium-graphite blocks with low U concentration; (3) LEU-COMP-THERM-032 - water moderated lattices of UO{sub 2} with stainless steel cladding, and intermediate U{sup 235} enrichment; (4) IEU-COMP-THERM-002 - water moderated lattices of annular UO{sub 2} with/without absorbers, and intermediate U{sup 235} enrichment; and (5) LEU-COMP-THERM-026 - water moderated lattices of UO{sub 2} at different pitches, and low U{sup 235} enrichment. In three of the five benchmarks (1, 3 and 5), modeling of the critical system at room temperature is conservative compared to modeling the system at elevated temperatures, i.e., a greater fissile mass is required at elevated temperature. Environmental Contamination: Lessons Learned from the Cleanup of Formerly Used Defense and Military Munitions Sites

Testimony issued by the Government Accountability Office with an abstract that begins "Under the Defense Environmental Restoration Program (DERP), the Department of Defense (DOD) has charged the Army Corps of Engineers (the Corps) with cleaning up 4,700 formerly used defense sites (FUDS) and active sites that were under its jurisdiction when they were initially contaminated. The 661-acre Spring Valley site in Washington, D.C is one such site. Like many other FUDS, the U.S. Army used the Spring Valley site during World War I for research and testing of chemical agents, equipment, and munitions. Most of the site is now privately owned and includes private residences, a hospital, and several commercial properties. The primary threats at the site are buried munitions, elevated arsenic in site soils, and laboratory waste; perchlorate was also found onsite. This testimony discusses GAO's past work relating to remediation efforts at FUDS and military munitions sites to provide context for issues at Spring Valley. Specifically, it addresses: (1) the impact that shortcomings in information and guidance can have on decision-making; (2) the impact that incomplete data can have on cost estimates and schedules; (3) how funding for a particular site may be influenced by overall program goals; …
