Accelerated Evolution of Conserved Noncoding Sequences in theHuman Genome

Genomic comparisons between human and distant, non-primatemammals are commonly used to identify cis-regulatory elements based onconstrained sequence evolution. However, these methods fail to detect"cryptic" functional elements, which are too weakly conserved amongmammals to distinguish from nonfunctional DNA. To address this problem,we explored the potential of deep intra-primate sequence comparisons. Wesequenced the orthologs of 558 kb of human genomic sequence, coveringmultiple loci involved in cholesterol homeostasis, in 6 nonhumanprimates. Our analysis identified 6 noncoding DNA elements displayingsignificant conservation among primates, but undetectable in more distantcomparisons. In vitro and in vivo tests revealed that at least three ofthese 6 elements have regulatory function. Notably, the mouse orthologsof these three functional human sequences had regulatory activity despitetheir lack of significant sequence conservation, indicating that they arecryptic ancestral cis-regulatory elements. These regulatory elementscould still be detected in a smaller set of three primate speciesincluding human, rhesus and marmoset. Since the human and rhesus genomesequences are already available, and the marmoset genome is activelybeing sequenced, the primate-specific conservation analysis describedhere can be applied in the near future on a whole-genome scale, tocomplement the annotation provided by more distant speciescomparisons.

Child Support Enforcement: More Focus on Labor Costs and Administrative Cost Audits Could Help Reduce Federal Expenditures

A letter report issued by the Government Accountability Office with an abstract that begins "Congress established federal standards for the child support enforcement program (CSE) in 1975. State agencies administer the program and the Office of Child Support Enforcement (OCSE) in the Department of Health and Human Services (HHS) oversees it. The CSE program provides several services, including collecting child support payments from noncustodial parents--those who are not the primary caregivers--and distributing these payments to families. Generally, the federal government reimburses state agencies 66 percent of their costs for administering the CSE program. GAO determined (1) how total net federal expenditures for administrative costs changed from fiscal year 2000 to fiscal year 2004; (2) the categories of costs that contributed most to administrative costs in recent years; and (3) steps state agencies have taken to manage costs, and steps OCSE has taken to help state agencies and ensure federal funds have been used appropriately. GAO analyzed program data, surveyed all 54 state agencies and visited 6, interviewed program officials, and reviewed laws, policies, and reports."

On June 28, 2006, the Bush Administration announced its proposal to sell 36 F-16 C/D Block 50/52 Falcon combat aircraft to Pakistan at an estimated case value of $3 billion. Some believe that these sales are partly an effort to reward the Pakistani Government for the role it has played in support of U.S.-led anti-terrorism efforts, and this consideration is noted in the text of the formal notification of the F-16 sales. Combat aircraft are considered "essential for conducting surprise attacks or initiating large-scale offensive operations." Therefore, the transfer of combat aircraft can be a significant policy decision, especially to a region with known tensions and territorial disputes. It is currently unclear what long-term effects a potential sale of combat aircraft to South Asia might have on U.S. political relations with Pakistan and India, or the political relationship between them.

A letter report issued by the Government Accountability Office with an abstract that begins "The Cheyenne Mountain Operations Center houses numerous complex computer systems for tracking air, missile, and space events that could threaten homeland security or undermine military operations in theater. To ensure this mission can be met, the systems require ongoing upgrades. The most recent upgrade program--the Combatant Commanders' Integrated Command and Control System (CCIC2S)--was initiated in 2000. Given the critical missions supported by Cheyenne Mountain systems, GAO initiated a review to (1) determine the status of the CCIC2S program in terms of meeting its cost, schedule, and performance goals; (2) gauge the extent to which DOD has followed best practices in managing program requirements; and (3) assess DOD's control and oversight mechanisms for CCIC2S."

Gamma-Ray Production Cross Sections in Multiple Channels for Neutron Induced Reaction on 48ti for en=1 to 200 Mev

Prompt {gamma}-ray production cross sections were measured on a {sup 48}Ti sample for incident neutron energies from 1 MeV to 200 MeV. Partial {gamma}-ray cross sections for transitions in {sup 45-48}Ti, {sup 45-48}Sc, and {sup 43-45}Ca were determined. The observation of about 130 transitions from 11 different isotopes in the present work provides a demanding test of reaction model calculations, and is the first study in this mass region to extract partial {gamma}-ray cross sections for many different reaction channels over a wide range of incident neutron energies. The neutrons were produced by the Los Alamos National Laboratory spallation neutron source located at the LANSCE/WNR facility. The prompt-reaction {gamma} rays were detected with the large-scale Compton-suppressed GErmanium Array for Neutron Induced Excitations (GEANIE). Event neutron energies were determined by the time-of-flight technique. The {gamma}-ray excitation functions were converted to partial {gamma}-ray cross sections and then compared with model calculations using the enhanced GNASH reaction code. Compound nuclear, pre-equilibrium emission and direct reaction mechanisms are included. Overall the model calculations of the partial {gamma}-ray cross sections are in good agreement with measured values.
