Deletion of ultraconserved elements yields viable mice

Ultraconserved elements have been suggested to retainextended perfect sequence identity between the human, mouse, and ratgenomes due to essential functional properties. To investigate thenecessities of these elements in vivo, we removed four non-codingultraconserved elements (ranging in length from 222 to 731 base pairs)from the mouse genome. To maximize the likelihood of observing aphenotype, we chose to delete elements that function as enhancers in amouse transgenic assay and that are near genes that exhibit markedphenotypes both when completely inactivated in the mouse as well as whentheir expression is altered due to other genomic modifications.Remarkably, all four resulting lines of mice lacking these ultraconservedelements were viable and fertile, and failed to reveal any criticalabnormalities when assayed for a variety of phenotypes including growth,longevity, pathology and metabolism. In addition more targeted screens,informed by the abnormalities observed in mice where genes in proximityto the investigated elements had been altered, also failed to revealnotable abnormalities. These results, while not inclusive of all thepossible phenotypic impact of the deleted sequences, indicate thatextreme sequence constraint does not necessarily reflect crucialfunctions required for viability.

Design and Evaluation of Ionic Liquids as Novel CO2 Absorbents

This is the final report for project DE-FG26-04NT42122 'Design and Evaluation of Ionic Liquids as Novel CO{sub 2} Absorbents'. The objective of this 'breakthrough concepts' project was to investigate the feasibility of using ionic liquids for post-combustion CO{sub 2} capture and obtain a fundamental understanding of the solubility of CO{sub 2} and other components present in flue gas in ionic liquids. Our plan was to obtain information on how composition and structure of ionic liquid molecules affected solubility and other important physical properties via two major efforts: synthesis and experimental measurements and molecular simulation. We also planned to perform preliminary systems modeling study to assess the economic viability of a process based on ionic liquids. We accomplished all the milestones and tasks specified in the original proposal. Specifically, we carried out extensive quantum and classical atomistic-level simulations of a range of ionic liquids. These calculations provided detailed information on how the chemical composition of ionic liquids affects physical properties. We also learned important factors that govern CO{sub 2} solubility. San Luis National Wildlife Refuge Complex to meetReclamation s obligations for Level 4 water supply under the CentralValley Project Improvement Act. Hydrogeological assessment of the EastBear Creek Unit of the San Luis National Wildlife Refuge was conductedusing a combination of field investigations and a survey of availableliterature from past US Geological Survey Reports and reports by localgeological consultants. Conservative safe yield estimates made using theavailable data show that the East Bear Creek Unit may have sufficientgroundwater resources in the shallow groundwater aquifer to meet aboutbetween 25 percent and 52 percent of its current Level II and between 17percent and 35 percent of its level IV water supply needs. The rate ofsurface and lateral recharge to the Unit and the design of the well fieldand the layout and capacity of pumped wells will decide both thepercentage of annual needs that the shallow aquifer can supply andwhether this yield is sustainable without affecting long-term aquiferquality. In order to further investigate the merits of pumping the nearsurface aquifer, which appears to have reasonable water quality for usewithin the East Bear Creek Unit -- monitoring of the potential sources ofaquifer recharge and the installation of a pilot shallow well would bewarranted. Iraq: U.S. Military Operations

Iraq’s chemical, biological, and nuclear weapons programs, together with Iraqi long-range missile development and support for al-Queda terrorism, were the primary justifications put forward for military action. On March 17, 2003, President Bush issued an ultimatum demanding that Saddam Hussein and his sons depart from Iraq within 48 hours. On March 19, offensive operations began with air strikes against Iraqi leadership positions. By April 15, after 27 days of operations, coalition forces were in relative control of all major Iraqi cities and Iraqi political and military leadership had disintegrated. On May 1, 2003, President Bush declared an end to major combat operations. There was no use of chemical or biological (CB)weapons, and no CB or nuclear weapons stockpiles or production facilities have been found.

Radiogenic and Stable Isotope and Hydrogeochemical Investigation of Groundwater, Pajarito Plateau and Surrounding Areas, New Mexico

From October 2004 through February 2006, Los Alamos National Laboratory, the New Mexico Environment Department-Department of Energy Oversight Bureau, and the United States Geological Survey conducted a hydrochemical investigation. The purpose of the investigation was to evaluate groundwater flow paths and determine groundwater ages using tritium/helium-3 and carbon-14 along with aqueous inorganic chemistry. Knowledge of groundwater age and flow paths provides a technical basis for selecting wells and springs for monitoring. Groundwater dating is also relevant to groundwater resource management, including aquifer sustainability, especially during periods of long-term drought. At Los Alamos, New Mexico, groundwater is either modern (post-1943), submodern (pre-1943), or mixed (containing both pre- and post-1943 components). The regional aquifer primarily consists of submodern groundwater. Mixed-age groundwater results from initial infiltration of surface water, followed by mixing with perched alluvial and intermediate-depth groundwater and the regional aquifer. No groundwater investigation is complete without using tritium/helium-3 and carbon-14 dating methods to quantify amounts of modern, mixed, and/or submodern components present in samples. Computer models of groundwater flow and transport at Los Alamos should be calibrated to groundwater ages for perched intermediate zones and the regional aquifer determined from this investigation. Sampling and Analysis Plan Waste Treatment Plant Seismic Boreholes Project.

This sampling and analysis plan (SAP) describes planned data collection activities for four entry boreholes through the sediment overlying the Saddle Mountains Basalt, up to three new deep rotary boreholes through the Saddle Mountains Basalt and sedimentary interbeds, and one corehole through the Saddle Mountains Basalt and sedimentary interbeds at the Waste Treatment Plant (WTP) site. The SAP will be used in concert with the quality assurance plan for the project to guide the procedure development and data collection activities needed to support borehole drilling, geophysical measurements, and sampling. This SAP identifies the American Society of Testing Materials standards, Hanford Site procedures, and other guidance to be followed for data collection activities. Revision 3 incorporates all interim change notices (ICN) that were issued to Revision 2 prior to completion of sampling and analysis activities for the WTP Seismic Boreholes Project. This revision also incorporates changes to the exact number of samples submitted for dynamic testing as directed by the U.S. Army Corps of Engineers. Revision 3 represents the final version of the SAP.
