Progress Report: DE-FG03-97ER20274, ''Microbial Production of Isoprene''

We have discovered that microorganisms produce and emit the hydrocarbon isoprene (2-methyl-1,3-butadiene), and have suggested that if isoprene-producing enzymes and their genes can be harnessed, useful hydrocarbon-producing systems might be constructed. The main goal of the proposed work is to establish the biochemical mechanism and regulation of isoprene formation in the bacterial system, Bacillus subtilis. Specific objectives of the proposed work are the following: (A) to characterize the physiological regulation of isoprene formation in B. subtilis; (B) to characterize mutations in B. subtilis 168 that suppress isoprene formation, clone these genes, and determine how isoprene and isoprenoid carbon flow are regulated; and (C) to test ''overflow'' and ''signaling'' models for Bacillus isoprene formation. We are also pursuing the isolation and cloning of B. subtilis isoprene synthase, which we believe may be a regulatory enzyme.

The 107th Congress is considering trade issues with implications for the U.S. agricultural sector. Trade in agricultural commodities and food products affects farm income and rural employment, and it also generates economic activity beyond the farm gate. With agricultural export sales the equivalent of one-quarter of farm income, some policymakers view U.S. efforts to develop market opportunities overseas as vital to the sector’s financial health. Decisions taken by the Bush Administration, and actions taken by Congress, thus will affect the outlook for agricultural trade.

Determination of Constraint-Modified J-R Curves for Carbon Steel Storage Tanks

The fracture toughness testing of A285 carbon steel storage tank material was carried out. A series of single edge-notched bend (SENB) specimens with various constraint levels were used. The variation of crack tip constraint was achieved by changing the ratio of the initial notch length to the specimen depth. The test data show that the J-R curves are specimen-dependent, which is known as the constraint effect. A two-parameter fracture methodology is adopted to construct a constraint-modified J-R curve, which is a function of the constraint parameter, A2, while J remains the loading parameter. This additional fracture parameter is derived from a closed form solution and can be extracted from the finite element analysis for a specific crack configuration. Using this set of SENB test data, a mathematical expression for the J-R curve of the A285 carbon steel can be developed. It is shown that the predicted J-R curves match well with the SENB data over an extensive amount of crack growth. In addition, this expression of the constraint modified J-R curve is used to predict the test data of a compact tension specimen (CT), and good agreement can be achieved. Characterization of Phase and Emulsion Behavior, Surfactant Retention, and Oil Recovery for Novel Alcohol Ethoxycarboxylate Surfactant

Electrical conductivity measurements for middle, bottom, and top phases, as well as bottom/middle, and middle/bottom conjugate pair phases of the NEODOX 23-4/dodecane/10mM water system were continued from the previous reporting period. Electrical conductivity of the mixture decreased as the fraction of volume of the middle phase was increased and vice versa. Also inversion phenomena was observed. Following this, more emulsion studies at various temperatures were progresses. A theoretical model to predict the conductivity measurements using Maxwell equations was developed and sensitivity analyses to test the performance of the model was completed. Surtek, Golden, CO, our industrial partner in this project, investigated the suitability of the surfactant for enhanced oil recovery employing coreflooding techniques and observed lower surfactant and hydrocarbon recovery for NEODOX 23-4.

Study of metal dusting phenomenon and development of materials resistant to metal dusting.

The deposition of carbon from carbonaceous gaseous environments is prevalent in many chemical and petrochemical processes such as reforming systems, syngas production systems, iron reduction plants, and others. One of the major consequences of carbon deposition is the degradation of structural materials by a phenomenon known as metal dusting. There are two major issues of importance in metal dusting. First is formation of carbon and subsequent deposition of carbon on metallic materials. Second is the initiation of metal dusting degradation of the alloy. Details are presented on a research program that is underway at Argonne National Laboratory to study the metal dusting phenomenon from a fundamental scientific base involving laboratory research in simulated process conditions and field testing of materials in actual process environments. The project has participation from the US chemical industry, alloy manufacturers, and the Materials Technology Institute, which serves the chemical process industry.

Migrating birds : assessment of impact on 915-MHz radar wind profiler performance at the Atmospheric Radiation Measurement Program's southern great plains.

The U. S. Department of Energy's Atmospheric Radiation Measurement Program is running a small network of 915-MHz radar wind profilers (RWPs) at its Southern Great Plains Cloud and Radiation Testbed site in northern Oklahoma and southern Kansas. Seasonal migration of passerines may cause significant interference with the operation of 915-MHz RWPs. The extent of this ''bird jamming'' depends on the radar's parameters, the place of deployment, the season, and the time of day. This poster presents a new diagnostic method for detecting possible bird contamination in RWP data, along with an evaluation of the method using a three-year data set for two RWPs.
