Self-organization of engineered epithelial tubules by differential cellular motility (open access)

Self-organization of engineered epithelial tubules by differential cellular motility

Patterning of developing tissues arises from a number of mechanisms, including cell shape change, cell proliferation, and cell sorting from differential cohesion or tension. Here, we reveal that differences in cell motility can also lead to cell sorting within tissues. Using mosaic engineered mammary epithelial tubules, we found that cells sorted depending on their expression level of the membrane-anchored collagenase matrix metalloproteinase (MMP)-14. These rearrangements were independent of the catalytic activity of MMP14 but absolutely required the hemopexin domain. We describe a signaling cascade downstream of MMP14 through Rho kinase that allows cells to sort within the model tissues. Cell speed and persistence time were enhanced by MMP14 expression, but only the latter motility parameter was required for sorting. These results indicate that differential directional persistence can give rise to patterns within model developing tissues.

Utilizing ZFS for the Storage of Acquired Data (open access)

Utilizing ZFS for the Storage of Acquired Data

Every day, the amount of data that is acquired from plasma experiments grows dramatically. It has become difficult for systems administrators to keep up with the growing demand for hard drive storage space. In the past, project storage has been supplied using UNIX filesystem (ufs) partitions. In order to increase the size of the disks using this system, users were required to discontinue use of the disk, so the existing data could be transferred to a disk of larger capacity or begin use of a completely new and separate disk, thus creating a segmentation of data storage. With the application of ZFS pools, the data capacity woes are over. ZFS provides simple administration that eliminates the need to unmount to resize, or transfer data to a larger disk. With a storage limit of 16 Exabytes (1018), ZFS provides immense scalability. Utilizing ZFS as the new project disk file system, users and administrators can eliminate time wasted waiting for data to transfer from one hard drive to another, and also enables more efficient use of disk space, as system administrators need only allocate what is presently required. This paper will discuss the application and benefits of using ZFS as an alternative …

Advantages of High Tolerance Measurements in Fusion Environments Applying Photogrammetry (open access)

Advantages of High Tolerance Measurements in Fusion Environments Applying Photogrammetry

Photogrammetry, a state-of-the-art technique of metrology employing digital photographs as the vehicle for measurement, has been investigated in the fusion environment. Benefits of this high tolerance methodology include relatively easy deployment for multiple point measurements and deformation/distortion studies. Depending on the equipment used, photogrammetric systems can reach tolerances of 25 microns (0.001 in) to 100 microns (0.004 in) on a 3-meter object. During the fabrication and assembly of the National Compact Stellarator Experiment (NCSX) the primary measurement systems deployed were CAD coordinate-based computer metrology equipment and supporting algorithms such as both interferometer-aided (IFM) and absolute distance measurementbased (ADM) laser trackers, as well as portable Coordinate Measurement Machine (CMM) arms. Photogrammetry was employed at NCSX as a quick and easy tool to monitor coil distortions incurred during welding operations of the machine assembly process and as a way to reduce assembly downtime for metrology processes.
