Understanding Virulence in the Brucellae and Francisellae: Towards Efficacious Treatments for Two Potential Biothreat Agents

Francisella tularensis, Yersinia pestis and Brucellae species are highly infectious pathogens classified as select agents by the Centers for Disease Control and Prevention (CDC) with the potential for use in bioterrorism attacks. These organisms are known to be facultative intracellular pathogens that preferentially infect human monocytes. As such, understanding how the host responds to infection with these organisms is paramount in detecting and combating human disease. We have compared the ability of fully virulent strains of each pathogen and their non-pathogenic near neighbors to enter and survive inside the human monocytic cell line THP-1 and have quantified the cellular response to infection with the goal of identifying both unique and common host response patterns. We expanded the scope of these studies to include experiments with pathogenic and non-pathogenic strains of Y. pestis, the causative agent of plague. Nonpathogenic strains of each organism were impaired in their ability to survive intracellularly compared with their pathogenic counterparts. Furthermore, infection of THP-1 cells with pathogenic strains of Y. pestis and F. tularensis resulted in marked increases in the secretion of the inflammatory chemokines IL-8, RANTES, and MIP-1{beta}. ASSESSMENT OF THE POTENTIAL FOR HYDROGEN GENERATION DURING GROUTING OPERATIONS IN THE R- AND P-REACTOR VESSELS

The R- and P-reactor buildings were retired from service and are now being prepared for deactivation and decommissioning (D&D). D&D activities will consist primarily of immobilizing contaminated components and structures in a grout-like formulation. Aluminum corrodes very rapidly when it comes in contact with the alkaline grout materials and as a result produces hydrogen gas. To address this potential deflagration/explosion hazard, the Materials Science and Technology Directorate (MS&T) of the Savannah River National Laboratory (SRNL) has been requested to review and evaluate existing experimental and analytical studies of this issue to determine if any process constraints on the chemistry of the fill material and the fill operation are necessary. Various options exist for the type of grout material that may be used for D&D of the reactor vessels. The grout formulation options include ceramicrete (pH 6-8), low pH portland cement + silica fume grout (pH 10.4), or Portland cement grout (pH 12.5). The assessment concluded that either ceramicrete or the silica fume grout may be used to safely grout the P-reactor vessel. FIRST INTERIM STATUS REPORT: MODEL 9975 LIFE EXTENSION PACKAGE TESTING

Three 9975 packages have been instrumented and exposed to bounding storage conditions to help identify the extent to which laboratory test results for fiberboard, O-rings and other components apply within a full-scale package. To date, changes observed in the fiberboard overpack and Oring seals are generally consistent with the behavior of laboratory samples. The lead shield within each package has developed a corrosion layer of lead carbonate, consistent with other 9975 packages. However, the morphology of the corrosion layer is different within these packages, possibly as a result of the severity of the conditioning environments beyond normal service conditions. Conditioning of the second package, LE2, has been discontinued due to the degree of degradation of the fiberboard, and its components have received a final examination. Additional testing will be performed on samples removed from the fiberboard assembly to verify its final condition. The other two packages will continue in test. This is an interim status report for experiments carried out per Task Technical Plan WSRC-TR-2005-00014 [1], which is part of the comprehensive 9975 package surveillance program [2]. Hydraulic Property and Soil Textural Classification Measurements for Rainier Mesa, Nevada Test Site, Nevada

This report presents particle size analysis, field-saturated hydraulic conductivity measurements, and qualitative descriptions of surficial materials at selected locations at Rainier Mesa, Nevada. Measurements and sample collection were conducted in the Rainier Mesa area, including unconsolidated sediments on top of the mesa, an ephemeral wash channel near the mesa edge, and dry U12n tunnel pond sediments below the mesa. Particle size analysis used a combination of sieving and optical diffraction techniques. Field-saturated hydraulic conductivity measurements employed a single-ring infiltrometer with analytical formulas that correct for falling head and spreading outside the ring domain. These measurements may prove useful to current and future efforts at Rainier Mesa aimed at understanding infiltration and its effect on water fluxes and radionuclide transport in the unsaturated zone.

PEP Run Report for Simulant Shakedown/Functional Testing

Pacific Northwest National Laboratory (PNNL) has been tasked by Bechtel National Inc. (BNI) on the River Protection Project-Waste Treatment Plant (RPP-WTP) project to perform research and development activities to resolve technical issues identified for the Pretreatment Facility (PTF). The Pretreatment Engineering Platform (PEP) was designed, constructed, and operated as part of a plan to respond to issue M12, "Undemonstrated Leaching Processes." The PEP is a 1/4.5-scale test platform designed to simulate the WTP pretreatment caustic leaching, oxidative leaching, ultrafiltration solids concentration, and slurry washing processes. The PEP replicates the WTP leaching processes using prototypic equipment and control strategies. The PEP also includes non-prototypic ancillary equipment to support the core processing. Two operating scenarios are currently being evaluated for the ultrafiltration process (UFP) and leaching operations. The first scenario has caustic leaching performed in the UFP-2 ultrafiltration feed vessels (i.e., vessel UFP-VSL-T02A in the PEP; and vessels UFP-VSL-00002A and B in the WTP PTF). The second scenario has caustic leaching conducted in the UFP-1 ultrafiltration feed preparation vessels (i.e., vessels UFP-VSL-T01A and B in the PEP; vessels UFP-VSL-00001A and B in the WTP PTF). Predicting detection probabilities for gas mixtures over HSI backgrounds

Detecting and identifying weak gaseous plumes using thermal image data acquired by airborne detectors is an area of ongoing research. This contribution investigates the relative detectability of gas mixtures over different backgrounds and a range of plume temperatures that are warmer and cooler than the ground. The focus of this analysis to support mission planning. When the mission is intended to collect evidence of particular chemicals, the analysis presented is this report can be used to determine conditions under which useful data can be acquired. Initial analyses can be used to determine whether LWIR is useful for the anticipated gas, temperature, and background combination.

Final Technical Report

The Center of Excellence for Hazardous Materials Management award was used to establish the organization and initiate investigations of hazardous waste issues along the U.S.-Mexico border. Scientific investigations conducted during the execution of this grant contributed significant data and established new sampling protocols to the dimension, frequency and severity of hazardous materials (e.g., heavy metals) along the U.S.-Mexico border. Additionally, new protocols and assessments with distinct Homeland Security implications were embedded thus establishing a baseline that will be significant for related investigations in the future.

Medicare Managed Care: Observations about Medicare Cost Plans

A letter report issued by the Government Accountability Office with an abstract that begins "Medicare cost plans--managed care plans paid based on the reasonable costs of delivering Medicare-covered services--enroll a small number of beneficiaries compared to Medicare Advantage (MA), Medicare's managed care program in which the plans accept financial risk if their costs exceed fixed payments received for each enrolled beneficiary. Despite the small enrollment, industry representatives stated that cost plans provide a managed care option in areas that traditionally had few or no MA plans. Current law allows existing cost plans to continue operating unless specific MA plans of sufficient enrollment serve the same area. In such cases, the cost plan must discontinue serving that area beginning in 2011. The Medicare Improvements for Patients and Providers Act of 2008 (MIPPA) required the Government Accountability Office (GAO) to examine issues related to the conversion of Medicare cost plans to MA plans. In response, GAO (1) determined the MA options available to beneficiaries in cost plans, (2) described key differences for beneficiaries between cost plans, MA plans, and Medicare fee-for-service (FFS); (3) determined the extent to which organizations offering cost plans also offer MA plans; and (4) described concerns cost plans …

SECONDARY WASTE/ETF (EFFLUENT TREATMENT FACILITY) PRELIMINARY PRE-CONCEPTUAL ENGINEERING STUDY

This pre-conceptual engineering study is intended to assist in supporting the critical decision (CD) 0 milestone by providing a basis for the justification of mission need (JMN) for the handling and disposal of liquid effluents. The ETF baseline strategy, to accommodate (WTP) requirements, calls for a solidification treatment unit (STU) to be added to the ETF to provide the needed additional processing capability. This STU is to process the ETF evaporator concentrate into a cement-based waste form. The cementitious waste will be cast into blocks for curing, storage, and disposal. Tis pre-conceptual engineering study explores this baseline strategy, in addition to other potential alternatives, for meeting the ETF future mission needs. Within each reviewed case study, a technical and facility description is outlined, along with a preliminary cost analysis and the associated risks and benefits.

Survival Rates of Juvenile Salmonids Passing Through the Bonneville Dam and Spillway in 2008

This report describes a 2008 acoustic telemetry survival study conducted by the Pacific Northwest National Laboratory for the Portland District of the U.S. Army Corps of Engineers. The study estimated the survival of juvenile Chinook salmon and steelhead passing Bonneville Dam (BON) and its spillway. Of particular interest was the relative survival of smolts detected passing through end spill bays 1-3 and 16-18, which had deep flow deflectors immediately downstream of spill gates, versus survival of smolts passing middle spill bays 4-15, which had shallow flow deflectors.

Systems Analysis of an Advanced Nuclear Fuel Cycle Based on a Modified UREX+3c Process

The research described in this report was performed under a grant from the U.S. Department of Energy (DOE) to describe and compare the merits of two advanced alternative nuclear fuel cycles -- named by this study as the “UREX+3c fuel cycle” and the “Alternative Fuel Cycle” (AFC). Both fuel cycles were assumed to support 100 1,000 MWe light water reactor (LWR) nuclear power plants operating over the period 2020 through 2100, and the fast reactors (FRs) necessary to burn the plutonium and minor actinides generated by the LWRs. Reprocessing in both fuel cycles is assumed to be based on the UREX+3c process reported in earlier work by the DOE. Conceptually, the UREX+3c process provides nearly complete separation of the various components of spent nuclear fuel in order to enable recycle of reusable nuclear materials, and the storage, conversion, transmutation and/or disposal of other recovered components. Output of the process contains substantially all of the plutonium, which is recovered as a 5:1 uranium/plutonium mixture, in order to discourage plutonium diversion. Mixed oxide (MOX) fuel for recycle in LWRs is made using this 5:1 U/Pu mixture plus appropriate makeup uranium. A second process output contains all of the recovered uranium except the …

The Institute for Multiscale Modeling of Biological Interactions (IMMBI) has two primary goals: Foster interdisciplinary collaborations among faculty and their research laboratories that will lead to novel applications of multiscale simulation and modeling methods in the biological sciences and engineering; and Building on the unique biophysical/biology-based engineering foundations of the participating faculty, train scientists and engineers to apply computational methods that collectively span multiple time and length scales of biological organization. The success of IMMBI will be defined by the following: Size and quality of the applicant pool for pre-doctoral and post-doctoral fellows; Academic performance; Quality of the pre-doctoral and post-doctoral research; Impact of the research broadly and to the DOE (ASCR program) mission; Distinction of the next career step for pre-doctoral and post-doctoral fellows; and Faculty collaborations that result from IMMBI activities. Specific details about accomplishments during the three years of DOE support for IMMBI have been documented in Annual Progress Reports (April 2005, June 2006, and March 2007) and a Report for a National Academy of Sciences Review (October 2005) that were submitted to DOE on the dates indicated. An overview of these accomplishments is provided.

Optical People Counting for Demand Controlled Ventilation: A Pilot Study of Counter Performance

This pilot scale study evaluated the counting accuracy of two people counting systems that could be used in demand controlled ventilation systems to provide control signals for modulating outdoor air ventilation rates. The evaluations included controlled challenges of the people counting systems using pre-planned movements of occupants through doorways and evaluations of counting accuracies when naive occupants (i.e., occupants unaware of the counting systems) passed through the entrance doors of the building or room. The two people counting systems had high counting accuracy accuracies, with errors typically less than 10percent, for typical non-demanding counting events. However, counting errors were high in some highly challenging situations, such as multiple people passing simultaneously through a door. Counting errors, for at least one system, can be very high if people stand in the field of view of the sensor. Both counting system have limitations and would need to be used only at appropriate sites and where the demanding situations that led to counting errors were rare.
