Carbachol- and ACPD-Induced Phosphoinositide Responses in the Developing Rat Neocortex

Signal transduction via the phosphoinositide (PI) second messenger system has key roles in the development and plasticity of the neocortex. The present study localized PI responses to individual cortical layers in slices of developing rat somatosensory cortex. The acetylcholine agonist carbachol and the glutamate agonist trans-1-amino-1,3-cyclopentanedicarboxylic acid (ACPD) were used to stimulate PI turnover. The PI responses were compared to the distribution of the corresponding PI-linked receptors in order to investigate the regional ontogeny of PI coupling to receptors in relation to neural development. The method for assessing PI turnover was modified from Hwang et al. (1990). This method images the PI response autoradiographically through the localizaton of [3H]cytidine that has been incorporated into the membrane-bound intermediate, cytidine diphosphate diacylglycerol. In each age group (postnatal days 4-30), carbachol resulted in more overall labeling than ACPD. For both agonists, the response peaked on postnatal day 10 (P10) and was lowest in the oldest age group. The laminar distribution of the carbachol PI response from P4-P16 corresponded fairly well with the laminar distribution of [3H]quinuclidinyl benzilate binding (Fuchs, 1995). Comparison of GPS Point Selection Methods for GIS Area Measurement of Small Jurisdictional Wetlands

U.S. Army Corps of Engineers (USACE) regulates fill of jurisdictional waters of the United States including wetlands. Recent USACE regulations set a threshold of impacts to wetlands at one-half acre. Impact area can be determined by Global Positioning System (GPS) measurement of wetland boundary and Geographic Information System (GIS) calculation of impact area. GPS point selection methods include (1) equal time interval, (2) transect and (3) intuition. Four two-acre shapes were measured with each GPS method and brought into GIS for area calculation. Analysis of variance and Root Mean Square Error analyses determine that the transect method is an inferior point selection method in terms of accuracy and efficiency.

A Novel Mechanism for Site-Directed Mutagenesis of Large Catabolic Plasmids Using Natural Transformation

Natural transformation is the process by which cells take up DNA from the surrounding medium under physiological conditions, altering the genotype in a heritable fashion. This occurs without chemical or physical treatment of the cells. Certain Acinetobacter strains exhibit a strong tendency to incorporate homologous DNA into their chromosomes by natural transformation. Transformation in Acinetobacter exhibits several unique properties that indicate this system's superiority as a model for transformation studies or studies which benefit from the use of transformation as an experimental method of gene manipulation. Pseudomonas putida is the natural host of TOL plasmids, ranging between 50 kbp and 300 kbp in size and encoding genes for the catabolism of toluene, meta-toluate, and xylene. These very large, single-copy plasmids are difficult to isolate, manipulate, or modify in vitro. In this study, the TOL plasmid pDKR1 was introduced into Acinetobacter calcoaceticus strains and genetically engineered utilizing natural transformation as part of the process. Following engineering by transformation, the recombinant DNA molecule was returned to the native genetic background of the original host P. putida strain. The Developmental Physiology of the Zebrafish: Influence of Environment and Cardiovascular Attributes

Temperature effects on the development of the zebrafish embryos and larvae and adults were examined. It was found that the earlier in development a temperature change was performed on an embryo, the more significant the change in survival and/or subsequent development. Thus, viable temperature ranges for zebrafish widened significantly as development proceeded. Adults reared and bred at 25oC produced embryos that were significantly more successful at the lower range of rearing temperatures compared to embryos produced from adults reared at 28oC. The majority of this study focused on the physiological effects of swim training during development in the zebrafish. The earlier in development the zebrafish larvae were trained, the greater the mortality. Trained free swimming larvae had a significantly higher routine oxygen consumption after 11 days of training, and a higher mass specific routine metabolic rate after 8 and 11 days of training. Trained free swimming larvae consumed significantly less oxygen during swimming and were more efficient at locomotion, compared to control larvae. Training enhanced survival during exposure to extreme hypoxia in all age groups. Spatial and Temporal Patterns of Areal and Volumetric Phytoplankton Productivity of Lake Texoma

Phytoplankton productivity of Lake Texoma was measured for one year from August 1999 to August 2000 for four stations, using the oxygen change method and laboratory incubation. Mean values of the photosynthetic parameters, PBmax and alphaB ranged from 4.86 to 46.39 mg O2.mg Chl-1.hr-1 for PBmax and 20.06 to 98.96 mg O2.mg Chl-1.E-1.m2 for alphaB. These values were in the range to be expected for a highly turbid, temperate reservoir. Estimated gross annual areal productivity ranged from 594 g C.m2.yr-1 (P.Q. = 1.2), at a station in the Washita River Zone to 753 g C.m2.yr-1 at a station in the Red River Zone, of the reservoir. Gross annual areal productivity at Station 17, in the Main Lake Zone, was 708 g C.m2.yr-1. Gross areal and volumetric productivity showed distinct seasonal variation with Photosynthetically Available Radiation (PAR) and temperature. Trophic status estimated on a station-by-station basis, using net productivity values derived from gross productivity and respiration estimates, was mesotrophic for all the stations, though one station approached eutrophy. Net productivity values ranged from 0.74 to 0.91 g C. m-2.d-1. A Possible Role of Ascorbate in Boron Deficient Radish (Raphanus sativa L. cv. Cherry Belle)

The most apparent symptom of boron deficiency in higher plants is a cessation of growth. Deficiency causes a reduction in ascorbate concentration and the absorption of nutrient ions. Addition of ascorbate temporarily relieves deficiency symptoms. In boron sufficient plants the addition of ascorbate to media causes an increased uptake of nutrients. In an attempt to discover if ascorbate addition to deficient plants causes increased ion uptake, radish plants were grown hydroponically in four different strengths of boron solution. A colorimetric assay for phosphorus was performed both before and after supplementation. Results, however, were inconclusive.

Life History and Secondary Production of Caenis latipennis Banks (Ephemeroptera: Caenidae) in Honey Creek, Oklahoma

A study of the life history and secondary production of Caenis latipennis, a caenid mayfly, was conducted on Honey Creek, OK. from August 1999 through September 2000. The first instar nymph was described. Nymphs were separated into five development classes. Laboratory egg and nymph development rates, emergence, fecundity, voltinism, and secondary production were analyzed. C. latipennis eggs and nymphs take 132 and 1709 degree days to develop. C. latipennis had an extended emergence with five peaks. Females emerged, molted, mated, and oviposited in an estimated 37 minutes. Mean fecundity was 888.4 ± 291.9 eggs per individual (range 239 -1576). C. latipennis exhibited a multivoltine life cycle with four overlapping generations. Secondary production was 6,052.57 mg/m2/yr.

Purification of Aspartate Transcarbamoylase from Moraxella (Branhamella) catarrhalis

The enzyme, aspartate transcarbamoylase (ATCase) from Moraxella (Branhamella) catarrhalis, has been purified. The holoenzyme has a molecular mass of approximately 510kDa, harbors predominantly positive charges and is hydrophobic in nature. The holoenzyme possesses two subunits, a smaller one of 40 kDa and a larger one of 45 kDa. A third polypeptide has been found to contribute to the overall enzymatic activity, having an approximate mass of 55 kDa. The ATCase purification included the generation of cell-free extract, streptomycin sulfate cut, 60 °C heat step, ammonium sulfate cut, dialysis and ion, gel-filtration and hydrophobic interaction chromatography. The enzyme's performance throughout purification steps was analyzed on activity and SDS-PAGE gradient gels. Its enzymatic, specific activities, yield and fold purification, were also determined.

An examination of the riparian bottomland forest in north central Texas through ecology, history, field study, and computer simulation

This paper explores the characterization of a riparian bottomland forest in north central Texas in two ways: field study, and computer simulation with the model ZELIG. First, context is provided in Chapter One with a brief description of a southern bottomland forest, the ecological services it provides, and a history of bottomland forests in Texas from the nineteenth century to the present. A report on a characterization study of the Lake Ray Roberts Greenbelt forest comprises Chapter Two. The final chapter reviews a phytosocial study of a remnant bottomland forest within the Greenbelt. Details of the ZELIG calibration process follow, with a discussion of ways to improve ZELIG's simulation of bottomland forests.

Plastidial carbonic anhydrase in cotton (Gossypium hirsutum L.): characterization, expression, and role in lipid biosynthesis

Recently, plastidial carbonic anhydrase (CA, EC 4.2.1.1) cDNA clones encoding functional CA enzymes were isolated from a nonphotosynthetic cotton tissue. The role of CA in photosynthetic tissues have been well characterized, however there is almost no information for the role of CA in nonphotosynthetic tissues. A survey of relative CA transcript abundance and enzyme activity in different cotton organs revealed that there was substantial CA expression in cotyledons of seedlings and embryos, both nonphotosynthetic tissues. To gain insight into the role(s) of CA, I examined CA expression in cotyledons of seedlings during post-germinative growth at different environmental conditions. CA expression in cotyledons of seedlings increased from 18 h to 72 h after germination in the dark. Seedlings exposed to light had about a 2-fold increase in CA activities when compared with seedlings kept in the dark, whereas relative CA transcript levels were essentially the same. Manipulation of external CO2 environments [zero, ambient (350 ppm), or high (1000 ppm)] modulated coordinately the relative transcript abundance of CA (and rbcS) in cotyledons, but did not affect enzyme activities. Palmitoyl-acyl Carrier Protein Thioesterase in Cotton (Gossypium hirsutum L.): Biochemical and Molecular Characterization of a Major Mechanism for the Regulation of Palmitic Acid Content

The relatively high level of palmitic acid (22 mol%) in cottonseeds may be due in part to the activity of a palmitoyl-acyl carrier protein (ACP) thioesterase (PATE). In embryo extracts, PATE activity was highest at the maximum rate of reserve accumulation (oil and protein). The cotton FatB mRNA transcript abundance also peaked during this developmental stage, paralleling the profiles of PATE enzyme activity and seed oil accumulation. A cotton FatB cDNA clone was isolated by screening a cDNA library with a heterologous Arabidopsis FatB probe (Pirtle et al., 1999, Plant and Cell Physiology 40: 155-163). The predicted amino acid sequence of the cotton PATE preprotein had 63% identity to the Arabidopsis FatB thioesterase sequence, suggesting that the cotton cDNA clone probably encoded a FatB-type thioesterase. When acyl-CoA synthetase-minus E. coli mutants expressed the cotton cDNA, an increase in 16:0 free fatty acid content was measured in the culture medium. In addition, acyl-ACP thioesterase activity assays in E. coli lysates revealed that there was a preference for palmitoyl-ACP over oleoyl-ACP in vitro, indicating that the cotton putative FatB cDNA encoded a functional thioesterase with a preference for saturated acyl-ACPs over unsaturated acyl-ACPs (FatA). Use of geographic information systems for assessing ground water pollution potential by pesticides in central Thailand

This study employed geographic information systems (GIS) technology to evaluate the vulnerability of groundwater to pesticide pollution. The study area included three provinces (namely, Kanchana Buri, Ratcha Buri, and Suphan Buri) located in the western part of central Thailand. Factors used for this purpose were soil texture, percent slope, primary land use, well depth, and monthly variance of rainfall. These factors were reclassified to a common scale showing potential to cause groundwater contamination by pesticides. This scale ranged from 5 to 1 which means high to low pollution potential. Also, each factor was assigned a weight indicating its influence on the movement of pesticides to groundwater. Well depth, the most important factor in this study, had the highest weight of 0.60 while each of the remaining factors had an equal weight of 0.10. These factors were superimposed by a method called “arithmetic overlay” to yield a composite vulnerability map of the study area. Maps showing relative vulnerability of groundwater to contamination by pesticides were produced. Pecan Creek flows southeast through the City of Denton, Texas. Characterized as an urban watershed, the basin covers approximately 63.5 km2. Pecan Creek is an intermittent stream that receives nonpoint runoff from urban landuses, and the City of Denton's wastewater treatment plant, Pecan Creek Water Reclamation Plant, discharges effluent to the stream. Downstream from the City of Denton and the wastewater treatment plant, Pecan Creek flows about 6,000 m through agricultural, pasture, and forested landscapes into Copas Cove of Lake Lewisville, creating backwater conditions. Pecan Creek water quality and chemistry were monitored from August 1997 to October 2001. Water quality was influenced by seasonal, spatial, climatic, and diurnal dynamics. Wastewater effluent discharged from the Pecan Creek Water Reclamation Plant had the greatest influence on water quality of the stream and backwaters. Water quality monitoring of Pecan Creek demonstrated that dissolved oxygen standards for the protection of aquatic life were being achieved. Water quality modeling of Pecan Creek was completed to assess future increases in effluent flow from the Pecan Creek Water Reclamation Plant. Water quality modeling indicated that dissolved oxygen standards would not be achieved at the future effluent flow of 21 MGD and at NPDES permitted loadings. Model results …

Mechanisms of rapid receptive field reorganization in rat spinal cord

Rapid receptive field (RF) reorganization of somatosensory neurons in the rat dorsal horn was examined using extracellular single unit recording. Subcutaneous injection of lidocaine into RFs of dorsal horn neurons results in expansion of their RFs within minutes. The expanded RFs appear adjacent to or/and proximal to original RFs. Out of 63 neurons tested, 36 (58%) show RF reorganization. The data suggest that dorsal horn of spinal cord is one of the initial sites for RF reorganization. The neural mechanisms of this effect are not well understood. We propose that changes in biophysical properties (membrane conductance, length constant) of the neurons resulting from lidocaine injection contribute to RF reorganization. Iontophoretic application of glutamate onto dorsal horn neurons that show lidocaine induced RF's expansion were used to test the model. Application of glutamate produced reduction of reorganized RFs in 9 of 20 (45%) tested cells. Application of NBQX produced no effect on either original or expanded RFs indicate that RF shrinkage effects of glutamate involve NMDA receptors. The results are consistent with the prediction of the proposed model. Neuronal Network Analyses in vitro of Acute Individual and Combined Responses to Fluoxetine and Ethanol

Embryonic murine neuronal networks cultured on microelectrode arrays were used to quantify acute electrophysiological effects of fluoxetine and ethanol. Spontaneously active frontal cortex cultures showed highly repeatable, dose-dependent sensitivities to both compounds. Cultures began to respond to fluoxetine at 3 µM and were shut off at 10-16 µM. EC50s mean ± S.D. for spike and burst rates were 4.1 ± 1.5 µM and 4.5 ± 1.1 µM (n=14). The fluoxetine inhibition was reversible and without effect on action potential wave shapes. Ethanol showed initial inhibition at 20 mM, with spike and burst rate EC50s at 52.0 ± 17.4 mM and 56.0 ± 17.0 mM (n=15). Ethanol concentrations above 100 -140 mM led to cessation of activity. Although ethanol did not change the shape and amplitude of action potentials, unit specific effects were found. The combined application of ethanol and fluoxetine was additive. Ethanol did not potentiate the effect of fluoxetine.

Use of Satellite Imagery and GIS to Model Brood-Rearing Habitat for Rio Grande Wild Turkey Populations Occurring in the Western Cross Timbers Region of Texas

Remote sensing and GIS have become standard tools for evaluating spatial components of wildlife habitats. These techniques were implemented to evaluate Rio Grande wild turkey (Meleagris gallopavo) poult-rearing habitat in the Western Cross Timbers region of Texas. Texas Parks and Wildlife (TPWD) random roving turkey counts for 1987-1989 and 1998-2000 were selected, indicating locations where hens with poults were observed. Satellite imagery from 1988 and 1999 was classified and then processed with Patch Analyst. To add robustness, stream, road and census population densities were also evaluated for each turkey location. Analysis of the 1988 canopy cover image, comparing observed locations with randomly-selected habitat cells (N = 20) indicated significant differences (p <.05) for patch edge variables. Mean patch edge was significantly greater for habitat locations where hens with poults were observed than for those selected at random. Spatial data for 1999 did not indicate a significant difference (p < .05) between sampling groups (observed vs. random, N = 30). Significant differences (p <.05) did occur for turkey locations observed in both 1988 and 1999 (N = 7). Recovery of the Fish Population of a Municipal Wastewater Dominated, North Texas Creek After a Major Chlorine Disturbance

This study evaluated the effects of a major chlorine disturbance on fish communities in Pecan creek by the City of Denton's Pecan Creek Water Reclamation Plant. Fish communities in Pecan Creek were sampled using a depletion methodology during February, April, July, and November, 1999. February and April sampling events showed that the fish communities were severely impacted by the chlorine. Sampling during July and November showed fish communities recovered in Pecan Creek. The first-twenty minutes of shocking and seining data were analyzed to mirror an equal effort methodology. This methodology was compared to the depletion methodology to see if the equal effort methodology could adequately monitor the recovery of Pecan Creek after the chlorine disturbance. It was determined that the equal effort methodology was capable of monitoring the recovery of Pecan Creek, but could not accurately represent the fisheries community as well as the depletion method. These data using the twenty-minute study were compared to a previous study. Results of this study were similar to those found in a previous study, although fish communities were more severely impacted and took longer to recover.

Use of GIS and Remote Sensing Technologies to Study Habitat Requirements of Ocelots, Leopardus pardalis, in south Texas

The goals of this study were to use Geographic Information Systems (GIS) and remote sensing technologies to gain a better understanding of habitat requirements of a population of ocelots in south Texas, and then apply this knowledge to form a predictive model to locate areas of suitable habitat in Willacy and Cameron counties, Texas. Satellite imagery from August 1991 and August 2000 were classified into four land cover types: closed canopy, open canopy, water, and urban/barren. These classified images were converted into digital thematic maps for use in resource utilization studies and modeling. Location estimates (762 from 1991 and 406 from 2000) were entered into a GIS in order to extract information about home range and resource selection. Each animal's home range was calculated using both Minimum Convex Polygon (MCP) and Kernel home range estimators (95% and 50%). Habitat parameters of interest were: soil, land cover, human density, road density, and distance to closest road, city and water body. Ocelots were found to prefer closed canopy and avoid open canopy land cover types. Ocelots preferred soils known to support thorn scrub, an indication of the importance of this habitat. Assessing the Spatial and Temporal Distribution of MTBE and BTEX Compounds in Lake Lewisville, Texas February 1999 - February 2000

The spatial and temporal distribution of Methyl Tertiary-Butyl Ether (MTBE) and BTEX (Benzene, Toluene, Ethylbenzene, Xylenes) compounds were assessed in a multipurpose reservoir, Lake Lewisville, Texas between February 1999 and February 2000. Concentrations of MTBE ranged from 0.0 - 16.7 mg/L. Levels of MTBE in the lake were related to watercraft. BTEX concentrations were never detected above 2.0 mg/L during the sampling period. Finished drinking water from Denton and the Upper Trinity Regional Water District (UTRWD) Treatment Plants were also tested for MTBE and BTEX. MTBE and BTEX were not detected in UTRWD water samples. Denton's finished water samples never exceeded 2.2 mg/L for MTBE and BTEX was not detected except for one replicate of 1.1 mg/L toluene.

Requirements for Cell-Free Cyanide Oxidation by Pseudomonas Fluorescens NCIMB 11764

The involvement of cyanide oxygenase in the metabolism of pyruvate and a-ketoglutarate-cyanohydrin was investigated and shown to occur indirectly by the consumption of free cyanide arising from the cyanohydrins via chemical dissociation. Thus, free cyanide remains the substrate, for which the enzyme displays a remarkably high affinity (Kmapp,4 mM). A model for cyanide utilization is therefore envisioned in which the substrate is initially detoxified by complexation to an appropriate ligand followed by enzymatic oxidation of cyanide arising at sublethal levels via chemical dissociation. Putative cyanide oxygenase in cell extracts consumed both oxygen and NADH in equimolar proportions during cyanide conversion to CO2 and NH3 and existed separately from an unknown heat-stable species responsible for the nonenzymatic cyanide-catalyzed consumption of oxygen. Evidence of cyanide inhibition and nonlinear kinetics between enzyme activity and protein concentration point to a complex mechanism of enzymatic substrate conversion.

Influence of copper on resistance of Lumbricus terrestris to bacterial challenge

Earthworms, Lumbricus terrestris, were challenged orally and intracoelomically with two bacterial species, Aeromonas hydrophila and Pseudomonas aeruginosa, and mortality rates were observed. Neither were found to be particularly pathogenic at injected doses of up to 108 bacteria per earthworm. The influence of Cu++ (as CuSO4) on the earthworm's response to bacterial challenge was investigated by exposing earthworms to sublethal levels of Cu++ prior to bacterial challenge. Exposure at sublethal concentrations up to 3 m g/cm2 did not have a pronounced influence on host resistance to challenge as measured by earthworm mortality. Cu++ increased the earthworm's ability to agglutinate rabbit erythrocytes, indicating that Cu++ exposure caused coelomocyte death, autolysis and release of agglutinins into the coelom, possibly explaining resistance to bacterial challenge.

Production and Characterization of a Novel Extracellular Polysaccharide Produced by Paenibacillus velaei, Sp. Nov

Paenibacillus velaei, sp. nov. is a soil bacterium capable of producing an unusually large amount of exopolysaccharide (EPS). The EPS contains glucose, mannose, galactose and fucose in a molar ratio of 4:2:1:1. The molecular weight of the EPS is higher than 2x106. The viscosity of 1% EPS is 1300 cP when measured at a shear rate of 1 sec-1. Physiological parameters for optimal production of the EPS were studied and it was found that 1.4 g dry weight per 1 l of medium was produced when the bacteria were grown at 30EC and the pH adjusted at 7± 0.2 in a medium containing glucose as the carbon source. Growing the bacteria on different carbon sources did not alter the quantity or the composition of the EPS produced. No toxicity effects were observed in mice or rats when EPS was administered in amounts ranging from 20 to 200 mg per kg body weight. The data obtained from physical, chemical and biological properties suggest that the EPS may be employed in several industrial and environmental applications. BioInformatics, Phylogenetics, and Aspartate Transcarbamoylase

In this research, the necessity of understanding and using bioinformatics is demonstrated using the enzyme aspartate transcarbamoylase (ATCase) as the model enzyme. The first portion of this research focuses on the use of bioinformatics. A partial sequence of the pyrB gene found in Enterococcus faecalis was submitted to GenBank and was analyzed against the contiguous sequence from its own genome project. A BLAST (Basic Local Alignment Search Tool; Atschul, et al., 1990) was performed in order to hypothesize the remaining portion of the gene from the contiguous sequence. This allowed a global comparison to other known aspartate transcarbamoylases (ATCases) and once deduced, a translation of the sequence gave the stop codon and thus the complete sequence of the open reading frame. When this was complete, upstream and downstream primers were designed in order to amplify the gene from genomic DNA. The amplified product was then sequenced and used later in phylogenetic analyses concerning the evolution of ATCase. The second portion of this research involves taking multiple ATCase nucleotide sequences and performing phenetic and phylogenetic analyses of the archaea and eubacter families. From these analyses, ancestral relationships which dictate both structure and function were extrapolated from the data and discussed.

Characterizing the Municipal Solid Waste Stream in Denton, Texas

Forty-two commercially collected dumpsters from Denton’s Municipal Solid Waste Stream were emptied, sorted and weighed to characterize the material types and make preliminary recycling policy recommendations. The general composition of Denton’s solid waste stream was not significantly different from the composition of the nation’s solid waste stream. Fifty-eight percent of the observed waste stream was recyclable. Paper made up the largest portion of recyclable materials and the "grocery" source category had more paper than any of the other five categories. Based on these findings, an incrementally aggressive approach is recommended to reduce certain types of wastes observed in the waste stream. This would include a Pay-As-You-Throw Program followed by an Intermediate Processing Center that can be converted to a Materials Recovery Facility.
