2010 Census: Census Bureau Has Improved the Local Update of Census Addresses Program, but Challenges Remain

A letter report issued by the Government Accountability Office with an abstract that begins "The Department of Commerce's (Commerce) U.S. Census Bureau (Bureau) seeks updated information on the addresses and maps of housing units and group quarters from state, local, and tribal governments through the Local Update of Census Addresses (LUCA) Program. Prepared under the Comptroller General's authority, this report assesses (1) the status of the LUCA Program, (2) the Bureau's response to prior recommendations by GAO and others and new challenges related to the program, and (3) the Bureau's plans for conducting the program in areas affected by hurricanes Katrina and Rita. GAO reviewed LUCA program documents, met with and surveyed participants in the LUCA Dress Rehearsal, and interviewed Bureau officials and local officials in the Gulf Coast region."

2010 Census: Counting Americans Overseas as Part of the Census Would Not Be Feasible

Testimony issued by the Government Accountability Office with an abstract that begins "The U.S. Census Bureau (Bureau) has typically excluded from the census private citizens residing abroad, but included overseas members of the military, federal civilian employees, and their dependents (in the 1990 and 2000 Censuses, these individuals were included in the numbers used for apportioning Congress). The Bureau recently tested the practicality of counting all overseas Americans. GAO was asked to testify on the test's initial results. Our statement is based on our published reports, one of which is being released at today's hearing."

21st Century Challenges: Performance Budgeting Could Help Promote Necessary Reexamination

Testimony issued by the Government Accountability Office with an abstract that begins "As part of its work to improve the management and performance of the federal government, GAO monitors progress and continuing challenges in performance budgeting and the Administration's related initiatives, such as the Program Assessment Rating Tool (PART). In light of the nation's long-term fiscal imbalance and other emerging 21st century challenges, we have also reported that performance budgeting can help facilitate a needed reexamination of what the federal government does, how it does it, who does it, and how it is financed in the future. GAO remains committed to working with Congress and the Administration to help address these important and complex issues."

21st Century Locomotive Technology: Quarterly Technical Status Report 11 DOE/AL68284-TSR11

The fuel injection hardware on the single cylinder research engine was upgraded and performance trends were quantified. The effects of fuel injection rate shapes were studied. Long-term cycling of subscale battery cell assemblies has identified aging trends. Follow-up vibration testing of an instrumented COTS battery was performed. Optimal trip planning algorithms were implemented and demonstrated in the interactive, real-time simulation environment.

21st Century Locomotive Technology: Quarterly Technical Status Report 12 DOE/AL68284-TSR12

A unit pump system was installed on the single cylinder engine. The performance entitlement of the high-pressure common rail system at notch 4 and notch 8 was determined, and data was also compared with a production design multi-cylinder engine. Vibration testing of a hybrid-bus-design battery revealed extensive insulation wear. The vendor initiated design activities to address vibration. A cell-level test program was initiated to study battery current flow at room temperature and develop limits. The robustness and sub-optimal simplified implementation of fuel optimization algorithms was studied.

2nd Annual DOE-ERSP PI Meeting: Abstracts (open access)

2nd Annual DOE-ERSP PI Meeting: Abstracts

Welcome to the annual 2007 Environmental Remediation Sciences Program (ERSP) Principal Investigators (PIs) meeting. The purpose of this meeting is to bring together all of the lead PIs and key Co-PIs in the program to share and review the results of funded research from the past year. This meeting allows program managers from the Environmental Remediation Sciences Division (ERSD) within the Office of Biological and Environmental Research (OBER) to gauge the progress and significance of the funded research, and it is also an important venue to showcase ERSP research to interested parties within DOE and other invited federal agency representatives. Additionally, these meetings should serve as an opportunity for funded PIs to view their research in the context of the entire ERSP portfolio. Past ERSP meetings have been very important venues for detailed discussion of research results among PIs, development of new research ideas, fostering new collaborations and discussion with ERSD program managers on future research efforts and/or initiatives within the program. In short, these meetings are an important resource for both program managers and PIs. There will be only one ERSP PI meeting for 2007. In years past, ERSD has sponsored two PI meetings, one in the spring and …

[3H]Azidodantrolene photoaffinity labeling, synthetic domain peptides and monoclonal antibody reactivity identify the dantrolene binding sequence on RyR1 (open access)

[3H]Azidodantrolene photoaffinity labeling, synthetic domain peptides and monoclonal antibody reactivity identify the dantrolene binding sequence on RyR1

Dantrolene is a drug that suppresses intracellular Ca2+ release from sarcoplasmic reticulum in normal skeletal muscle and is used as a therapeutic agent in individuals susceptible to malignant hyperthermia. Though its precise mechanism of action has not been elucidated, we have identified the N-terminal region (amino acids 1-1400) of the skeletal muscle isoform of the ryanodine receptor (RyR1), the primary Ca2+ release channel in sarcoplasmic reticulum, as a molecular target for dantrolene using the photoaffinity analog [3H]azidodantrolene(1). Here, we demonstrate that heterologously expressed RyR1 retains its capacity to be specifically labeled with [3H]azidodantrolene,indicating that muscle specific factors are not required for this ligand-receptor interaction. Synthetic domain peptides of RyR1, previously shown to affect RyR1 function in vitro and in vivo, were exploited as potential drug binding site mimics and used in photoaffinity labeling experiments. Only DP1 and DP1-2, peptide s containing the amino acid sequence corresponding to RyR1 residues 590-609, were specifically labeled by [3H]azidodantrolene. A monoclonal anti-RyR1 antibody which recognizes RyR1 and its 1400 amino acid N-terminal fragment, recognizes DP1 and DP1-2 in both Western blots and immunoprecipitation assays, and specifically inhibits [3H]azidodantrolene photolabeling of RyR1 and its N-terminal fragment in sarcoplasmic reticulum. Our results indicate that synthetic domain …

3rd year final contractor report for: U.S. Department of Energy Stewardship Science Academic Alliances Program Project Title: Detailed Measurements of Rayleigh-Taylor Mixing at Large and Small Atwood Numbers (open access)

3rd year final contractor report for: U.S. Department of Energy Stewardship Science Academic Alliances Program Project Title: Detailed Measurements of Rayleigh-Taylor Mixing at Large and Small Atwood Numbers

This project had two major tasks: Task 1. The construction of a new air/helium facility to collect detailed measurements of Rayleigh-Taylor (RT) mixing at high Atwood number, and the distribution of these data to LLNL, LANL, and Alliance members for code validation and design purposes. Task 2. The collection of initial condition data from the new Air/Helium facility, for use with validation of RT simulation codes at LLNL and LANL. This report describes work done in the last twelve (12) months of the project, and also contains a summary of the complete work done over the three (3) life of the project. As of April 1, 2006, the air/helium facility (Task 1) is now complete and extensive testing and validation of diagnostics has been performed. Initial condition studies (Task 2) is also comp lete. Detailed experiments with air/helium with Atwood numbers up to 0.1 have been completed, and Atwood numbers of 0.25. Within the last three (3) months we have been able to successfully run the facility at Atwood numbers of 0.5. The progress matches the project plan, as does the budget. We have finished the initial condition studies using the water channel, and this work has been accepted for …
