Liquid Effluent Monitoring Information System (LEMIS) System Construction (open access)

Liquid Effluent Monitoring Information System (LEMIS) System Construction

The liquid effluent sampling program is part of the effort to minimize adverse environmental impact during the cleanup operation at the Hanford Site. Of the 33 Phase I and Phase II liquid effluents, all streams actively discharged to the soil column will be sampled. The Liquid Effluent Monitoring Information System (LEMIS) is being developed as the organized information repository facility in support of the liquid effluent monitoring requirements of the Tri-Party Agreement. It is necessary to provide an automated repository into which the results from liquid effluent sampling will be placed. This repository must provide for effective retention, review, and retrieval of selected sample data by authorized persons and organizations. This System Construction document is the aggregation of the DMR P+ methodology project management deliverables. Together they represent a description of the project and its plan through four Releases, corresponding to the definition and prioritization of requirements defined by the user.

105-DR Large Sodium Fire Facility closure activities evaluation report (open access)

105-DR Large Sodium Fire Facility closure activities evaluation report

This report evaluates the closure activities at the 105-DR Large Sodium Fire Facility. The evaluation compares these activities to the regulatory requirements and closure plan requirements. The report concludes that the areas identified in the closure plan can be clean closed. This report summarizes and evaluates the closure activities performed in support of partial closure of the 105-DR Large Sodium Fire Facility (LSFF). This evaluation will be used in assessing the condition of the 105-DR LSFF for the purpose of meeting the partial clean closure conditions described in the 105-DR Large Sodium Fire Facility Closure Plan (DOE-RL 1995). Based on the evaluation of the decontamination activities, sampling activities, and sample data, it is has been determined that the partial clean closure conditions for the 105-DR LSFF have been met.

Challenges in biotechnology at LLNL: from genes to proteins (open access)

Challenges in biotechnology at LLNL: from genes to proteins

This effort has undertaken the task of developing a link between the genomics, DNA repair and structural biology efforts within the Biology and Biotechnology Research Program at LLNL. Through the advent of the I.M.A.G.E. (Integrated Molecular Analysis of Genomes and their Expression) Consortium, a world-wide effort to catalog the largest public collection of genes, accepted and maintained within BBRP, it is now possible to systematically express the protein complement of these to further elucidate novel gene function and structure. The work has ensued in four phases, outlined as follows: (1) Gene and System selection; (2) Protein expression and purification; (3) Structural analysis; and (4) biological integration. Proteins to be expressed have been those of high programmatic interest. This includes, in particular, proteins involved in the maintenance of genome integrity, particularly those involved in the repair of DNA damage, including ERCC1, ERCC4, XRCC2, XRCC3, XRCC9, HEX1, APN1, p53, RAD51B, RAD51C, and RAD51. Full-length cDNA cognates of selected genes were isolated, and cloned into baculovirus-based expression vectors. The baculoviral expression system for protein over-expression is now well-established in the Albala laboratory. Procedures have been successfully optimized for full-length cDNA clining into expression vectors for protein expression from recombinant constructs. This includes the …

Modeling the wind-fields of accidental releases with an operational regional forecast model (open access)

Modeling the wind-fields of accidental releases with an operational regional forecast model

The Atmospheric Release Advisory Capability (ARAC) is an operational emergency preparedness and response organization supported primarily by the Departments of Energy and Defense. ARAC can provide real-time assessments of atmospheric releases of radioactive materials at any location in the world. ARAC uses robust three-dimensional atmospheric transport and dispersion models, extensive geophysical and dose-factor databases, meteorological data-acquisition systems, and an experienced staff. Although it was originally conceived and developed as an emergency response and assessment service for nuclear accidents, the ARAC system has been adapted to also simulate non-radiological hazardous releases. For example, in 1991 ARAC responded to three major events: the oil fires in Kuwait, the eruption of Mt. Pinatubo in the Philippines, and the herbicide spill into the upper Sacramento River in California. ARAC`s operational simulation system, includes two three-dimensional finite-difference models: a diagnostic wind-field scheme, and a Lagrangian particle-in-cell transport and dispersion scheme. The meteorological component of ARAC`s real-time response system employs models using real-time data from all available stations near the accident site to generate a wind-field for input to the transport and dispersion model. Here we report on simulation studies of past and potential release sites to show that even in the absence of local meteorological …
