Temperature-sensitive Post-translational Regulation of Plant Omega-3 Fatty-acid Desaturases Is Mediated by the Endoplasmic Reticulum-associated Degradation Pathway

Article expressing two closely related plant FAD3 genes in yeast cells and found that their enzymes produced significantly different amounts of omega-3 fatty acids and that these differences correlated to differences in rates of protein turnover. The findings indicate that Fad3 protein abundance is regulated by a combination of cis-acting degradation signals and the ubiquitin-proteasome pathway and that modulation of Fad3 protein amounts in response to temperature may represent one mechanism of homeoviscous adaptation in plants.

Visualization of Lipid Droplet Composition by Direct Organelle Mass Spectrometry

Article reporting an approach that combines the visualization of individual lipid droplets (LDs), microphase extraction of lipid components from droplets, and the direct identification of lipid composition by nanospray mass spectrometry, even to the level of a single LD. The triacylglycerol (TAG) composition of LDs from several plant sources (mature cotton (Gossypium hirsutum) embryos, roots of cotton seedlings, and Arabidopsis thaliana seeds and leaves) were examined by direct organelle mass spectrometry and revealed the heterogeneity of LDs derived from different plant tissue sources.

Lipoxygenase-mediated Oxidation of Polyunsaturated N-Acylethanolamines in Arabidopsis

Article hypothesizing that lipoxygenase (LOX) participated in the metabolism of PU-NAEs through the formation of NAE-oxylipins. The data suggests that Arabidopsis LOXs indeed compete with FAAH to metabolize PU-NAEs during seedling establishment. Identification of endogenous amide-conjugated oxylipins suggests potential significance of these metabolites in vivo, and FAAH mutants may offer opportunities to address this in the future.

Upper lethal temperatures of Northern Bobwhite embryos and the thermal properties of their eggs

This article explores the upper lethal temperature for Bobwhite eggs. Results find that thermal tolerances of Northern Bobwhite embryos are high, indicating an adaptation to the naturally occurring temperature extremes in the Bobwhite's environment. However, the results also indicate that high temperatures could have plausibly contributed to the population decline observed in the Bobwhite's semi-arid range.

In situ cardiac performance of Atlantic cod (Gadus morhua) at cold temperatures: long-term acclimation, acute thermal challenge and the role of adrenaline

This article measures the cardiac performance of Newfoundland Atlantic cod (Gadus morhua). The results: (1) show that maximum performance of the isolated cod heart is not compromised by exposure to cold temperatures; and (2) support data from other studies, which show that, in contrast to salmonids, cod cardiac performance/myocardial contractility is not dependent upon humoral adrenergic stimulation.

RNA helicase DP103 and TAK1: a new connection in cancer

Article on new markers of tumor metastasis. The authors found that expression of DP103 was significantly upregulated in the metastatic basal subtype of breast cancers in 3 independent cohorts. This elevated DP103 expression correlated with metastatic breast cancer gene signatures and was strongly associated with patient survival. This study provides the first evidence that the RNA helicase DEAD-box protein DP103 is an NF-kB target that could form part of a positive feedback loop contributing to DP103- mediated regulation of TAK1 kinase activity on the major NF-kB kinase IKK2, thus implicating DP103 in the maintenance of this oncogenic signaling arm in human cancer.

A novel benzimidazole derivative, MBIC inhibits tumor growth and promotes apoptosis via activation of ROS-dependent JNK signaling pathway in hepatocellular carcinoma

This study further explores a prior screening program carried out using MTT assay that identified a series of novel benzimidazole derivatives, among which Methyl 2-(5-fluoro-2-hydroxyphenyl)-1H-benzo[d]imidazole-5-carboxylate (MBIC) showed highest anticancer efficacy compared to that of chemotherapeutic agent, cisplatin. In this article, the authors find that MBIC inhibited cell viability in different hepatocellular carcinoma (HCC) cell lines without exerting significant cytotoxic effects on normal liver cells. The results of this study demonstrate that MBIC may inhibit cell proliferation via ROS-mediated activation of the JNK signaling cascade in HCC cells.
