MPSA 2004 Final Report

This proposal was to support a portion of the costs of Methods in Protein Structure Analysis 2004 (MPSA2004). MPSA2004 was the 15th in the series of MPSA international conferences on protein structure analysis that began in 1974. MPSA2004 was held on the campus of the University of Washington, Seattle, WA August 29 through September 2, 2004. Twenty-four internationally recognized speakers gave invited presentations; additional participants were chosen to present short talks in the 10 topics that were addressed. The aim of MPSA conferences is to communicate the latest, cutting-edge techniques in protein structure analysis and proteomics through science success stories as told by the scientific leaders who developed the technologies. Biotechnology vendors are present to explain currently available commercial technology through workshops and demonstrations booths. The overall aim is to provide a forum for exchanging the latest methods and ideas in protein structure analysis and proteomics to current and future practitioners. The conference supported the missions of the DOE Office of Science and the Office of Biological and Environmental Research (BER) in at least two ways: by enabling the above forum, and by encouraging young researchers who might otherwise not attend to meet leading researchers and to learn about the …

The BIOMOL grant was for 'Local System Support for PDB Biological Unit Search and Display' to augment Rasmol's [Bernstein 2000] [Sayle, Milner-White 1995] existing macromolecular display functions with new capabilities by taking advantage of recent increases in local computing power in order to move functionality that is now scattered among various local and remote systems into one local package. Work included new algorithms for molecular surface display, an extended format for Protein Data Bank Entries, work on issues relating to the integration of multiple diffraction images formats.

GFOC Project results: High Temperature / High Pressure, Hydrogen Tolerant Optical Fiber

Tests results are given for exposure of multimode optical fiber to high temperatures (300 deg. C) and high partial pressure (15 bar) hydrogen. These results demonstrate that fluorine down doped optical fibers are much more hydrogen tolerant than traditional germanium doped multimode optical fibers. Also demonstrated is the similar hydrogen tolerance of carbon coated and non-carbon coated fibers. Model for reversible H2 impact in fiber versus T{sup o}C and H2 pressure is given. These results have significant impact for the longevity of use for distributed temperature sensing applications in harsh environments such as geothermal wells.

Nano Structured Activated Carbon for Hydrogen Storge

Development of a nanostructured synthetic carbons materials that have been synthesized by thermal-decomposition of aromatic rich polyether such as poly(ether ether ketone) (PEEK) is reported. These polymers based nanostructured carbons efficacious for gas adsorption and storage and have Brunauer-Emmett-Teller (BET) surface area of more than 3000 m2/g, and with average pore diameter of < 2nm. Surface-area, pore characteristics, and other critical variables for selecting porous materials of high gas adsorption capacities are presented. Analysis of the fragments evolved under various carbonization temperatures, and the correlation between the activation and carbonization temperatures provides a mechanistic perspective of the pore evolution during activation. Correlations between gas (N2 and H2) adsorption capacity and porous texture of the materials have been established. The materials possess excellent hydrogen storage properties, with hydrogen storage capacity up to 7.4 wt% (gravimetric) and ~ 45 g H2 L-1 (volumetric) at -196oC and 6.0 MPa.

Interpretation of Flow Logs from Nevada Test Site Boreholes to Estimate Hydraulic conductivity Using Numerical Simulations Constrained by Single-Well Aquifer Tests

Hydraulic conductivities of volcanic and carbonate lithologic units at the Nevada Test Site were estimated from flow logs and aquifer-test data. Borehole flow and drawdown were integrated and interpreted using a radial, axisymmetric flow model, AnalyzeHOLE. This integrated approach is used because complex well completions and heterogeneous aquifers and confining units produce vertical flow in the annular space and aquifers adjacent to the wellbore. AnalyzeHOLE simulates vertical flow, in addition to horizontal flow, which accounts for converging flow toward screen ends and diverging flow toward transmissive intervals. Simulated aquifers and confining units uniformly are subdivided by depth into intervals in which the hydraulic conductivity is estimated with the Parameter ESTimation (PEST) software. Between 50 and 150 hydraulic-conductivity parameters were estimated by minimizing weighted differences between simulated and measured flow and drawdown. Transmissivity estimates from single-well or multiple-well aquifer tests were used to constrain estimates of hydraulic conductivity. The distribution of hydraulic conductivity within each lithology had a minimum variance because estimates were constrained with Tikhonov regularization. AnalyzeHOLE simulated hydraulic-conductivity estimates for lithologic units across screened and cased intervals are as much as 100 times less than those estimated using proportional flow-log analyses applied across screened intervals only. Method and Apparatus for In-Process Sensing of Manufacturing Quality

A method for determining the quality of an examined weld joint comprising the steps of providing acoustical data from the examined weld joint, and performing a neural network operation on the acoustical data determine the quality of the examined weld joint produced by a friction weld process. The neural network may be trained by the steps of providing acoustical data and observable data from at least one test weld joint, and training the neural network based on the acoustical data and observable data to form a trained neural network so that the trained neural network is capable of determining the quality of a examined weld joint based on acoustical data from the examined weld joint. In addition, an apparatus having a housing, acoustical sensors mounted therein, and means for mounting the housing on a friction weld device so that the acoustical sensors do not contact the weld joint. The apparatus may sample the acoustical data necessary for the neural network to determine the quality of a weld joint.

Policy Analysis of Produced Water Issues Associated With in-Situ Thermal Technologies

Commercial scale oil shale and oil sands development will require water, the amount of which will depend on the technologies adopted and the scale of development that occurs. Water in oil shale and oil sands country is already in scarce supply, and because of the arid nature of the region and limitations on water consumption imposed by interstate compacts and the Endangered Species Act, the State of Utah normally does not issue new water rights in oil shale or oil sands rich areas. Prospective oil shale and oil sands developers that do not already hold adequate water rights can acquire water rights from willing sellers, but large and secure water supplies may be difficult and expensive to acquire, driving oil shale and oil sands developers to seek alternative sources of supply. Produced water is one such potential source of supply. When oil and gas are developed, operators often encounter ground water that must be removed and disposed of to facilitate hydrocarbon extraction. Water produced through mineral extraction was traditionally poor in quality and treated as a waste product rather than a valuable resource. A light reflection technique suitable for development into an automated surface quality certification system was investigated to determine if reflected light intensity distributions could be corre]ated with surface defect depths. Reflected laser light intensity distributions from pit and scratch defects on highly reflective spheres were studied with a commercial multi-element photodetector. It was found that the intensity distributions --Goll&#x27;lf be correlated with depths of pits and scratches in a size range of concern on highly reflective&#x27; spheres.
