Metabolic Design and Control for Production in Prokaryotes

Prokaryotic life on earth is manifested by its diversity and omnipresence. These microbes serve as natural sources of a large variety of compounds with the potential to serve the ever growing, medicinal, chemical and transportation needs of the human population. However, commercially viable production of these compounds can be realized only through significant improvement of the native production capacity of natural isolates. The most favorable way to achieve this goal is through the genetic manipulation of metabolic pathways that direct the production of these molecules. While random mutagenesis and screening have dominated the industrial production of such compounds in the past our increased understanding of microbial physiology over the last five decades has shifted this trend towards rational approaches for metabolic design. Major drivers of this trend include recombinant DNA technology, high throughput characterization of macromolecular cellular components, quantitative modeling for metabolic engine ring, targeted combinatorial engineering and synthetic biology. In this chapter we track the evolution of microbial engineering technologies from the black box era of random mutagenesis to the science and engineering-driven era of metabolic design.

Influence Of Coal Combustion Flue Gas Desulfurization Waste On Element Uptake By Maize (Zea Mays L.)

A greenhouse study was conducted to determine the effect of coal combustion flue gas desulfurization (FGD) waste from a coal combustion electric power facility on element uptake by maize (Zea mays L.). Unweathered FGD was applied to an Orangeburg Series (Typic Paleudult) soil with an initial soil pH salt of 4.90. The FGD was added at 0, 1, 2, 4, 6, 8, and 10 per cent by weight. The test plant, maize, was harvested after 6 weeks of growth. Within 56 days of the FGD application, all rates of FGD significantly increased pH in the soil and the soil leachate above 6.0. The elemental concentration of the maize tissues indicated a characteristic elevation of B, Se, Mo, and As. However, no visual symptoms of toxicity of B or other elements in plants were observed. Increasing level of FGD caused a steady decline in dry weight, with the highest treatment producing plants which had approximately half the biomass of the control plants. Due to elevated concentrations of B and other elements and due to adverse yield effects measured on plants, unweathered FGD would not be a suitable amendment for 6-week old maize on this soil.

Measurement of Simulated Waste Glass Viscosity

A new high-temperature glass viscometer instrument was established and evaluated using a simulated waste glass in a comparative test with eight other laboratories and viscometers. The unit has distinct advantages in physical size, the amount of glass required for testing, and the simplicity of operation. These advantages can be important for work with radioactive materials. Results from the comparison indicate excellent accuracy and repeatability.

SOME PHYSICAL AND ENGINEERING ASPECTS OF HIGH CURRENT EBIS.

Some applications of an Electron Beam Ion Source (EBIS) require intensities of highly charged ions significantly greater than those which have been achieved in present EBIS sources. For example, the ion source for the Relativistic Heavy Ion Collider (RHIC) at Brookhaven National Laboratory (BNL) must be capable of generating 3x10{sup 9} ions of Au{sup 35+} or 2 x 10{sup 9} ions of U{sup 45+} per pulse [1]. In this case, if the fraction of ions of interest is 20% of the total ion space charge, the total extracted charge is {approx}{approx} 5 x 10{sup 11}. It is also desirable to extract these ions in a 10 ps pulse to allow single turn injection into the first synchrotron. Requirements for an EBIS which could meet the needs of the LHC at CERN are similar ({approx} 1.5 x 10{sup 9} ions of Pb{sup 54+} in 5.5 {micro}s). This charge yield is about an order of magnitude greater than that achieved in existing EBIS sources, and is what is meant here by ''high current''. The Carbelle prototype house is a new design produced under the U.S. Department of Energy's Building America program. Working with other members of the Consortium for Advanced Residential Buildings, Mitchell Homes developed the Carbelle as an energy-efficient upgrade to one of their standard models. By treating all design aspects of the house as a system and involving all stakeholders in the process, Mitchell expects to decrease on-site energy use for space heating and cooling by as much as 40% compared to their typical construction.
