The Genetics of Pigmentation in Corynebacterium poinsettiae ATCC 9682

Corynebacterium poinsettiae mutant strains blocked in carotenoid biosynthesis were obtained by treatment with the mutagen N-methyl-N1-nitro-N-nitrosoguanidine. Additional carotenoid (Crt) mutant strains were obtained from a previous study conducted in our laboratory. Fifty-nine Crt mutants affected in carotenoid biosynthesis were examined by a normal phase high performance liquid chromatography (HPLC) system. Pigment extracts of Crt mutants and C. poinsettiae wild type strains were resolved by an isocratic system with hexane:acetone:dicholoromethane, 11.35:1.73:1.00 (by vol.) as the eluting solvent. In addition to the five major peaks, twelve minor peaks were observed in the wild type C. poinsettiae strain used in this study. Crt mutant and wild type strain peak heights were measured from the individual chromatograms and the peak height data set created was analyzed using the Statistical Analysis System program to perform a cluster analysis. The cluster analysis revealed five carotenoid mutant groups. Carotenoid pigments which accumulated or were absent in each of the cluster groups are reported. Cluster group 1 mutants (CrtA) are blocked in the dehydrogenase(s) which is(are) responsible for the dehydrogenations between phytoene and lycopene. Imagery Technology: Effects on a Chronic Pain Population

The effects of a computer program (Health Imagery Technology Systems, HITS) designed to promote attitude and cognitive changes through elicitation of evoked response potentials were evaluated with chronic pain patients. A treatment and control group were used for comparison (52 patients, 22 females, 32 males, mean ages 47). Wechsler Adult Intelligence Scale-Revised subtests, a Semantic Differential scale, the Health Attribution Test, an imagery protocol, the McCoy-Lawlis Pain Drawing, and the Zung Depression scale were used at admission and discharge to measure change. A pre- post-mood thermometer was used with the treatment group. The hypotheses that the treatment group would show significant changes on these measures were tested with a two group repeated measures analysis of variance design. No significant changes were noted for either group on the intellectual measures, on health attitudes, or reports of pain. The similarities subscale showed significant within group variance (F = 5.46, p < .023). One bipolar adjective pair indicated significant differences (F = 4.79, p < .035), possibly a result of chance. One of seven imagery measures suggested a significant improvement in strength of imagery for the treatment group (F = 18.2, p < .00008). This study examines nursing students' adoption of inferential sets in a clinical situation. The investigation determines (1) the particular inferential set(s) nursing students adopt toward a patient in a clinical situation; (2) the particular inferential set(s) adopted by sophomore and senior nursing students in a clinical situation; and (3) whether or not inferential sets adopted by the sophomore and senior nursing students differ. Sophomore and senior nursing students at a woman's university in Texas were asked to complete a research tool designed to determine inferential set adoption.

An Interpretive and Postulational Model for Perception and Adoption of Innovation

The problem with which this research is dealing is the lack of any explanatory model which explains both the perception and the adoption of new products. One objective of this study is to advance a new conceptual framework concerning both the perception and the adoption of new products. The second objective of this study is to evaluate this new framework theoretically and empirically. Bunge's evaluative criteria are used to evaluate the new model theoretically while Hunter, Schmidt, and Jackson's meta-analysis technique is used to evaluate the model empirically. An extensive review of literature pertaining to the definition of innovation, the adoption process, and innovativeness is included in the second chapter. Chapter three covers research plan and methods. The new model and its assumptions are presented in chapter four. The results of both theoretical and empirical investigations of the new model are reported in chapter five. Finally, chapter six includes a discussion of the main findings and provides some suggestions for future research. An interpretive and postulational model is introduced in this study. The model is built on three main assumptions and contains thirty-one different theoretical constructs. Those constructs are bounded together by forty-six theoretical propositions. The Missae De Beata Virgine C. 1500-1520: A Study of Transformation From Monophonic to Polyphonic Modality

While musical sources and documents from throughout the Middle Ages reveal that mode was an enduring and consciously derived trait of monophonic chant, modality in later polyphony shares neither the historical span nor the theoretical clarity of its monophonic counterpart. Modern theorists are left with little more than circumstantial evidence of the early development of modality in polyphony. This study attempts to shed light on the problem by detailed analysis of a select body of paraphrase masses from the early sixteenth century. First, it correlates the correspondence between the paraphrased voice and the original chant, establishing points of observation that become the basis of melodic analysis. Then, these points are correlated with known rules of counterpoint. Exceptions are identified and examined for their potential to place emphasis on individual mode-defining pitches. A set of tools is derived for quantifying the relative strength of cadential actions. Levels of cadence are defined, ranging from full, structural cadences to surfacelevel accentuations of individual pitches by sixth-to-octave dyadic motions. These cadence levels are traced through the Missae de beata virqine repertoire from c. 1500-1520, a repertoire that includes masses of Josquin, Brumel, La Rue, Isaac, and Rener. This investigation traces the history and development of music theory in Mexico from the date of the first Mexican treatise available (1776) to the early second half of the nineteenth century (1866). This period of ninety years represents an era of special importance in the development of music theory in Mexico. It was during this time that the old modal system was finally abandoned in favor of the new tonal system and that Mexican authors began to pen music treatises which could be favorably compared with the imported European treatises which were the only authoritative source of instruction for serious musicians in Mexico.

A Neglected Clarinet Concerto by Ludwig August Lebrun: A Performing Edition with Critical Commentary: A Lecture Recital, Together with Three Other Recitals

The present study makes available a modern performing edition of an eighteenth-centyry clarinet concerto. Written by the Mannheim oboist and composer Ludwig August Lebrun, the Concerto in B-flat for solo clarinet and orchestra has existed solely as a set of manuscript parts for over 200 years. The following chapters present biographical information on Ludwig August Lebrun as an oboist and composer of the late eighteenth century, the historical background of Lebrun's Concerto in B-flat. a thematic and harmonic analysis of the concerto's three movements, and a summary of the procedures followed in preparing the present edition of orchestral parts and piano reduction. Contemporaneous sources which provided pertinent performance practice information in the areas of articulation and ornamentation are also discussed. A copy of the piano reduction and orchestral performing parts are included in the appendices.

Orlando di Lasso's Missa Ad Imitationem Moduli Doulce Memoire: An Examination of the Mass and its Model

Orlando di Lasso is regarded as one of the great polyphonic masters of the Renaissance. An international composer of both sacred and secular music, his sacred works have always held an important place in the choral repertory. Especially significant are Lasso's Parody Masses, which comprise the majority of settings in this genre. The "Missa Ad Imitatiomem Moduli Doulce Memoire" and its model, the chanson "Doulce Memoire" by Sandrin, have been selected as the subject of this lecture recital. In the course of this study, the two works have been compared and analyzed, focusing on the exact material which has been borrowed from the chanson. In addition to the borrowed material, the longer movements, especially the Gloria and the Credo, exhibit considerable free material. This will be considered in light of its relation to the parody sections. Chapter One gives an introduction to the subject of musical parody with definitions of parody by several contemporary authors. In addition, several writers of the sixteenth century, including Vicentino, Zarlino, Ponzio, and Cerone are mentioned. The Perceptions of Public School Principals in the State of Texas Concerning Selected Court Decisions

The problem of this study was to determine the extent of agreement or disagreement by public school principals in the State of Texas with fifty selected court decisions. The population of the study consisted of all the public school principals in the State of Texas. From this population, a subject of 600 potential respondents was chosen for participation in the study upon the recommendation of the doctoral advisory committee using a random sampling technique. The instrument used in this study was a law questionnaire that was developed in conjunction with Dr. Roosevelt Washington of North Texas State University. The first part of the law questionnaire requested background information from the respondents which was used for the independent variables of the study. The second part of the law questionnaire contained fifty summarized court decisions to which principals were asked to indicate their level of agreement or disagreement.

Prediction of Business Failure as a Criterion for Evaluating the Usefulness of Alternative Accounting Measures

This study examines the usefulness of general price level information (GPL) and current cost information (CC) originally provided by SFAS No. 33 as compared to historical cost information (HC) in predicting bankruptcy. The study also examines the usefulness of GPL data versus CC data when each supplements HC data. In addition, this study tests the usefulness of the three types of information systems combined in one model (HC, GPL, and CC) versus HC data in predicting bankruptcy. The study focuses on the predictability of business failure using financial ratios as predictors. A comparison of these predictors is made in order to identify the accounting system that yields a better prediction of bankruptcy. Two multivariate statistical techniques, multiple discriminant analysis (MDA) and logistic regression analysis (LRA), are used to derive the ex—post classification and the ex-ante prediction results. Six functions are developed, based on ratios computed with HC, CC, GPL, the combined HC and GPL, the combined HC and CC, and the combined HC, GPL, and CC. The resulting functions are used to classify 40 firms as failed or nonfailed. The analysis is repeated for three time bases—one, two, and three years before bankruptcy. Pretherapy Religious Value Information its Influence on Stated Perceptions of and Willingness to See a Counselor

This study sought to determine the influence of pretherapy religious value information upon potential clients' (a) perceptions of a counselor, (b) willingness to see a counselor and (c) confidence of counselor helpfulness. Two hundred and ten undergraduate college students volunteered for the study. Subjects were randomly assigned to one of three treatment groups and given varying amounts and types of written information about a counselor. Group 1 received just the counselor's credentials. Group 2 received the same information plus statements about the counselor's beliefs about counseling and his therapeutic approach. Group 3 received the same information as group 2 plus a statement of the counselor's religious values. Subjects then viewed a short video tape of the counselor in a counseling session. Results of statistical treatment of dependent variables indicated that subjects' perceptions of the counselor, willingness to see the counselor, and confidence of counselor helpfulness were not influenced by the written information, including the statement of religious values that the subjects received before viewing the video tape of the counselor. Implications and recommendations for further research are discussed.

A Quasi-Experimental Study of Behavior in the Professional Negotiation Process: An Analysis of the Nigerian Setting

The problem investigated by this study was that of understanding types of behaviors exhibited by participants in negotiations processes and impact of behaviors on collective bargaining in Nigeria. The study's three purposes were to describe the nature and extent of interpersonal conflict that occurs in collective bargaining, to determine the consequences that stem from such conflict, and to suggest the behaviors and performances during the face-to-face negotiations that should exist to enhance labor-management relations in Nigerian public education. This study examined behavior in negotiations by using simulation, i.e., a quasiexperimental method. Four outcomes of negotiations--time required to reach agreement, terms of agreements, verbal behavior exhibited during negotiations, and satisfaction derived by negotiators in negotiations—were examined.

Questions Used by Teachers with Skilled and Less Skilled Readers

This study described the way teachers used questions with skilled and less skilled readers during reading instruction. The cognitive level and functions of questions were analyzed based on data collected through direct observation within the natural environment of the classroom. In addition, the patterns of questioning which included wait-time and sequencing of questions were identified and reported. Twenty sixth grade teachers randomly selected from a metropolitan school district were observed while instructing skilled readers and less skilled readers. Data collected during non-participatory observation of reading instruction through audiotape recordings, a low-inference observation instrument, and field notes were analyzed using the chisquare statistic, log-linear analysis, and descriptive statistics. Each question/response/response loop which occurred during the eighty observations was analyzed as to the cognitive level and function of the question, designation and wait-time of the student's response, the appropriateness, type, and length of the student's response, and the content of the teacher's response. Within the limitations of this study, the following conclusions have been formulated. 1. Teachers use different cognitive levels of questions for particular functions as dictated by the specific needs and characteristics of the students in the skill level. 2. Radial Compression High Performance Liquid Chromatography as a Tool for The Measurement of Endogenous Nucleotides in Bacteria

High performance liquid chromatography was used to measure ribonucleoside triphosphates in microbial samples. Anion exchange columns in a radial compression module were used to separate and quantify purine and pyrimidine ribonucleotides. Endogenous ribonucleoside triphosphates were extracted from Escherichia coli and pseudomonas aeruginosa using three different solvents, namely trifluorocetic acid (TFA; 0.5M), trichloroacetic acid (TCA; 6 per cent w/v) and formic acid (1.0M) Extracts were assayed for uridine 5'-triphosphate (ATP), and guanosine 5'-triphosphate (GTP) by using anion exchange radial compression high performance (pressure) liquid chromatography. The three extraction produres were compared for yield of triphosphates. E. coli, the TFA extraction procedure was more sensitive and reliable than TCA and formic acid extraction procedures, but , in P. aeruginosa, the best yields of ATP and GTP were obrained following extraction with TFA. Yields of UTP and CTP increased when extraction was performed in TCA. These data illustrate that different extraction produres produce different measures for different triphosphates, a point often overlooked.

The Relationship of Continuing Professional Education and Pastoral Tenure Among Southern Baptist Pastors

This study examined the relationship between the continuing professional education practices of Southern Baptist pastors and their tenure in their pastoral positions. A survey questionnaire was designed to collect data about the pastors' continuing education and pastoral tenure. A stratified sample of 500 pastors was selected from the 36,235 Southern Baptist churches according to church membership. A total of 217 (43 percent) of the questionnaires was returned and used in the data analysis. Thirty percent of the pastors reported spending no days in continuing education during the past year. Sixty-two percent spent five days or more in continuing professional education. Tenure was not significantly linked to continuing education among the pastors. A personal perception of ministerial competency was positively related to continuing education. The majority of pastors felt accountable to God and themselves in terms of their competency. They strongly resisted any move toward certification or accreditation. The majority of the pastors did not view mobility as an alternative to becoming involved in continuing education. However, the reasons reported for mobility could be interpreted as needs assessment addressed by professional growth. The purpose of this study was to determine whether or not the introduction of a regular librarian-centered reading guidance program as an integral part of the entire school program would improve the reading attitudes and habits of elementary school students and increase the reading achievement scores on a standardized test of elementary school students. In addition, the reading attitudes of students were compared with reading achievement scores to assess any relationship between the two.

The problem of this study was the roles of the general assembly agencies of the Presbyterian Church in the U.S.A., the Presbyterian Church in the U.S., and the United Presbyterian Church of North America in the development of Barber- Scotia College, Knoxville College, Johnson C. Smith University, Stillman College, and Mary Holmes College. The historical records of these three churches for the period from 1866 to 1983 were examined to analyze the factors surrounding the establishment of the five colleges, the differences and similarities in the administrative practices of the general assembly agencies charged with operating the colleges, the relationships of the colleges to the churches in the transition from dependent mission schools to independent colleges, and to identify way in which the Presbyterian Church (U.S.A.) may improve its support of Black higher education. The Presbyterian Churches established the mission schools to meet the religious, educational, and economical needs of the emancipated Black slaves. Though the three 2 churches had differences over the issues of slavery and doctrine, the administrative systems developed for the operations of the schools were very similar. Silenes and Silenoids in the Chemistry of Cyclopentadienylsilanes

Evidence is presented that apparent silene products obtained from the metalation of cyclopentadienyldimethyl - chlorosilane either with tert-butyl1ithium or with methylenetriphenylphosphorane actually arise from the metalated starting material, a silenoid, rather than from a silafulvene intermediate. Trimethylmethoxysi1ane is shown to be an effective trap for dimethylsilafulvene. A new dimethylsilafulvene precursor, bis(dimethylmethoxysi1yl) cyclopentadiene, which gives high yields of dimethyldimethoxysi1ane and the silafulvene at temperatures as low as 240°C is reported.

A Study of the Demographic Status, Actual Role and Ideal Role of the Elementary School Assistant Principal in Texas

This study analyzes the status and job functions of the 546 elementary school assistant principals in Texas during the 1985-1986 school year. It is concerned with the status of the position and the degree of responsibility assigned to the assistant principal for forty-eight specific job functions in the actual and ideal practice. A sample of 125 assistant principals, 125 supervising principals and a population of 135 superintendents whose districts employ assistant principals were included in the study. All three groups completed the role survey instrument. The results were analyzed by using the one-sample chi-square test to determine whether significant differences existed among the perceptions of the superintendents, principals, and assistant principals at the .01 level. The return rate was 60.8 percent for the assistant principals and principals and 84.6 percent for the superintendents.

A Study of the Job Satisfaction of Female Principals and Vice-Principals in Texas Public Schools

This study is designed to determine the degree to which female principals and vice-principals in Texas derive job satisfaction from their work and to identify those factors, both negative and positive, which affect the job satisfaction of these principals and vice-principals. A single questionnaire was used to collect the data for this study. Usable questionnaires were returned by 331 vice-principals and 504 principals. These represented 336 school districts throughout the state. Respondents were compared as to their view of their overall job satisfaction, the importance assigned to intrinsic and extrinsic job facets, their satisfaction with intrinsic and extrinsic job facets, and the differences in facet satisfaction connected with various personal characteristics.
