Forest Landscape Dynamics: a Semi-Markov Modeling Approach

A transition model (MOSAIC) is used to describe forest dynamics at the landscape scale. The model uses a semi-Markov framework by considering transition probabilities and Erlang distributed holding times in each transition. Parameters for the transition model are derived from a gap model (ZELIG). This procedure ensures conceptual consistency of the landscape model with the fine scale ecological detail represented by the forest gap model. Spatial heterogeneity in the transition model is driven by maps of terrain with characteristics contained in a Geographic Information System (GIS) database. The results of the transition model simulations, percent cover forest type maps, are exported to grid-maps in the GIS. These cover type maps can be classified and used to describe forest dynamics using landscape statistics metrics. The linkage model-GIS enhances the transition model spatial analytical capabilities. A parameterization algorithm was developed that takes as input gap model tracer files which contain the percent occupation of each cover type through time. As output, the algorithm produces a file that contains the parameter values needed for MOSAIC for each one of the possible transitions. Managing Cattail (Typha latifolia) Growth in Wetland Systems

Nutrient availability, water depth, competition, and soil management effects on cattail (Typha latifolia) growth in wetland systems were examined. Soluble reactive phosphorous (SRP), nitrate-nitrogen (NO3-N), and ammonia-nitrogen (NH3-N) removals were tested at a constructed wetland receiving municipal wastewater effluent. Over all, no significant differences in nutrients occurred between diverse planted and cattail areas. T. latifolia seeds, under the canopy of Eleochoris macrostachya, had low seed germination. Established stands of emergent vegetation can prevent cattail colonization and spread. Germination of T. latifolia at various water depths was tested, and depth impacts on cattail seedling growth and survival were ascertained using various moist soil management techniques in three ponds. Water levels at 0cm and >40cm can adversely impact cattail establishment.

Phosphorus Retention and Fractionation in Masonry Sand and Light Weight Expanded Shale Used as Substrate in a Subsurface Flow Wetland

Constructed wetlands are considered an inefficient technology for long-term phosphorus (P) removal. The P retention effectiveness of subsurface wetlands can be improved by using appropriate substrates. The objectives of this study were to: (i) use sorption isotherms to estimate the P sorption capacity of the two materials, masonry sand and light weight expanded shale; (ii) describe dissolved P removal in small (2.7 m3) subsurface flow wetlands; (iii) quantify the forms of P retained by the substrates in the pilot cells; and (iv) use resulting data to assess the technical and economic feasibility of the most promising system to remove P. The P sorption capacity of masonry sand and expanded shale, as determined with Langmuir isotherms, was 60 mg/kg and 971 mg/kg respectively. In the pilot cells receiving secondarily treated wastewater, cells containing expanded shale retained a greater proportion of the incoming P (50.8 percent) than cells containing masonry sand (14.5 percent). After a year of operation, samples were analyzed for total P (TP) and total inorganic P (TIP). Subsamples were fractionated into labile-P, Fe+Al-bound P, humic-P, Ca+Mg-bound P, and residual-P. The objectives of this project were to measure changes in frequency of occurrence of submerged macrophytes over the first two growing seasons following stocking with triploid grass carp at two fish per acre and to measure differences in macrophyte biomass between areas excluded from herbivory and adjacent control sites after 16 months following establishment of exclosures. The project also seeks to measure concentrations of fluridone following the herbicide treatment, and to compare two methods of aquatic vegetation sampling.

General Biology Lecture and Laboratory Curriculum Outline in a Two or Four-Year College

In July of 1999, I wrote to 24 Texas junior and community colleges (and one four-year institution) describing my thesis agenda of a general biology lecture and laboratory syllabus for introductory biology students. I requested the titles and authors of the general biology textbooks and laboratory manuals they were currently using, the publishers of these texts, and the edition of said texts. I then contacted publishers of the various textbooks who, in turn, directed me to the Dallas-area representatives for further inquiries. I assimilated the various authors' general biology topics into a two-semester syllabus of lecture and one semester of laboratory. The document is not a text manuscript, but an all-inclusive listing of a general biology syllabus broken down by subject.

Development of an In Vitro Protoplast Culture System for Albizia Lebek (L.) Benth., an Economically Important Leguminous Tree

An in vitro system of generating protoplasts from their callus cultures was established. The friable callus was more productive in terms of producing protoplasts than the green compact callus. The concentration of the various cell wall degrading enzymes had an effect on the viability of the protoplasts in the medium. The protoplast system developed from the experiments was stable and could be used for the transformation experiments of Albizia lebek and for other plant improvement practices.

The Role of Rainfed Farm Ponds in Sustaining Agriculture and Soil Conservation in the Dry High Valley Region of Cochabamba, Bolivia: Design Considerations and Post Impoundment Analysis

Lack of sufficient water for irrigation is a major problem in and around the valleys surrounding the town of Aiquile, Cochabamba Bolivia. In addition, much of the region is undergoing desertification compounded by drought, deforestation, bad traditional agricultural practices, over grazing and a "torrential" rainfall pattern leading to severe soil erosion and low agricultural production. Between 1992 and 1994, the author constructed a network of 24 small, mostly rainfed farm ponds to increase agricultural production and alleviate soil erosion and land-use problems by improving cover conditions. A 5-year post-impoundment analysis was carried out in 1998. The analysis examined current pond conditions, design criteria, irrigation water / crop production increases and the alleviation of land-use problems. Current pond conditions fell into four distinct categories with only 25 percent of the ponds being deemed as "functioning well." The project increased irrigation in the region and improved cover conditions in 66 percent of the pond sites.

The Isolation and Characterization of a Hitherto Undescribed Gram-Negative Bacterium

A unique undosciribed gramnegative rod is extensively characterized in this study. The cells of this unusual water isolate measure 1.2 X 6.5 microns, The most distinguishing characteristic of the bacterium is a polar tuft of 35-40 flagella that aggregate to function as a single organelle which is visible under phase contrast. Aging cells deposit poly- -hydroxybutyric acid granules which are bound by an inclusion membrane made up of four distinct layers. It also possesses an unusual exterior membrane outside the cell wall which contains large fibrils of protein running at a slight angle to the longitudinal axis of the cell. The guanosine-cytosine ratio was found to be 62.2$. The organism's taxonomic position was further investigated by immunological, morphological, and biochemical methods. It was found to be most closely akin to members of the genus Pseudo onas, although somewhat divergent from other species classified in this genus. After careful evaluation of the findings obtained during this study, the new bacterium was subsequently named Pseudomonas multiflagella.

Opthalmic Use Of Sodium Cephalothin: An In Vivo Comparison

A rabbit keratoconjunctivities model was used to evaluate ophthalmic formulations containing 1 percent sodium cephalothin in silicon oil, a 1 percent sodium cephalothin aqueous solution, and a 0.3 percent gentamicin sulfate solution. Rabit eyes were inoculated intracorneally with Pseudomonas aeruginosa, Staphylococcus aureus, or Streptococcus pneumoniae, After topical treatment, none of the antibiotic formulations were effective in the P. aeruginosa model; all three showed good activity against S. aureus, and against S. pneumoniae, the caphalothin formulations were more effective than gentamicin.In a related stability study, the cephalothin potency of the silicon formulation was maintained for 16 weeks at 4, 25, and 450 C These studies suggest that sodium cephalothin can be formulated as an effective and stable ophthalmic dosage form.

Genetic Variation in a Population of the Plains Woodrat Neotoma micropus

Neotoma micropus from Jack County, Texas, were studied over a 9-month period. Loci from blood and saliva were used to determine genetic variation within the population. Deviations from Hardy-Weinberg equilibrium were found at one locus. The average temporal F over all seven loci was 0.040. Genetic structuring was subtle, fluctuated on a seasonal basis, and was due to differential migration or predation on genotypes. Heterozygotes tended to move more than homozygotes, and a greater proportion of heterozygotes were lost from the population during each season. Genetic variation was maintained in the population by immigrant individuals. This differential in dispersal of genotypes fits current models of reorganization within the genome of populations.

Comparative Toxicity Responses in Earthworms Lumbricus Terrestris and Eisenia Foetida to Cadmium Nitrate and Chlordane Using Artificial Soil and Filter Paper Exposures

This research compares LC50 and LD50 of earthworms, Lumbricus terrestris and Eisenia foetida exposed to cadmium nitrate and chlordane using 48-h contact filter paper (FP) and 14-d artificial soil (AS) protocols. Both LC50 and LD50 showed that chlordane was more toxic than cadmium in both species regardless of the exposure. The reference toxicant 2-chloroacetamide using the standardized 48-h FP exposure was used to assess the general response of the earthworm prior to toxicity experiments. A glucose test was developed as an internal standard to assess homogeneity of mixtures among both replicates and dilutions. Accuracy of dilutions is assessed by the slope of a regression line relating nominal dilution to observed internal standard concentration. Precision of replicate preparation is assessed by among replicate variance.

Estimated Extent and Fate of Chlorinated Solvent Contamination in the Soil of the Naval Air Station, Dallas, Texas

This thesis estimates the spatial extent of chlorinated solvent contamination of the soil at the Naval Air Station, Dallas, then estimates the fate and transport of these contaminants, over time, using the Soil Transport and Fate database and the Vadose-Zone Interactive Processes (VIP) modeling software. Geostatistical analysis identifies two areas with serious chlorinated solvent contamination. Fate and transport modeling estimates that this contamination will degrade and disperse from the soil phase to below regulatory limits within one year, although there is a risk of groundwater contamination. Contaminants are estimated to persist in the water and air phases of the soil. Further sampling is recommended to confirm the results of this study.

A Vegetative Analysis of and Distribution of the Grasses of North Central Texas

Accurate identification is fundamental in any study of grasses by farmers, ranchers, range specialists, ecologists, or students interested in the changes taking place in the grass habitat. Frequently it is convenient, and sometimes it is necessary, to identify the grasses by their vegetative characters. Some are readily recognized at a glance by their habitat and certain characters well known to the experienced observer. In other cases, identification is more difficult; and, perhaps with a few, it is impossible to be certain of the species from vegetative characters. However, this may also be true when the characters of the floral parts alone are considered. The inflorescence, used in most keys and descriptions, is often available only for a short period of time. Identification by the characters of the vegetation can be used throughout the growing season, even if grazing or mowing has removed or prevented the development of the floral parts. There have been other studies of grass identification related to vegetative characters, but they have been local and have not covered North Central Texas. Adenylate Energy Charge Determinations of Soil Bacteria Grown in Soil Extract Medium

The adenylate energy charge values of twenty bacteria isolated from soil and cultured in a medium consisting of soil and distilled water were determined by the luciferin-luciferase bioluminescense method. The purpose of this study was to examine the growth and energy charge values of these organisms in soil extract medium, and to determine what effect the addition of glucose has on their energy charge values. Three of the organisms employed in this study showed energy charge values similar to those reported for bacteria grown in enriched media. The remainder of the isolates demonstrated low energy charge values, and scant growth in the soil medium.

The following is a complete syllabus for a college level genetics course. The syllabus contains lecture outlines and notes for each chapter, along with a list of transparencies needed. The quizzes and exams are prepared and placed at the beginning of the syllabus. The beginning of the course will consist of a lecture to introduce the students to the basics of genetics, followed by many applications of genetics. The process of cell division will be mastered by the students, as well as Mendelian genetics, quantitative genetics, chromosome mapping, and inheritance. The replication, synthesis, and organization of DNA are also discussed within the lectures. The final topics that will be covered using this syllabus are genetics of cancer and immunology and population genetics. These topics are essential for a detailed genetics course. The syllabus is written in great detail, and will require a full semester to be completed. The book used in association with this syllabus is Essentials of Genetics by William S. Klug and Michael R. Cummings.

The Use of Similarity Indicies to Assess the Spatial Differences of the Phytoplankton Communities in Lake Ray Roberts, Texas

A study of the phytoplankton communities of Lake Ray Roberts was conducted at three sampling sites on two sampling dates during the summer of 1995, assessing both community structure and chlorophyll a concentrations. Eight similarity indices were then used to characterize and compare the communities of these sites. Both community similarity and chlorophyll a concentrations were evaluated using a minimum detectable difference equation to determine the amount of change that must occur in these parameters in order to be deemed statistically significant. The Bray-Curtis Index was shown to be the most adequate index evaluated, and was subsequently used in conjunction with bootstrap analysis to determine the similarity between the three sampling sites.

Biological and Toxicological Responses Resulting from Dechlorination of a Major Municipal Wastewater Treatment Plant Discharge to the Trinity River

Federal regulations such as the Clean Water Act (P.L. 92-500), and its amendments, direct the Environment Protection Agency (EPA) to implement programs to control the releases of conventional pollutants and toxics into the waterways of the United States. The EPA began requiring treatment plants to conduct toxicity tests (biomonitoring) of their effluent discharges. To control toxicity caused by chlorination of wastewater discharges, the EPA also began requiring some treatment facilities to dechlorinate their wastewater before discharging. This research was funded by the EPA to document the changes that occurred in the Trinity River from the dechlorination of the effluent from Ft. Worth's Village Creek municipal wastewater treatment plant. The study occurred over a two year period beginning in August 1990. A wide variety of biological field assessments and toxicological assays were used to measure various responses. Seven river stations, covering approximately twenty river miles, and the treatment plant effluent were assessed. Two of the river stations were upstream from the treatment plant and used as reference sites. The remaining five river stations were downstream from the treatment plant, spread out over seventeen river miles. Molecular Cloning and Functional Analysis of Transposable Mercury Resistance Genes Encoded by the OCT Plasmid

Translocation of a 17.1 kilobase region of the OCT plasmid encoding mercury resistance (mer) in Pseudomonas putida was shown to occur in a recombination-deficient host with plasmid PP1 serving as a recipient replicon. The frequency of transposition in Pseudomonas was estimated at 10^3 -10 -^2, but undetectable in Escherichia soli. ' DNA comprising all of mr as well as subregions there of were cloned and subjected to DNA sequence analysis. Like other transposons, mer was found to contain inverted repeat sequences at its termini. These were similar to, but not identical to the inverted repeat structures found in the prototypical mercury resistance transposon Tn501 from E. aeruginosa.

Properties of Normal Rat Kidney Cells Transformed by a Temperature-Sensitive Mutant (LA31) of Rous Sarcoma Virus

The basis of this investigation is to characterize growth property differences in normal versus virally transformed cells. Using a temperature-sensitive mutant of Rous sarcoma virus, the cells' transformation state is regulated by the growth temperature; at 33°C the cells are transformed, while at 39°C the cells have normal characteristics. The morphology of NRK cells is elongated and fibroblastic; when transformed the cells are rounded. Normal cells grow to a monolayer and stop, while transformed cells grow to saturation densities greater than just a monolayer amount. Transformed cells can form foci when grown in mixture with normal cells. Normal cells must be in contact with the culture vessel in order to grow, but transformed cells lack anchorage dependence for growth.

Ethylbromo Acetate as an Olfactory Probe

Olfactory transduction mechanisms are best studied when specific blocking agents are available with which to modify cellular responses to odorant stimulation. This study is an electrophysiological investigation of functional group-specific olfactory acceptor mechanisms using ethylbromo acetate (EtBrAc), a World War I war gas which inhibits olfactory responses to odorant stimulation. The major findings of this investigation show that (1) vaporous EtBrAc is found to be a quick and effective inhibitor of electroolfactogram (EOG) responses to odorant stimulation, (2) isoamyl acetate is shown to protect EOG responses to a wide variety of odorants, inclusive of itself, from the inhibition effect of EtBrAc treatment, and (3) amine acceptor sites exist which are resistant to the inhibitory effects fo EtBrAc treatment.

Studies on the Purification and Phosphorylation of Phosphofructokinase from Ascaris suum

A new procedure has been developed to concentrate the phosphofructokinase from muscle of Ascaris suum with minimum loss of activity. By utilizing this method, 50 ml fraction was concentrated to a final volume of 3 ml in about 1.5 h without loss in enzyme activity. The concentrated enzyme had a specific activity of 64 units per mg. Ascaris muscle-cuticle was incubated in 50 1M solutions of either acetylcholine, serotonin, y-aminobutyric acid, levamisole, or saline alone. Phosphate analysis of the isolated phosphofructokinase from each incubation revealed that the enzyme contained the following moles of phosphate per subunit: 2.9 (acetylcholine), 2.2 (serotonin), 2.0 (y-aminobutyric acid), 1.5 (levamisole), and 3.4 (salne alone). The present study did not establish a direct correlation between degree of phosphorylation and phosphofructokinase activity. Phosphofructokinase from muscle of Ascaris suum appears to contain several phosphorylation sites, and one of these sites is required to be phosphorylated in order for the enzyme to exhibit maximum activity under physiological conditions.

Isolation and Partial Characterization of Pigment Mutants of Corynebacterium poinsettie ATCC 9682

Carotenoid pigments were extracted from Corynebacteriuma poinsettiae (wild type) ATCC 9682, and from 108 mutants obtained by exposure of a streptomycin resistant strain of C. poinsettiae to ultra-violet light irradiation and N-methyl- N'-nitro-N-nitrosoguanidine. The pigments were characterized by their absorption maxima, Rf-values, and partition ratios in petroleum ether and methanol. Thin layer chromatography was used to compare pigments of the wild type with those of the mutants. Possible biosynthetic pathways in carotenoid synthesis of the wild type were postulated on the basis of the observed genetic blocks. Mutants were found which suggested the existence of a linear pathway in carotenoid synthesis from the aliphatic C4 0 molecule to the bi-cyclic C50-diol. Other mutants suggested possible alternative pathways in the biosynthesis of these pigments or the presence of intermediates not detectable by thin layer chromatography.

Effect of Colchicine on Neuronal Excitabilty

The abundance of microtubules in receptive dendrites suggests they may function in sensory transduction. Responses of frog muscle spindle receptors and joint receptors is inhibited within 25 minutes by 50 mM colchicine, a microtubuledisrupting agent. The inhibition is reversible upon removal of colchicine, and the time course of recovery is comparable to that of inhibition. Frog olfactory responses are briefly inhibited by washing the olfactory mucosa with perfusion fluid. Colchicine accentuates the inhibition and substantially retards the rate of recovery in a dose-dependent fashion. Colchicine does not affect axonal conduction, nor the oxygen uptake of isolated crab or frog leg nerves. The inhibitory action of colchicine is therefore an effect on the electrical excitability of the receptive dendrites or soma, and not an effect on axonal conduction.
