The Effects of Long Term Moderate Ethanol Intake on the Immune Response in Rats

Using a rat model, the effects of a single dose or six to twelve months of daily oral administration of ethanol on the immune system were determined. The rats were challenged with sheep red blood cells after the various dosing periods to elicit an immune response. Immune system responsiveness was determined by means of white blood cell counts and differentials, antibody titers, and T-cell numbers. No deleterious effects of the ethanol on the immune response were seen, while the female alcohol-fed rats showed a significant increase in T-Cell numbers, white blood cell counts, and lymphocytes over the sham group.

Molecular Cloning and Functional Analysis of Transposable Mercury Resistance Genes Encoded by the OCT Plasmid

Translocation of a 17.1 kilobase region of the OCT plasmid encoding mercury resistance (mer) in Pseudomonas putida was shown to occur in a recombination-deficient host with plasmid PP1 serving as a recipient replicon. The frequency of transposition in Pseudomonas was estimated at 10^3 -10 -^2, but undetectable in Escherichia soli. ' DNA comprising all of mr as well as subregions there of were cloned and subjected to DNA sequence analysis. Like other transposons, mer was found to contain inverted repeat sequences at its termini. These were similar to, but not identical to the inverted repeat structures found in the prototypical mercury resistance transposon Tn501 from E. aeruginosa.

Microbiological Treatment of Wastewater from a Wood-Preserving Plant

This research investigates interacting biological, chemical, and physical factors affecting the efficiency of microbiological wastewater treatment at the W. J. Smith Wood- Preserving Company in Denison, Texas. The treatment process consisted of collecting exhaust boiler water containing unidentified boiler treatment compounds, steam condensate contaminated with preservatives and wood extracts, plant process waters, and rainfall runoff from plant grounds. With a 5-minute residence time, wastewater was passed over 2 oxidation towers in series, each containing approximately 47,000 square feet of surface area. Suspended solids were removed from the wastewater before discharge. Various amino acids such as serine, aspartate, cysteine, phenylalanine, alanine, proline, glycine, histidine, and tyrosine significantly stimulated phenol degradation in the laboratory. The plant wastewater contained approximately 0.1 mg/l of several of the stimulatory amino acids. It was assumed that these concentrations provided maximal stimulation in the field situation. The plant wastewater also contained sufficient nitrogen to permit the organisms to degrade up to 100 mg phenol/1 of water examined. Amino acids in the wastewater probably serve as a source of microbial nutrition. Toxicity of the wastewater to fish was not caused by the presence of phenol, phenol degradation products, or traces of pentachlorophenol. The wastewater was rendered non-toxic by …

Properties of Normal Rat Kidney Cells Transformed by a Temperature-Sensitive Mutant (LA31) of Rous Sarcoma Virus

The basis of this investigation is to characterize growth property differences in normal versus virally transformed cells. Using a temperature-sensitive mutant of Rous sarcoma virus, the cells' transformation state is regulated by the growth temperature; at 33°C the cells are transformed, while at 39°C the cells have normal characteristics. The morphology of NRK cells is elongated and fibroblastic; when transformed the cells are rounded. Normal cells grow to a monolayer and stop, while transformed cells grow to saturation densities greater than just a monolayer amount. Transformed cells can form foci when grown in mixture with normal cells. Normal cells must be in contact with the culture vessel in order to grow, but transformed cells lack anchorage dependence for growth.

Ethylbromo Acetate as an Olfactory Probe

Olfactory transduction mechanisms are best studied when specific blocking agents are available with which to modify cellular responses to odorant stimulation. This study is an electrophysiological investigation of functional group-specific olfactory acceptor mechanisms using ethylbromo acetate (EtBrAc), a World War I war gas which inhibits olfactory responses to odorant stimulation. The major findings of this investigation show that (1) vaporous EtBrAc is found to be a quick and effective inhibitor of electroolfactogram (EOG) responses to odorant stimulation, (2) isoamyl acetate is shown to protect EOG responses to a wide variety of odorants, inclusive of itself, from the inhibition effect of EtBrAc treatment, and (3) amine acceptor sites exist which are resistant to the inhibitory effects fo EtBrAc treatment.

Studies on the Purification and Phosphorylation of Phosphofructokinase from Ascaris suum

A new procedure has been developed to concentrate the phosphofructokinase from muscle of Ascaris suum with minimum loss of activity. By utilizing this method, 50 ml fraction was concentrated to a final volume of 3 ml in about 1.5 h without loss in enzyme activity. The concentrated enzyme had a specific activity of 64 units per mg. Ascaris muscle-cuticle was incubated in 50 1M solutions of either acetylcholine, serotonin, y-aminobutyric acid, levamisole, or saline alone. Phosphate analysis of the isolated phosphofructokinase from each incubation revealed that the enzyme contained the following moles of phosphate per subunit: 2.9 (acetylcholine), 2.2 (serotonin), 2.0 (y-aminobutyric acid), 1.5 (levamisole), and 3.4 (salne alone). The present study did not establish a direct correlation between degree of phosphorylation and phosphofructokinase activity. Phosphofructokinase from muscle of Ascaris suum appears to contain several phosphorylation sites, and one of these sites is required to be phosphorylated in order for the enzyme to exhibit maximum activity under physiological conditions.

Food Habits of Stoneflies (Plecoptera) in the Gunnison and Dolores Rivers, Colorado

Gut contents of 2,500 stonefly nymphs, comprising 10 species, from the Gunnison and Dolores Rivers, Colorado were examined from Dec., 1974-Oct., 1975. Perlidae species were carnivorous feeding primarily on chironomids, mayflies and caddisflies. Seasonal patterns of ingestion and preference varied among species and predator sizes and between rivers. Early instar polyphagous species utilized detritus in the fall, eventually shifting to carnivorous habits as they grew through winter-spring. Pteronarcids fed predominantly on detritus. Dietary overlap of predators was greatest in the Gunnison River, with subtle mechanisms such as prey species and size selectivity, temporal succession and seasonal shifts to detritus-plant material in some, providing reduction of competition. A more complete partitioning of prey resources was evident in the Dolores River.

Trophic Structure and Energy Flow in a Texas Pond

Annual energy flow and mean annual biocontent of eighteen compartments were determined for a 0.94 ha north central Texas pond ecosystem. Annual primary production was 7,780 kcal m^-2 yr^-2, and community production-to-respiration ratio was 1.49. One-third of annual primary production accumulated on the substrate as silt and sedimentation. Community production, production-respiration ratio, and biocontents of all compartments except aquatic insects were large in summer, small in winter. Biocontents of four trophic levels in the pond were all of the same order of magnitude, approximately 50 kcal m^-2. Suspended and benthic organic material forprimary consumers and terrestrial insects for tertiary consumers were substantial allochthanous energy imports into the pond system.

Drumming Behavior of Selected Nearctic Stoneflies (Plecoptera)

Drumming was recorded for 11 of 13 Nearctic stonefly species, representing 4 families. Both male and female signals were obtained from 5 species, and were either 2-way or 3-way communications. Signals were species-specific; those of males and females varied from 3-39 and 1-14 beats/ signal, respectively. Duration of male signals varied from 105-8,016 ms; those of females, except Perlinella drymo (1 beat), varied from 402-1318 ms. Signals among related taxa showed greatest similarities. Duration of male signals of Perlinella drymo became progressively shorter at each of 4 temperatures from 7-29 0C. Females of Perlinella drymo would only repeatedly answer male signals recorded at near their own temperature, and would not repeatedly answer recorded male signals of 8 other species.

The Effect of Intermediate Filament Inhibitors on Steroidogenesis and Cytoskeleton in Y-1 Mouse Adrenal Tumor Cells

When Y-1 mouse adrenal tumor cells were treated with sodium orthovanadate, an intermediate filament (IF) inhibitor in BHK21-F cells, there was no change in the amount of 20α-dihydroprogesterone produced. A neurofilament inhibitor, β, β'-iminodipropionitrile (IDPN), enhanced the ability of Y-1 cells to produce steroid in response to ACTH by acting on the plasma membrane. Electron microscopy of Y-1 cells extracted with Triton X-100 revealed that both vanadate and IDPN caused the aggregation of cytoskeletal and granular structures in the perinuclear area. The steroidogenic effects of IDPN suggest that the perinuclear aggrergation of cytoskeletal structures may result from the detachment of IF from the plasma membrane, while the reason for the cytoskeletal changes by vanadate is unknown.

Effects of Exogenous Steroids on the Adrenal Plasma Membrane Alteration of Steroidogenesis and Cell Morphology

Using cultured Y-1 mouse adrenal tumor cells which produce the steroid 20(-hydroxypregn-4-en-3-one (20-DHP), it was found that 10-5 M corticosterone and deoxycorticosterone increased basal and inhibited ACTH-induced 20-DHP production. The steroid effects were concentration-dependent, reversible, and specific since six other steroids did not stimulate steroidogenesis and varied in their ability to inhibit ACTH-induced steroidogenesis. Cytochalasin D inhibited steroid-stimulated 20-DHP production, suggesting a mechanism of steroid stimulation similar to that of ACTH. Steroidogenesis stimulated by cholera toxin, (Bu) 2 cAMP, or pregnenolone was not inhibited by exogenous steroid; corticosterone increased basal and inhibited ACTH-induced intracellular cAMP production. Steroids altered cell surface morphology. These findings suggest that steroids alter adrenal steroidogenesis by acting within the plasma membrane.

Isolation and Partial Characterization of Pigment Mutants of Corynebacterium poinsettie ATCC 9682

Carotenoid pigments were extracted from Corynebacteriuma poinsettiae (wild type) ATCC 9682, and from 108 mutants obtained by exposure of a streptomycin resistant strain of C. poinsettiae to ultra-violet light irradiation and N-methyl- N'-nitro-N-nitrosoguanidine. The pigments were characterized by their absorption maxima, Rf-values, and partition ratios in petroleum ether and methanol. Thin layer chromatography was used to compare pigments of the wild type with those of the mutants. Possible biosynthetic pathways in carotenoid synthesis of the wild type were postulated on the basis of the observed genetic blocks. Mutants were found which suggested the existence of a linear pathway in carotenoid synthesis from the aliphatic C4 0 molecule to the bi-cyclic C50-diol. Other mutants suggested possible alternative pathways in the biosynthesis of these pigments or the presence of intermediates not detectable by thin layer chromatography.

Effect of Colchicine on Neuronal Excitabilty

The abundance of microtubules in receptive dendrites suggests they may function in sensory transduction. Responses of frog muscle spindle receptors and joint receptors is inhibited within 25 minutes by 50 mM colchicine, a microtubuledisrupting agent. The inhibition is reversible upon removal of colchicine, and the time course of recovery is comparable to that of inhibition. Frog olfactory responses are briefly inhibited by washing the olfactory mucosa with perfusion fluid. Colchicine accentuates the inhibition and substantially retards the rate of recovery in a dose-dependent fashion. Colchicine does not affect axonal conduction, nor the oxygen uptake of isolated crab or frog leg nerves. The inhibitory action of colchicine is therefore an effect on the electrical excitability of the receptive dendrites or soma, and not an effect on axonal conduction.

Permeability of the Kidney Capillaries to Narrow-Range Macromolecular Dextran Fractions

Recent investigations into the permeability of the kidney capillaries have produced conflicting reports. This study was an attempt to better describe the permeability of the kidney capillaries by using narrow-range macromolecular dextran fractions in four molecular sizes: MW 61,400, MW 77,000, MW 118,000, and MW 147,000. Permeability was measured by dextran concentration differences in plasma and kidney lymph. Permeability decreased as the dextran molecular weight increased. Molecular weights 61,400 and 77,000 penetrated into the kidney lymph. Molecular weight 118,000 exhibited greater difficulty in penetrating to the lymph. The largest fraction penetrated into the kidney lymph with greatest difficulty. Plasma expansion by saline infusion increased the permeability of all dextran fractions.

Immunocompetence in the AKR Mouse

A model for the study of the relationship of immunity to cancer is found in AKR mice which harbor Gross virus. This genetically transmitted virus is present in a latent form for months before it spontaneously induces leukemia. Many investigators have demonstrated near normal humoral responses, but abnormal cellular immunity in the preleukemic animal. With increasing age, pathology of the disease is expressed, reflecting diminished immunity. In this study, the ontogeny of humoral antibodies of AKR/J and SWR/J mice was assayed by microagglutination techniques in response to thymus-independent, thymus-dependent, and solubilized antigens. Simultaneous injections of thymusdependent and -independent antigens provided data suggesting an impaired humoral response in the AKR mouse.

Failure of Azotobacter Vinelandii to Fix Nitrogen in Soil

It was found that Azotobacter vinelandii grown in a dialyzed soil medium did not fix nitrogen and had a much lower rate of respiration than when grown in Burk's nitrogen-free medium. When para-hydroxybenzoic acid served as the added oxidizable organic carbon source in dialyzed soil medium, the azotobacter grown in it were found to be unable to fix nitrogen. On the other hand, A. vinelandii fixed nitrogen when grown in soil supplemented with glucose. It was concluded that natural conditions in the soil are not conducive to nitrogen fixation by A. vinelandii.

Seasonal and Spatial Variability of the Microcrustacean Community in Lake Texoma, Texas and Oklahoma

Twenty-eight species of zooplankton were identified from Lake Texoma. Seasonal density of the overall microcrustacean community and seasonal cycles of individual species were compared with northern populations and any available literature from the Southwest. Cycles of occurrence and abundance were similar to those observed in northern populations but tended to occur earlier in the year due to higher temperatures. Spatial distributions within the reservoir were heavily influenced by nutrient and salt input from the Red River, which resulted in dense populations in the Red River Arm. In addition, during the summer, the microcrustacean community was restricted to the epilimnion due to anoxic conditions in the hypolimnion of the reservoir.

Stream water quality corridor assessment and management using spatial analysis techniques: Introduction, evaluation, and implementation of the WQCM model.

The rapid development of once-rural landscapes often produces detrimental effects on surface water quality entering local reservoirs through vulnerable stream channels. This study presents a methodology that incorporates geographic information systems (GIS) and remote sensing techniques for the creation of a stream corridor evaluation mechanism, coined the water quality corridor management (WQCM) model. Specifically, the study focuses on determining the viability of the WQCM model in assessing the stream corridor conditions within a northern Denton County pilot study region. These results will aid in the prediction and evaluation of the quality of stream water entering reservoirs that serve as the primary drinking water source for local municipalities.

Application of Cultured Neuronal Networks for Use as Biological Sensors in Water Toxicology and Lipid Signaling.

This dissertation research explored the capabilities of neuronal networks grown on substrate integrated microelectrode arrays in vitro to be applied to toxicological research and lipid signaling. Chapter 1 details the effects of chlorine on neuronal network spontaneous electrical activity and pharmacological sensitivity. This study demonstrates that neuronal networks can maintain baseline spontaneous activity, and respond normally to pharmacological manipulations in the present of three times the chlorine present in drinking water. The findings suggest that neuronal networks may be used as biological sensors to monitor the quality of water and the presence of novel toxicants that cannot be detected by conventional sensors. Chapter 2 details the neuromodulatory effects of N-acylethanolamides (NAEs) on the spontaneous electrical activity of neuronal networks. NAEs are a group of lipids that can mimic the effects of marijuana and can be derived from a variety of plant sources including soy lecithin. The most prominent NAEs in soy lecithin, palmitoylethanolamide (PEA) and linoleoylethanolamide (LEA), were tested individually and were found to significantly inhibit neuronal spiking and bursting activity. These effects were potentiated by a mixture of NAEs as found in a HPLC enriched fraction from soy lecithin. Evaluation of virulence in wild type and pyrimidine auxotrophs of Pseudomonas aeruginosa using the eukaryotic model system Caenorhabditis elegans.

The human opportunistic pathogen, Pseudomonas aeruginosa PAO1, has been shown to kill the nematode Caenorhabditis elegans. C. elegans has been a valuable model for the study of bacterial pathogenesis, and has reinforced the notion that common virulence and host defense mechanisms exist. Recently, the pyrimidine pathway was shown to regulate virulence levels. Therefore, mutations in the pyrimidine pathway of PAO1 showed decrease virulence in the nematode. When starving the nematode, bacterial resistance was also shown to increase. It was hypothesized that starvation induced the DAF pathway, which regulates the transcription of genes involved with the antibacterial defense mechanism. Further research will be conducted to test this theory by performing RNAi experiments for the genes functioning in the antibacterial defense mechanism.

Evaluation of City of Denton Sub-Watershed by Benthic Macroinvertebrate Field Experimental Approach

In this study, two different field experiments were designed to assess the relative influence of urbanization on benthic communities. During spring and summer, four urban and one reference sites from Denton County, Texas were selected for benthic macroinvertebrate evaluation. Statistically significant differences in colonized benthic macroinvertebrate taxa on artificial substrates were observed among the four urban sites and the reference site. Oligochaetes and chironomids were the dominant taxa at all sites. Identification of chironomid larvae at the subfamily and genus level to detect differences between sites had higher statistical power than the evaluation based on total chironomids. At the reference site, Caenis, Cladotanytarsus, Orthocladius, and Ceratopogonidae were the dominant taxa, while the urban sites were dominated by Dero, Physella, Ancylidae, Chironomus, Dicrotendipes, Glyptotendipes, Polypedilum, Pseudochironomus, Stenochironomus, and Tanytarsus. These differences may have been dependent upon differences in hydrologic regime and water quality between sites. Significant differences (ANOVA, p < 0.01) in water quality parameters (alkalinity, hardness, nitrates, phosphates, chlorides, sulfates, calcium, magnesium, potassium, and triazine) were found among water samples collected from the reference and urban sites. Morphological and physiological developmental consequences of parental effects in the chicken embryo (Gallus gallus domesticus) and the zebrafish larva (Danio rerio).

Cardiac, metabolic and growth response of early-stage chicken embryos to perturbations in yolk environment was investigated. Also, effects of parental hypoxia exposure on hypoxia resistance, thermal tolerance and body length of zebrafish larvae were investigated. In the first study, thyroxine, triiodothyronine and testosterone produced differential effects on heart rate and development rate of chicken embryos during the first 4 days of development. Triiodothyronine caused a dose-dependent increase in heart rate when applied at 40 or 70 hours of age, while thyroxine caused a dose-dependent increase in heart rate when applied at 40 hours only. Testosterone and propyl-thiouracil (deiodinase antagonist) did not have an effect on heart rate. Development rate was not changed by thyroxine, triiodothyronine, testosterone or propyl-thiouracil, which suggested that heart rate changes did not result from changes in embryo maturity. In the second study, chicken embryos exposed to yolks of different bird species during early-stage embryonic development showed changes in heart rate, mass-specific oxygen consumption and body mass that scaled with the egg mass, incubation period length, and yolk triiodothyronine and testosterone levels of the species from which yolk was derived. The Role of Thyroid Hormone on the Development of Endothermy in White Leghorn Chickens (Gallus gallus)

As chickens hatch, there is a rapid change in their physiology and metabolism associated with attaining endothermy. It is thought that thyroid hormones (TH) play a major role in regulating developmental changes at hatching. In birds, TH regulates skeletal muscle growth, which has a direct impact on the chick's ability to thermoregulate via shivering thermogenesis. To better understand the role of TH in the timing of hatching, development of thermogenic capacity, and metabolic rate, we manipulated plasma TH levels in chicken embryos beginning at 85% development (day 17 of a 21 day incubation) with either thyroperoxidase inhibitor methimazole (MMI) or supplemental triiodothyronine (T3). After TH manipulation, we characterized O2 consumption and body temperature in the thermal neutral zone and during gradual cooling. Externally pipped embryos and 1 day post hatch (dph) chicks were cooled from 35 to 15°C. Manipulation of TH altered the timing of hatching, accelerating hatching under hyperthyroid conditions and decelerating hatching with hypothyroid conditions. Cohen's d revealed a large effect size on body temperature (Tb) of EP embryos of hypothyroid animals when compared to euthyroid animals in environmental temperatures of 32°C to 15°C, which was not seen in 1dph animals. Genomic Island Discovery through Enrichment of Statistical Modeling with Biological Information

Horizontal gene transfer enables acquisition and dissemination of novel traits including antibiotic resistance and virulence among bacteria. Frequently such traits are gained through the acquisition of clusters of functionally related genes, often referred to as genomic islands (GIs). Quantifying horizontal flow of GIs and assessing their contributions to the emergence and evolution of novel metabolic traits in bacterial organisms are central to understanding the evolution of bacteria in general and the evolution of pathogenicity and antibiotic resistance in particular, a focus of this dissertation study. Methods for GI detection have also evolved with advances in sequencing and bioinformatics, however, comprehensive assessment of these methods has been lacking. This motivated us to assess the performance of current methods for identifying islands on broad datasets of well-characterized bacterial genomes and synthetic genomes, and leverage this information to develop a novel approach that circumvents the limitations of the current state-of-the-art in GI detection.
