Molecule-Specific Imaging Analysis of Carcinogens in Breast Cancer Cells Using Time-of-Flight Secondary Ion Mass Spectrometry

Imaging time-of-flight secondary ion mass spectrometry (TOF-SIMS) is used to study the localization of heterocyclic amines in MCF7 line of human breast cancer cells. The detection sensitivities of a model rodent mutagen, 2-amino-1-methyl-6-phenylimidazo[4,5-b]pyridine (PhIP) were determined. Following an established criteria for the determination of status of freeze-fracture cells, the distribution of PhIP in the MCF7 cells are reported.

Fast Tuner R&D for RIA

The limited cavity beam loading conditions anticipated for the Rare Isotope Accelerator (RIA) create a situation where microphonic-induced cavity detuning dominates radio frequency (RF) coupling and RF system architecture choices in the linac design process. Where most superconducting electron and proton linacs have beam-loaded bandwidths that are comparable to or greater than typical microphonic detuning bandwidths on the cavities, the beam-loaded bandwidths for many heavy-ion species in the RIA driver linac can be as much as a factor of 10 less than the projected 80-150 Hz microphonic control window for the RF structures along the driver, making RF control problematic. System studies indicate that for the low-{beta} driver linac alone, running the cavities with no fast tuner may cost 50% or more than an RF system employing a voltage controlled reactance (VCX) or other type of fast tuner. An update of these system cost studies, along with the status of the VCX work being done at Lawrence Livermore National Lab is presented.

Cryptic Translocation Identification in Human and Mouse using Several Telomeric Multiplex FISH (TM-FISH) Strategies

Experimental data published in recent years showed that up to 10% of all cases with mild to severe idiopathic mental retardation may result from small rearrangements of the subtelomeric regions of human chromosomes. To detect such cryptic translocations, we developed a ''telomeric'' multiplex FISH assay, using a set of previously published and commercially available subtelomeric probes. This set of probes includes 41 cosmid/PAC/P1 clones located from less than 100kb to about 1 Mb from the end of the chromosomes. Similarly, a published mouse probe set, comprised of BACs hybridizing to the closest known marker toward the centromere and telomere of each mouse chromosome, was used to develop a mouse-specific ''telomeric'' M-FISH. Three different combinatorial labeling strategies were used to simultaneously detect all human sub-telomeric regions on one slide. The simplest approach uses only three fluors, and can be performed in laboratories lacking sophisticated imaging equipment or personnel highly trained in cytogenetics. A standard fluorescence microscope equipped with only three filters is sufficient. Fluor-dUTPs and labeled probes can be custom-made, thus dramatically reducing costs. Images can be prepared using generic imaging software (Adobe Photoshop), and analysis performed by simple visual inspection.

Implementation of the work program of Budget Period 2 of the East Binger Unit (''EBU'') DOE Project continues. The drilling of new horizontal well EBU 63-2H, scheduled for this reporting period, was delayed due to a lack of rig availability. This well was spud near the end of the reporting period. EBU 59-1 was converted to injection service, and injection capacity at the plant was increased from 19 MMscf/d to 22 MMscf/d. Although other factors have temporarily limited the field's ability to fully utilize the increased capacity, injection in the pilot area has been increased over 2 MMscf/d since the start of the project. Nitrogen recycle within the pilot area has been reduced with the projects implemented to date. Prior to pilot development, nitrogen production was 56% of nitrogen injected. Currently, nitrogen production is only 26% of injection. Pilot area oil production has increased 90 bpd or 30%, with 160 bpd from new wells offset by a loss of 70 bpd from wells converted to injection.

Single well seismic imaging of a gas-filled hydrofracture

A single well seismic survey was conducted at the Lost Hills, Ca oil field in a monitoring well as part of a CO2 injection test. The source was a piezoelectric seismic source and the sensors were a string of hydrophones hanging below the source. The survey was processed using standard CMP reflection seismology techniques. A potential reflection event was observed and interpreted as being caused by a near vertical hydrofracture. The radial distance between the survey well and the hydrofracture is estimated from Kirchoff migration using a velocity model derived from cross well seismic tomography. The hydrofracture location imaged after migration agrees with the location of an existing hydrofracture.

Characterization of Fast Ion Absorption of the High Harmonic Fast Wave in the National Spherical Torus Experiment

Ion absorption of the high harmonic fast wave in a spherical torus is of critical importance to assessing the viability of the wave as a means of heating and driving current. Analysis of recent National Spherical Torus Experiment (NSTX) shots has revealed that under some conditions when neutral beam and radio-frequency (RF) power are injected into the plasma simultaneously, a fast ion population with energy above the beam injection energy is sustained by the wave. In agreement with modeling, these experiments find the RF-induced fast ion tail strength and neutron rate at lower B-fields to be less enhanced, likely due to a larger beta profile, which promotes greater off-axis absorption where the fast ion population is small. Ion loss codes find the increased loss fraction with decreased B insufficient to account for the changes in tail strength, providing further evidence that this is an RF interaction effect. Though greater ion absorption is predicted with lower k(sub)||, surprisingly little variation in the tail was observed, along with a small neutron rate enhancement with higher k(sub)||. QUANTIFICATION OF MERCURY IN FLUE GAS EMISSION USING BORON-DOPED DIAMOND ELECTROCHEMISTRY

In this project, we have attempted to develop a new technique utilizing Boron-doped diamond (BDD) films to electrochemically detect mercury dissolved in solution via the initial deposition of metallic mercury, followed by anodic linear sweep voltammetry in the range from 10-10{sup -10} M to 10{sup -5} M. Cyclic voltammetry (CV) and differential pulse voltammetry (DPV) techniques were employed. The extremely low background current for BDD electrodes compared to glassy carbon (GC) provides a strong advantage in trace metal detection. CV peak currents showed good linearity in the micromolar range. A detection level of 6.8 x 10{sup -10} M was achieved with DPV in 0.1 M KNO{sub 3} (pH = 1) for a deposition time of 20 minutes. Reproducible stripping peaks were obtained, even for the low concentration range. A comparison with GC shows that BDD is superior. Linear behavior was also obtained in the mercury concentration range from 10{sup -10} M to 10{sup -9} M.
