Transcriptome and Biochemical Analyses of Fungal Degradation of Wood
Lignocellulosic accounts for a large percentage of material that can be utilized for biofuels. The most costly part of lignocellulosic material processing is the initial hydrolysis of the wood which is needed to circumvent the lignin barrier and the crystallinity of cellulose. Enzymes will play an increased role in this conversion in that they potentially provide an alternative to costly and caustic high temperature and acid treatment. The increasing use of enzymes in biotechnology is facilitated by both continued improvements in enzyme technology but also in the discovery of new and novel enzymes. The present proposal is aimed at identifying the enzymes which are known to depolymerize woody biomass. Fundamental understanding of how nature gains access to cellulose and hemicellulose will impact all applications. Because fungi are the only known microbes capable of circumventing the lignin barrier, knowledge of the enzyme they use is of great potential for biofuel processing. Nature has evolved different fungal mechanisms for enzymatic hydrolysis of wood. Most notable are the white-rot fungi (wrf) and the brown-rot fungi (brf). Transfer of Physical and Hydraulic Properties Databases to the Hanford Environmental Information System - PNNL Remediation Decision Support Project, Task 1, Activity 6
This report documents the requirements for transferring physical and hydraulic property data compiled by PNNL into the Hanford Environmental Information System (HEIS). The Remediation Decision Support (RDS) Project is managed by Pacific Northwest National Laboratory (PNNL) to support Hanford Site waste management and remedial action decisions by the U.S. Department of Energy and one of their current site contractors - CH2M-Hill Plateau Remediation Company (CHPRC). The objective of Task 1, Activity 6 of the RDS project is to compile all available physical and hydraulic property data for sediments from the Hanford Site, to port these data into the Hanford Environmental Information System (HEIS), and to make the data web-accessible to anyone on the Hanford Local Area Network via the so-called Virtual Library.1 These physical and hydraulic property data are used to estimate parameters for analytical and numerical flow and transport models that are used for site risk assessments and evaluation of remedial action alternatives. In past years efforts were made by RDS project staff to compile all available physical and hydraulic property data for Hanford sediments and to transfer these data into SoilVision{reg_sign}, a commercial geotechnical software package designed for storing, analyzing, and manipulating soils data. Transportation Programs: Challenges Facing the Department of Transportation and Congress
Testimony issued by the Government Accountability Office with an abstract that begins "A safe, efficient, and convenient transportation system is integral to the health of our economy and quality of life. Our nation's vast transportation system of airways, railways, roads, transit systems, and waterways has served this need, yet is under considerable pressure due to increasing congestion and costs to maintain and improve the system. Calls for increased investment come at a time when traditional funding for transportation projects is increasingly strained. The authorizing legislation supporting transportation programs will soon expire. The Department of Transportation (DOT) implements national transportation policy and administers most federal transportation programs. DOT received funds for transportation infrastructure projects through the American Recovery and Reinvestment Act of 2009 to aid in economic recovery. DOT also requested $72.5 billion to carry out its activities for fiscal year 2010. This statement presents GAO's views on major challenges facing DOT and Congress as they work to administer recovery funds and reauthorize surface transportation and aviation programs. Transportation Security: Comprehensive Risk Assessments and Stronger Internal Controls Needed to Help Inform TSA Resource Allocation
A letter report issued by the Government Accountability Office with an abstract that begins "The Department of Homeland Security (DHS) has called for using risk-informed approaches to help prioritize its investments, develop plans, and allocate resources in a way that balances security and commerce. Within DHS, the Transportation Security Administration (TSA) is responsible for making risk-informed investments to secure the transportation system. Transuranic (TRU) Waste Repackaging at the Nevada Test Site
This paper describes the activities required to modify a facility and the process of characterizing, repackaging, and preparing for shipment the Nevada Test Site’s (NTS) legacy transuranic (TRU) waste in 58 oversize boxes (OSB). The waste, generated at other U.S. Department of Energy (DOE) sites and shipped to the NTS between 1974 and 1990, requires size-reduction for off-site shipment and disposal. The waste processing approach was tailored to reduce the volume of TRU waste by employing decontamination and non-destructive assay. As a result, the low-level waste (LLW) generated by this process was packaged, with minimal size reduction, in large sea-land containers for disposal at the NTS Area 5 Radioactive Waste Management Complex (RWMC). The remaining TRU waste was repackaged and sent to the Idaho National Laboratory Consolidation Site for additional characterization in preparation for disposal at the Waste Isolation Pilot Plant (WIPP), near Carlsbad, New Mexico. The DOE National Nuclear Security Administration Nevada Site Office and the NTS Management and Operating (M&O) contractor, NSTec, successfully partnered to modify and upgrade an existing facility, the Visual Examination and Repackaging Building (VERB). TRIDENT flyer plate Impact technique: comparison to gas gun plate impact technique
This report describes the details of a series of plate impact experiments that were conducted on a gas gun in an effort to validate a new technique for plate impact using the TRIDENT laser to launch thin flyers. The diagnostics fielded were VISAR and identical samples and impactors were used on both platforms. All experimenters agree that the VISAR results should have agreed between the two experimental platforms. The VISAR results did not agree across the platforms and experimenters offer explanations and implications for this outcome.

Troubled Asset Relief Program: March 2009 Status of Efforts to Address Transparency and Accountability Issues
A letter report issued by the Government Accountability Office with an abstract that begins "GAO's third report on the Troubled Asset Relief Program (TARP) follows up on recommendations from the January 28, 2009, report (GAO-09-296). It also reviews (1) the nature and purpose of activities that had been initiated under TARP as of March 27, 2009; (2) the Department of Treasury's Office of Financial Stability's (OFS) hiring efforts, use of contractors, and progress in developing an internal control system; and (3) TARP performance indicators. For this work, GAO reviewed signed agreements and other relevant documentation and met with officials from OFS, contractors, and federal agencies. As of March 27, 2009, Treasury had disbursed $303.4 billion of the $700 billion in TARP funds. Most of the funds (almost $199 billion) went to purchase preferred shares of 532 financial institutions under the Capital Purchase Program (CPP), Treasury's primary vehicle under TARP for stabilizing financial markets."
