Design of Genomic Signatures of Pathogen Identification & Characterization

This chapter will address some of the many issues associated with the identification of signatures based on genomic DNA/RNA, which can be used to identify and characterize pathogens for biodefense and microbial forensic goals. For the purposes of this chapter, we define a signature as one or more strings of contiguous genomic DNA or RNA bases that are sufficient to identify a pathogenic target of interest at the desired resolution and which could be instantiated with particular detection chemistry on a particular platform. The target may be a whole organism, an individual functional mechanism (e.g., a toxin gene), or simply a nucleic acid indicative of the organism. The desired resolution will vary with each program's goals but could easily range from family to genus to species to strain to isolate. The resolution may not be taxonomically based but rather pan-mechanistic in nature: detecting virulence or antibiotic-resistance genes shared by multiple microbes. Entire industries exist around different detection chemistries and instrument platforms for identification of pathogens, and we will only briefly mention a few of the techniques that we have used at Lawrence Livermore National Laboratory (LLNL) to support our biosecurity-related work since 2000. Conservation Laws for Coupled Hydro-mechanical Processes in Unsaturated Porous Media: Theory and Implementation

We develop conservation laws for coupled hydro-mechanical processes in unsaturated porous media using three-phase continuum mixture theory. From the first law of thermodynamics, we identify energy-conjugate variables for constitutive modeling at macroscopic scale. Energy conjugate expressions identified relate a certain measure of effective stress to the deformation of the solid matrix, the degree of saturation to the matrix suction, the pressure in each constituent phase to the corresponding intrinsic volume change of this phase, and the seepage forces to the corresponding pressure gradients. We then develop strong and weak forms of boundary-value problems relevant for 3D finite element modeling of coupled hydro-mechanical processes in unsaturated porous media. The paper highlights a 3D numerical example illustrating the advances in the solution of large-scale coupled finite element systems, as well as the challenges in developing more predictive tools satisfying the basic conservation laws and the observed constitutive responses for unsaturated porous materials.

Interview with Louise Young and Vivienne Armstrong, longtime activists in the Dallas lesbian community. The interview includes Armstrong's personal experiences of childhood in Knoxville, Tennessee, New Jersey, and California, as well as her decision to study nursing and settle in Denver, Colorado, and her coming out narrative. The interview also includes Young's personal experiences of childhood in Ada, Oklahoma, her education at East Central State University and the University of Colorado, and her coming out narrative. They talk about their meeting and early relationship, give descriptions of Denver's gay and lesbian communities, their involvement with various groups such as the Daughters of Bilitis and Gay Liberation Front, their decision to move to Dallas, their involvement with groups such as the National Organization for Women and Dallas Gay Political Caucus/Dallas Gay and Lesbian Alliance in various political campaigns, and Young's career at Texas Instruments and Raytheon and efforts to create more equitable human resources policies from within the corporations. Additionally, Armstrong and Young talk about the effects of the HIV-AIDS crisis on Dallas's gay and lesbian communities, their 2008 marriage, relationships with family members, and secrets to a long relationship.

NUCLEAR FORENSICS ANALYSIS CENTER FORENSIC ANALYSIS TO DATA INTERPRETATION

The Nuclear Forensics Analysis Center (NFAC) is part of Savannah River National Laboratory (SRNL) and is one of only two USG National Laboratories accredited to perform nuclear forensic analyses to the requirements of ISO 17025. SRNL NFAC is capable of analyzing nuclear and radiological samples from bulk material to ultra-trace samples. NFAC provides analytical support to the FBI's Radiological Evidence Examination Facility (REEF), which is located within SRNL. REEF gives the FBI the capability to perform traditional forensics on material that is radiological and/or is contaminated. SRNL is engaged in research and development efforts to improve the USG technical nuclear forensics capabilities. Research includes improving predictive signatures and developing a database containing comparative samples.

Captain John R. Hughes: Lone Star Ranger

Captain John R. Hughes, Lone Star Ranger is the first full and complete modern biography of a man who served as a Texas Ranger from 1887 until early 1915. He came to the attention of the Rangers after doggedly trailing horse thieves for nearly a year and recovering his stolen stock. After helping Ranger Ira Aten track down another fugitive from justice, Hughes then joined Company D of the Texas Rangers on Aten’s recommendation, intending to stay for only a few months; he remained in the service for nearly thirty years. When Sgt. Charles Fusselman was killed by bandits, Hughes took his place. When Captain Frank Jones was killed by bandits in 1893, Hughes was named captain of Company D. As captain, Hughes and his men searched the border and identified every bandit involved in the killing of Jones. They all received justice. Toward the end of his career Hughes became a senior captain based in Austin, and in 1915, having served as a captain and ranger longer than any other man, he retired from the force. His later years were happy ones, with traveling and visiting friends and relatives. He became a Texas icon and national celebrity, receiving more …
