Somatic cell genotoxicity at the glycophorin A locus in humans

We have developed an assay for detecting variant erythrocytes that occur as a result of in vivo allele loss at the glycophorin A (GPA) locus on chromosome 4 in humans. This gene codes for an erythroid- specific cell surface glycoprotein, and with our assay we are able to detect rare variant erythrocytes that have lost expression of one of the two GPA alleles. Two distinctly different variant cell types are detected with this assay. One variant cell type (called N{O}) is hemizygous. Our assay also detects homozygous variant erythrocytes that have lost expression of the GPA(M) allele and express the GPA(N) allele at twice the heterozygous level. The results of this assay are an enumeration of the frequency of N{O} and NN variant cell types for each individual analyzed. These variant cell frequencies provide a measure of the amount of somatic cell genotoxicity that has occurred at the GPA locus. Such genotoxicity could be the result of (1) reactions of toxic chemicals to which the individual has been exposed, or (2) high energy radiation effects on erythroid precursor cells, or (3) errors in DNA replication or repair in these cells of the bone marrow. As a consequence of the Chernobyl accident, data have been obtained throughout the Northern Hemisphere on the concentrations of radionuclides in air, vegetation, soil, water, and foodstuffs that could be important means of human exposure. At the IAEA's invitation, the traveler reviewed recently published data and handbook summaries. The traveler evaluated the need for revising the default values recommended in Chapter 5, Terrestrial and Aquatic Food Chain Transport,'' of IAEA Safety Series No. 57. All attempts at revision were made to keep the mathematical complexity of the models to a minimum without substantial underestimation of dose to critical population subgroups. The traveler also served as chairman of the Multiple Pathways Working Group of the Coordinated Research Program on VAMP. This group has been established to test predictions of models assessing multiple exposure pathways potentially leading to human exposure to {sup 137}Cs. Testing is carried out for major components of assessment models that predict deposition, environmental transport, food chain bioaccumulation, and subsequent uptake and retention in the human body and dose due to exposure to external gamma radiation.

DOE standard guidelines for use of probabilistic seismic hazard curves at Department of Energy sites

This Standard is intended to provide guidance in the use of the seismic hazard curves developed by the Lawrence Livermore National Laboratory (LLNL) and the Electric Power Research Institute (EPRI). Experience to-date has shown that application of these methodologies can yield significantly different results. In response to this issue, a Seismic Working Group (SWG) has been formed at the Department of Energy (DOE) Headquarters to coordinate the application of these methodologies within DOE in a consistent manner. The position developed by the SWG and contained in this Standard is intended for immediate use in developing seismic hazard estimates at DOE sites for the evaluation of new and existing, nuclear and non-nuclear DOE facilities. This Standard is needed not only to address the LLNL/EPRI issue but also to assure that state-of-the-art seismic hazard methods are incorporated into DOE standards as soon as possible. The DOE is currently involved in a joint program with the Nuclear Regulatory Commission and EPRI to evaluate these existing probabilistic seismic hazard methodologies and to develop recommendations for an improved methodology for the 1990`s. The final product of this effort is expected to result in more stable hazard estimates and will supersede this Standard in approximately two …

This document is the Health and Safety Plan (HASP) for the demonstration of IITRI`s EM Treatment Technology. In this process, soil is heated in situ by means of electrical energy for the removal of hazardous organic contaminants. This process will be demonstrated on a small plot of contaminated soil located in the Pit Area of Classified Burial Ground K-1070-D, K-25 Site, Oak Ridge, TN. The purpose of the demonstration is to remove organic contaminants present in the soil by heating to a temperature range of 85{degrees} to 95{degrees}C. The soil will be heated in situ by applying 60-Hz AC power to an array of electrodes placed in boreholes drilled through the soil. In this section a brief description of the process is given along with a description of the site and a listing of the contaminants found in the area.

This report presents results from studies of the uptake of Cu(II) and phenol by laboratory-type wetlands. The uptake of Cu(II) follows a tri-phasic behavior. The addition of peat was observed to have only a minimal effect on Cu(II) uptake. On the other hand, phenol sorption was favorably modified by addition of peat.

Mechanical and Cryogenic Teting of VLPC Flex Circuits Made by Litchfield, Cirexx, and Speedy Circuit

Four flex circuits were tested. They are categorized as: (1) Litchfield narrow; (2) Litchfield wide; (3) Cirexx wide; and (4) Speedy Circuit green. The cross sectional area of copper was calculated from trace resistance measurements. All the flex circuits had an acceptable amount of copper cross section. about 6 e-8 m{sup 2} for all except the Cirexx flex which had 4.5 e-8 m{sup 2}. The thickness of the ribbons was 0.006-inch for the Litchfield and about 0.004-inch for the Cirexx and Speed circuit. The room temperature resistivity ratio (RRR) was measured to be 15 for all the circuits. This was expected and means that the trace material resistivity at helium temperatures is 1/15th of what it is at room temperature. This is good from the standpoint of thermal heat load. Purer copper, with an RRR > 30 would result in an unacceptable heat load. The cryogenic cycling and bending tests to liquid nitrogen proved that both the Cirexx wide and Speedy circuit green were made of acceptable materials. Both Litchfield flex circuits had significant cracking of the coverlay when bent at cold temperatures and are not acceptable. The Materials Consultation Group of SRTC has completed a review of existing literature and data regarding the useable service life of Viton{reg_sign} GLT fluoroelastomer O-rings currently used in the Model 9975 packaging assemblies. Although the shipping and transportation period is normally limited to 2 years, it is anticipated that these packages will be used for longer-term storage of Pu-bearing materials in KAMS (K-Area Materials Storage) prior to processing or disposition in the APSF (Actinide Packaging and Storage Facility). Based on the service conditions and review of available literature, Materials Consultation concludes that there is sufficient existing data to establish the technical basis for storage of Pu-bearing materials using Parker Seals O-ring compound V835-75 (or equivalent) for up to 10 years following the 2-year shipping period. Although significant physical deterioration of the O-rings and release of product is not expected, definite changes in physical properties will occur. However, due to the complex relationship between elastomer formulation, seal properties, and competing degradation mechanisms, the actual degree of property variation and impact upon seal performance is difficult to predict. Therefore, accelerated aging and/or surveillance programs are recommended to validate the assumptions outlined in this report and to assess the long-term performance of O-ring …

Alloy 22 [UNS N06022] is now being considered for construction of high level waste containers to be emplaced at the potential repository at Yucca Mountain or elsewhere. In essence, this alloy is 21% Cr, 13% Mo, 4% Fe, 3% W, 2% Co, with the balance being Ni. Variants without tungsten are also being considered. Detailed mechanistic models are being developed to account for the corrosion of Alloy 22 surfaces in crevices that will inevitably form. Such occluded areas experience substantial decreases in pH, with corresponding elevations in chloride concentration. Other relevant materials will also be investigated: nickel-based alloys such as Alloys 825, 625, C-4, C-276 and 59; titanium-based alloys such as Grades 12, 7 and 16, carbon steels such as A516 Grade 55; stainless steels such as 304, 304L, 316, 316L and 316NG; various copper-based alloys; and any materials that would serve as crevice formers (rock, thermally-sprayed ceramics, etc.). Experimental work has been undertaken to validate the crevice corrosion model, including parallel studies with 304 stainless steel. The crevice corrosion model is described in detail in scientific notebooks of the Principal Investigator, as well as other publications.
