Advanced microinstrumentation for rapid DNA sequencing and large DNA fragment separation

Our efforts to develop novel technology for a rapid DNA sequencer and large fragment analysis system based upon gel electrophoresis are described. We are using microfabrication technology to build dense arrays of high speed micro electrophoresis lanes that will ultimately increase the sequencing rate of DNA by at least 100 times the rate of current sequencers. We have demonstrated high resolution DNA fragment separation needed for sequencing in polyacrylamide microgels formed in glass microchannels. We have built prototype arrays of microchannels having up to 48 channels. Significant progress has also been made in developing a sensitive fluorescence detection system based upon a confocal microscope design that will enable the diagnostics and detection of DNA fragments in ultrathin microchannel gels. Development of a rapid DNA sequencer and fragment analysis system will have a major impact on future DNA instrumentation used in clinical, molecular and forensic analysis of DNA fragments.

Copper-diamond composite substrates for electronic components

High-power density electronic components such as fast microprocessors and power semiconductors are often limited by inability to keep the device junctions below their max rated operating temperature. Present high power multichip module and single chip package designs use substrate materials such as Si nitride or copper tungsten with thermal conductivity in the range of 200 W/m{center_dot}K. We have developed a copper-diamond composite (Dymalloy) with a thermal conductivity of 420 W/m{center_dot}K, better than Cu, and an adjustable thermal expansion coefficient (TCE=5.5 ppM/C at 25 C), compatible with Si and GaAs. Because of the matched TCE, it is possible to use low thermal resistance hard die attach methods. The mechanical properties of the composite also make it attractive as an electronic component substrate material.

The effect of chemical composition on the PCT durability of mixed waste glasses from wastewater treatment sludges

An experimental program has been designed to examine the chemical durability of glass compositions derived from the vitrification of simulated wastewater treatment sludges. These sludges represent the majority of low-level mixed wastes currently in need of treatment by the US DOE. The major oxides in these model glasses included SiO{sub 2}, Al{sub 2}O{sub 3}, B{sub 2}O{sub 3}, Na{sub 2}O, CaO and Fe{sub 2}O{sub 3}. In addition, three minor oxides, BaO, NiO, and PbO, were added as hazardous metals. The major oxides were each varied at two levels resulting in 32 experimental glasses. The chemical durability was measured by the 7-Day Product Consistency Test (PCT). The normalized sodium release rates (NRR{sub Na}) of these glasses ranged from 0.01 to 4.99 g/m{sup 2}. The molar ratio of the glass-former to glass-modifier (F/M) was found to have the greatest effect on PCT durability. Glass-formers included SiO{sub 2}, Al{sub 2}O{sub 3}, and B{sub 2}O{sub 3}, while Na{sub 2}O, CaO, BaO, NiO, and PbO were glass-modifiers. As this ratio increased from 0.75 to 2.0, NRR{sub Na} was found to decrease between one and two orders of magnitude. Another important effect on NRR{sub Na} was the Na{sub 2}O/CaO ratio. Engineering development of advanced physical fine coal cleaning for premium fuel applications. Quarterly technical progress report 9, October 1, 1994--December 31, 1994

The primary goal of this project is the engineering development of two advanced physical fine coal cleaning processes, column flotation and selective agglomeration, for premium fuel applications. The project scope includes laboratory research and bench-scale testing on six coals to optimize these processes, followed by design, and construction of a 2-t/hr process development unit (PDU). The PDU will then be operated to generate 200 ton lots of each of three project coals, by each process. The project began in October, 1992 and is scheduled for completion by March, 1997. During Quarter 9 (October--December, 1995), parametric and optimization testing was completed for the Taggart, Sunnyside, and Indiana VII coal using a 12-inch Microcel{trademark} flotation column. The detailed design of the 2-t/hr PDU grinding, flotation, and dewatering circuits neared completion with the specification of the major pieces of capital equipment to be purchased for these areas. Selective agglomeration test work investigated the properties of various industrial grades of heptane for use during bench- and PDU-scale testing. It was decided to use a hydrotreated grade of commercial heptane due to its low cost and low concentration of aromatic compounds. The final Subtask 6.4 CWF Formulation Studies Test Plan was issued. Engineering task plan for flammable gas atmosphere mobile color video camera systems

This Engineering Task Plan (ETP) describes the design, fabrication, assembly, and testing of the mobile video camera systems. The color video camera systems will be used to observe and record the activities within the vapor space of a tank on a limited exposure basis. The units will be fully mobile and designed for operation in the single-shell flammable gas producing tanks. The objective of this tank is to provide two mobile camera systems for use in flammable gas producing single-shell tanks (SSTs) for the Flammable Gas Tank Safety Program. The camera systems will provide observation, video recording, and monitoring of the activities that occur in the vapor space of applied tanks. The camera systems will be designed to be totally mobile, capable of deployment up to 6.1 meters into a 4 inch (minimum) riser.

Iron/potassium perchlorate pellet burn rate measurements

A burn rate test having several advantages for low gas-producing pyrotechnic compacts has been developed. The technique involves use of a high speed video motion analysis system that allows immediate turnaround and produces all required data for rate computation on magnetic tape and becomes immediately available on the display screen. The test technique provides a quick method for material qualification along with data for improved reliability and function. Burn rate data has been obtained for both UPI and Eagle Pitcher Iron/Potassium Perchlorate blends. The data obtained for the UPI blends cover a range of composition, pellet density, and ambient (before ignition) pellet temperature. Burn rate data for the E-P blends were extended to include surface conditions or particle size as a variable parameter.

Perhaps the most common approximation in engineering is that, relative to its neighbors, a system component is structurally rigid. This paper presents a development of the rigid assumption for use in nonlinear, implicit finite element codes. In this method, computational economy is gained by condensing the size of the associated linear system of equations, eliminating the processing of rigid elements, and reducing the overall nonlinearity of the problem.

Robotic weld overlay coatings for erosion control. Quarterly technical progress report, October 1994--December 1994

Research is presently being conducted to develop a criteria for selecting weld overlay coatings for erosion mitigation in Circulated Fluidized Beds. Initially, eleven weld overlay alloys were selected for erosion testing based upon a literature review. All eleven coatings were deposited on 1018 steel substrates using the plasma arc welding process. Ten samples from each coating were prepared for erosion testing. The coating deposition and sample preparation procedures were described in the second quarterly report. All selected coatings were erosion tested at 400{degree}C and their erosion resistance was evaluated by determining the steady state erosion rate. In addition, the microstructure of each coating was characterized before and after the erosion tests. The results of the tests are discussed in the third quarterly report. No correlations were found between room temperature hardness of the weld overlay coatings and their erosion resistance at elevated temperature. During the last quarter tensile tests were performed at 400{degree}C for the Ultimet, Inconel-625, 316L SS, C-22, and Stellite-6 wrought alloys. The erosion tests for these materials at 400{degree}C are in progress. The results of mechanical and erosion tests will be used to correlate mechanical properties of selected wrought alloys such as tensile toughness, ductility, strain hardening …

Scalar wave diffraction from a circular aperture

The scalar wave theory is used to evaluate the expected diffraction patterns from a circular aperture. The standard far-field Kirchhoff approximation is compared to the exact result expressed in terms of oblate spheroidal harmonics. Deviations from an expanding spherical wave are calculated for circular aperture radius and the incident beam wavelength using suggested values for a recently proposed point diffractin interferometer. The Kirchhoff approximation is increasingly reliable in the far-field limit as the aperture radius is increased, although significant errors in amplitude and phase persist.

This document is a compilation of work performed as part of the Dome Load Control Project in 1994. Section 2 contains the calculations of the weight of the soil over the tank dome for each of the 75-feet-diameter waste-storage tanks located at the Hanford Site. The chosen soil specific weight and soil depth measured at the apex of the dome crown are the same as those used in the primary analysis that qualified the design. Section 3 provides reference dimensions for each of the tank farm sites. The reference dimensions spatially orient the tanks and provide an outer diameter for each tank. Section 4 summarizes the available soil surface elevation data. It also provides examples of the calculations performed to establish the present soil elevation estimates. The survey data and other data sources from which the elevation data has been obtained are printed separately in Volume 2 of this Supporting Document. Terminal ballistic experiments for the development of turbine engine blade containment technology

The ballistic experiments reported herein were conducted in three sets between October 1993 and November 1994. The first set of experiments examined the ballistic failure of annealed titanium plates. These experiments were performed in a manner consistent with earlier experiments conducted at United Technologies` Pratt and Whitney Division. The second set of experiments examined the ballistic performance of select aluminum and titanium alloys in single-plate and laminate form. In both sets of experiments, the failure modes of the targets were observed and catalogued. The third set of experiments evaluated underlying issues associated with geometric scaling. Blunt .30-and .50-caliber hard steel projectiles impacted on geometrically similar annealed titanium plates.

Utilization of a hydraulic barrier to control migration of a uranium plume

A uranium plume emanating from the U.S. Department of Energy`s Fernald Environmental Management Project (FEMP) in Fernald, Ohio had migrated off site and the leading edge of the plume had already mixed with an organic and inorganic plume emanating from two industries south of the FEMP. A method was needed to prevent the further southern migration of the plume, minimize any impacts to the geometry, concentrations, distribution or flow patterns of the organic and inorganic plumes emanating from the off-site industries, while meeting the ultimate cleanup goals for the FEMP. This paper discusses the use of a hydraulic barrier created to meet these goals by pumping a five well recovery system and the problems associated with the disposition of over 2 million gallons per day of water with low concentrations of uranium.

The Automated Bar Coding of Air Samples at Hanford (ABCASH) system provides an integrated data collection, sample tracking, and data reporting system for radioactive particulate air filter samples. The ABCASH plotting program provides a graphical trend report for ABCASH of the performance of air sample results. This document provides an operational guide for using the program. Based on sample location identifier and date range, a trend chart of the available data is generated. The trend chart shows radiological activity versus time. General indications of directional trend of the concentrations in air over time may be discerned. Comparison limit set point values are also shown as derived from the ABCASH data base.
