Using Cytochrome c{sub 3} to Make Selenium Nanowires

We report on a new method to make nanostructures, in this case selenium nanowires, in aqueous solution at room temperature. We used the protein cytochrome c{sub 3} to reduce selenate (SeO{sub 4}{sup 2{minus}}) to selenium (Se{sup 0}). Cytochrome c{sub 3} is known for its ability to catalyze reduction of metals including U{sup VI} {yields} U{sup IV}, Cr{sup VI} {yields} Cr{sup III}, Mo{sup VI} {yields} Mo{sup IV}, Cu{sup II} {yields} Cu{sup 0}, Pb{sup II} {yields} Pb{sup 0}, Hg{sup II} {yields} Hg{sup 0}. Nanoparticles of Se{sup 0} precipitated from an aqueous solution at room temperature, followed by spontaneous self-assembling into nanowires. Cytochrome c{sub 3} was extracted from the sulfate-reducing bacteria Desulfovibrio vulgaris (strain Holdenborough) and isolated by the procedure of DerVartanian and Legall.

Thin-film characterization and flaw detection. Progress report No. 7, February 1, 1993--January 31, 1994

Objective is to determine the elastic constants of thin films deposited on substrates, measure residual stresses, and detect and characterize defects in thin film substrate configurations. A line-focus acoustic microscope is being used to measure speed of surface acoustic waves (SAW) in thin film/substrate system. V(z) curves (record of transducer voltage output (V) with variation of distance z between acoustic lens and specimen) were calculated in terms of characteristic functions of acoustic lens and reflectance function of fluid-loaded specimen, and were compared with data for isotropic, anisotropic, and layered anisotropic materials. For thin film/substrate of known elasticity, theoretical surface acoustic wave velocities agree with measured V(z) curves for full range of wave propagation directions. Results have been obtained for homogeneous nitride films and transition-metal nitride superlattice films.

Optimizing the Delivery of Short-Lived Alpha Particle-Emitting Isotopes to Solid Tumors

The underlying hypothesis of this project was that optimal alpha emitter-based radioimmunotherapy (RAIT) could be achieved by pairing the physical half-life of the radioisotope to the biological half-life of the targeting vehicle. The project had two specific aims. The first aim was to create and optimize the therapeutic efficacy of 211At-SAPS-C6.5 diabody conjugates. The second aim was to develop bispecific-targeting strategies that increase the specificity and efficacy of alpha-emitter-based RAIT. In the performance of the first aim, we created 211At-SAPS-C6.5 diabody conjugates that specifically targeted the HER2 tumor associated antigen. In evaluating these immunoconjugates we determined that they were capable of efficient tumor targeting and therapeutic efficacy of established human tumor xenografts growing in immunodeficient mice. We also determined that therapeutic doses were associated with late renal toxicity, likely due to the role of the kidneys in the systemic elimination o f these agents. We are currently performing more studies focused on better understanding the observed toxicity. In the second aim, we successfully generated bispecific single-chain Fv (bs-scFv) molecules that co-targeted HER2 and HER3 or HER2 and HER4. DWPF GLASS BEADS AND GLASS FRIT TRANSPORT DEMONSTRATION

DWPF is considering replacing irregularly shaped glass frit with spherical glass beads in the Slurry Mix Evaporator (SME) process to decrease the yield stress of the melter feed (a non-Newtonian Bingham Plastic). Pilot-scale testing was conducted on spherical glass beads and glass frit to determine how well the glass beads would transfer when compared to the glass frit. Process Engineering Development designed and constructed the test apparatus to aid in the understanding and impacts that spherical glass beads may have on the existing DWPF Frit Transfer System. Testing was conducted to determine if the lines would plug with the glass beads and the glass frit slurry and what is required to unplug the lines. The flow loop consisted of vertical and horizontal runs of clear PVC piping, similar in geometry to the existing system. Two different batches of glass slurry were tested: a batch of 50 wt% spherical glass beads and a batch of 50 wt% glass frit in process water. No chemicals such as formic acid was used in slurry, only water and glass formers. The glass beads used for this testing were commercially available borosilicate glass of mesh size -100+200. Ion Acceleration from the Interaction of Ultra-Intense Lasers with Solid Foils

The discovery that ultra-intense laser pulses (I > 10{sup 18} W/cm{sup 2}) can produce short pulse, high energy proton beams has renewed interest in the fundamental mechanisms that govern particle acceleration from laser-solid interactions. Experiments have shown that protons present as hydrocarbon contaminants on laser targets can be accelerated up to energies > 50 MeV. Different theoretical models that explain the observed results have been proposed. One model describes a front-surface acceleration mechanism based on the ponderomotive potential of the laser pulse. At high intensities (I > 10{sup 18} W/cm{sup 2}), the quiver energy of an electron oscillating in the electric field of the laser pulse exceeds the electron rest mass, requiring the consideration of relativistic effects. The relativistically correct ponderomotive potential is given by U{sub p} = ([1 + I{lambda}{sup 2}/1.3 x 10{sup 18}]{sup 1/2} - 1) m{sub o}c{sup 2}, where I{lambda}{sup 2} is the irradiance in W {micro}m{sup 2}/cm{sup 2} and m{sub o}c{sup 2} is the electron rest mass. At laser irradiance of I{lambda}{sup 2} {approx} 10{sup 20} W {micro}m{sup 2}/cm{sup 2}, the ponderomotive potential can be of order several MeV. InGaAsN Solar Cells with 1.0eV Bandgap, Lattice Matched to GaAs

The design, growth by metal-organic chemical vapor deposition, and processing of an In{sub 0.07}Ga{sub 0.93}As{sub 0.98}N{sub 0.02} solar Al, with 1.0 ev bandgap, lattice matched to GaAs is described. The hole diffusion length in annealed, n-type InGaAsN is 0.6-0.8 pm, and solar cell internal quantum efficiencies > 70% arc obwined. Optical studies indicate that defects or impurities, from InGAsN doping and nitrogen incorporation, limit solar cell performance.

WxWindows Interface for CALE

wxWindows is an Open Source, platform independent, User Interface (UI) which has been in development for over eleven years (http://www.wxwindows.org). Currently wxWindows is actively supported for the Linux/Unix (X11, Motif and GTK+), Mac OS 9 and X, all Win32 OSes, MGL, and OS/2 operating systems. wxWindows is written in C++ using an object oriented programming framework; it is a reasonably lightweight API (called wxWidgets) sitting over the native graphics packages of the various platforms it supports. The original version of CALE was written for the basic target platform of Unix using X11 as the graphics package. There have been separate efforts to port the code to Mac OS 9, Mac OS X, Win32, Windows Services for Unix (SFU) and CygWin. Each of these used a variety of different graphical interface approaches and build/make systems. For instance Windows SFU and CygWin could still only use X11 graphics. So could the Win32 version, if a X11 server library and client software were installed. A native Win32 version of CALE was contemplated, but never started. The Macintosh versions were completed but never widely distributed to the users. Given the growing code version support issues, and the slow deviation from the portable code model …

MODIS Airborne simulator (MAS) Final Report for CLASIC

The MAS was flown aboard the NASA ER-2 for the CLASIC field experiment, and for all data collected, provided calibrated and geolocated (Level-1B) radiance data for it’s 50 spectral bands (ranging in wavelength for 0.47 to 14.3 µm). From the Level-1B data, as directed in the Statement of Work, higher order (Level-2) data products were derived. The Level-2 products include: a) cloud optical thickness, b) cloud effective radius, c) cloud top height (temperature), d) cloud fraction, e) cloud phase products. Preliminary Level-1B and Level-2 products were provided during the field experiment (typically within one or two days of data collection). Final version data products were made available in December 2008 following considerable calibration analysis. Data collection, data processing (to Level-2), and discussion of the calibration work are summarized below.

On the Performance of an Algebraic Multigrid Solver on Multicore Clusters

Algebraic multigrid (AMG) solvers have proven to be extremely efficient on distributed-memory architectures. However, when executed on modern multicore cluster architectures, we face new challenges that can significantly harm AMG's performance. We discuss our experiences on such an architecture and present a set of techniques that help users to overcome the associated problems, including thread and process pinning and correct memory associations. We have implemented most of the techniques in a MultiCore SUPport library (MCSup), which helps to map OpenMP applications to multicore machines. We present results using both an MPI-only and a hybrid MPI/OpenMP model.

A New Class of Non-Linear, Finite-Volume Methods for Vlasov Simulation

Methods for the numerical discretization of the Vlasov equation should efficiently use the phase space discretization and should introduce only enough numerical dissipation to promote stability and control oscillations. A new high-order, non-linear, finite-volume algorithm for the Vlasov equation that discretely conserves particle number and controls oscillations is presented. The method is fourth-order in space and time in well-resolved regions, but smoothly reduces to a third-order upwind scheme as features become poorly resolved. The new scheme is applied to several standard problems for the Vlasov-Poisson system, and the results are compared with those from other finite-volume approaches, including an artificial viscosity scheme and the Piecewise Parabolic Method. It is shown that the new scheme is able to control oscillations while preserving a higher degree of fidelity of the solution than the other approaches.

Probabilistic Analysis of Rechargeable Batteries in a Photovoltaic Power Supply System

We developed a model for the probabilistic behavior of a rechargeable battery acting as the energy storage component in a photovoltaic power supply system. Stochastic and deterministic models are created to simulate the behavior of the system component;. The components are the solar resource, the photovoltaic power supply system, the rechargeable battery, and a load. Artificial neural networks are incorporated into the model of the rechargeable battery to simulate damage that occurs during deep discharge cycles. The equations governing system behavior are combined into one set and solved simultaneously in the Monte Carlo framework to evaluate the probabilistic character of measures of battery behavior.

High Level Trigger Configuration and Handling of Trigger Tables in the CMS Filter Farm

The CMS experiment at the CERN Large Hadron Collider is currently being commissioned and is scheduled to collect the first pp collision data in 2008. CMS features a two-level trigger system. The Level-1 trigger, based on custom hardware, is designed to reduce the collision rate of 40 MHz to approximately 100 kHz. Data for events accepted by the Level-1 trigger are read out and assembled by an Event Builder. The High Level Trigger (HLT) employs a set of sophisticated software algorithms, to analyze the complete event information, and further reduce the accepted event rate for permanent storage and analysis. This paper describes the design and implementation of the HLT Configuration Management system. First experiences with commissioning of the HLT system are also reported.

Observation of Localized Corrosion of Ni-Based Alloys Using Coupled Orientation Imaging Microscopy and Atomic Force Microscopy

We present a method for assessing the relative vulnerabilities of distinct classes of grain boundaries to localized corrosion. Orientation imaging microscopy provides a spatial map which identifies and classifies grain boundaries at a metal surface. Once the microstructure of a region of a sample surface has been characterized, a sample can be exposed to repeated cycles of exposure to a corrosive environment alternating with topographic measurement by an atomic force microscope in the same region in which the microstructure had been mapped. When this procedure is applied to Ni and Ni-based alloys, we observe enhanced attack at random grain boundaries relative to special boundaries and twins in a variety of environments.

Direct numerical simulations of type Ia supernovae flames I: The landau-darrieus instability

Planar flames are intrinsically unstable in open domains due to the thermal expansion across the burning front--the Landau-Darrieus instability. This instability leads to wrinkling and growth of the flame surface, and corresponding acceleration of the flame, until it is stabilized by cusp formation. We look at the Landau-Darrieus in stability for C/O thermonuclear flames at conditions relevant to the late stages of a Type Ia supernova explosion. Two-dimensional direct numerical simulations of both single-mode and multi-mode perturbations using a low Mach number hydrodynamics code are presented. We show the effect of the instability on the flame speed as a function of both the density and domain size, demonstrate the existence of the small scale cutoff to the growth of the instability, and look for the proposed breakdown of the non-linear stabilization at low densities. The effects of curvature on the flame as quantified through measurements of the growth rate and computation of the corresponding Markstein number. While accelerations of a few percent are observed, they are too small to have any direct outcome on the supernova explosion.
