Analyses of inter- and intra-patient variation in the V3 loop of the HIV-1 envelope protein

The third hypervariable domain of the HIV-1 gp120 envelope protein (V3) has been the focus of intensive sequencing efforts. To date, nearly one thousand V3 loop sequences have been stored in the HIV sequence database. Studies have revealed that the V3 loop elicits potent type-specific immune responses, and that it plays a significant role in cell tropism and fusion . The immunogenic tip of the loop can serve as a type-specific neutralizing antibody epitope, as well as a cytotoxic T-cell epitope. A helper T-cell epitope that lies within the amino terminal half of the V3 loop has also been characterized. Despite the richness of the immunologic response to this region, its potential for variation makes it an elusive target for vaccine design. Analyses of sibling sequence sets (sets of viral sequences derived from one person) show that multiple forms of the immunogenic tip of the loop are found within most HIV-1 infected individuals. Viral V3 sequences obtained from epidemiologically unlinked individuals from North America and Europe show extensive variation. This document is an end-of-contract report, prepared by Hydro Geo Chem for Los Alamos National Laboratory under contract number 9-XDD-6329F-1. The ultimate goal of this work is to characterize historical infiltration and unsaturated flow in the Yucca Mountain area of the Nevada Test Site. Work on this contract has focused on using chloride, bromide, stable chlorine isotopes, and chlorine-36 distributions to evaluate the depth of infiltration in the unsaturated zone. Effort in support of this work has included developing analytical procedures, exploring ways in which to separate the. meteoric component from the rock component, and meeting quality assurance requirements.

The following calculations arranged in a spreadsheet format derive the flowrate from both ECN relieving devices. In this case it is assumed that the ECN is full of liquid argon and it is in its steady state cooling mode. One of the other cryostats is assumed to be cooling down while the other is being filled with LAr. Other assumptions in this analysis include: (1) Pressure in the cryostat is 19.75 psig (1.16X(MAWP+FV)). (2) Gaseous Nitrogen is concurrently flowing in the vent piping at a rate of 3477 lb/hr. This is derived from 0.3 gpm required for ECN steady state conditions, 4 gpm required for cooldown (max.), and 5 gpm required for filling with LAr (max.). (3) Mixture mass flows are at their maximum at the junction of the relief device outlets on the ECN (GN2 mass flow actually increases gradually at junctions toward the ECS and there is a short segment of piping between the GAr outlets and the condenser exhaust). Field trip guide to selected outcrops, Arbuckle Mountains, Oklahoma

The Arbuckle Mountains, named for Brigadier General Matthew Arbuckle, are located in south-central Oklahoma. The formations that comprise the Arbuckle Mountains have been extensively studied for hydrocarbon source rock and reservoir rock characteristics that can be applied to the subsurface in the adjacent Anadarko and Ardmore basins. Numerous reports and guidebooks have been written concerning the Arbuckle Mountains. A few important general publications are provided in the list of selected references. The purpose of this handout is to provide general information on the geology of the Arbuckle Mountains and specific information on the four field trip stops, adapted from the literature. The four stops were at: (1) Sooner Rock and Sand Quarry; (2) Woodford Shale; (3) Hunton Anticline and Hunton Quarry; and (4) Tar Sands of Sulfur Area. As part of this report, two papers are included for more detail: Paleomagnetic dating of basinal fluid migration, base-metal mineralization, and hydrocarbon maturation in the Arbuckle Mountains, Oklahoma and Laminated black shale-bedded chert cyclicity in the Woodford Formation, southern Oklahoma.

The overall objective of the program was to improve the understanding of retrograde reactions and their dependencies on coal rank and structure, and/or coal modifications and reaction conditions. Because retrograde reactions are competitive with bond breaking reactions, an understanding of both is required to shift the competition in favor of the latter. Related objectives were to clarify the conflicting observations reported in literature on such major topics as the role of oxygen groups in retrograde reactions and to provide a bridge from very fundamental studies on pure compounds to phenomenological studies on actual coal. This information was integrated into the FG-DVC model, which was improved and extended to the liquefaction context.

Mixed waste treatment with a mediated electrochemical process

The process described in this paper is intended to convert mixed waste containing toxic organic compounds (not heavy metals) to ordinary radioactive waste, which is treatable. The process achieves its goal by oxidizing hydrocarbons to CO{sub 2} and H{sub 2}O. Other atoms that may be present in the toxic organic generally are converted to nonhazardous anions such as sulfate and phosphate. This electro chemical conversion is performed at conditions of temperature and pressure that are just moderately above ambient conditions. Gaseous hydroxides and oxyhydroxides that are formed by many radionuclides during incineration cannot form in this process. 1 ref., 3 figs.

Molecular Design Concept for X-Ray Laser Research

The goal of this program is the construction of an x-ray laser in the kilovolt regime. Recent experimental results indicate that a new technique for the generation of strong amplification of x-ray wavelengths is feasible. It involves the combination of (1) a new ultrahigh brightness subpicosecond laser technology and (2) a recently discovered unique mode of strong-field interaction, particularly applicable to molecules. A concept of molecular x-ray laser design emerges from these considerations which matches the mode of excitation to the structure of the molecular system. The molecular approach enables the combination of very highly electronically excited conditions with an environment characteristic of dense cold matter, a general situation exceptionally conducive to x-ray amplification. Both high efficiency and wavelength tunability are intrinsic features of this method. 19 refs., 4 figs.

Moving granular-bed filter development program topical report. Base contract test plan

The Westinghouse Science & Technology Center has proposed a novel moving granular-bed filter concept, the Standleg Moving Granular-Bed Filter (S-MGBF) system, that overcomes the inherent deficiencies of the current state-of-the-art moving granular-bed filter technology. The S-MGBF system combines two unique features that make it highly effective for use in advanced coal-fueled power plants. First, the S-MGBF system applies pelletization technology to generate filter pellets from the power plant solid waste materials, and uses these pellets as a ``once-through`` filtering media to eliminate the need for costly, complex, and large filter media recycling equipment. This pelletizing step also generates a more environmentally acceptable solid waste product and provides the potential to incorporate gas-phase contaminant sorbents into the filtering media. Secondly, the S-MGBF system passes these pellets and the flyash laden power plant gas through a highly compact S-MGBF that uses cocurrent gas-pellet contacting in an arrangement that greatly simplifies and enhances the distribution of dirty gas to the moving bed and the disengagement of clean gas from the moving bed.

Catalytic studies of the earlier investigators also (3) show similar influence on the product selectivity and indicate reproducible performance is critically dependent on the control and rigor of the preparation technique. To compliment the NMR results, we have made a thorough investigation of the Hysteresis character of the Cu/Co catalysts with the metal ratio varying from 0.2 to 4.0.

Oxidation of hazardous waste in supercritical water: A comparison of modeling and experimental results for methanol destruction

Recent experiments at Sandia National Laboratories conducted in conjunction with MODEC Corporation have demonstrated successful clean- up of contaminated water in a supercritical water reactor. These experiments targeted wastes of interest to Department of Energy production facilities. In this paper we present modeling and experimental results for a surrogate waste containing 98% water, 2% methanol, and parts per million of chlorinated hydrocarbons and laser dyes. Our initial modeling results consider only methanol and water. Experimental data are available for inlet and outlet conditions and axial temperature profiles along the outside reactor wall. The purpose of our model is to study the chemical and physical processes inside the reactor. We are particularly interested in the parameters that control the location of the reaction zone. The laboratory-scale reactor operates at 25 MPa., between 300 K and 900 K; it is modeled as a plug-flow reactor with a specified temperature profile. We use Chemkin Real-Gas to calculate mixture density, with the Peng-Robinson equation of state. The elementary reaction set for methanol oxidation and reactions of other C{sub 1} and C{sub 2} hydrocarbons is based on previous models for gas-phase kinetics. Results from our calculations show that the methanol is 99.9% destroyed at 1/3 …

Shock-interface interaction: Current research on the Richtmyer- Meshkov problem

The basis for the study of the evolution of a shocked interface stems from the question of the Rayleigh-Taylor (RT) instability (Taylor 1950). Starting in the late 18th century, the stability of an interface submitted to gravitational forces was investigated for the case in which the density of one of the materials across the interface was negligible compared to the other. Taylor analyzed the case in which the Atwood number (ratio of the difference of the densities to their sum) is less than 1, and the acceleration of the system is constant. He determined that the interface was unstable to small perturbations only if the direction of the acceleration normal to the interface coincides with that of the density gradient. Richtmyer (1960) extended Taylor's analysis to the case of an implusive acceleration. His results implied that the interface would be unstable irrespective of the relative orientation of the velocity impulse and the density gradient. His predictions were verified experimentally by Meshkov (1969), and the Richtmyer-Meshkov (RM) instability became a subject of research in its own right.
