Genetic Modification of Fatty Acid Profiles in Cotton

The industrial uses of cottonseed oil are limited by its fatty acid composition. Genetic modification of cotton lipid profiles using seed-specific promoters could allow cotton growers to produce valuable new oils in the seed without adverse effects on fiber quality and yield, therefore making this crop more commercially profitable. Transgenic cotton callus harboring a diverged fatty acid desaturase gene (FADX) from Momordica charantia was characterized for production of alpha-eleostearic acid (conjugated double bonds: 18:3 D9 cis, 11 trans, 13 trans), not normally found in cotton. Gas chromatography (GC) in conjunction with mass spectrometry (MS) confirmed production of alpha-eleostearic acid in the transgenic cotton tissues. A second series of transformation experiments introduced the cotton fatty acid thioesterase B (FATB) cDNA, fused to the seed-specific oleosin promoter into cotton to promote the over-expression of FATB, to generate cotton with increased palmitate in the cottonseed. PCR amplification, as well as fatty acid analysis by gas chromatography, confirmed introduction of the FATB cDNA in transgenic tissues. Collectively, these results demonstrate the feasibility of manipulating the fatty acid composition in cotton via transgenic approaches and form the basis for continued efforts to create novel oils in cottonseed.

A Study of Breath Management as Treated by Four Major American Vocal Pedagogues: Appelman, Reid, Vennard, and Miller

Trained musicians cannot use the same breath process in daily living as for singing. Also, the normal breath cycle applied to speech is not efficient. Therefore, students who are learning to sing need to know proper breathing techniques. In this thesis, I will describe the breathing process and the correct way to breathe while singing, based on studies of four American pedagogues; Appleman, Reid, Vennard and Miller. To understand the breathing process for singing, it is necessary to study and understand the anatomical system and the mechanics of the respiratory system. Therefore, the first chapter contains anatomical system of breath management. Then, in the second chapter, the specific breath management techniques of four American pedagogues will be examined and compared. Three of them, Appelman, Vennard, and Miller, suggested some exercises in order to develop correct and efficient breathing habits.

Temperature tolerances and predation susceptibilities of transgenic and wildtype zebra danios, Danio rerio.

Both the upper and lower temperature tolerances of red fluorescent protein transgenic zebra danios and wildtype zebra danios, Danio rerio, were significantly different via two different methods; however, all differences are small (< 1°C) and probably not ecologically important. The U.S. geographic distributions of both transgenic and wildtype zebra danios will not be restricted by their upper thermal tolerances, but will be limited to the southern and western portions of the U.S. by their lower thermal tolerances. Largemouth bass did not preferentially prey upon transgenic zebra danios compared to wildtype danios or wildtypes relative to a native fish. If transgenic or wildtype zebra danios are released into southern or western U.S. waters, it is possible they could be eliminated by predation.

An exploration of parental sensitivity and child cognitive and behavioral development.

The current study attempted to show the relationship of paternal sensitivity and maternal sensitivity and their possible influences on child cognitive and behavioral development. This study used data collected as part of the National Institute for Child Health and Human Development (NICHD) Study of Early Child Care, which is a longitudinal, multi-site study. Correlation and regression analyses were computed to examine relationships between the variables at child age 6 and 36 months. Results indicated paternal sensitivity was a significant positive predictor of child cognitive abilities and a negative predictor of both fathers' reports of children's externalizing and internalizing behaviors. Maternal sensitivity was a significant negative predictor of mothers' reports of children's externalizing behaviors. Interpretations of these results and directions for future research are discussed.

The Effects of Two Types of Consequence Delivery on Task Acquisition.

The effects of two consequence delivery methods on task acquisition were evaluated within a multi-element design. A typical 3 year-old child and a 4 year-old child with autism participated in this study. The task for both children was to select a picture after the experimenter said its name. The consequence in one condition consisted of the experimenter handing the edible item to the children. The consequence in the other condition consisted of the children retrieving the edible item directly from the apparatus, located in a crevice underneath each picture. Results show slightly quicker acquisition in the condition where children retrieved the edible consequence. However, it is possible that other variables had greater influence on the task acquisition.

Truancy Intervention: A Study of Dallas Independent School District Participants in the Dallas Challenge Truancy and Class C Enforcement Center

This research examines agency data for participants in a truancy intervention program. Previous literature provides mostly descriptive information and fails to examine the effects of truancy intervention efforts. The analysis provides a profile of truant participants referred to the program and factors that correlate to successful completion of the program requirements. The results will be applicable to the study program as evaluation research and will be generalized for application to other truancy programs. Directions for future truancy research will be suggested based on the need to continue to evaluate truancy reduction efforts.

Education for Education's Sake? Exposing the Arts District of Downtown Dallas

This thesis discusses the relatively new approach of art education, by paralleling it to Marxist ideology on art. The Dallas Arts District is one example of a city where museum art education is in conflict: being adopted more vigorously by some and with less acceptance by others. In order to provide a glimpse into the museum ideology of downtown Dallas, previous schools of thought regarding the role of curators and the introduction of educators into museums will be detailed, as well as conflicts between these two factions. The following questions will be addressed: Is museum art education truly a movement which strives to infuse the American culture with a greater appreciation of art? Is there a link to overcoming Marx's key issue of class? How is the movement affecting the Dallas Arts District and to what extent is museum art education being utilized within this forum? Is the emphasis toward museum art education greater in Dallas than in other large cities across the United States, and if so, how has that affected the cities' patrons?

The Recorder Tutors in 't Uitnement Kabinet (open access)

The Recorder Tutors in 't Uitnement Kabinet

Paulus Matthysz, a prominent music printer in Amsterdam during the seventeenth century, published Jacob van Eyck's Der Fluyten Lust-hof and a collection entitled 't Uitnement Kabinet. Three extant copies of Lust-hof include a tutor Vertoninge...op de Handt-fluit, presumably by Matthysz, and a tutor by Gerband van Blanckenburgh, Onderwyzinge...op deHandt-Fluyt. Their content is not correlated with Lust-hof, and they were presumably designed for inclusion in the Kabinet II. Confusion over the tutors' conception has led to published misinformation jeopardizing their historical worth. The casual generalizations regarding the two tutors can be refuted by reestablishing the interrelationship between the tutors and the two collections. This paper employs a comprehensive study into their origins in order to rectify how the tutors are referenced in the twenty-first century.

Effects of Fluency and Accuracy-Only Training on Acquisition and Retention of Letter Naming by Individuals with Traumatic Brain Injury

This study examines the effects of accuracy-only training and fluency training on retention of material learned. Two adolescent participants with traumatic brain injuries were taught to name 2 sets of lowercase Greek letters. Each of the 2 sets consisted of 7 letters. Practice and rate of reinforcement were controlled for in this study. Fluency trained letters showed higher retention (percent correct during retention checks) than the accuracy-only trained letters.

Effect of Early Childhood Teacher Characteristics on Classroom Practice, Quality, and Child Abilities

The National Institute of Child Health and Human Development (NICHD) is funding and supporting a longitudinal study of Early Child Care. Beginning in 1991, data was collected from ten sites across the United States and included 1,364 families with a newborn child. This study used the NICHD Early Child Care data set to investigate characteristics of teachers that provide childcare in a daycare-like setting or childcare centers. Specifically, the relationship between early childhood teacher endorsement of developmentally appropriate belief systems and teacher education in early childhood were examined to determine their potential influence on the teachers' developmentally appropriate classroom practices, global rating of classroom quality, and child cognitive abilities. These relationships were examined at two time periods- at child age 36 months and child age 54 months. The results indicated that many of these relationships were significant. Interestingly, many of the significant findings were present only at child age 54 months.

Teaching Two Children with Autism to Follow a Computer-Mediated Activity Schedule Utilizing Microsoft® PowerPoint® Presentation Software

Children with autism typically exhibit deficits in behavior and also in visual processing. Development and implementation of visually-cued instructional procedures, combined with electronic technology, have been used successfully to teach children with autism complex behavior chains. This study used photographic activity schedules on computer slideshow software to teach two children with autism to follow computer-mediated cues and engage in four play activities, and to transition between each activity in their homes without the presence of a trained behavior therapist. Results of this study demonstrated that these technologies can be utilized in children's homes to promote computer-mediated play behavior while eliminating the necessary cost of a home behavior therapist to prompt and supervise such activities.

Organizational development: A comparison of individual and organizational level change.

Organizational change and development (OCD) has been studied by researchers to identify the effectiveness of change initiatives. Because of the broad scope of interventions in OCD, these studies have covered a range of areas including multiple interventions and the methodological rigor used by researchers. However, few have looked at organizational versus individual change within an organization, to examine whether individual change is more effective than organizational change. The purpose of this study is to determine if organizational change occurs in a top down or bottom up manner. A meta-analysis was conducted using 238 field experiments. Each study was coded for intervention and organizational outcome and for individual or organizational level variables. Effect sizes were calculated for each study, each level, and each level by intervention and outcome measure. Results indicate that while OCD interventions overall had a moderate effect size, the level of intervention or outcome was not a moderating variable.

Stories: Strange Men and Thinking Girls

What is the boundary between fiction and nonfiction? What happens if the line between the two is crossed? Can we possibly recall events in our lives exactly as they happened? In creative nonfiction, such as memoir, the audience expects the writer to recall things exactly as they happened, with no embellishments, re-ordering, additions, or subtractions. It seems as if authors of creative nonfiction are bound to be questioned about events, nitpicked on details, challenged on memories, and accused of portraying real-life people the "wrong" way. Yet when the writer creates fiction, it seems to go the other way: readers like to think there are parallels between an author and her stories. Readers congratulate themselves for finding the similarities between the two, and instead of focusing on the crafted story at hand, try to search out which parts are "true" and which are embellished. Does any of this matter, though; don't all stories tell a kind of truth? We have an insatiable urge to classify, to "know" the truth, but truth isn't merely a recollection of cold facts; likewise, a story isn't any less true if it's fiction.

Booze, Boomtowns, and Burning Crosses: The Turbulent Governorship of Pat M. Neff of Texas, 1921-1925

Pat M. Neff served as governor of Texas from 1921 to 1925, a period marked by political conflict between rural conservatives and urban progressives. Neff, a progressive, found himself in the middle of this conflict. Neff supported prohibition, declared martial law in the oil boomtown of Mexia, and faced the rise of the Ku Klux Klan as a political force in Texas. Though often associated with the Klan, Neff did not approve of the organization and worked against it whenever possible. During the Railroad Shopmen's Strike of 1922, Neff stalled the federal government in its demand he send troops to Denison just long enough to win re-nomination. William Jennings Bryan mentioned Neff as a possible candidate for the presidency in 1924, but he pursued a back-door strategy that alienated his political base among Texas Democrats.

A Content Analysis of Mozambican Newspapers' Coverage of the 2004 Presidential Election

This study focuses on the amount of coverage given by four major tabloid newspapers-Demos, Zambeze, Savana and Domingo-to the candidates of the major political parties Renamo and Frelimo, during the 2004 presidential race. The number of stories of both parties in those newspapers were counted and calculated by chi-square to determine how much one party was covered than the other identifying signs of balance or bias. The research showed that there was a significant result of 42 percent of likelihood that stories in the four newspapers would either be about Frelimo or Renamo. However, the study also revealed that Frelimo was the party covered most often by Demos, Zambeze and Savana while Renamo was covered most often by Domingo.

Clarifying Variables associated with Problem Behaviors Using Structured Descriptive Assessment

This study evaluated the utility of a structured descriptive assessment (SDA) as an alternative method of functional assessment. Initially, an analogue functional analysis, conducted to assess the problem behavior of two adults with developmental disabilities, produced inconclusive results. Subsequently, SDAs was conducted in the individuals' natural environment with the direct-contact caregivers acting as therapists. This assessment manipulated antecedent variables similarly to the analogue functional analysis but allowed for consequences to occur naturally. The results from SDAs suggested that problem behaviors, for both participants, were occasioned by removal of personal items and maintained by their return. Treatments based on the results of SDAs were implemented in a reversal design and resulted in a notable reduction in the occurrences of problem behavior for both participants. These outcomes suggest that SDA procedures may be useful when results from the analogue functional analysis are inconclusive.

Shoreline Erosion at Mad Island Marsh Preserve, Matagorda County, Texas

The Nature Conservancy of Texas (TNC) is concerned with the amount of shoreline erosion taking place at its Mad Island Marsh Preserve (MIMP), located in Matagorda Bay, Texas. The MIMP is a 7,100 acre nature preserve that borders the Gulf Intracoastal Waterway and is eroded by waves generated by barge traffic. TNC is concerned that erosion will shorten Mad Island Bayou which may increase the salinity of Mad Island Lake; with detrimental effects on lake and marsh habitats. This study uses GPS technology to map the current shoreline and GIS to determine ten year erosion rates (1995 - 2005). Results show that erosion is occurring at various rates along the shoreline as well as along the oxbow bend in Mad Island Bayou.

Mutation of Polaris, an Intraflagellar Transport Protein, Shortens Neuronal Cilia

Primary cilia are non-motile organelles having 9+0 microtubules that project from the basal body of the cell. While the main purpose of motile cilia in mammalian cells is to move fluid or mucus over the cell surface, the purpose of primary cilia has remained elusive for the most part. Primary cilia are shortened in the kidney tubules of Tg737orpk mice, which have polycystic kidney disease due to ciliary defects. The product of the Tg737 gene is polaris, which is directly involved in a microtubule-dependent transport process called intraflagellar transport (IFT). In order to determine the importance of polaris in the development of neuronal cilia, cilium length and numerical density of cilia were quantitatively assessed in six different brain regions on postnatal days 14 and 31 in Tg737orpk mutant and wildtype mice. Our results indicate that the polaris mutation leads to shortening of cilia as well as decreased percentage of ciliated neurons in all brain regions that were quantitatively assessed. Maintainance of cilia was especially affected in the ventromedial nucleus of the hypothalamus. Furthermore, the polaris mutation curtailed cilium length more severely on postnatal day 31 than postnatal day 14. Measuring Team Meeting Success: Does Everyone Really Need to Participate?

Facilitators are encouraged to get all meeting attendees to participate in the meeting. There is the assumption made that, if they do participate, then this participation will increase the group's general satisfaction of the meeting. Also, knowing the factors that can increase the probability of a successful meeting has been a focus of previous research, yet attendee participation has not been studied. The current research study empirically examines participation's effect on meeting evaluations. This study is a field experiment conducted in a team-based organization, where successful meetings are critical. Data was collected on the amount of participation of team members in their weekly team meeting and their evaluations of the meeting. After running correlations and a principal components analysis, this study found a relationship between participation and meeting evaluations. A scale of meeting success was also created.

Environmental Virtue Education: Ancient Wisdom Applied

The focus of environmental philosophy has thus far heavily depended on the extension of rights to nonhuman nature. Due to inherent difficulties with this approach to environmental problems, I propose a shift from the contemporary language of rights and duties to the concept of character development. I claim that a theory of environmental virtue ethics can circumvent many of the difficulties arising from the language of rights, duties, and moral claims by emphasizing the cultivation of certain dispositions in the individual moral agent. In this thesis, I examine the advantages of virtue ethics over deontological and utilitarian theories to show the potential of developing an ecological virtue ethic. I provide a preliminary list of ecological virtues by drawing on Aristotle's account of traditional virtues as well as on contemporary formulations of environmental virtues. Then, I propose that certain types of rules (rules of thumb) are valuable for the cultivation of environmental virtues, since they affect the way the moral agent perceives a particular situation. Lastly, I offer preliminary formulations of these rules of thumb.

Embedded Within Landscapes: Agrarian Philosophy and Sustainable Agriculture

Small-scale, conservation-based agrarianism provides a model for sustainable human habitation within heterogeneous landscapes. Thoreau's Transcendentalism and the historical roots of American Agrarianism are explored as influences for wilderness preservation and the New Agrarian movement. Idealizing a distant wilderness too often means overlooking the ecological and socio-economic environment where people live. Middle landscapes between nature and culture, or between wilderness and cities, can either increase or reduce ecological and social functioning within the landscape matrix. Managing middle landscapes by agrarian principles helps move both nature and culture towards ecological, economic, and social sustainability. This thesis ends with a discussion of agrarian themes, such as supporting decentralized local economies and increasing community connectivity, applied in urban, rural, and wilderness landscapes.

Do Shared S-minus Functions Among Stimuli Lead to Equivalence?

We examined the claim that equivalence classes contain all positive elements in a reinforcement contingency by asking whether negative stimuli in a reinforcement contingency will also form an equivalence class, based on their shared function as S-minus stimuli. In Experiment 1, 5 subjects were tested for equivalence for positive and negative stimuli. Testing of positive stimuli preceded testing of negative stimuli. Two of five subjects demonstrated equivalence for positive stimuli, and three subjects demonstrated equivalence for negative stimuli. In Experiment 2, order of testing was reversed. Four of six subjects demonstrated equivalence for positive stimuli, and none demonstrated equivalence for negative stimuli. In Experiment 3, positive and negative stimuli were tested together. Only one of five subject demonstrated equivalence for positive and negative stimuli. These data suggest that negative stimuli may enter an equivalence class, and so Sidman paradigm should be expanded. Order of testing was found as a meaningful variable.

Shaping: From art to science.

This study evaluated the effectiveness of a procedure for teaching a caregiver to shape vocal language in a young child with autism. A multiple baseline design was employed to assess caregiver use of shaping procedures, child vocal language progress, and the social validity of the procedures. Following baseline and introductory sessions, the coach and caregiver reviewed video from the previous session and the coach gave descriptive feedback to the caregiver about her performance. Following the review of the videotaped segment, procedures to increase skills were selected and practiced. Rates of responsive opportunity arrangement, model presentation, responsive model delivery, and responsive event delivery, as well as the child's rate of requests, vocalizations, diversity of vocalizations, and social validity were measured. Data suggested that the procedures effectively taught the skill of shaping to a caregiver, which in turn seemed to produce increases in the child's vocal responding.
