Accelerated Stem Growth Rates and Improved Fiber Properties of Loblolly Pine: Functional Analysis Of CyclinD from Pinus taeda

A sustained supply of low-cost, high quality raw materials is essential for the future success of the U.S. forest products industry. To maximize stem (trunk) growth, a fundamental understanding of the molecular mechanisms that regulate cell divisions within the cambial meristem is essential. We hypothesize that auxin levels within the cambial meristem regulate cyclin gene expression and this in turn controls cell cycle progression as occurs in all eukaryotic cells. Work with model plant species has shown that ectopic overexpression of cyclins promotes cell division thereby increasing root growth > five times. We intended to test whether ectopic overexpression of cambial cyclins in the cambial zone of loblolly pine also promotes cell division rates that enhance stem growth rates. Results generated in model annual angiosperm systems cannot be reliably extrapolated to perennial gymnosperms, thus while the generation and development of transgenic pine is time consuming, this is the necessary approach for meaningful data. We succeeded in isolating a cyclin D gene and Clustal analysis to the Arabidopsis cyclin D gene family indicates that it is more closely related to cyclin D2 than D1 or D3 Using this gene as a probe we observed a small stimulation of cyclin D expression …

Accurate and efficient radiation transport in optically thick media -- by means of the Symbolic Implicit Monte Carlo method in the difference formulation

The equations of radiation transport for thermal photons are notoriously difficult to solve in thick media without resorting to asymptotic approximations such as the diffusion limit. One source of this difficulty is that in thick, absorbing media thermal emission is almost completely balanced by strong absorption. In a previous publication [SB03], the photon transport equation was written in terms of the deviation of the specific intensity from the local equilibrium field. We called the new form of the equations the difference formulation. The difference formulation is rigorously equivalent to the original transport equation. It is particularly advantageous in thick media, where the radiation field approaches local equilibrium and the deviations from the Planck distribution are small. The difference formulation for photon transport also clarifies the diffusion limit. In this paper, the transport equation is solved by the Symbolic Implicit Monte Carlo (SIMC) method and a comparison is made between the standard formulation and the difference formulation. The SIMC method is easily adapted to the derivative source terms of the difference formulation, and a remarkable reduction in noise is obtained when the difference formulation is applied to problems involving thick media.

Active Control for Statistically Stationary Turbulent PremixedFlame Simulations

The speed of propagation of a premixed turbulent flame correlates with the intensity of the turbulence encountered by the flame. One consequence of this property is that premixed flames in both laboratory experiments and practical combustors require some type of stabilization mechanism to prevent blow-off and flashback. The stabilization devices often introduce a level of geometric complexity that is prohibitive for detailed computational studies of turbulent flame dynamics. Furthermore, the stabilization introduces additional fluid mechanical complexity into the overall combustion process that can complicate the analysis of fundamental flame properties. To circumvent these difficulties we introduce a feedback control algorithm that allows us to computationally stabilize a turbulent premixed flame in a simple geometric configuration. For the simulations, we specify turbulent inflow conditions and dynamically adjust the integrated fueling rate to control the mean location of the flame in the domain. We outline the numerical procedure, and illustrate the behavior of the control algorithm on methane flames at various equivalence ratios in two dimensions. The simulation data are used to study the local variation in the speed of propagation due to flame surface curvature.

Adaptive Management and Planning Models for Cultural Resources in Oil and Gas Fields in New Mexico and Wyoming

In 2002, Gnomon, Inc., entered into a cooperative agreement with the U.S. Department of Energy (DOE) National Energy Technology Laboratory (NETL) for a project entitled, Adaptive Management and Planning Models for Cultural Resources in Oil and Gas Fields in New Mexico and Wyoming (DE-FC26-02NT15445). This project, funded through DOE’s Preferred Upstream Management Practices grant program, examined cultural resource management practices in two major oil- and gas-producing areas, southeastern New Mexico and the Powder River Basin of Wyoming (Figure 1). The purpose of this project was to examine how cultural resources have been investigated and managed and to identify more effective management practices. The project also was designed to build information technology and modeling tools to meet both current and future management needs. The goals of the project were described in the original proposal as follows: Goal 1. Create seamless information systems for the project areas. Goal 2. Examine what we have learned from archaeological work in the southeastern New Mexico oil fields and whether there are better ways to gain additional knowledge more rapidly or at a lower cost. Goal 3.
