Enhanced geothermal systems (EGS) using CO2 as working fluid - Anovelapproach for generating renewable energy with simultaneoussequestration of carbon

Responding to the need to reduce atmospheric emissions of carbon dioxide, Donald Brown (2000) proposed a novel enhanced geothermal systems (EGS) concept that would use CO{sub 2} instead of water as heat transmission fluid, and would achieve geologic sequestration of CO{sub 2} as an ancillary benefit. Following up on his suggestion, we have evaluated thermophysical properties and performed numerical simulations to explore the fluid dynamics and heat transfer issues in an engineered geothermal reservoir that would be operated with CO{sub 2}. We find that CO{sub 2} is superior to water in its ability to mine heat from hot fractured rock. CO{sub 2} also has certain advantages with respect to wellbore hydraulics, where larger compressibility and expansivity as compared to water would increase buoyancy forces and would reduce the parasitic power consumption of the fluid circulation system. While the thermal and hydraulic aspects of a CO{sub 2}-EGS system look promising, major uncertainties remain with regard to chemical interactions between fluids and rocks. An EGS system running on CO{sub 2} has sufficiently attractive features to warrant further investigation.

Chemical imaging of biological materials by NanoSIMS using isotopic and elemental labels

The NanoSIMS 50 combines unprecedented spatial resolution (as good as 50 nm) with ultra-high sensitivity (minimum detection limit of {approx}200 atoms). The NanoSIMS 50 incorporates an array of detectors, enabling simultaneous collection of 5 species originating from the same sputtered volume of a sample. The primary ion beam (Cs{sup +} or O{sup -}) can be scanned across the sample to produce quantitative secondary ion images. This capability for multiple isotope imaging with high spatial resolution provides a novel new approach to the study of biological materials. Studies can be made of sub-regions of tissues, mammalian cells, and bacteria. Major, minor and trace element distributions can be mapped on a submicron scale, growth and metabolism can be tracked using stable isotope labels, and biogenic origin can be determined based on composition. We have applied this technique extensively to mammalian and prokaryotic cells and bacterial spores. The NanoSIMS technology enables the researcher to interrogate the fate of molecules of interest within cells and organs through elemental and isotopic labeling. Biological applications at LLNL will be discussed.

Long Pulse High Performance Plasma Scenario Development for the National Spherical Torus Experiment

The National Spherical Torus Experiment [Ono et al., Nucl. Fusion, 44, 452 (2004)] is targeting long pulse high performance, noninductive sustained operations at low aspect ratio, and the demonstration of nonsolenoidal startup and current rampup. The modeling of these plasmas provides a framework for experimental planning and identifies the tools to access these regimes. Simulations based on neutral beam injection (NBI)-heated plasmas are made to understand the impact of various modifications and identify the requirements for (1) high elongation and triangularity, (2) density control to optimize the current drive, (3) plasma rotation and/or feedback stabilization to operate above the no-wall limit, and (4) electron Bernstein waves (EBW) for off-axis heating/current drive (H/CD). Integrated scenarios are constructed to provide the transport evolution and H/CD source modeling, supported by rf and stability analyses. Important factors include the energy confinement, Zeff, early heating/H mode, broadening of the NBI-driven current profile, and maintaining q(0) and qmin>1.0. Simulations show that noninductive sustained plasmas can be reached at IP=800 kA, BT=0.5 T, 2.5, N5, 15%, fNI=92%, and q(0)>1.0 with NBI H/CD, density control, and similar global energy confinement to experiments. This Work Plan outlines the scope of work for targeted sampling at Webber, Kansas (Figure 1.1). This activity is being conducted at the request of the Kansas Department of Health and Environment (KDHE), in accordance with Section V of the Intergovernmental Agreement between the KDHE and the Commodity Credit Corporation of the U.S. Department of Agriculture (CCC/USDA). Data obtained in this sampling event will be used to (1) evaluate the current status of previously detected contamination at Webber and (2) determine whether the site requires further action. This work is being performed on behalf of the CCC/USDA by the Environmental Science Division of Argonne National Laboratory. Argonne is a nonprofit, multidisciplinary research center operated by the University of Chicago for the U.S. Department of Energy (DOE). The CCC/USDA has entered into an interagency agreement with DOE, under which Argonne provides technical assistance to the CCC/USDA with environmental site characterization and remediation at its former grain storage facilities. Development of a Performance-Based Industrial Energy Efficiency Indicator for Corn Refining Plants.

Organizations that implement strategic energy management programs have the potential to achieve sustained energy savings if the programs are carried out properly. A key opportunity for achieving energy savings that plant managers can take is to determine an appropriate level of energy performance by comparing their plant's performance with that of similar plants in the same industry. Manufacturing facilities can set energy efficiency targets by using performance-based indicators. The U.S. Environmental Protection Agency (EPA), through its ENERGY STAR{reg_sign} program, has been developing plant energy performance indicators (EPIs) to encourage a variety of U.S. industries to use energy more efficiently. This report describes work with the corn refining industry to provide a plant-level indicator of energy efficiency for facilities that produce a variety of products--including corn starch, corn oil, animal feed, corn sweeteners, and ethanol--for the paper, food, beverage, and other industries in the United States. Consideration is given to the role that performance-based indicators play in motivating change; the steps needed to develop indicators, including interacting with an industry to secure adequate data for an indicator; and the actual application and use of an indicator when complete. A program called ``Innovative Billing?? has been developed to provide individualized energy information for a mass audience?the entireresidential customer base of an electric or gas utility. Customers receive a graph on the bill that compares that customer?s consumption with othersimilar customers for the same month. The program aims to stimulate customers to make ef?ciency improvements. To group as many as severalmillion customers into small ``comparison groups??, an automated method must be developed drawing solely from the data available to the utility.This paper develops and applies methods to compare the quality of resulting comparison groups.A data base of 114,000 customers from a utility billing system was used to evaluate Innovative Billing comparison groups, comparing fouralternative criteria: house characteristics (?oor area, housing type, and heating fuel); street; meter read route; billing cycle. Also, customers wereinterviewed to see what forms of comparison graphs made most sense and led to fewest errors of interpretation. We ?nd that good qualitycomparison groups result from using street name, meter book, or multiple house characteristics. Other criteria we tested, such as entire cycle, entiremeter book, or single house characteristics such as ?oor area, resulted in poor quality comparison groups. Genetic Diversity Among Botulinum Neurotoxin Producing Clostridial Strains

Clostridium botulinum is a taxonomic designation for many diverse anaerobic spore forming rod-shaped bacteria which have the common property of producing botulinum neurotoxins (BoNTs). The BoNTs are exoneurotoxins that can cause severe paralysis and even death in humans and various other animal species. A collection of 174 C. botulinum strains were examined by amplified fragment length polymorphism (AFLP) analysis and by sequencing of the 16S rRNA gene and BoNT genes to examine genetic diversity within this species. This collection contained representatives of each of the seven different serotypes of botulinum neurotoxins (BoNT A-G). Analysis of the16S rRNA sequences confirmed earlier reports of at least four distinct genomic backgrounds (Groups I-IV) each of which has independently acquired one or more BoNT serotypes through horizontal gene transfer. AFLP analysis provided higher resolution, and can be used to further subdivide the four groups into sub-groups. Sequencing of the BoNT genes from serotypes A, B and E in multiple strains confirmed significant sequence variation within each serotype. Four distinct lineages within each of the BoNT A and B serotypes, and five distinct lineages of serotype E strains were identified. Subtask 1.18 - A Decision Tool for Watershed-Based Effluent Trading

Handling produced water in an economical and environmentally sound manner is vital to coalbed methane (CBM) development, which is expected to increase up to 60% in the next 10-15 years as the demand for natural gas increases. Current produced water-handling methods (e.g., shallow reinjection and infiltration impoundments) are too costly when implemented on a well-by-well basis. A watershed-based effluent credit trading approach may be a means of managing produced water at reduced cost while meeting or surpassing water quality regulations. This market-based approach allows for improved water quality management by enabling industrial, agricultural, and municipal discharge facilities to meet water quality permit requirements by purchasing pollutant reduction credits from other entities within the same watershed. An evaluation of this concept was conducted for the Powder River Basin (PRB) of Montana and Wyoming by the Energy & Environmental Research Center (EERC). To conduct this assessment, the EERC collected and evaluated existing water quality information and developed the appropriate tools needed to assess the environmental and economic feasibility of specific trading scenarios. Evaluation of 2004 Toyota Prius Hybrid Electric Drive System

The 2004 Toyota Prius is a hybrid automobile equipped with a gasoline engine and a battery- and generator-powered electric motor. Both of these motive-power sources are capable of providing mechanical-drive power for the vehicle. The engine can deliver a peak-power output of 57 kilowatts (kW) at 5000 revolutions per minute (rpm) while the motor can deliver a peak-power output of 50 kW over the speed range of 1200-1540 rpm. Together, this engine-motor combination has a specified peak-power output of 82 kW at a vehicle speed of 85 kilometers per hour (km/h). In operation, the 2004 Prius exhibits superior fuel economy compared to conventionally powered automobiles. To acquire knowledge and thereby improve understanding of the propulsion technology used in the 2004 Prius, a full range of design characterization studies were conducted to evaluate the electrical and mechanical characteristics of the 2004 Prius and its hybrid electric drive system. These characterization studies included (1) a design review, (2) a packaging and fabrication assessment, (3) bench-top electrical tests, (4) back-electromotive force (emf) and locked rotor tests, (5) loss tests, (6) thermal tests at elevated temperatures, and most recently (7) full-design-range performance testing in a controlled laboratory environment. Biosynthesis of the Cyclotide MCoTI-II using an Engineered Intein

Cyclotides are an emerging family of naturally occurring circular mini-proteins ({approx}30-40 amino acids) characterized by six conserved Cys residues (forming 3 disulfide bridges) that create a topologically unique structure designated as a cyclic cysteine knot (CCK). The cysteine knot motif, which is embedded within the macrocylic backbone, is described as two disulfide bridges that form a ring that is penetrated by the third disulfide bridge. The cyclic backbone and CCK motif together confer cyclotides with a remarkable stability and resistance to proteolytic, chemical, and thermal degradation. Further, cyclotides are functionally diverse and display a wide range of functions including uterotonic activity, trypsin inhibition, cytotoxicity, neurotensin binding, anti-HIV, antimicrobial, and insecticidal activity. Together, these characteristics make cyclotides attractive candidates for both drug design and agricultural applications, both in their native forms and as molecular scaffolds for the incorporation of novel bioactivities. [1] The ability to manipulate production of cyclotides within biological systems is critical for mutagenesis studies, production of grafted products, and the mass production of cyclotides with novel activities. Targeting the tumor microenvironment

Despite some notable successes cancer remains, for the most part, a seemingly intractable problem. There is, however, a growing appreciation that targeting the tumor epithelium in isolation is not sufficient as there is an intricate mutually sustaining synergy between the tumor epithelial cells and their surrounding stroma. As the details of this dialogue emerge, new therapeutic targets have been proposed. The FDA has already approved drugs targeting microenvironmental components such as VEGF and aromatase and many more agents are in the pipeline. In this article, we describe some of the 'druggable' targets and processes within the tumor microenvironment and review the approaches being taken to disrupt these interactions.

This research effort examines the post-detonation environmental, safety, health and operational aspects of experimental explosive tests with mercury. Specific experimental information is necessary for the evaluation of post-detonation by-products in comparison with those potentially resulting from mercury-bearing material accumulation in biomass accumulation areas, such as landfills, from batteries, electrical switches, thermometers, and fluorescent lights (Lindberg et al 2001). This will assist in determining appropriate abatement techniques for cleaning the work environment and environmental mitigation to determine waste stream components and risk assessment protocol. Determination of the by-products for personal protection equipment and personal exposure monitoring parameters are also part of this experimental work.

A Markov-Chain Monte-Carlo Based Method for Flaw Detection in Beams

A Bayesian inference methodology using a Markov Chain Monte Carlo (MCMC) sampling procedure is presented for estimating the parameters of computational structural models. This methodology combines prior information, measured data, and forward models to produce a posterior distribution for the system parameters of structural models that is most consistent with all available data. The MCMC procedure is based upon a Metropolis-Hastings algorithm that is shown to function effectively with noisy data, incomplete data sets, and mismatched computational nodes/measurement points. A series of numerical test cases based upon a cantilever beam is presented. The results demonstrate that the algorithm is able to estimate model parameters utilizing experimental data for the nodal displacements resulting from specified forces.

The long-planned and much-needed merger of EnviroCAT into the Materials Research Collaborative Access Team (MR-CAT) will provide dedicated state-of-the-art facilities that are critical to research on a broad range of issues in environmental sciences. These CATs will focus on developing a bending magnet (BM) beamline for x-ray absorption fine structure (XAFS) and micro x-ray analysis of environmental samples through integration with existing insertion device (ID) capabilities in XAFS, micro x-ray analysis, and x-ray scattering. In addition, the expanded MR-CAT will serve as the hub of personnel and laboratory infrastructure support for molecular environmental science and biogeochemical science at the Advanced Photon Source (APS). In conjunction with the merger of EnviroCAT into MR-CAT, the US Environmental Protection Agency (EPA) will become a member institution of MR-CAT, joining the present members (University of Notre Dame, Illinois Institute of Technology, University of Florida, British Petroleum, and Argonne's Chemical Engineering and Biosciences Division). The motivation for blending capabilities meeting the needs of EnviroCAT users into the MR-CAT facilities is the explosion of synchrotron-radiation-based research in the field known as molecular environmental science (MES). The complete chloroplast genome sequence of Pelargonium xhortorum: Or ganization and evolution of the largest and most highlyrearranged chloroplast genome of land plants

The chloroplast genome of Pelargonium e hortorum has beencompletely sequenced. It maps as a circular molecule of 217,942 bp, andis both the largest and most rearranged land plant chloroplast genome yetsequenced. It features two copies of a greatly expanded inverted repeat(IR) of 75,741 bp each, and consequently diminished single copy regionsof 59,710 bp and 6,750 bp. It also contains two different associations ofrepeated elements that contribute about 10 percent to the overall sizeand account for the majority of repeats found in the genome. Theyrepresent hotspots for rearrangements and gene duplications and include alarge number of pseudogenes. We propose simple models that account forthe major rearrangements with a minimum of eight IR boundary changes and12 inversions in addition to a several insertions of duplicated sequence.The major processes at work (duplication, IR expansion, and inversion)have disrupted at least one and possibly two or three transcriptionaloperons, and the genes involved in these disruptions form the core of thetwo major repeat associations. Sculpting the shape of semiconductor heteroepitaxial islands: fromdots to rods

In the Ge on Si model heteroepitaxial system, metal patterns on the silicon surface provide unprecedented control over the morphology of highly ordered Ge islands. Island shape including nanorods and truncated pyramids is set by the metal species and substrate orientation. Analysis of island faceting elucidates the prominent role of the metal in promoting growth of preferred facet orientations while investigations of island composition and structure reveal the importance of Si-Ge intermixing in island evolution. These effects reflect a remarkable combination of metal-mediated growth phenomena that may be exploited to tailor the functionality of island arrays in heteroepitaxial systems.

IMG provides graphicalviewers, summaries and occurrence profile tools for comparing genes,pathways and functions (terms) across specific genomes. Quantifying the Diffusion of a Fluid through Membranes by RemoteDetection MRI

We present a method to measure self-diffusion acrossmembranes without the need for concentration or pressure gradients.Hyperpolarized xenon in combination with remote detection of NMR allowsthe measurement of membrane permeation, even in the gas phase. Theresulting images allow quantification of the amount of fluid diffusedthrough the membrane, and represent an alternative, potentially moreprecise way of measuring a membrane diffusion coefficient. The use ofremote detection of NMR allows for non-invasive signal encoding coupledto sensitive detection, making this approach ideal for the study ofdiffusion in intact devices such as fuel cells or separationsystems.

Modeling the Integrated Performance of Dispersion and Monolithic U-Mo Based Fuels

The evaluation and prediction of integrated fuel performance is a critical component of the Reduced Enrichment for Research and Test Reactors (RERTR) program. The PLATE code is the primary tool being developed and used to perform these functions. The code is being modified to incorporate the most recent fuel/matrix interaction correlations as they become available for both aluminum and aluminum/silicon matrices. The code is also being adapted to treat cylindrical and square pin geometries to enhance the validation database by including the results gathered from various international partners. Additional modeling work has been initiated to evaluate the thermal and mechanical performance requirements unique to monolithic fuels during irradiation.
