Transportation Research: Opportunities for Improving the Oversight of DOT's Research Programs and User Satisfaction with Transportation Statistics

A letter report issued by the Government Accountability Office with an abstract that begins "The Department of Transportation's (DOT) research, development, and technology (RD&T) budget totaled $1.1 billion in fiscal year 2005. DOT's Research and Innovative Technology Administration (RITA)--which includes the Bureau of Transportation Statistics (BTS)--oversees DOT's RD&T activities. GAO examined (1) how RITA's responsibilities for overseeing DOT's RD&T activities differ from those of its predecessor, the Research and Special Programs Administration (RSPA); (2) RITA's practices for coordinating, facilitating, and reviewing RD&T activities; (3) the progress DOT has made in implementing GAO's 2003 recommendations on how to improve the coordination and evaluation of RD&T activities; and (4) how BTS identifies and monitors how well it serves its users. To address these issues, GAO reviewed relevant documentation and interviewed officials from RITA, BTS, and three operating administrations."

The Border Patrol implemented the Border Safety Initiative (BSI) in 1998 with the intention of reducing injuries and preventing deaths among migrants that attempt to cross the border illegally. GAO assessed: (1) Trends in the numbers, locations, causes, and characteristics of border-crossing deaths. (2) Differences among the Border Patrol sectors in implementing the BSI methodology. (3) The extent to which existing data allow for an evaluation of the effectiveness of the BSI and other efforts to prevent border-crossing deaths."

Biosynthesis of the Cyclotide MCoTI-II using an Engineered Intein

Cyclotides are an emerging family of naturally occurring circular mini-proteins ({approx}30-40 amino acids) characterized by six conserved Cys residues (forming 3 disulfide bridges) that create a topologically unique structure designated as a cyclic cysteine knot (CCK). The cysteine knot motif, which is embedded within the macrocylic backbone, is described as two disulfide bridges that form a ring that is penetrated by the third disulfide bridge. The cyclic backbone and CCK motif together confer cyclotides with a remarkable stability and resistance to proteolytic, chemical, and thermal degradation. Further, cyclotides are functionally diverse and display a wide range of functions including uterotonic activity, trypsin inhibition, cytotoxicity, neurotensin binding, anti-HIV, antimicrobial, and insecticidal activity. Together, these characteristics make cyclotides attractive candidates for both drug design and agricultural applications, both in their native forms and as molecular scaffolds for the incorporation of novel bioactivities. [1] The ability to manipulate production of cyclotides within biological systems is critical for mutagenesis studies, production of grafted products, and the mass production of cyclotides with novel activities. My adviser's hope is to achieve this capability by employing recombinant DNA expression techniques to generate large combinatorial libraries of cyclotides. HWMA/RCRA Closure Plan for the CPP-648 Radioactive Solid and Liquid Waste Storage Tank System (VES-SFE-106)

This Hazardous Waste Management Act/Resource Conservation and Recovery Act closure plan for the Radioactive Solid and Liquid Waste Storage Tank System located in the adjacent to the Sludge Tank Control House (CPP-648), Idaho Nuclear Technology and Engineering Center, Idaho National Laboratory, was developed to meet the interim status closure requirements for a tank system. The system to be closed includes a tank and associated ancillary equipment that were determined to have managed hazardous waste. The CPP-648 Radioactive Solid and Liquid Waste Storage Tank System will be "cleaned closed" in accordance with the requirements of the Hazardous Waste Management Act/Resource Conservation and Recovery Act as implemented by the Idaho Administrative Procedures Act and 40 Code of Federal Regulations 265. This closure plan presents the closure performance standards and methods of acheiving those standards for the CPP-648 Radioactive Solid and Liquid Waste Storage Tank System.

CH-TRU Waste Content Codes (CH-TRUCON)

The CH-TRU Waste Content Codes (CH-TRUCON) document describes the inventory of the U.S. Department of Energy (DOE) CH-TRU waste within the transportation parameters specified by the Contact-Handled Transuranic Waste Authorized Methods for Payload Control (CH-TRAMPAC). The CH-TRAMPAC defines the allowable payload for the Transuranic Package Transporter-II (TRUPACT-II) and HalfPACT packagings. This document is a catalog of TRUPACT-II and HalfPACT authorized contents and a description of the methods utilized to demonstrate compliance with the CH-TRAMPAC. A summary of currently approved content codes by site is presented in Table 1. The CH-TRAMPAC describes "shipping categories" that are assigned to each payload container. Multiple shipping categories may be assigned to a single content code. A summary of approved content codes and corresponding shipping categories is provided in Table 2, which consists of Tables 2A, 2B, and 2C. Table 2A provides a summary of approved content codes and corresponding shipping categories for the "General Case," which reflects the assumption of a 60-day shipping period as described in the CH-TRAMPAC and Appendix 3.4 of the CH-TRU Payload Appendices. Longitudinal Double Spin Asymmetry in Inclusive Jet Production at STAR

This contribution reports on the first measurement of the longitudinal double-spin asymmetry A{sub LL} for the inclusive production of jets in polarized proton-proton collisions at {radical}s = 200 GeV. The data were collected with STAR at RHIC in the years 2003 and 2004, and correspond to a sampled integrated luminosity of 0.3 pb{sup -1} with beam polarizations up to 45%. The results on A{sub LL} cover jet transverse momenta 5 < p{sub T} < 17 GeV/c and agree with perturbative QCD evaluations based on deep-inelastic scattering parametrizations for the gluon polarization in the proton. The results disfavor large positive gluon polarization in the polarized proton.

Modeling and Optimization of Direct Chill Casting to Reduce Ingot Cracking

Approximately 68% of the aluminum produced in the United States is first cast into ingots prior to further processing into sheet, plate, extrusions, or foil. The direct chill (DC) semi-continuous casting process has been the mainstay of the aluminum industry for the production of ingots due largely to its robust nature and relative simplicity. Though the basic process of DC casting is in principle straightforward, the interaction of process parameters with heat extraction, microstructural evolution, and development of solidification stresses is too complex to analyze by intuition or practical experience. One issue in DC casting is the formation of stress cracks [1-15]. In particular, the move toward larger ingot cross-sections, the use of higher casting speeds, and an ever-increasing array of mold technologies have increased industry efficiencies but have made it more difficult to predict the occurrence of stress crack defects. The Aluminum Industry Technology Roadmap [16] has recognized the challenges inherent in the DC casting process and the control of stress cracks and selected the development of 'fundamental information on solidification of alloys to predict microstructure, surface properties, and stresses and strains' as a high-priority research need, and the 'lack of understanding of mechanisms of cracking as a function …

The effect of temporal pulse shape on optical damage

The conditions under which optical materials are susceptible to laser-induced damage is a topic which has been the subject of considerable study. Laser parameters such as wavelength and temporal pulse duration have been studied extensively. Until this work the effect of temporal pulse shape has not been considered. We present here data from a simple single-parameter model and a supporting experiment which predicts that a Flat-In-Time-pulse will produce damage at approximately 80% of the fluence of a Gaussian pulse of the same FWHM duration.

Nanoplasmonic molecular ruler for nuclease activity and DNAfootprinting

We have constructed a nanoplasmonic molecular ruler, which can perform label-free and real-time monitoring of DNA length changes and perform DNA footprinting. The ruler was created by tethering double-stranded DNA to single Au nanoparticles. The scattering spectra of Au-DNA nanoconjugates showed red-shifted peak plasmon resonance wavelength dependent on DNA length, which can be measured with sub-nanometer axial resolution, averaging {approx}1.24 nm peak wavelength shift per DNA base pair. The spectra of individual Au-DNA nanoconjugates in the presence of nuclease showed a time-resolved dependence on the reaction dynamics, allowing quantitative, kinetic and real-time measurement of nuclease activity. The ruler was further developed into a new DNA footprinting platform. We showed the specific binding of a protein to DNA and the accurate mapping of its footprint. This work promises a very fast and convenient platform for mapping DNA-protein interactions, for nuclease activity monitoring, and for other DNA size-based methods.

Using Cable Suspended Submersible Pumps to Reduce Production Costs to Increase Ultimate Recovery in the Red Mountain Field of the San Juan Basin Region

A joint venture between Enerdyne LLC, a small independent oil and gas producer, and Pumping Solutions Inc., developer of a low volume electric submersible pump, suspended from a cable, both based in Albuquerque, New Mexico, has re-established marginal oil production from Red Mountain Oil Field, located in the San Juan Basin, New Mexico by working over 17 existing wells, installing cable suspended submersible pumps ( Phase I ) and operating the oil field for approximately one year ( Phase II ). Upon the completion of Phases I and II ( Budget Period I ), Enerdyne LLC commenced work on Phase III which required additional drilling in an attempt to improve field economics ( Budget Period II ). The project was funded through a cooperative 50% cost sharing agreement between Enerdyne LLC and the National Energy Technology Laboratory (NETL), United States Department of Energy, executed on April 16, 2003. The total estimated cost for the two Budget Periods, of the Agreement, was $1,205,008.00 as detailed in Phase I, II & III Authorization for Expenditures (AFE). This report describes tasks performed and results experienced by Enerdyne LLC during the three phases of the cooperative agreement.

With financial support from the U.S. Department of Energy (DOE), Layman Energy Associates, Inc. (LEA) has completed a program of geothermal exploration at the Truckhaven area in Imperial County, California. The exploratory work conducted by LEA included the following activities: compilation of public domain resource data (wells, seismic data, geologic maps); detailed field geologic mapping at the project site; acquisition and interpretation of remote sensing imagery such as aerial and satellite photographs; acquisition, quality control and interpretation of gravity data; and acquisition, quality control and interpretation of resistivity data using state of the art magnetotelluric (MT) methods. The results of this exploratory program have allowed LEA to develop a structural and hydrologic interpretation of the Truckhaven geothermal resource which can be used to guide subsequent exploratory drilling and resource development. Of primary significance, is the identification of an 8 kilometer-long, WNW-trending zone of low resistivity associated with geothermal activity in nearby wells. The long axis of this low resistivity zone is inferred to mark a zone of faulting which likely provides the primary control on the distribution of geothermal resources in the Truckhaven area. Abundant cross-faults cutting the main WNW-trending zone in its western half may indicate elevated fracture permeability …

HYDROCARBON-DEGRADING BACTERIA AND SURFACTANT ACTIVITY

Fate of benzene ethylbenzene toluene xylenes (BTEX) compounds through biodegradation was investigated using two different bacteria, Ralstonia picketti (BP-20) and Alcaligenes piechaudii (CZOR L-1B). These bacteria were isolated from extremely polluted petroleum hydrocarbon contaminated soils. PCR and Fatty Acid Methyl Ester (FAME) were used to identify the isolates. Biodegradation was measured using each organism individually and in combination. Both bacteria were shown to degrade each of the BTEX compounds. Alcaligenes piechaudii biodegraded BTEXs more efficiently while mixed with BP-20 and individually. Biosurfactant production was observed by culture techniques. In addition 3-hydroxy fatty acids, important in biosurfactant production, was observed by FAME analysis. In the all experiments toluene and m+p- xylenes were better growth substrates for both bacteria than the other BTEX compounds. In addition, the test results indicate that the bacteria could contribute to bioremediation of aromatic hydrocarbons (BTEX) pollution increase biodegradation through the action by biosurfactants.

Small Mammal Sampling in Mortandad and Los Alamos Canyons, 2005

As part of an ongoing ecological field investigation at Los Alamos National Laboratory, a study was conducted that compared measured contaminant concentrations in sediment to population parameters for small mammals in the Mortandad Canyon watershed. Mortandad Canyon and its tributary canyons have received contaminants from multiple solid waste management units and areas of concern since establishment of the Laboratory in the 1940s. The study included three reaches within Effluent and Mortandad canyons (E-1W, M-2W, and M-3) that had a spread in the concentrations of metals and radionuclides and included locations where polychlorinated biphenyls and perchlorate had been detected. A reference location, reach LA-BKG in upper Los Alamos Canyon, was also included in the study for comparison purposes. A small mammal study was initiated to assess whether potential adverse effects were evident in Mortandad Canyon due to the presence of contaminants, designated as contaminants of potential ecological concern, in the terrestrial media. Study sites, including the reference site, were sampled in late July/early August. Species diversity and the mean daily capture rate were the highest for E-1W reach and the lowest for the reference site. Charge state dependence of cathodic vacuum arc ion energy andvelocity distributions

In the literature, conflicting conclusions are reported concerning the charge state dependence of cathodic arc ion energy and velocity distributions. It appears that data from electrostatic energy analyzers indicate charge state dependence of ion energy, whereas time-of-flight methods support charge state independence of ion velocity. Here we present charge-state-resolved ion energy distributions and calculate the corresponding ion velocity distributions in aluminum vacuum arc plasma. We show that the conflicting conclusions reported in the literature for the two different characterization techniques may originate from the commonly employed data interpretation of energy and velocity, in which peak values and average values are not carefully distinguished.

Simulating Electron Clouds in High-Current Ion Accelerators with Solenoid Focusing

Contamination from electrons is a concern for the solenoid-focused ion accelerators being developed for experiments in high-energy-density physics (HEDP). These electrons are produced directly by beam ions hitting lattice elements and intercepting diagnostics, or indirectly by ionization of desorbed neutral gas, and they are believed responsible for time dependence of the beam radius, emittance, and focal distance seen on the Solenoid Transport Experiment (STX) at Lawrence Berkeley National Laboratory. The electrostatic particle-in-cell code WARP has been upgraded to included the physics needed to simulate electron-cloud phenomena. We present preliminary self-consistent simulations of STX experiments suggesting that the observed time dependence of the beam stems from a complicated interaction of beam ions, desorbed neutrals, and electrons.

Under-sampling in a Multiple-Channel Laser Vibrometry System

Laser vibrometry is a technique used to detect vibrations on objects using the interference of coherent light with itself. Most vibrometry systems process only one target location at a time, but processing multiple locations simultaneously provides improved detection capabilities. Traditional laser vibrometry systems employ oversampling to sample the incoming modulated-light signal, however as the number of channels increases in these systems, certain issues arise such a higher computational cost, excessive heat, increased power requirements, and increased component cost. This thesis describes a novel approach to laser vibrometry that utilizes undersampling to control the undesirable issues associated with over-sampled systems. Undersampling allows for significantly less samples to represent the modulated-light signals, which offers several advantages in the overall system design. These advantages include an improvement in thermal efficiency, lower processing requirements, and a higher immunity to the relative intensity noise inherent in laser vibrometry applications. A unique feature of this implementation is the use of a parallel architecture to increase the overall system throughput. This parallelism is realized using a hierarchical multi-channel architecture based on off-the-shelf programmable logic devices (PLDs).

Draft Guidance: Response, Restoration, and Recovery Checklist for Biologically Contaminated Facilities

The Checklist for Facility Response, Restoration, and Recovery presented in this document is principally focused on the Consequence Management Phase of a biothreat agent (i.e., Bacillus anthracis) release at a large facility, such as an airport or subway. Information in this document conforms to the National Response Plan (NRP) (DHS 2004) and the National Incident Management System (NIMS 2004). Under these two guidance documents, the personnel responsible for managing biological response and recovery efforts--that is, the decision-makers--are members of an Incident Command (IC), which is likely to transition to a Unified Command (UC) in the event of a biological warfare agent attack. A UC is used when more than one agency has incident jurisdiction or when incidents cross political jurisdictions. The location for primary, tactical-level command and management is referred to as the Incident Command Post (ICP), as described in the NRP. Thus, regardless of whether an IC or an UC is used, the responsible entities are located at an ICP. Agencies work together through designated members of the UC to establish their designated Incident Commanders at a single ICP and to establish a common set of objectives and strategies and a single Incident Action Plan.
