Characterization of a 1,4-. beta. -D-glucan synthase from Dictyostelium discoideum

Various aspects of research concerning Dictyostelium discoideum are presented. The initial focus of this project was upon: the characterization of potential probes for the cellulose synthase (antibody and nucleic acid), the determination of the cultural induction conditions of cellulose synthesis, the solubilization of the enzyme activity, the development of a non-inhibitory disruption buffer, the generation and isolation of mutant strains deficient in cellulose synthesis, and the development of the capability to determine the degree of polymerization of the in vitro product. I have briefly summarized our most significant findings with only selected data sets being shown in this report in the interest of brevity.

Evaluation of Gas-Reburning and Low NOx Burners on a Wall Fired Boiler

Low NO{sub x} burners operate on the principle of delayed mixing between the coal fuel and burner air, so that less NO{sub x} is formed. Gas reburning is a combustion modification technique that consists of firing 80--85 percent of the fuel corresponding to the total heat release in the lower furnace. Reduction of NO{sub x} to molecular nitrogen (N{sub 2}) is accomplished via the downstream injection of the remaining fuel requirement in the form of natural gas (which also reduces the total SO{sub x} emissions). In a third stage, burnout air is injected at the lower temperatures in the upper furnace to complete the combustion process without generating significant additional NO{sub x}. The specific goal of this project is to demonstrate NO{sub x} emission reductions of 75 percent or more as a result of combing Low NO{sub x} Burners and Gas Reburning on a utility boiler having the design characteristics mentioned above. A Host Site Agreement has been signed by EER and a utility company in the State of Colorado: Public Service Company of Colorado (Cherokee Unit No. 3, 172 MW{sub e}) front wall fired boiler near Denver.

Air gun test evaluation

A mechanical shock testing apparatus is used for testing the response of components subject to large accelerations in hostile environments. The test acceleration is provided by the impact of a bullet against a plate on which the component to be tested is mounted. This report describes a series of experiments that were performed to determine the dependence of the air gun test apparatus performance on incremental changes in the hardware configurations, changes in the pressure used to drive the bullet, and different accelerometers. The effect of variation of these experimental factors on the measured acceleration was determined using a Taguchi screening experimental design. Experimental settings were determined that can be used to operate the tester with a measured output within acceleration specifications.

Role of structure in ion movement of glasses

Can the structure of a melt much above the glass transition temperature (Tg) determine the structure and (transport) properties of glass In other words, are there extremely long lasting configurations in a melt which would affect ion transport in the glassy state A major effort during the past year has been to address these very fundamental questions. An important consideration here is to separate the effect of the variable melt structure from that of a variable cooling rate in the glass transformation range. For this reason our experiment consists of preparing 0.3 Na{sub 2}O--0.7 B{sub 2}O{sub 3} glasses from the melt which is first equilibrated at 1400 C and then annealed at 850 C for 0 to 180 minutes. From 850 C variously annealed melts are quenched to the glassy state by following identical procedure. If the structure (as reflected in ion transport) of 1400 C melt relaxes to that of 850 C in {approximately} minutes, we may expect to observe variations in the conductivity time in the ns range.

Under the Phase II, Alternative Energy Resources Development (AERD) project of the United States Agency for International Development (USAID) and the Government of India (GOI), five collaborative coal projects have been initiated in the areas of: (1) NO{sub x}/SO{sub x} control from coal-fired power plants, (2) slagging combustor development for high-ash Indian coals, (3) characterization of Indian coals for combustion and gasification, (4) diagnostic studies for prediction of power plant life expectancy, and (5) environmental and natural resource analysis of coal cycle. The Pittsburgh Energy Technology Center (PETC) has the implementation responsibility for these projects. The Indian collaborative institutions identified for these projects are the Bharat Heavy Electricals Ltd. (BHEL), Trichy, (Projects 1--4), and the Tata Energy Research Institute (TERI) for Project 5. The Oak Ridge National Laboratory (ORNL) is providing cross-cut technical coordination and support for these five projects.

Spin motion of electrons in the SLC linac

It is generally expected that the depolarizing effects of the linear accelerator RF fields will be small. Recently Bill Atwood raised the question whether this conclusion is still correct in view of the fact that the particles in the SLC spend a larger fraction of their time at phase angles off crest'' due to BNS damping; since radial fields are in quadrature with the accelerating field this might imply that depolarizing effects are larger. On the other hand, because of the smaller emittance of the SLC relative to the earlier linac radial excursions would be smaller. The anticipation is therefore that the depolarizing effect will again be negligible but it might be worthwhile to update the early calculations of SLAC TN-63-97 revised. This paper discusses these updates.

Enchancing the use of coals by gas reburning-sorbent injection

The objective of this project is to evaluate and demonstrate a cost effective emission control technology for acid rain precursors, oxides of nitrogen (NO{sub x}) and sulfur (SO{sub x}), on two coal fired utility boilers in Illinois. The units selected are representative of pre-NSPS design practices: tangential and cyclone fired. Work on a third unit, wall fired, has been stopped because of funding limitations. The specific objectives are to demonstrate reductions of 60 percent in NO{sub x} and 50 percent in SO{sub x} emissions, by a combination of two developed technologies, gas reburning (GR) and sorbent injection (SI). With GR, about 80--85 percent of the coal fuel is fired in the primary combustion zone. The balance of the fuel is added downstream as natural gas to create a slightly fuel rich environment in which NO{sub x} is converted to N{sub 2}. The combustion process is completed by overfire air addition. SO{sub x} emissions are reduced by injecting dry sorbents (usually calcium based) into the upper furnace. The sorbents trap SO{sub x} as solid sulfates that are collected in the particulate control device.

Hydrocarbons associated with brines from geopressured wells

The purpose of this research is to determine the concentration of the cryocondensates in fluids of the various USDOE Geopressured wells as a function of production volume, to correlate the production of these compounds with reservoir and well production characteristics, to precisely measure solubilities of cryocondensates components in water and sodium chloride solutions (brines) as a function of ionic strength and temperature and the component's distribution coefficients between these solutions and oil, to develop models of the reservoir which are consistent with the data obtained, to monitor the wells for the production of aliphatic oils and relate any such production with the data obtained, and to develop a harsh environment pH probe for use in well brines. Results are summarized.

Molecular mapping of chromosomes 17 and X

Progress toward the construction of high density genetic maps of chromosomes 17 and X has been made by isolating and characterizing a relatively large set of polymorphic probes for each chromosome and using these probes to construct genetic maps. We have mapped the same polymorphic probes against a series of chromosome breakpoints on X and 17. The probes could be assigned to over 30 physical intervals on the X chromosome and 7 intervals on 17. In many cases, this process resulted in improved characterization of the relative locations of the breakpoints with respect to each other and the definition of new physical intervals. The strategy for isolation of the polymorphic clones utilized chromosome specific libraries of 1--15 kb segments from each of the two chromosomes. From these libraries, clones were screened for those detecting restriction fragment length polymorphisms. The markers were further characterized, the chromosomal assignments confirmed and in most cases segments of the original probes were subcloned into plasmids to produce probes with improved signal to noise ratios for use in the genetic marker studies. The linkage studies utilize the CEPH reference families and other well-characterized families in our collection which have been used for genetic disease linkage work. …

Wind system documentation

Atmospheric transport and diffusion models have been developed by the Environmental Technology Section (ETS) of the Savannah River Technology Center to calculate the location and concentration of toxic or radioactive materials during an accidental release at the Savannah River Site (SRS). The output from these models has been used to support initial on-site and off-site emergency response activities such as protective action decision making and field monitoring coordination. These atmospheric transport and diffusion models have been incorporated into an automated computer-based system called the (Weather Information and Display) System and linked to real-time meteorological and radiological monitoring instruments to provide timely information for these emergency response activities (Hunter, 1990). This report documents various aspects of the WIND system.

Radioiodine in the Savannah River Site environment

Radioiodine, which is the collective term for all radioactive isotopes of the element iodine, is formed at the Savannah River Site (SRS) principally as a by-product of nuclear reactor operations. Part of the radioiodine is released to the environment during reactor and reprocessing operations at the site. The purpose of this report is to provide an introduction to radioiodine production and disposition, its status in the environment, and the radiation dose and health risks as a consequence of its release to the environment around the Savannah River Plant. A rigorous dose reconstruction study is to be completed by thee Center for Disease Control during the 1990s.

Confined zone dispersion flue gas desulfurization demonstration

This is the sixth quarterly report for this project and it covers work performed on Phase 3a of the project from February 1, 1992 through April 30, 1992. Extension of the parametric test period through June 1992 provides an opportunity to regain most of the schedule slippage, but only if the modifications needed for continuous operation of the CZD system are installed concurrent with the extended test period. These modifications include automation of the lime preparation and transfer system, automatic injection control, and related instrumentation and controls as necessary to integrate the operation of the CZD system with Seward Station Boiler No. 15. Early installation of these modifications would permit testing, debugging and adjustment of the automatic control system during the parametric test period. Results of current testing indicate that considerable testing and adjustment will be required to optimize operation of the CZD system after it is automated for continuous operation. Therefore, we intend to incorporate in Phase 3a(parametric testing) the system modifications needed for continuous automatic operation that were originally included in Phase 3b. Phase 3b would then be limited only to the one-year continuous demonstration.

Flue gas conditioning for improved particle collection in electrostatic precipitators

It is concluded that the laboratory tests should be conducted at high levels of SO[sub 3] such that the resulting resistivity is in the range of 10[sup 7]--10[sup 8] ohm-cm. There are several reasons leading to this conclusion. At SO[sub 3] concentrations of 30 ppM and greater, the curves for both dew point and resistivity are relatively flat so that changes in gas phase SO[sub 3] will have minimal impact on particle characteristics. In addition, the electrostatic forces are relatively flat in this range so that changes in flue gas conditions will that result in a change in resistivity by up to two orders of magnitude will have little effect on the magnitude of reentrainment. Finally, at the very low resistivity conditions, reentrainment will be the highest. Since the purpose of the laboratory resistivity tests is to determine the relative ability of the various additives to reduce resistivity, the greater the reentrainment, the easier it will be to measure an improvement. Tests were conducted by first operating at baseline conditions with no additives and then repeating the test with additives. The research was in the area of solid cluster compounds containing metal clusters. Two classes of materials were studied: those based on solutions formed by dissolving LiMo[sub 3]Se[sub 3] in polar solvents, and those based on M[sub 6]X[sub 8] clusters. The cluster phases were synthesized and their properties examined. Nb[sub 6]I[sub 11] and Mo[sub 6]X[sub 8] were also studied.

Study of Pu consumption in light water reactors: Evaluation of GE advanced boiling water reactor plants, compilation of Phase 1C task reports

This report summarizes the evaluations conducted during Phase 1C of the Pu Disposition Study have provided further results which reinforce the conclusions reached during Phase 1A & 1B: These conclusions clearly establish the benefits of the fission option and the use of the ABWR as a reliable, proven, well-defined and cost-effective means available to disposition the weapons Pu. This project could be implemented in the near-term at a cost and on a schedule being validated by reactor plants currently under construction in Japan and by cost and schedule history and validated plans for MOX plants in Europe. Evaluations conducted during this phase have established that (1) the MOX fuel is licensable based on existing criteria for new fuel with limited lead fuel rod testing, (2) that the applicable requirements for transport, handling and repository storage can be met, and (3) that all the applicable safeguards criteria can be met.

Surfactant studies for bench-scale operation. Second quarterly technical progress report, October 1, 1992--December 31, 1992

A phase II study has been initiated to investigate surfactant-assisted coal liquefaction, with the objective of quantifying the enhancement in liquid yields and product quality. This report covers the second quarter of work. The major accomplishments were (1) completion of coal liquefaction autoclave reactor runs with Illinois No. 6 coal at processing temperatures of 300, 325, and 350{degrees}C, and pressures of 1800 psig, (2) analysis of the filter cake and the filtrate obtained from the treated slurry in each run, and (3) correlation of the coal conversions and the liquid yield quality to the surfactant concentration. An increase in coal conversions and upgrading of the liquid product quality due to surfactant addition was observed for all runs.

This Act was substantially amended for the first time in 1992 by passage of the Energy Policy Act (EPACT). The report also includes a discussion of the issues which led to the amendment of PUHCA and projections of the impact of these changes on the electric industry. The report should be of use to Federal and State regulators, trade associations, electric utilities, independent power producers, as well as decision-makers in Congress and the Administration.
