Development of an In Vitro Protoplast Culture System for Albizia Lebek (L.) Benth., an Economically Important Leguminous Tree

An in vitro system of generating protoplasts from their callus cultures was established. The friable callus was more productive in terms of producing protoplasts than the green compact callus. The concentration of the various cell wall degrading enzymes had an effect on the viability of the protoplasts in the medium. The protoplast system developed from the experiments was stable and could be used for the transformation experiments of Albizia lebek and for other plant improvement practices.

Analyses of Particulate Contaminants in Semiconductor Processing Fluids

Particle contamination control is a critical issue for the semiconductor industry. In the near future, this industry will be concerned with the chemical identities of contaminant particles as small as 0.01 pm in size. Therefore, analytical techniques with both high chemical sensitivity and spatial resolution are required. Transmission electron microscopy (TEM) provides excellent spatial resolution and yields structural and compositional information. It is rarely used, however, due to the difficulty of sample preparation. The goals of this research are to promote the use of TEM as an ultrafine particle analysis tool by developing new sample preparation methods, and to exploit the new TEM techniques for analysis of particles in semiconductor processing fluids. A TEM methodology for the analysis of particulate contaminants in fluids with an elemental detectability limit as low as 0.1 part per trillion (ppt), and a particle concentration detectability limit as low as 1 particle/ml for particles greater than 0.2 pm was developed and successfully applied to the analysis of particles in HF, H202, de-ionized (DI) water, and on the surface of an electronic device. Dalbergia and Albizia: Plantlet Production via Tissue Culture, Karyological Evaluation, and Seed Anatomy with Scanning Electron Microscopy

A publication by the National Academy of Sciences, USA (1979) outlined some of the research need for a great variety of economically important woody species whose remaining genetic resources need urgently to be collected and conserved. A viable regeneration system was established via tissue and cell suspension culture for Albizia falcataria and A. lebbeck, two important wood yielding leguminous tree species. The culture medium was standardized after several trials to obtain callus from the leaflet explants of these two tree species. The optimum use of casein hydrolysate (w/v) and coconut milk (v/v) in addition to 6-Benzylaminopurine and Indole-3-butyric acid could induce morphogenesis and somatic embryogenesis in the cultured tissue. This reports the first observation on somatic embryogenesis ofA. lebbeck using leaflets as the explants. Scanning Electron Microscopy and histological studies were done on the different stages plant development following standard techniques. Embryogenesis in suspension culture followed regeneration of plantlets in A. lebbeck. In A.falcaaria the regenerative process followed via organogenesis from the shoot buds developed on the leaf explants. After hardening the regenerated plants were transferred to the greenhouse. Biogeographic Relationships of Pocket Gophers (Geomys breviceps and Geomys bursarius) in the Southeastern Portion of Their Ranges

This research utilized population genetic analyses (protein starch-gel electrophoresis and DNA sequencing of the cytochrome b mtDNA gene), host-parasite specificity (lice coevolution), remote sensing of satellite data, and geographic information systems (GIS) to characterize newly discovered populations of pocket gophers (genus: Geomys) in Arkansas. These populations are isolated and occur in seemingly unsuitable habitat in the Ozark Mountains of Arkansas. Analyses of electrophoretic and ectoparasite data suggested the populations in the Ozark Mountains represented isolates allied to Geomys bursarius, a species not known to occur in Arkansas. Comparison of mitochondrial DNA sequence data of the cytochrome b gene with that of other taxa and morphometric analyses confirmed that these populations are most closely allied to G. bursarius occurring to the north in Missouri. Moreover, these mtDNA sequence analyses indicated a degree of differentiation typical of that between other subspecies of pocket gophers. Therefore, these populations represent a distinct genetic entity in an intermediate stage of speciation and should be designated as a new subspecies, Geomys bursarius ozarkensis. Estimated Extent and Fate of Chlorinated Solvent Contamination in the Soil of the Naval Air Station, Dallas, Texas

This thesis estimates the spatial extent of chlorinated solvent contamination of the soil at the Naval Air Station, Dallas, then estimates the fate and transport of these contaminants, over time, using the Soil Transport and Fate database and the Vadose-Zone Interactive Processes (VIP) modeling software. Geostatistical analysis identifies two areas with serious chlorinated solvent contamination. Fate and transport modeling estimates that this contamination will degrade and disperse from the soil phase to below regulatory limits within one year, although there is a risk of groundwater contamination. Contaminants are estimated to persist in the water and air phases of the soil. Further sampling is recommended to confirm the results of this study.

Role of α-Keto Acids In Cyanide Detoxification and Assimilation by Pseudomonas Bacteria

Cyanide was rapidly removed when added to culture supernatants of seven different Pseudomonas. The ability to remove cyanide was correlated with the accumulation of α-keto acids (pyruvate and α-ketoglutarate). These compounds react with cyanide forming less toxic cyanohydrins, thus conferring a mechanism for bacterial cyanide tolerance. When added to growth media the α-keto acids were shown also to serve as effective cyanide antagonists. While all bacteria tested accumulated α-keto acids, only those capable of utilizing cyanide as a nutritional nitrogen source were able to metabolize cyanohydrins. In P. fluorescens NCIMB 11764, the same enzyme (cyanide oxygenase) shown previously to be involved in cyanide metabolism appears responsible for cyanohydrin transformation. Keto acid excretion is believed to represent a new mechanism of bacterial cyanide detoxification with further enzymatic metabolism of the cyanohydrins helping to explain how cyanide can satisfy the nitrogen requirement in cyanide-utilizing bacteria.

Search for Surrogate Marker(s) of Immunity Following Vaccination with Experimental Vaccine (Autoclaved Leishmania Major + Bacille Calmette-Guérin) in Human Volunteers

Cutaneous leishmaniasis (CL) is usually a self-limiting lesion on the skin while visceral leishmaniasis is a progressive, systemic disease with high mortality even if treated. The problem associated with treatment and vector control justifies a search for an effective vaccine which seems to be the only practical means to control the disease. The aim of this study is to identify immunological surrogate marker(s) associated with protection against Leishmania infection. The results indicate that a single dose of ALM+BCG induced Thl-like response but the level of such response is not sufficient for full protection. Accordingly, further evaluation of the vaccine is necessary other strategies multiple injections or changing the adjutant.

This document consists of the following elements: (1) an examination of Healey Willan's background in the English Cathedral tradition; (2) a study of the formation of his compositional style based on his knowledge of English composers and treatises of the day; (3) a look at the German influences on his compositional style; and (4) an analysis showing how he merged these styles into a unified whole to create a monumental work for the organ. Included are musical examples of English and German keyboard works compared with examples from Willan's introduction, Passacaglia and Fugue in order to illustrate the use of similar compositional devices and stylistic traits. Also included is a discussion of the evidence of Willan's individuality as a composer in spite of numerous derivative sources.

The goal of this study is to increase understanding of information problems as they are revealed in interactions among users and search intermediaries during information retrieval. Specifically, this study seeks to investigate: (a) how interaction between users and search intermediaries reveals aspects of user information problems; (b) to explore the concept of representation with respect to information problems in interactive information retrieval; and (c) how user and search intermediaries focus on aspects of user information problems during the course of searches. This project extends research on interactive information retrieval, and presents a theoretical framework that synthesizes rational and non-rational questions concerning mental representation as it pertains to user's understanding of information problems.

Content and Musical Language in the Piano Sonata of Sofia Gubaidulina, and Three Recitals with Works by Bach, Beethoven, Mozart, Chopin, Schumann, Debussy, and Rachmaninov

Sofia Gubaidulina is one of the leading composers in the contemporary music world. Her compositional interests have been stimulated by the exploration of and improvisation with rare folk and ritual instruments, and by a deep-rooted belief in the mystical properties of music. Gubaidulina is the author of orchestral and choral works, compositions for solo instruments, chamber music, as well as electronics music. Gubaidulina's Piano Sonata sums up the composer's thinking within her piano music, and at the same time projects the development of her spiritual vision within other genres that are to come. The analytical approach in this paper is based on the correlation between each of the elements of the musical material (form, rhythm, sound, etc.) and its contextual meaning in terms of musical dramaturgy. Set-theory is applied to the analysis of motivic components of the work. The traditional form is just the basis for the original intonational structure within a modern musical idiom. Varieties of rhythmic patterns, as well as an unconventional sound production, make this work breath with an impetuous power. Neurophysiological and Behavioral Correlates of Language Processing and Hemispheric Specialization

The purpose of this study was to examine language organization in the brain by using a series of three tasks concurrent with event-related potentials (ERPs) to investigate both hemispheric differences and interhemispheric reactions. Overall, the findings from this study support a relative rather than absolute hemispheric specialization for language processing. Despite an overall RVF (LH) advantage, both hemispheres were capable of performing the tasks and benefited from semantic priming.

Inspired by Chinese poems, the overall characteristics of the work reflect the assimilation of several non-Western musical and philosophical influences such as the use of pentatonic scale patterns, the principle of embellishing a single note, and the application of the I-Ching in dealing with active instrumental passages over a long-sustained computer music drone. Traditional Western compositional techniques such as aleatory counterpoint, serialism, and moment form are also employed in the treatment of thematic material, developmental processes and formal design.

Group Decision-Making in Computer-Supported Cooperative Work Environments

Computer-Support Cooperative Work (CSCW) reflects the change in emphasis from using computers to solve problems to using computers to facilitate human interactions. Most studies, however, have focused on the use of the technology rather than on the human-human interaction (HHI) in these environments due to: the varied perspectives of the investigators; and the lack of a consistent variables. Although numerous studies exist on a variety of products, only limited research has been conducted with the most prevalent of the technologies in the marketplace, Lotus Notes™. This field study, conducted using Lotus Notes™, operationalizes a model proposed, but not tested, for the study of group decision-making in CSCW environments put forth by Kraemer and Pinsonneault (1990). This study examines the use of CSCW in the group decision-making process, the participation rate for group decision-making in CSCW environments, and the criteria for determining quality in group decisions in CSCW environments. The study also proposes a new perspective for examining technology using the human context, recommends extensions for the group study framework and explores areas for future research.

Classification of Denial in Sex Offenders; an Investigation of Response Styles

Standard psychological assessment instruments have not produced consistent results by which decisions can be made regarding the appropriate placement and legal disposition of an individual who has committed a sexual offense. The purpose of the present study was to systematically investigate deception and dissimulation as measured by three assessment instruments commonly utilized with sex offenders. A denial classification system was utilized in order to classify offenders into categories based on their level of admission to the legal system. The four group classification system did not produce significant differences on all measures of deception and dissimulation. Contrary to previous research, admitters were found to respond more defensively than deniers on one of the assessment instruments. In addition, partial deniers were identified as responding significantly differently from both admitters and deniers on a separate instrument. The differences found suggest that sex offenders' level of deception is multifaceted. Difficulties in identifying classificatory strategies and implications for theoretical conceptions of denial within this population are discussed.

The effectiveness of prevention programs is often assessed by examining program effects of a variety of factors that are known to elevate or reduce risk for delinquent involvement. It is necessary to identify whether differential programming is required for significantly represented offender subpopulations, including those with emotional/behavioral disorders (E/BD) or learning disabilities (LD). This study sought to determine the relevance of specific individual, family, and school-risk factors for delinquency across three categorical subtypes ofjuvenile offenders: (a) those with E/BD, (b) those with LD, and (c) those offenders considered nondisabled.

The laboratory notebook is a summation of all the experiments and procedures used in the six-credit hour Advanced Microbial and Molecular Biology (BIOL 5160) course offered during the summer semester at the University of North Texas. This class is a team taught effort by Professors O'Donovan and Kunz. The course is constructed as an intensive practice exercise to teach the student about gene mutations, biosynthetic pathways, preparation and analysis of plasmid DNA, and many other topics included in the notebook.

The Gender of Time in the Eighteenth-century English Novel

This study takes a structuralist approach to the development of the novel, arguing that eighteenth-century writers build progressive narrative by rendering abstract, then conflating, literary theories of gendered time that originate in the Renaissance with seventeenth-century scientific theories of motion. I argue that writers from the Renaissance through the eighteenth century generate and regulate progress-as-product in their narratives through gendered constructions of time that corresponded to the generation and regulation of economic, political, and social progress brought about by developing capitalism.
