Life History and Case Building Behaviors of Phylloicus ornatus (Banks) (Trichoptera: Calamoceratidae) In Two Spring Fed Tributaries in the Central Edwards Plateau Bioregion of Texas

The life history and case-making behaviors of Phylloicus ornatus from two springfed first order streams in the Edwards Plateau Bioregion of Texas were studied from January 1998 to November 1999. Field larval, pupal and adult samples and laboratory rearings indicated a multivoltine cycle. First instars differ from late instars in number of labral setae and in having a unique spur-like claw on each lateral hump. Larval development was asynchronous with second through fifth instars and pupae present most months. First instars were present April through July, October and November. Case making of first instar and case reconstruction of later instars extracted from their cases was documented by videophotography.

Phototoxic Effects of Titanium Dioxide Nanoparticles on Daphnia Magna

Titanium dioxide nanoparticles (TiO2-NP) are one of the most abundantly utilized nanomaterials in the world. Studies have demonstrated the mechanism of acute toxicity in TiO2-NP to be the production of reactive oxygen species (ROS) leading to oxidative stress and mortality in exposed organisms. It has also been demonstrated that the anatase crystalline conformation is capable of catalyzing the cleavage of water molecules to further increase the concentration of ROS in the presence of ultraviolet radiation. This photoenhanced toxicity significantly lowers the toxicity threshold of TiO2-NP to environmentally relevant concentrations (ppb). The goal of this study was to determine whether dietary uptake and accumulation of TiO2-NP in the aquatic filter feeder Daphnia magna resulted in photoenhanced toxicity. D. magna and S. caprincornatum were exposed to aqueous solutions of 20ppm and 200ppm TiO2-NP for 24hrs and then transferred to clean moderately hard water. Samples were taken at various time points, dried, and TiO2 quantified using ICP-MS. Toxicity assays were run on D. magna using three TiO2-NP (20ppm, 200ppm) exposure protocols and two ultraviolet radiation treatments. The Effect of Menthol on Nicotine Metabolism: a Cross Species Evaluation

The effect of menthol on nicotine metabolism was examined in liver S9 fractions of four different species and in the in vivo mouse model. The purpose of this study was to investigate three parameters: (1) biotransformation of nicotine to cotinine in various species (human, mouse, rat and trout) using in vitro methods; (2) to determine if the addition of menthol with nicotine altered biotransformation of nicotine to cotinine; (3) and to assess similar parameters in an in vivo mouse model. The major findings of this study include: (1) mice appear to metabolize nicotine, over time, in a manner similar to humans; (2) menthol decreased cotinine production, over time, after a single dose in mice; and (3) menthol increased cotinine production, over time, after repeated doses, in mice.

Shortened in Vivo Bioconcentration Factor Testing in Cyprinus Carpio

Bioconcentration factor testing serves as the most valuable surrogate for the assessment of bioaccumulation. The assessment of potentially harmful chemicals is crucial to not only the health of aquatic environments, but to humans as well. Chemicals that possess the ability to persist in the environment or that have the potential to bioaccumulate, pose a greater risk to organisms that are exposed to these chemicals. The Organization for Economic Cooperation and Development Guideline 305 outlines specific protocols to run an accurate and reliable aquatic flow-through test. However, since its adoption in 1996, very few changes have been made to accommodate the endeavor to lowering the amount of test species to run one of these said tests. Running an aquatic flow-through test, according to 305, takes much time and money as well as numerous amounts of fish. Such burdens can be eliminated through simple modifications to the standard protocols. In this study, we propose an abbreviated study design for aquatic bioconcentration testing which effectively alleviates the burdens of running a flow-through test. Four chemicals were used individually to evaluate the usefulness of the proposed shortened design; 4-Nonyphenol, Chlorpyrifos, Musk Xylene, and DDT. Effects of Airway Pressure, Hypercapnia, and Hypoxia on Pulmonary Vagal Afferents in the Alligator (Alligator Misssissippiensis)

The American alligator (Alligator mississippiensis) is an aquatic diving reptile with a periodic breathing pattern. Previous work has identified pulmonary stretch receptors (PSR), both rapidly- and slowly-adapting, and intrapulmonary chemoreceptors (IPCs) that modulate breathing patterns in alligators. The purpose of the present study was to identify the effects of prolonged lung inflation and deflation (simulated dives) on PSR and/or IPC firing characteristics in the alligator. The effects of airway pressure, hypercapnia, and hypoxia on dynamic and static responses of pulmonary stretch receptors (PSR) were studied in juvenile alligators (mean mass = 246 g) at 24°C. Receptor activity appeared to be a mixture of slowly-adapting PSRs (SARs) and rapidly-adapting PSRs (RARs) with varying thresholds and degrees of adaptation, but no CO2 sensitivity. Dives were simulated in order to character receptor activity before, during, and after prolonged periods of lung inflation and deflation. Some stretch receptors showed a change in dynamic response, exhibiting inhibition for several breaths after 1 min of lung inflation, but were unaffected by prolonged deflation. For SAR, the post-dive inhibition was inhibited by CO2 and hypoxia alone. Immunohistochemistry of the Gills of the Channel Catfish Ictalurus Punctatus: Cells and Neurochemicals That May Be Involved in the Control of Cardioventilatory Reflexes

In teleost fishes the neurochemicals involved in sensing and responding to hypoxia are unresolved. Serotonergic branchial neuroepithelial cells (NECs) are putative O2 chemoreceptors believed to be homologous to the neural crest (NC) derived APUD (amine-precursor uptake and decarboxylation) pulmonary NECs and carotid body type-1 glomus cells. Branchial NECs contain serotonin (5-HT), thought to be central to the induction of the hypoxic cardioventilatory reflexes. However, application of 5-HT in vivo does not elicit cardioventilatory reflexes similar to those elicited by hypoxia. But previous in vitro neural recordings from glossopharyngeal (IX) afferents innervating O2 chemoreceptors in the trout gill show the same discharge response to hypoxic conditions as does that of acetylcholine (ACh) application. This evidence strongly supports the cholinergic hypothesis of chemoreceptor impulse origin rather than a serotonergic-induced impulse origin model. We therefore hypothesized that NECs contain ACh among other neurochemicals in cells belonging to the APUD series. Although serotonergic branchial NECs did not colocalize with ACh using immunohistochemical methods, several populations of ACh and/or tyrosine hydroxylase (TH) (catecholaminergic) positive, dopamine (DA) negative, cells were found throughout the second gill arch of the channel catfish Ictalurus punctatus. Responses of Pristina leidyi Smith 1896 (Naididae: Oligochaeta) to Cadmium, Vanadium, and Some Environmental Factors

Concern over sediment toxicity has increased the need for toxicity test information with organisms that inhabit sediments. Oligochaetes are exposed to toxicants through feeding and direct body contact with aquatic sediments. Chronic testing with oligochaetes has historically focused on tubificids with test lengths of one year or more to encompass several generations. Most naidid oligochaetes have generation times of three to seven days and could provide chronic information in a matter of weeks. The cosmopolitan distributed naidid, Pristina leidyi, was evaluated for use as a toxicity test organism. Results of research conducted includes culture methods, effects of temperature on reproduction, growth rates in a reference sediment, acute toxicity tests, and chronic toxicity tests.

Toxicity of Polychlorinated Biphenyls (Aroclor 1254) on the Earthworm Eisenia foetida

Objectives were to: (1) assess toxicity of polychlorinated biphenyls on Eisenia foetida, in terms of survival (LC5O/LD5O), and suppression of coelomocytes to form secretory (SR) and erythrocyte rosettes (ER) with, and to phagocytize rabbit erythrocytes; and (2) compare results with those of Lumbricus terrestris to assess relative sensitivities to PCB. Using 5-d filter paper contact exposure protocol, LC50 and LD50 were 30.4 cg/cm2 and 4450 cg/g dry mass, respectively. Nominal PCB exposure concentrations of 5.0 and 10.0 pg/cm2 resulted in tissue levels of 1389 and 2895 pg/g dry mass causing a significant reduction in SR formation by 18 and 52%, respectively. ER formation and phagocytosis were reduced significantly (52 and 61%) only at the higher tissue concentration. Compared to reported data on lethality and immunomodulation in L. terrestris, E. foetida appears to be more resistant.

Degradation of Phenolic Acids by Azotobacter Species Isolated from Sorghum Fields

Sorghum plants excrete phenolic acids which reduce subsequent crop yields. These acids accumulate in field soil by combining with soil and clay particles to form stable complexes which remain until degraded by bacterial metabolism. The amount of phenolic acids in soil samples were obtained by gas chromatography measurements, while Azotobacter populations were obtained by plate counts in 40 sorghum field samples from Denton County, Texas. One can conclude that increasing the Azotobacter population in the soil increased the degradation rate of phenolic acids proportionally. It is proposed that seed inoculation will introduce selected strains of Azotobacter into the soil. The presence of Azotobacter should increase crop size in subsequent plantings.

The Reproductive Consequences of Carriers of Methylenebisacrylamide-Induced Balanced Reciprocal Translocations in Mus Musculus

N,N'-methylenebisacrylamide (MBA) was studied because of its effectiveness in inducing heritable translocations in germ cells of male mice. The health impact of translocations was studied through anatomical analysis of the progeny of semisterile translocation carriers. As expected, the semisterility of translocation carriers resulted primarily from embryonic death during periimplantation stages due to unbalanced chromosome sperm segregants. Among conceptuses that survived to mid- and late-gestation stages, there was an increased incidence of developmental anomalies including fetal death and phenotypic defects. These abnormalities are associated with unbalanced chromosome complements that allow survival to the later stages of development.

Characterization of the OCT Plasmid-Encoded Mercury Resistance Genetic Locus in Pseudomonas putida

A 17.1 Kb genetic element encoding for mercury resistance (OCT-Hg^r) was shown to translocate from its original location on the OCT plasmid to the resistance plasmid, RPl, in Pseudomonas putida. Analysis of RPl-Hg^r recombinant plasmids revealed that insertion of mercury resistance genes into RPl could occur at a variety of sites, with all recombinants having common EcoRI restriction fragments of 9.4, 3.8, 2.3, and 1.6 Kb, derived from the insertion. Hybridization analysis suggested the existence of extensive homology between this insertion and the prototypic mercury resistance transposon, Tn501, as well as the location of a similar merA sequence. Although the overall size was shown to be quite different from Tn501, striking physical similarities are shared between these two elements.

Genetic Differentiation of the Geomys Pocket Gopher Complex of Texas

Genetic variation was analyzed for populations of seven taxa comprising four cytotypes of the Geomys bursarius chromosome complex, including G. b. major, G. b. knoxjonesi, and the Edwards Plateau taxa, G. b. llanensis and G. b. texensis. Genetic relationships of the Edwards Plateau gophers with other taxa and between themselves were examined. Genetic similarity, number of fixed allelic differences, and ectoparasite distribution indicate the Edwards Plateau gophers are a distinct gene pool. Isolation of the Edwards Plateau taxa precludes contact zone analysis. However, genetic differentiation is typical of that between other species of Geomys, and the Edwards Plateau taxa should be recognized as G. texensis. Distributions of allelic frequencies indicate little justification in retaining the subspecific status of the Edwards Plateau forms.

Development of an In Vitro Protoplast Culture System for Albizia Lebek (L.) Benth., an Economically Important Leguminous Tree

An in vitro system of generating protoplasts from their callus cultures was established. The friable callus was more productive in terms of producing protoplasts than the green compact callus. The concentration of the various cell wall degrading enzymes had an effect on the viability of the protoplasts in the medium. The protoplast system developed from the experiments was stable and could be used for the transformation experiments of Albizia lebek and for other plant improvement practices.

A Multimedia Atlas of Dissection for Comparative Anatomy of the Vertebrates

This interactive multimedia content is part of a Master's thesis regarding how traditional methods of teaching the laboratory course for Comparative Anatomy of the Vertebrates could be improved by applying current computer technology to construct an interactive, multimedial atlas of dissection. Five specimens used in comparative anatomy courses at most institutions were chosen as representative members of the Phylum Chordata: amphioxus, lamprey, dogfish shark, mud puppy, and cat. Specimens were dissected according to the modified method of Wischnitzer, 1993, and each stage was photographed with a Kodak DC120 digital zoom camera. These images were processed on a Power Macintosh 7600 computer with Adobe Photoshop v. 5.0. The atlas was constructed from these images using Macromedia Authorware v. 4.0.3. Each image contains a series of interactive objects that display a highlight and descriptive text as the cursor passes over each object.

Investigation of Inhibitory Influences in Neuronal Monolayer Networks Cultured from Mouse Spinal Cord

The effects of the inhibitory neurotransmitters gammaamino butyric acid (GABA) and glycine were characterized on spontaneous activity recorded from mouse spinal cord cultures. The GABA concentration which completely inhibited burst activity was chosen as a quantifiable measure of culture drug response and was used to 1) assess interculture and intraculture variability, 2) determine the influence of culture age and initial activity on GABA responses, and 3) compare the GABA responses between networks obtained from whole spinal cord and ventral half spinal cord. Results showed that 1) no significant variability existed either within or among cultures, 2) the initial culture activity directly affected GABA responses, 3) the culture age had no effect on GABA responses, and 4) there was no significant difference in GABA responses between the two spinal cord tissues.

Development and Validation of a Ramping Treadmill Protocol for the On-Line Measurement of Four Aerobic Parameters

Previously, Whipp et. al. (J. Appl. Physiol.: Respirat. Environ. Exerc. Physiol. 50(1):217-221, 1981) demonstrated the feasibility of determining four parameters of aerobic function, identified as maximum oxygen uptake (μVO_2), VO_2 at anaerobic threshold (θan), the time constant for oxygen uptake kinetics (rVO_2) and work efficiency (η), using a short duration ramped bicycle ergometer exercise test. Because of the importance of being able to measure these parameters on a variety of measurement instruments a short duration ramping treadmill protocol has been developed. The ability of this protocol to determine the four aerobic parameters has been validated against conventional methods. The results of this investigation indicate that μVO_2, θan, rVO_2 and, η may be obtained from a single, short-duration ramping treadmill test.

Endogenous Nucleotide Pools in Growing Cells of Azotobacter Vinelandii

The objective of this investigation was to examine the changes in the nucleotide pools of Azotobacter vinelandii during the growth cycle. Endogenous ribonucleotides were extracted from A. vinelandii using trichloroacetic acid (TCA; 12% w/v). The 5' mono-, di- and triphosphates of adenine, guanine, uracil and cytosine were separated and quantified by anion-exchange high performance liquid chromatography. Results indicated that the adenylate energy charge of A. vinelandii paralleled the growth rate during exponential phase and that it declined rapidly as the stationary phase was reached. In addition, the amount of each nucleotide in A. vinelandii tended to increase in the logarithmic phase and decrease in the stationary phase in a similar manner to the energy charge.

The Effects of Long Term Modernate Ethanol Intake on Plasma Levels of ACTH, Beta Endorphin, and Corticosterone in Rats

The effects of single injections and daily oral administration of ethanol on plasma levels of ACTH, beta endorphin, and corticosterone in response to cold stress were examined. The long-term experimental animals were given 0.25 ml of 28% ethanol or water orally once a day, five days a week, for fourteen months. Plasma levels of ACTH, beta endorphin, and corticosterone were lower in alcohol-treated rats as compared with water-treated rats when exposed to cold stress. The effects of a single injection of ethanol significantly elevated plasma levels of all three hormones. Mortality in sham-treated males was higher than ethanol-treated.

The Effect of N, N Bis (ethylene)-P (1-adamantyl) Phosphonic Diamide on Rous Sarcoma Virus

The drug, N,N bis (ethylene)-P (1-adamantyl) phosphonic diamide inhibits focus formation of Rous Sarcoma Virus in tissue culture. Transformation of chick cells was inhibited when the drug was added to chick cells prior to infection. The drug did not inhibit the transformation of Normal Rat Kidney Cells infected with RSV, when the cells were grown at non-permissive temperatures and shifted to permissive temperatures upon addition of the drug. Nor did the drug revert cells transformed at permissive temperatures. These studies indicated that the inhibition of RSV is in the early stage of viral growth, possible penetration or uncoating.

An Evaluation of the Short-Term Embryo-Larval and Seven-Day Larval Test Methods for Estimating Chronic Toxicity of Zinc to the Fathead Minnow (Pimephales promelas)

Chronic toxicity of zinc to Pimephales promelas was estimated by conducting replicate static and static-renewal short-term embryo-larval tests and static-renewal seven-day larval tests. The two test methods were highly reproducible. Daily renewal of test solutions had little effect on the toxicity of zinc, however, the stage of development at which exposure was initiated affected the sensitivity of the toxic endpoints measured. The most sensitive and reproducible endpoint in the embryo-larval tests was survival of viable (non-deformed) larvae and in the seven-day larval test was growth of the larvae, which was slightly more sensitive than the embryo-larval test endpoint. The estimated MATC of 0.18 and 0.15 mg/L mean total and mean soluble zinc, respectively, compared well with published results. Because of its advantages and similar sensitivity, the short-term embryo-larval test was recommended for estimating chronic toxicity.

Opthalmic Use Of Sodium Cephalothin: An In Vivo Comparison

A rabbit keratoconjunctivities model was used to evaluate ophthalmic formulations containing 1 percent sodium cephalothin in silicon oil, a 1 percent sodium cephalothin aqueous solution, and a 0.3 percent gentamicin sulfate solution. Rabit eyes were inoculated intracorneally with Pseudomonas aeruginosa, Staphylococcus aureus, or Streptococcus pneumoniae, After topical treatment, none of the antibiotic formulations were effective in the P. aeruginosa model; all three showed good activity against S. aureus, and against S. pneumoniae, the caphalothin formulations were more effective than gentamicin.In a related stability study, the cephalothin potency of the silicon formulation was maintained for 16 weeks at 4, 25, and 450 C These studies suggest that sodium cephalothin can be formulated as an effective and stable ophthalmic dosage form.

Diphenyloxazole Metabolism by Aryl Hydrocarbon Hydroxylase

2,5-Diphenyloxazole (PPO) was tested as a potential alternate inducer for the aryl hydrocarbon hydroxylase (AHH) system. Its apparent lack. of carcinogenicity and toxicity provide a possible system for investigation of enzyme systems related to chemical carcinogenesis without exposure of the researcher to potent carcinogenic compounds. These studies found PPO to be an inducer of AHH in cultured human lymphocytes. When PPO was utilized as a substrate for the AHH assay system, the major metabolites produced were strongly fluorescent. A simple fluorometric assay was developed which employed PPO as the substrate and which measured constitutive activity more efficiently than similar assays using benzo(a)pyrene as the substrate. Quantitation of both basal and induced lymphocyte AHH metabolism of PPO may be applicable to human population studies and may provide a tool to determine possible genetic variables with respect to carcinogen metabolism related to cancer risk.

Investigations have shown that the hypothalamus and pituitary respond to decreases in body temperature by stimulating thyroid release of T_3 and T_4 . This study was designed to bypass the control of the hypothalamus and pituitary gland and investigate the direct effect of temperature on the thyroid gland. Hypothermia was by an in vivo isolated perfusion of the thyroid gland. Radio-immunoassay was used to measure T_3 and T_4 concentrations. Significant increases were observed in animals perfused between 36º and 25ºc. These results indicate that the thyroid gland is directly effected by decreased temperature and that it is capable of exerting control over body temperature independent of the hypothalamus and pituitary gland. Lower perfusion temperatures produced no significant increases.

Nutritional Requirements of Corynebacterium poinsettiae

In a minimal medium supplemented with glucose and yeast extract, the optimum pH for the growth of C. poinsettias was found to be 7.5. The organism requires thiamine, biotin, and pantothenic acid for growth. No absolute requirement was found for any amino acid, purine or pyrimidine although an amino acid mixture was stimulatory. Casamino acids could be substituted for the synthetic amino acid mixture. Yeast extract provided an additional factor(s) necessary for maximal growth. The results suggest that the unknown factor found in yeast extract might be purified by a combination of solvent extraction, and adsorption and elution from charcoal.
