Testing and Evaluation of Environmental Fate Models Using Aquatic Microcosms and Three Organic Chemicals

Two compartment (sediment and water) flow-through model ecosystems were constructed to investigate the compartmentalization of different organic chemicals. Lindane, naphthalene, and mirex were pumped into the systems and resultant compartmental chemical concentrations determined. Steady state concentrations were compared to those predicted by two environmental fate models - EXAMS (Exposure Analysis Modeling System) and SLSA (Simplified Lake and Stream Analysis) which were developed by EPA-Athens, Georgia and HydroQual, Inc., respectively.

The Stoneflies (Plecoptera) of the Ozark and Ouachita Mountains

Collections of stoneflies (Plecoptera) were made at 603 stream sites from Nov. 1983 - May 1988 in the Ozark-Ouachita Mountain region, in relation to physiographic and vegetational characteristics. Examination of approximately 9000 vials from these collections, supplemented with material from major museums and other collectors, revealed 88 stonefly species in 8 families and 24 genera. Pearson's measure of association (R) showed there was a significant association between species present and each of the tested variables.

Investigation of Pyrimidine Salvage Pathways to Categorize Indigenous Soil Bacteria of Agricultural and Medical Importance and Analysis of the Pyrimidine Biosynthetic Pathway's Enzyme Properties for Correlating Cell Morphology to Function in All Phases of Growth

This dissertation comprises three parts and is presented in two chapters. Chapter 1 concerns Arthrobacter, a bacterium with an intriguing growth cycle. Whereas most bacteria exist as either a rod or coccus, this bacterium shares the rod/coccus lifestyle. It therefore seemed important to examine the growth regulatory pathways from the rod and coccus. The committed step, that catalyzed by aspartate transcarbamoylase (ATCase), in the pyrimidine biosynthetic pathway was chosen. The ATCase in Arthrobacter is like the well known Pseudomonas enzyme except that it has an active dihydroorotase (DHOase) associated. Included in Chapter 1 is the description of a microorganism, Burkholderia cepacia, whose ATCase has characteristics that are at once reminiscent of bacteria, mammals, and fungi. It differs in size or aggregation based on environmental conditions. In addition, it has an active DHOase associated with the ATCase, like Arthrobacter. B. cepacia is important both medically and for bioremediation. Since B. cepacia is resistant to most antibiotics, its unique ATCase is a prime target for inhibition. Estimating Postmortem Interval Using VNIR Spectroscopy on Human Cortical Bone

Postmortem interval (PMI) estimation is a necessary but often difficult task that must completed during a death investigation. The level of difficulty rises as time since death increases, especially with the case of skeletonized remains (long PMI). While challenging, a reliable PMI estimate may be of great importance for investigative direction and cost-savings (e.g. suspect identification, tailoring missing persons searches, non-forensic remains exclusion). Long PMI can be estimated by assessing changes in the organic content of bone (i.e. collagen), which degrades and is lost as the PMI lengthens. Visible-near infrared (VNIR) spectroscopy is one method that can be used for analyzing organic constituents, including proteins, in solid specimens. A 2013 preliminary investigation using a limited number of human cortical bone samples suggested that VNIR spectroscopy could provide a fast, reliable technique for assessing PMI in human skeletal remains. Clear separation was noted between "forensic" and "archaeological" specimen spectra within the near-infrared (NIR) bands. The goal of this research was to develop reliable multivariate classification models that could assign skeletal remains to appropriate PMI classes (e.g. "forensic" and "non-forensic"), based on NIR spectra collected from human cortical bone. Genetics Lecture and Laboratory Syllabus for a Junior-Level Course

The following is a complete syllabus for a college level genetics course. The syllabus contains lecture outlines and notes for each chapter, along with a list of transparencies needed. The quizzes and exams are prepared and placed at the beginning of the syllabus. The beginning of the course will consist of a lecture to introduce the students to the basics of genetics, followed by many applications of genetics. The process of cell division will be mastered by the students, as well as Mendelian genetics, quantitative genetics, chromosome mapping, and inheritance. The replication, synthesis, and organization of DNA are also discussed within the lectures. The final topics that will be covered using this syllabus are genetics of cancer and immunology and population genetics. These topics are essential for a detailed genetics course. The syllabus is written in great detail, and will require a full semester to be completed. The book used in association with this syllabus is Essentials of Genetics by William S. Klug and Michael R. Cummings.

Acute and Sublethal Impacts of Crude Oil Photo-Induced Toxicity in an Early Life Stage Marine Fish (Sciaenops ocellatus) and Invertebrate (Americamysis bahia)

We investigated the modifying effects of ultraviolet (UV) light and chemical dispersant (Corexit 9500A) on crude oil toxicity in juvenile mysids (≤ 24 h) (Americamysis bahia) and larval red drum (24-72 hpf) (Sciaenops ocellatus). These results demonstrate that crude oil toxicity significantly increases with co-exposure to environmentally relevant UV levels in both species, indicating photo-induced toxicity. This toxicity was further exacerbated by the application of chemical dispersants which increased the dissolution and concentration of oil-derived polycyclic aromatic hydrocarbons (PAHs) in test solutions. To better understand the mechanisms and initiating events of this observed photo-induced toxicity, the incidence of apoptotic cell death and global transcriptomic changes were assessed in larval red drum (24-72 hpf) following co-exposure to crude oil and UV. These results showed that co-exposure to UV and low concentrations of crude oil (<1 µg/L ∑PAH50) induced apoptotic cell death in skin and eye tissue and altered transcriptomic pathways related to visual processing and dermatological disease. To link these cellular and molecular impacts of photo-induced toxicity to apical endpoints of ecological performance, sublethal impacts to growth, metabolic rate, and visually mediated behaviors were explored in larval red drum at 2 developmental stages. Aquatic Vegetation Nutrient Budgets and Sedimentation in a Southwestern Reservoir

During four growing seasons, aquatic vascular plant production and distribution were studied in Pat Mayse Lake, Texas, a 2425 hectare oligo-mesotrophic reservoir. The dominant macrophyte population was Myriophyllum spicatum L. Growth rates and regrowth rates of mechanically harvested Myriophyllum beds were found to be dissimilar. Based on estimates of watermilfoil nutrient content, there were insufficient nutrients in the entire population to alter the trophic status of this reservoir should all of the nutrients be instantaneously released. Sediments were the primary nutrient (nitrogen and phosphorus) sink. Bank erosion and solids transport from the watershed appear to contribute most of the sediments and a lake-wide mean sedimentation rate of 2.5 cm/year was estimated from sediment trap and core sample data.
