Acute and Sublethal Impacts of Crude Oil Photo-Induced Toxicity in an Early Life Stage Marine Fish (Sciaenops ocellatus) and Invertebrate (Americamysis bahia)

We investigated the modifying effects of ultraviolet (UV) light and chemical dispersant (Corexit 9500A) on crude oil toxicity in juvenile mysids (≤ 24 h) (Americamysis bahia) and larval red drum (24-72 hpf) (Sciaenops ocellatus). These results demonstrate that crude oil toxicity significantly increases with co-exposure to environmentally relevant UV levels in both species, indicating photo-induced toxicity. This toxicity was further exacerbated by the application of chemical dispersants which increased the dissolution and concentration of oil-derived polycyclic aromatic hydrocarbons (PAHs) in test solutions. To better understand the mechanisms and initiating events of this observed photo-induced toxicity, the incidence of apoptotic cell death and global transcriptomic changes were assessed in larval red drum (24-72 hpf) following co-exposure to crude oil and UV. These results showed that co-exposure to UV and low concentrations of crude oil (<1 µg/L ∑PAH50) induced apoptotic cell death in skin and eye tissue and altered transcriptomic pathways related to visual processing and dermatological disease. To link these cellular and molecular impacts of photo-induced toxicity to apical endpoints of ecological performance, sublethal impacts to growth, metabolic rate, and visually mediated behaviors were explored in larval red drum at 2 developmental stages. Supply Chain Transparency from a Stakeholder's Perspective: Analyzing the Risks and Benefits of Supply Chain Information Disclosure

Supply chain transparency is principally focused on a company's efforts toward disclosing information about their products, and their supply chain operations to the public. Essay 1 is a conceptual paper that examines the risks of disclosing supply chain mapping information to consumers and proposes an approach to developing risk mitigation strategies. This essay also develops a set of supply chain mapping conventions that support the development of an agility-focused supply chain map. Essay 2 employs an experimental design methodology to examine the impact of disclosing the ethnicity of a supplier on consumers' behaviors, while also capturing the extent to which a consumers' ethnic identity and prosocial disposition influence their behaviors. Finally, also using an experimental design, Essay 3 analyzes consumer outcomes based on disclosing no, partial, and full supply chain transparency information, and accounts for heterogenous consumer traits such as the importance of information to a consumer and their perceived quality of information. Collectively, these essays advance the body of knowledge that seeks to understand the risks and benefits of supply chain transparency, by conceptually identifying risks and proposing an approach to minimize the risks associated with supply chain transparency, and by illuminating the conditions that prompt favorable consumer outcomes.

Development of Biomimetic Human Lung Alveolus Chip

The potential of physiologically relevant in vitro cell culture models for studying physiological and pathophysiological phenomena has been widely recognized as replacements for animal and conventional in vitro models. To create models that accurately replicate the structure and function of tissues and organs, it is essential to comprehend the biophysical and mechanical features of the extracellular matrix (ECM) and incorporate them into the in vitro cell culture models. Therefore, we first aimed to investigate how nanotopography can modulate cell behaviors by studying cell behaviors on nanostructures of various aspect ratios on a cobalt-chromium-molybdenum (CoCrMo) alloy surface. We also explored the impact of nanofibrous membranes on the formation of alveolar epithelium, which is critical for lung alveolar interstitium chips. In addition, we investigated the effect of mechanical stretch on cell behaviors and focused on how the dimensionality of the stretch affects cell behaviors. To create physiologically relevant in vitro models based on our findings, we engineered a stem cell niche using a combination of nanofibrous membranes, mechanical stretch, and a soft substrate, and evaluated its impact on stem cell behaviors. Finally, we created a biomimetic human lung interstitium chip for application in physiological and pathophysiological in vitro studies.

A historical-cultural approach is a powerful tool in uncovering deeper understandings of the Chinese way of policy making and strategy on issues such as the South China Sea. The key research questions include: first, is there any historical legitimacy in China's sovereignty claim over the South China Sea islands? Second, do Beijing's South China Sea policies in various periods have any regularity or pattern, and how did they serve China's grand strategies at the time? By utilizing extensive Chinese and English primary sources and other sources, this study conducts a comprehensive and in-depth analysis of the South China Sea issue from the framework of Chinese strategic culture.

This study uses a mixed methods approach to investigate how individuals perceive nature and engage with Texas state park (TSP) programs and resources while also identifying major barriers that visitors perceive/encounter when visiting TSPs. This study looks through the anthropological lens by using theoretical frameworks such as habitus, presentation of the social self, space and place, as well as communities of practice (CoP), to better understand the factors that influence the establishment and maintenance of an individual's relationship to nature and participation in related practices. This study illustrates how an individual's relationship to nature is influenced by experiences in early life that involve activities, landscape or bioregion, and social factors. Relationships with nature are strengthened through social support especially when CoPs are involved. By understanding park visitor experiences through motivations and limitations to participating in the outdoors, parks can expand engagement tactics, foster existing and create new CoP related to nature that aid in the introduction and adoption of outdoor learning and experiences creating lifelong stewards. There has been a rapid surge toward "organic" products devoid of GMOs, MSGs, and other common compounds found in processed foods that continue to indicate an association with an increased risk for disease. These consumers seek nutrients and vitamins that are lacking in their diet and lifestyle in the form of nutraceuticals for disease prevention and treatment as well as overall lifestyle enhancement. However, these products generally lack clinical evidence as well as legal definition. Due to this ambiguity, nutraceuticals are neither considered a food product nor a pharmaceutical product. Furthermore, due to their alleged natural properties allowing for safe, therapeutic effects, nutraceuticals are being eagerly sought after by consumers in the place of pharmaceuticals. Additionally, since nutraceutical substances are "naturally" derived, there is a general lack of regulation regarding the manufacturing and distribution process. This mismanagement leads to lack of quality assurance (QA) and quality control (QC) protocols strictly implemented to define appropriate production and storage parameters. In 2016, the Colombian Constitutional Court recognized the Atrato River as a subject of rights based on the theory of biocultural rights. This dissertation analyzes a new legal concept that aims to defend the rights to a good life for humans and other-than-human co-inhabitants who share river ecosystems, focusing on the case of the Atrato River in Colombia. The 3Hs framework of biocultural ethics is adopted to interconnect complex and interrelated historical, biophysical, cultural, and political dimensions. With this analysis, broader biocultural approaches are suggested. They could be valuable for understanding and implementing biocultural rights in other world regions. Moreover, it could transform the current situation that destroys biocultural diversity toward public policies that favor more just and sustainable forms of co-inhabiting biocultural diversity. A primary limitation of the implementation of biocultural rights is the context of a "failed state," in which the Colombian State is subject to severe problems of corruption, illegal mining, conflicts between legal and illegal armed groups, and drug trafficking. Re-Envisioning the Future: A Research Study about Increased Plastic Pollution from Desalination Plants and Environmental Education in Texas

This study examines the relationship between proposed desalination plants and increased plastic pollution along the Texas Gulf Coast. It specifically focuses on their expected impact on communities in the area and was conducted for Society of Native Nations. The goal was to gather information about environmental ideologies and experiences from different environmental experts and scientists to educate community members and inform policy recommendations. The study relied on semi structured interviews and archival research to understand how environmental experts and scientists envision the future, how they interpret the impact of desalination plants as related to plastic pollution. Ideas that guided this research include decolonial methodologies, political ecology, Indigenous research agendas, environmental justice and knowledge, cultural hybridity, and the anthropology of the borderlands. This research provides actionable steps and recommendations to improve environmental education in Texas Gulf Coast communities on the U.S./Mexico border and to reduce plastic pollution in order to ensure that these communities have ample amounts of water supply without relying on desalination plants.

Influence of Hypoxia on Acute Lead Toxicity and Calcium Homeostasis in Early Life Stage Zebrafish (Danio rerio)

The purpose of this study was to investigate the effects of Pb and hypoxia co-exposure on Pb toxicity and Ca homeostasis in early life stage (ELS) zebrafish (Danio rerio). Previous evidence indicates that exposure of ELS zebrafish to hypoxia (~20% air saturation) reduces Ca uptake, likely through down-regulation of the apical epithelial Ca channel (ECaC). Considering that Pb and Ca are known antagonists and compete for uptake pathways, it was hypothesized that co-exposure of Pb with hypoxia would decrease Pb toxicity by reducing Pb uptake (likely mediated through a reduced number of ECaCs). However, it was shown that at 96 hpf, whole body accumulation of both Pb and Ca was lower at 40% air saturation compared to 100% and 20% air saturation. This result closely aligned with the 96h LC50 results which showed the highest mortality of zebrafish at 40% compared to the other air saturation levels. This suggests that toxicity is likely the result of exacerbated hypocalcemia at 40% air saturation due to both Pb competition for Ca binding to Ca uptake channels/transporters, such as ECaC, and potentially reduced expression of such channels/transporters in response to this level of hypoxia. CO2 Transport and Acid-Base Status during Fluctuations in Metabolic Status in Reptiles

Reptiles can often experience perturbations that greatly influence their metabolic status (e.g., temperature, exercise, digestion, and ontogeny). The most common cause of fluctuations in metabolic status in post-embryonic reptiles is arguably digestion and physical activity (which will be further referred to as exercise). The objective of this thesis is to determine the mechanisms involved in CO2 transport during digestion, determine the mechanisms that allow for the maintenance of acid-base homeostasis during digestion, and observing the effect of an understudied form of exercise in semi-aquatic reptiles on the regulation of metabolic acidosis and base deficit. This dissertation provided evidence for potentially novel and under investigated mechanisms for acid-base homeostasis (e.g., small intestine and tissue buffering capacity; Chapters 3 & 4), while also debunking a proposed hypothesis for the function of an anatomical feature that still remains a mystery to comparative physiologist (Chapter 2). This thesis is far from systematic and exhaustive in its approach, however, the work accomplished in this dissertation has become the foundation for multiple distinct paths for ecologically relevant investigations of the regulation of metabolic acidosis/alkalosis in reptiles.

Development and Application of Novel Computer Vision and Machine Learning Techniques

The following thesis proposes solutions to problems in two main areas of focus, computer vision and machine learning. Chapter 2 utilizes traditional computer vision methods implemented in a novel manner to successfully identify overlays contained in broadcast footage. The remaining chapters explore machine learning algorithms and apply them in various manners to big data, multi-channel image data, and ECG data. L1 and L2 principal component analysis (PCA) algorithms are implemented and tested against each other in Python, providing a metric for future implementations. Selected algorithms from this set are then applied in conjunction with other methods to solve three distinct problems. The first problem is that of big data error detection, where PCA is effectively paired with statistical signal processing methods to create a weighted controlled algorithm. Problem 2 is an implementation of image fusion built to detect and remove noise from multispectral satellite imagery, that performs at a high level. The final problem examines ECG medical data classification. PCA is integrated into a neural network solution that achieves a small performance degradation while requiring less then 20% of the full data size.

Community First: An Ethnographic Approach to Understanding Local Perceptions of Sustainability in the Age of Neoliberalism

This work describes ethnographic research completed in order to understand how local community members in Denton, Texas define, conceptualize, and speak about sustainability. The goal of this research is to encourage a more representative approach to sustainability initiatives within the City of Denton by uniting community ideas with local governance. Data for this study was collected through semi-structured interviews with residents, participant observation at community meetings, and quantitative survey analysis. Through the use of a Foucauldian framework for analysis, in conjunction with David Harvey's "entrepreneurial city," and work done in the field of environmental justice, this study highlights a potential link between neoliberal approaches to city governance and community perceptions of sustainability. This research concludes by calling for more representation of all community members within local sustainability initiatives, and provides several suggestions for how this can be achieved.

The Dallas Story: The North American Aviation Plant during World War II

During the Second World War the United States mobilized its industrial capacity to become the great "Arsenal of Democracy," as vehicles, ships, and small arms flowed out of American factories. Perhaps the most impressive accomplishment was the mobilization of the aviation industry, which grew rapidly after the war began in Europe. In 1940 the United States produced 24,600,000 pounds of airframe. By 1943 this figure had grown exponentially, with 760,926,600 airframe pounds produced. This was achieved through the cooperation of the United States government and the aviation industry. It required creative techniques in funding and manufacturing, and the construction of expansion facilities throughout the country, including Dallas, Texas. The city was selected as the site of a factory operated by North American Aviation. This plant produced some 18,784 aircraft in all, making it one of the most prolific in the country. This dissertation is a study of the North American factory in Dallas. It begins with decisions leading to President Franklin D. Roosevelt's call for 50,000 aircraft in May of 1940. From there the focus moves to the selection of Dallas as a location, the construction and opening of the factory, its operation, its relations with the local community, and …

In the Tall Grass West of Town: Racial Violence in Denton County during the Rise of the Second Ku Klux Klan

The aim of this thesis is to narrate and analyze lynching and atypical violence in Denton County, Texas, between 1920 and 1926. Through this intensive study of a rural county in north Texas, the role of law enforcement in typical and systemic violence is observed and the relationship between Denton County Officials and the Ku Klux Klan is analyzed. Chapter 1 discusses the root of the word lynching and submits a call for academic attention to violence that is unable to be categorized as lynching due to its restrictive definition. Chapter 2 chronicles known instances of lynching in Denton County from its founding through the 1920s including two lynchings perpetrated by Klavern 136, the Denton County Klan. Chapter 3 examines the relationship between Denton County Law Enforcement and the Klan. In Chapter 4, seasons of violence are identified and applied to available historical records. Chapter 5 concludes that non-lynching violence, termed "disappearances," occurred and argues on behalf of its inclusion within the historiography of Jim Crow Era criminal actions against Black Americans. In the Prologue and Epilogue, the development and dissolution of the St. John's Community in Pilot Point, Texas, is narrated.

The Administration of Unemployment Relief by the State of Texas during the Great Depression, 1929-1941

During the Great Depression, for the first time in its history, the federal government provided relief to the unemployed and destitute through myriad New Deal agencies. This dissertation examines how "general relief" (direct or "make-work") from federal programs—primarily the Emergency Relief and Construction Act (ERCA) and Federal Emergency Relief Administration (FERA)—was acquired and administered by the government of Texas through state administrative agencies. These agencies included the Chambers of Commerce (1932-1933), Unofficial Texas Relief Commission (1933), Texas Rehabilitation and Relief Commission (1933), Official Texas Relief Commission (1933-1934), Texas Relief Commission Division of the State Board of Control (1934), and the Department of Public Welfare (1939). Overall, the effective administration of general relief in the Lone Star State was undermined by a political ideology that persisted from, and was embodied by, the "Redeemer" Constitution of 1876.

An Evaluation of Long-Term Air Quality Trends in North Texas using Statistical and Machine Learning Techniques

While ozone design values have decreased since 2000, the values measured in Denton Airport South (DEN), an exurban region in the northwest tip of the Dallas-Fort Worth (DFW) metroplex, remains above those measured in Dallas Hinton (DAL) and Fort Worth Northwest (FWNW), two extremely urbanized regions; in addition, all three sites remained in nonattainment of National Ambient Air Quality Standards (NAAQS) ozone despite reductions in measured NOx and CO concentrations. The region's inability to achieve ozone attainment is tied to its concentration of total non-methane organic compounds (TNMOC). The mean concentration of TNMOC measured at DAL, FWNW, and DEN between 2000 and 2018 were 67.4 ± 1.51 ppb-C, 89.31 ± 2.12 ppb-C, and 220.69 ± 10.36 ppb-C, respectively. Despite being the least urbanized site of the three, the TNMOC concentration measured at DEN was over twice as large as those measured at the other two sites. A factor-based source apportionment analysis using positive matrix factorization technique showed that natural gas was a major contributing source factor to the measured TNMOC concentrations at all three sites and the dominant source factor at DEN. Creeks and Open Spaces: Ned Fritz's Environmental Crusades

Edward C. Fritz was one of the most influential environmentalists in Texas history. Although he took a circuitous route to environmental activism, Fritz evolved into a powerful force fighting on behalf of Texan nature. Participating in substantial actions throughout the second half of the twentieth century, Fritz's contributions to environmental activism resulted in the successful preservation of thousands of acres and multiple wildlife species. Fritz parlayed his legal background into effective activism, beginning his career with a successful lobbying campaign for protection of Harris Hawks. He led the campaign to stop a decades old plan for canalization of the Trinity River. The creation of COST combined Fritz's environmental focus with the concerns of economic conservatives to prevent a billion dollar government funded project that would have significantly altered the river. Fritz then led a cadre who took over efforts to establish a preserve in the Big Thicket national forest. He oversaw the foundation of a protected area far larger than original expectations, capitalizing on the growing awareness of environmental issues in the 1970s. Fritz's interest in the Big Thicket led to a fight against the Forest Service's practice of clearcutting and its effect on Red Cockaded Woodpeckers. Through litigation and …

This dissertation aims to examine the relationship between interorganizational collaboration and the implementation of urban greening policy. Specifically, it discusses bonding and bridging relationships that explain a successful interorganizational collaboration, and to what extent these factors explain the perception of success in the implementation of public programs. The effects of risks of collaboration on the implementation of urban greening policy are also studied. To frame the analysis, this dissertation uses Feiock's institutional collective action (ICA) framework, which aims to understand successful interorganizational collaboration and policy implementation across sectors. The collection of data was carried out in Riyadh City in Saudi Arabia, which is located in the center of Saudi Arabia. In this study, the unit of analysis is the networks of relationships among organizations that work with the Royal Commission for Riyadh City (RCRC). The data were obtained from 44 organizations collaborating to implement urban greening projects in Riyadh City and were collected over 17 days from June 15, 2019 to July 2, 2019. Framing a Sacred Fight: Framing Analysis and Collective Identity of the #noDAPL Movement

The #noDAPL movement was an Indigenous-led environmental social movement occurring between 2015 and 2017, in which the Standing Rock Sioux and other American Indian tribes comprising the Oceti Sakowin garnered support to oppose the 1,172-mile Dakota Access Pipeline. Pipeline opponents agreed that the pipeline's construction posed a threat to the health and safety of tribal members and other residents of the area and that the pipeline's path crossed previously-designated tribal treaty boundaries, compromising tribal sovereignty. In this body of work, I utilize Facebook data from the Sacred Stone Camp Facebook page to locate and identify collective action frames and core framing tasks, adhering to social movement framing theory. Further, I provide insight into the movement's most used collective action frames and how their use enabled to movement to maintain occupation at protest camps along the Missouri River, garner resources from participants and gain international social support. I also draw on concepts of pan-Indianism and supratribalism to discuss indigenous collective identity, as well as concepts like relational values and Indigenous traditional knowledge to better assess the nuances of Indigenous environmental activism and how this movement evoked discussions of modern day settler colonialism.

Irrigation Methods and Their Effects on Irrigation Water Efficiency in High Tunnels

Improving water efficiency is and will continue to be a top concern to meet the world food production demands for a growing population. By having a clear understanding of water efficiencies, communities will be able to address these concerns from an economic standpoint and use more productive methods to grow food and limit water consumption. This study examines the water efficiencies of three irrigation methods over a single growing season in southeastern Oklahoma. Two crops, tomatoes and cucumbers, were grown using drip irrigation, a self-wicking container, and a non-circulating hydroponics barrel. Results at the end of the season showed the drip irrigation method had the highest water efficiency in terms of yield of product over water applied for both crops. The drip irrigation method also had the lowest associated set up costs and second lowest time requirements after the hydroponics method. These results were found to be consistent with other studies that compared drip irrigation to other irrigation methods and showed drip to have the highest water efficiencies.

The Rise and Fall of the Greenback Party in Texas: Economic Change and Political Dissent in the Post-Civil War Era

In 1873, a financial crisis plunged the United States into a deep economic depression that exacerbated a number of post-war economic issues. By the late 1870s, political dissent centered primarily on financial issues merged into the Greenback movement, which represented a loose coalition of reformers calling for economic relief based on the expanded use of greenbacks (paper currency issued by the United States Treasury during the Civil War). The Greenback Party emerged as a direct response to federal financial policies, but in Texas, it also provided a broad political platform for those opposed to the policies of "Redeemer/Bourbon" Democrats. The Greenback Party of Texas brought together a wide range of dissenters, including disgruntled Democrats, ousted Republicans, and many different economic and social reformers. From 1876, when the first Greenback clubs appeared in Texas, to the Greenback Party's virtual disappearance after the election of 1884, the Texas Greenbackers reached across boundaries of section, race, class, and sometimes gender; brought together farmers, workers, and professionals; Southerners and Northerners, white and black; former Confederates and former Unionists; native-born Americans and immigrants; and received sizable support from multiple counties in the northern, eastern, and central part of the state.
