Development of Microorganisms With Improved Transport and Biosurfactant Activity for Enhanced Oil Recovery Annual Report

Diverse microorganisms were screened for biosurfactant production and anaerobic growth at elevated salt concentrations to obtain candidates most suitable for microbial oil recovery. Seventy percent of the 205 strains tested, mostly strains of Bacillus mojavensis, Bacillus subtilis, Bacillus licheniformis, and Bacillus sonorensis, produced biosurfactants aerobically and 41% of the strains had biosurfactant activity greater than Bacillus mojavensis JF-2, the current candidate for oil recovery. Biosurfactant activity varied with the percentage of the 3-hydroxy-tetradecanoate isomers in the fatty acid portion of the biosurfactant. Changing the medium composition by incorporation of different precursors of 3-hydroxy tetradecanoate increased the activity of biosurfactant. The surface tension and critical micelle concentration of 15 different, biosurfactant-producing Bacillus strains was determined individually and in combination with other biosurfactants. Some biosurfactant mixtures were found to have synergistic effect on surface tension (e.g. surface tension was lowered from 41 to 31 mN/m in some cases) while others had a synergistic effect on CMD-1 values. We compared the transport abilities of spores from three Bacillus strains using a model porous system to study spore recovery and transport. Material Property Estimation for Direct Detection of DNAPL using Integrated Ground-Penetrating Radar Velocity, Imaging and Attribute Analysis

The focus of this project is direct detection of DNAPLs, specifically chlorinated solvents, via material property estimation from multi-fold surface ground-penetrating radar (GPR) data. We combine state-of-the-art GPR processing methodology with quantitative attribute analysis and material property estimation to determine the location and extent of residual and/or pooled DNAPL in both the vadose and saturated zones. An important byproduct of our research is state-of-the-art imaging which allows us to pinpoint attribute anomalies, characterize stratigraphy, identify fracture zones, and locate buried objects.

Big Sky Carbon Sequestration Partnership Quarterly Report: July-September 2004

The Big Sky Carbon Sequestration Partnership, led by Montana State University, is comprised of research institutions, public entities and private sectors organizations, and the Confederated Salish and Kootenai Tribes and the Nez Perce Tribe. Efforts under this Partnership fall into four areas: evaluation of sources and carbon sequestration sinks; development of GIS-based reporting framework; designing an integrated suite of monitoring, measuring, and verification technologies; and initiating a comprehensive education and outreach program. At the first two Partnership meetings the groundwork was put in place to provide an assessment of capture and storage capabilities for CO{sub 2} utilizing the resources found in the Partnership region (both geological and terrestrial sinks), that would complement the ongoing DOE research. During the third quarter, planning efforts are underway for the next Partnership meeting which will showcase the architecture of the GIS framework and initial results for sources and sinks, discuss the methods and analysis underway for assessing geological and terrestrial sequestration potentials. The meeting will conclude with an ASME workshop. The region has a diverse array of geological formations that could provide storage options for carbon in one or more of its three states. The increasing importance of Prompt Gamma-ray ActivationAnalysis (PGAA) in a broad range of applications is evident, and has beenemphasized at many meetings related to this topic (e.g., TechnicalConsultants' Meeting, Use of neutron beams for low- andmedium-fluxresearch reactors: radiography and materialscharacterizations, IAEA Vienna, 4-7 May 1993, IAEA-TECDOC-837, 1993).Furthermore, an Advisory Group Meeting (AGM) for the Coordination of theNuclear Structure and Decay Data Evaluators Network has stated that thereis a need for a complete and consistent library of cold- and thermalneutron capture gammaray and cross-section data (AGM held at Budapest,14-18 October 1996, INDC(NDS)-363); this AGM also recommended theorganization of an IAEA CRP on the subject. The International NuclearData Committee (INDC) is the primary advisory body to the IAEA NuclearData Section on their nuclear data programmes. At a biennial meeting in1997, the INDC strongly recommended that the Nuclear Data Section supportnew measurements andupdate the database on Neutron-induced PromptGamma-ray Activation Analysis (21st INDC meeting, INDC/P(97)-20). As aconsequence of the various recommendations, a CRP on "Development of aDatabase for Prompt Gamma-ray Neutron Activation Analysis (PGAA)" wasinitiated in 1999.
