Report for the NGFA-5 project.

The objective of this project is to provide DHS a comprehensive evaluation of the current genomic technologies including genotyping, TaqMan PCR, multiple locus variable tandem repeat analysis (MLVA), microarray and high-throughput DNA sequencing in the analysis of biothreat agents from complex environmental samples. To effectively compare the sensitivity and specificity of the different genomic technologies, we used SNP TaqMan PCR, MLVA, microarray and high-throughput illumine and 454 sequencing to test various strains from B. anthracis, B. thuringiensis, BioWatch aerosol filter extracts or soil samples that were spiked with B. anthracis, and samples that were previously collected during DHS and EPA environmental release exercises that were known to contain B. thuringiensis spores. The results of all the samples against the various assays are discussed in this report.

"Adlam and Allen and Davis, Lust, and Schluter have studied nonlinear plane-waves, propagating normal to the magnetic field, in a cold plasma. One solution of particular interest is a solitary wave, or single pulse. We present a method for solving the analogous problem for a plasma with finite temperature, in the limiting case where the amplitude of the wave is small and where, consequently, the width of the waver is very large."

A review of plutonium environmental data with a bibliography for use in risk assessments

Plutonium fueled radioisotopic heat sources find space, terrestrial, and undersea applications to generate electrical power. Such systems under postulated accident conditions could release radioactivity into the environment resulting in risks to the general population in the form of radiological doses and associated health effects. The evaluation of the radiological impact of postulated scenarios involving releases of activity into the environment includes identification of postulated accident release modes, including the probability of release and the release location; source term definition, including the activity of each radionuclide released and the corresponding chemical form and particle size distribution; analysis of the environmental behavior of the released radioactivity to determine the concentrations in environmental media (air, soil, and water) as a function of time; and analysis of the interaction between the environmental concentrations and man, leading to ingestion, inhalation, and external doses through each environmental exposure pathway. 443 refs., 2 figs., 4 tabs.

Low Odor, High Yield Kraft Pulping

In laboratory cooks pure oxygen was profiled into the circulation line of a batch digester during two periods of the cooking cycle: The first injection occurred during the heating steps for the purpose of in-situ generation of polysulfide. This chip treatment was studied to explore stabilization against alkaline induced carbohydrate peeling and to increase pulp yield. Under optimum conditions small amounts of polysulfide were produced with yield increase of about 0.5% These increases fell below earlier reports suggesting that unknown differences in liquor composition may influence the relative amounts of polysulfide and thiosulfate generated during the oxidation. Consequently, further studies are required to understand the factors that influence the ratios of those two sulfur species.

Final Technical Report

Most prokaryotes of interest to DOE are poorly understood. Even when full genomic sequences are available, the function of only a small number of gene products are clear. The critical question is how to best infer the most probable network architectures in cells that are poorly characterized. The project goal is to create a computational hypothesis testing (CHT) framework that combines large-scale dynamical simulation, a database of bioinformatics-derived probable interactions, and numerical parallel architecture data-fitting routines to explore many “what if ?” hypotheses about the functions of genes and proteins within pathways and their downstream effects on molecular concentration profiles and corresponding phenotypes. From this framework we expect to infer signal transduction pathways and gene expression networks in prokaryotes. Detailed mechanistic models of E. Coli have been developed that directly incorporate DNA sequence information. The CHT framework is implemented in the NIEngine network inference software. NIEngine has been applied to recover gene regulatory networks in E. coli to assess performance. Application to Shewanel la oneidensi and other organism of interest DOE will be conducted in partnership with Jim Collin's Lab at Boston University and other academic partners. KINETICS OF Mn-BASED SORBENTS FOR HOT COAL GAS DESULFURIZATION

Mixed manganese oxide sorbents have been investigated for high-temperature removal of hydrogen sulfide (the primary sulfur bearing compound) from hot coal gases. The sorbents were screened by thermodynamic equilibrium considerations for sulfidation. Preliminary experimental work using thermogravimetric analysis (TGA) indicated titania to be a superior substrate than alumina. Four formulations showing superior reactivity in a TGA were then tested in an ambient pressure fixed-bed reactor to determine steady state H 2 S concentrations, breakthrough times and effectiveness of the sorbent when subjected to cyclic sulfidation and regeneration testing. Eight tests were conducted with each test consisting of five cycles of sulfidation and regeneration. Sulfidation occurred at 600 o C using a simulated coal gas at an empty-bed space velocity of approximately 12,000 per hour. Manganese-based sorbents with molar ratios > 1:1 Mn:Substrate were effective in reducing the H 2 S concentration in simulated coal gases to less than 100 ppmv over five cycles. Actual breakthrough time for formulation C6-2-1100 was as high as 73% of breakthrough time based on wt% Mn in sorbent at 600 o C. Regeneration tests determined that loaded pellets can be essentially completely regenerated in an air/steam mixture at 750 o C with minimal sulfate formation. …

Aspects of safety and reliability for fusion magnet systems first annual report

General systems aspects of fusion magnet safety are examined first, followed by specific detailed analyses covering structural, thermal, electrical, and other aspects of fusion magnet safety. The design examples chosen for analysis are illustrative and are not intended to be definitive, since fusion magnet designs are rapidly evolving. Included is a comprehensive collection of design and operating data relating to the safety of existing superconducting magnet systems. The remainder of the overview lists the main conclusions developed from the work to date. These should be regarded as initial steps. Since this study has concentrated on examining potential safety concerns, it may tend to overemphasize the problems of fusion magnets. In fact, many aspects of fusion magnets are well developed and are consistent with good safety practice. A short summary of the findings of this study is given.

PATHFINDER ATOMIC POWER PLANT FINAL REPORT MOISTURE DE-ENTRAINMENT TESTS IN TWO- AND FOUR-INCH DIAMETER TEST SECTIONS

Tests are described to determine the amount of entrained moisture occurring in the steam of the Pathfinder Power Plant as design parameters were varied. It was found that the height of the moisture separator and the velocity of the steam should be adjusted so that the permissible steam velocity for the particular height is not exceeded. If this is done, the amount of moisture entering the separator should be less than 1%, an amount of moisture that should not be difficult to remove with moisture separators. (J.R.D.)

Cerro Grande Fire Impact to Water Quality and Stream Flow near Los Alamos National Laboratory: Results of Four Years of Monitoring

In May 2000, the Cerro Grande fire burned about 7400 acres of mixed conifer forest on the Los Alamos National Laboratory (LANL), and much of the 10,000 acres of mountainside draining onto LANL was severely burned. The resulting burned landscapes raised concerns of increased storm runoff and transport of contaminants by runoff in the canyons traversing LANL. The first storms after the fire produced runoff peaks that were more than 200 times greater than prefire levels. Total runoff volume for the year 2000 increased 50% over prefire years, despite a decline in total precipitation of 13% below normal and a general decrease in the number of monsoonal thunderstorms. The majority of runoff in 2000 occurred in the canyons at LANL south of Pueblo Canyon (70%), where the highest runoff volume occurred in Water Canyon and the peak discharge occurred in Pajarito Canyon. This report describes the observed effects of the Cerro Grande fire and related environmental impacts to watersheds at and near Los Alamos National Laboratory (LANL) for the first four runoff seasons after the fire, from 2000 through 2003. Vadose Zone Transport Field Study FY 2003 Test Plan

Conceptual models have been identified as one of the sources of uncertainty in the interpretation and prediction of contaminant migration through the vadose zone at Hanford. Current conceptual models are limited partly because they often do not account for the random heterogeneity that occurs under the extremes of very nonlinear flow behavior typical of the Hanford vadose zone. Over the last two years significant progress has been made in characterizing physical heterogeneity and in the development of techniques for incorporating this heterogeneity into predictive and inverse models for field-scale subsurface flow. One of the remaining pieces of the puzzle is the impact of heterogeneity on the distribution of reactive contaminants. Reactive transport occurs over a wide range of spatial and temporal scales. However, the manner in which the various subsurface physical and chemical processes interact to influence transport is not very well understood. Hydrogeologic characterization and model analysis, however, have traditionally focused on measurement of physical properties and predicting the effects of variability in these properties on flow and transport. As a result, the role of geochemical heterogeneity on solute transport has remained largely unexplored. An evaluation of three coal feeder concepts (lockhopper, Fuller Kinyon screw and Foster Miller pocket feeder) for use in the ICGG first stage gasifier was made. For the sake of consistency, all feed concepts were evaluated on the same basis, that is, feeding a nominal 100 Tons per hour (TPH) against a back pressure of 50 psig. Also, all feed systems were assumed to have similar atmospheric storage hoppers and final injection systems. Accordingly, no further consideration of these support systems was made. This survey was accomplished by reviewing the literature, interviewing vendors, contacting applicable facilities and performing necessary original analysis. The emphasis of the survey was on the test and industrial experience of each system. The results of this survey which are summarized on Table I show that the lockhopper system still should be considered as the baseline system because it is a proven system with extensive operating experience. On the other hand, if definitive tests can prove conclusively that the Fuller Kinyon screw pump will deliver coal against 50 psig back pressure with acceptable efficiencies and coal particle attrition characteristics, it would be the preferred system because it is the lowest cost and easiest to maintain. Characterization of Vadose Zone Sediment: Borehole 299-E33-46 Near Tank B-110 in the B-BX-BY Waste Management Area.

This report presents vadose sediment characterization data that improves understanding of the nature and extent of past releases in the B tank farm. A vertical borehole, located approximately 15 ft (5 m) from the northeast edge of single-shell tank 241-B-110 was drilled to a total depth of 264.4 ft bgs, the groundwater table was encountered at 255.8 ft bgs. During drilling, a total of 3 two-ft long, 4-inch diameter split-spoon core samples were collected between 10 and 254 ft bgs-an average of every 7.5 ft. Grab samples were collected between these core sample intervals to yield near continuous samples to a depth of 78.3 m (257 ft). Geologic logging occurred after each core segment was emptied into an open plastic container, followed by photographing and sub-sampling for physical and chemical characterization. In addition, 54 out of a total of 120 composite grab samples were opened, sub-sampled, logged, and photographed. Immediately following the geologic examination, the core and selected grab samples were sub-sampled for moisture content, gamma-emission radiocounting, tritium and strontium-90 determinations, total carbon and inorganic carbon content, and 8 M nitric acid extracts (which provide a measure of the total leachable sediment content of contaminants) and one-to-one sediment to water …

Physics and numerics of the tensor code (incomplete preliminary documentation)

The present TENSOR code is a descendant of a code originally conceived by Maenchen and Sack and later adapted by Cherry. Originally, the code was a two-dimensional Lagrangian explicit finite difference code which solved the equations of continuum mechanics. Since then, implicit and arbitrary Lagrange-Euler (ALE) algorithms have been added. The code has been used principally to solve problems involving the propagation of stress waves through earth materials, and considerable development of rock and soil constitutive relations has been done. The code has been applied extensively to the containment of underground nuclear tests, nuclear and high explosive surface and subsurface cratering, and energy and resource recovery. TENSOR is supported by a substantial array of ancillary routines. The initial conditions are set up by a generator code TENGEN. ZON is a multipurpose code which can be used for zoning, rezoning, overlaying, and linking from other codes. Linking from some codes is facilitated by another code RADTEN. TENPLT is a fixed time graphics code which provides a wide variety of plotting options and output devices, and which is capable of producing computer movies by postprocessing problem dumps. The Smart Grid: An Estimation of the Energy and CO2 Benefits

This report articulates nine mechanisms by which the smart grid can reduce energy use and carbon impacts associated with electricity generation and delivery. The quantitative estimates of potential reductions in electricity sector energy and associated CO2 emissions presented are based on a survey of published results and simple analyses. This report does not attempt to justify the cost effectiveness of the smart grid, which to date has been based primarily upon the twin pillars of cost-effective operation and improved reliability. Rather, it attempts to quantify the additional energy and CO2 emission benefits inherent in the smart grid’s potential contribution to the nation’s goal of mitigating climate change by reducing the carbon footprint of the electric power system.

Development of Microorganisms with Improved Transport and Biosurfactant Activity for Enhanced Oil Recovery: Final Report

The project had three objectives: (1) to develop microbial strains with improved biosurfactant properties that use cost-effective nutrients, (2) to obtain biosurfactant strains with improved transport properties through sandstones, and (3) to determine the empirical relationship between surfactant concentration and interfacial tension and whether in situ reactions kinetics and biosurfactant concentration meets appropriate engineering design criteria. Here, we show that a lipopeptide biosurfactant produced by Bacillus mojavensis strain JF-2 mobilized substantial amounts of residual hydrocarbon from sand-packed columns and Berea sandstone cores when a viscosifying agent and a low molecular weight alcohol were present. The amount of residual hydrocarbon mobilized depended on the biosurfactant concentration. Tertiary oil recovery experiments showed that 10 to 40 mg/l of JF-2 biosurfactant in the presence of 0.1 mM 2,3-butanediol and 1 g/l of partially hydrolyzed polyacrylamide (PHPA) recovered 10-40% of residual oil from Berea sandstone cores. Even low biosurfactant concentrations (16 mg/l) mobilized substantial amounts of residual hydrocarbon (29%). The bio-surfactant lowered IFT by nearly 2 orders of magnitude compared to typical IFT values of 28-29 mN/m. Increasing the salinity increased the IFT with or without 2,3-butanediol present.
