Interim Report on SNP analysis and forensic microarray probe design for South American hemorrhagic fever viruses, tick-borne encephalitis virus, henipaviruses, Old World Arenaviruses, filoviruses, Crimean-Congo hemorrhagic fever viruses, Rift Valley fever

The goal of this project is to develop forensic genotyping assays for select agent viruses, enhancing the current capabilities for the viral bioforensics and law enforcement community. We used a multipronged approach combining bioinformatics analysis, PCR-enriched samples, microarrays and TaqMan assays to develop high resolution and cost effective genotyping methods for strain level forensic discrimination of viruses. We have leveraged substantial experience and efficiency gained through year 1 on software development, SNP discovery, TaqMan signature design and phylogenetic signature mapping to scale up the development of forensics signatures in year 2. In this report, we have summarized the whole genome wide SNP analysis and microarray probe design for forensics characterization of South American hemorrhagic fever viruses, tick-borne encephalitis viruses and henipaviruses, Old World Arenaviruses, filoviruses, Crimean-Congo hemorrhagic fever virus, Rift Valley fever virus and Japanese encephalitis virus.

Evaluation of the hazard associated with fabricating beryllium copper alloys

Beryllium-copper alloys should be considered toxic materials and proper controls must be used when they are machined, heated, or otherwise fabricated. Air samples should be taken for each type of fabrication to determine the worker's exposure and the effectiveness of the controls in use. It has been shown that aerosols containing beryllium are generated during the four methods of fabrication tested, and that these aerosols can be reduced through local exhaust to undetectable levels. Considering the acute, chronic and possibly carcinogenic effects of exposure to beryllium, effective controls should be required because they are feasible both technologically and economically. The health hazards and control measures are reviewed.

Tank 241-AY-102 Data Report

This data report discusses the methods and philosophy used to characterize two samples of sludge and one sample of drainable liquid from tank 241-AY-102 at the Hanford Site. Archived samples of sludge and drainable liquid from tank 241-AY-102 were characterized in the laboratory in order to evaluate analytical methods for testing tank waste and determine the composition and leaching characteristics of this material. The tests included physical characterization, quantitative analysis of waste composition, and short-term water leach and acid digestion of the waste material. The water leach tests were conducted over time periods of one day, two weeks, and one month to determine if contact time had an impact on leachability. Comparisons of the results of the water leach tests with the acid digestions allow for an estimation of the water leachable percentage of an element. The average water leachability of Tc-99 was measured at 20% over the one-month time frame of the leach tests. There did not appear to be any temporal change in water leachability by comparing the results of the three sampling intervals. Approximately 24 to 48% of the total Cs-137 in the sludge is water leachable over a time period of one day to one month. …

A comparison of the experimental subsonic pressure distributions about several bodies of revolution with pressure distributions computed by means of the linearized theory

"An analysis is made of the effects of compressibility on the pressure coefficients about several bodies of revolution by comparing experimentally determined pressure coefficients with corresponding pressure coefficients calculated by the use of the linearized equations of compressible flow. The results show that the theoretical methods predict the subsonic pressure-coefficient changes over the central part of the body but do not predict the pressure-coefficient changes near the nose. Extrapolation of the linearized subsonic theory into the mixed subsonic-supersonic flow region fails to predict a rearward movement of the negative pressure-coefficient peak which occurs after the critical stream Mach number has been attained" (p. 1125).

Restoration of Secondary Containment in Double Shell Tank (DST) Pits

Cracks found in many of the double-shell tank (DST) pump and valve pits bring into question the ability of the pits to provide secondary containment and remain in compliance with State and Federal regulations. This study was commissioned to identify viable options for maintain/restoring secondary containment capability in these pits. The basis for this study is the decision analysis process which identifies the requirements to be met and the desired goals (decision criteria) that each option will be weighed against. A facilitated workshop was convened with individuals knowledgeable of Tank Farms Operations, engineering practices, and safety/environmental requirements. The outcome of this workshop was the validation or identification of the critical requirements, definition of the current problem, identification and weighting of the desired goals, baselining of the current repair methods, and identification of potential alternate solutions. The workshop was followed up with further investigations into the potential solutions that were identified in the workshop and through other efforts. These solutions are identified in the body of this report. Each of the potential solutions were screened against the list of requirements and only those meeting the requirements were considered viable options. Near-frictionless carbon coatings for spark-ignited direct-injected fuel systems. Final report, January 2002.

This report describes an investigation by the Tribology Section of Argonne National Laboratory (ANL) into the use of near-frictionless carbon (NFC) coatings for spark-ignited, direct-injected (SIDI) engine fuel systems. Direct injection is being pursued in order to improve fuel efficiency and enhance control over, and flexibility of, spark-ignited engines. SIDI technology is being investigated by the Partnership for a New Generation of Vehicles (PNGV) as one route towards meeting both efficiency goals and more stringent emissions standards. Friction and wear of fuel injector and pump parts were identified as issues impeding adoption of SIDI by the OTT workshop on ''Research Needs Related to CIDI and SIDI Fuel Systems'' and the resulting report, Research Needs Related to Fuel Injection Systems in CIDI and SIDI Engines. The following conclusions were reached: (1) Argonne's NFC coatings consistently reduced friction and wear in existing and reformulated gasolines. (2) Compared to three commercial DLC coatings, NFC provided the best friction reduction and protection from wear in gasoline and alternative fuels. (3) NFC was successfully deposited on production fuel injectors. (4) Customized wear tests were performed to simulate the operating environment of fuel injectors. Background Radioactivity in River and Reservoir Sediments near Los Alamos, New Mexico

As part of its continuing Environmental Surveillance Program, regional river and lake-bottom sediments have been collected annually by Los Alamos National Laboratory (the Laboratory) since 1974 and 1979, respectively. These background samples are collected from three drainage basins at ten different river stations and five reservoirs located throughout northern New Mexico and southern Colorado. Radiochemical analyses for these sediments include tritium, strontium-90, cesium-137, total uranium, plutonium-238, plutonium-239,-240, americium-241, gross alpha, gross beta, and gross gamma radioactivity. Detection-limit radioactivity originates as worldwide fallout from aboveground nuclear weapons testing and satellite reentry into Earth's atmosphere. Spatial and temporal variations in individual analyte levels originate from atmospheric point-source introductions and natural rate differences in airborne deposition and soil erosion. Background radioactivity values on sediments reflect this variability, and grouped river and reservoir sediment samples show a range of statistical distributions that appear to be analyte dependent. Traditionally, both river and reservoir analyte data were blended together to establish background levels. In this report, however, we group background sediment data according to two criteria. These include sediment source (either river or reservoir sediments) and station location relative to the Laboratory (either upstream or downstream). Particle physics aims to understand the universe around us. The Standard Model of particle physics describes the basic structure of matter and forces, to the extent we have been able to probe thus far. However, it leaves some big questions unanswered. Some are within the Standard Model itself, such as why there are so many fundamental particles and why they have different masses. In other cases, the Standard Model simply fails to explain some phenomena, such as the observed matter-antimatter asymmetry in the universe, the existence of dark matter and dark energy, and the mechanism that reconciles gravity with quantum mechanics. These gaps lead us to conclude that the universe must contain new and unexplored elements of Nature. Most of particle and nuclear physics is directed towards discovering and understanding these new laws of physics. These questions are best pursued with a variety of approaches, rather than with a single experiment or technique. Particle physics uses three basic approaches, often characterized as exploration along the cosmic, energy, and intensity frontiers. Each employs different tools and techniques, but they ultimately address the same fundamental questions. Highly Dispersed Pseudo-Homogeneous and Heterogeneous Catalysts Synthesized via Inverse Micelle Solutions for the Liquefaction of Coal

The mission of this project was to use inverse micelle solutions to synthesize nanometer sized metal particles and test the particles as catalysts in the liquefaction of coal and other related reactions. The initial focus of the project was the synthesis of iron based materials in pseudo-homogeneous form. The frost three chapters discuss the synthesis, characterization, and catalyst testing in coal liquefaction and model coal liquefaction reactions of iron based pseudo-homogeneous materials. Later, we became interested in highly dispersed catalysts for coprocessing of coal and plastic waste. Bifunctional catalysts . to hydrogenate the coal and depolymerize the plastic waste are ideal. We began studying, based on our previously devised synthesis strategies, the synthesis of heterogeneous catalysts with a bifunctional nature. In chapter 4, we discuss the fundamental principles in heterogeneous catalysis synthesis with inverse micelle solutions. In chapter 5, we extend the synthesis of chapter 4 to practical systems and use the materials in catalyst testing. Finally in chapter 6, we return to iron and coal liquefaction now studied with the heterogeneous catalysts.

Radar-cross-section reduction of wind turbines. part 1.

In recent years, increasing deployment of large wind-turbine farms has become an issue of growing concern for the radar community. The large radar cross section (RCS) presented by wind turbines interferes with radar operation, and the Doppler shift caused by blade rotation causes problems identifying and tracking moving targets. Each new wind-turbine farm installation must be carefully evaluated for potential disruption of radar operation for air defense, air traffic control, weather sensing, and other applications. Several approaches currently exist to minimize conflict between wind-turbine farms and radar installations, including procedural adjustments, radar upgrades, and proper choice of low-impact wind-farm sites, but each has problems with limited effectiveness or prohibitive cost. An alternative approach, heretofore not technically feasible, is to reduce the RCS of wind turbines to the extent that they can be installed near existing radar installations. This report summarizes efforts to reduce wind-turbine RCS, with a particular emphasis on the blades. The report begins with a survey of the wind-turbine RCS-reduction literature to establish a baseline for comparison. The following topics are then addressed: electromagnetic model development and validation, novel material development, integration into wind-turbine fabrication processes, integrated-absorber design, and wind-turbine RCS modeling. Selection of Sampling Pumps Used for Groundwater Monitoring at the Hanford Site

The variable frequency drive centrifugal submersible pump, Redi-Flo2a made by Grundfosa, was selected for universal application for Hanford Site groundwater monitoring. Specifications for the selected pump and five other pumps were evaluated against current and future Hanford groundwater monitoring performance requirements, and the Redi-Flo2 was selected as the most versatile and applicable for the range of monitoring conditions. The Redi-Flo2 pump distinguished itself from the other pumps considered because of its wide range in output flow rate and its comparatively moderate maintenance and low capital costs. The Redi-Flo2 pump is able to purge a well at a high flow rate and then supply water for sampling at a low flow rate. Groundwater sampling using a low-volume-purging technique (e.g., low flow, minimal purge, no purge, or micropurges) is planned in the future, eliminating the need for the pump to supply a high-output flow rate. Under those conditions, the Well Wizard bladder pump, manufactured by QED Environmental Systems, Inc., may be the preferred pump because of the lower capital cost.

Task 1.5 Genomic Shift and Drift Trends of Emerging Pathogens

The Lawrence Livermore National Laboratory (LLNL) Bioinformatics group has recently taken on a role in DTRA's Transformation Medical Technologies Initiative (TMTI). The high-level goal of TMTI is to accelerate the development of broad-spectrum countermeasures. To achieve those goals, TMTI has a near term need to conduct analyses of genomic shift and drift trends of emerging pathogens, with a focused eye on select agent pathogens, as well as antibiotic and virulence markers. Most emerging human pathogens are zoonotic viruses with a genome composed of RNA. The high mutation rate of the replication enzymes of RNA viruses contributes to sequence drift and provides one mechanism for these viruses to adapt to diverse hosts (interspecies transmission events) and cause new human and zoonotic diseases. Additionally, new viral pathogens frequently emerge due to genetic shift (recombination and segment reassortment) which allows for dramatic genotypic and phenotypic changes to occur rapidly. Bacterial pathogens also evolve via genetic drift and shift, although sequence drift generally occurs at a much slower rate for bacteria as compared to RNA viruses. However, genetic shift such as lateral gene transfer and inter- and intragenomic recombination enables bacteria to rapidly acquire new mechanisms of survival and antibiotic resistance. Radiochemical detector cross section sensitivity studies in the A approx. = 90 mass region

The use of radiochemical detectors in the A approx. = 90 mass region to measure 14 MeV neutron fluences is investigated from the standpoint of cross section sensitivities. Specifically, /sup 90/Zr and /sup 89/Y neutron-induced cascades leading to the production of /sup 89/Zr, /sup 88/Zr, /sup 88/Y, and /sup 87/Y are studied in a one-energy-group approximation, and the sensitivities of the measured ratios R/sub Z/ = /sup 88/Zr//sup 89/Zr and R/sub Y/ = /sup 87/Y//sup 88/Y to the input cross sections are delineated. The most sensitive cross section in each cascade is the cross section that dominates the production of the final isotope in the chain. Only one of the isomeric levels in /sup 90/Zr contributes importantly to the Zr cascade. Burnback (n,..gamma..) and burnup (n,p) reactions are also considered. The (n,..gamma..) effects are small, but (n,p) effects can be substantial due to low energy enhancements of (n,p) cross sections in the neutron-deficient isotopes.

Predicting tropospheric ozone and hydroxyl radical in a global, three-dimensional, chemistry, transport, and deposition model

Two of the most important chemically reactive tropospheric gases are ozone (O{sub 3}) and the hydroxyl radical (OH). Although ozone in the stratosphere is a necessary protector against the sun`s radiation, tropospheric ozone is actually a pollutant which damages materials and vegetation, acts as a respiratory irritant, and is a greenhouse gas. One of the two main sources of ozone in the troposphere is photochemical production. The photochemistry is initiated when hydrocarbons and carbon monoxide (CO) react with nitrogen oxides (NO{sub x} = NO + NO{sub 2}) in the presence of sunlight. Reaction with the hydroxyl radical, OH, is the main sink for many tropospheric gases. The hydroxyl radical is highly reactive and has a lifetime on the order of seconds. Its formation is initiated by the photolysis of tropospheric ozone. Tropospheric chemistry involves a complex, non-linear set of chemical reactions between atmospheric species that vary substantially in time and space. To model these and other species on a global scale requires the use of a global, three-dimensional chemistry, transport, and deposition (CTD) model. Scaling up of Carbon Exchange Dynamics from AmeriFlux Sites to a Super-Region in the Eastern United States

The primary objective of this project was to evaluate carbon exchange dynamics across a region of North America between the Great Plains and the East Coast. This region contains about 40 active carbon cycle research (AmeriFlux) sites in a variety of climatic and landuse settings, from upland forest to urban development. The core research involved a scaling strategy that uses measured fluxes of CO{sub 2}, energy, water, and other biophysical and biometric parameters to train and calibrate surface-vegetation-atmosphere models, in conjunction with satellite (MODIS) derived drivers. To achieve matching of measured and modeled fluxes, the ecosystem parameters of the models will be adjusted to the dynamically variable flux-tower footprints following Schmid (1997). High-resolution vegetation index variations around the flux sites have been derived from Landsat data for this purpose. The calibrated models are being used in conjunction with MODIS data, atmospheric re-analysis data, and digital land-cover databases to derive ecosystem exchange fluxes over the study domain.
