Developing Informatics Tools and Methods Utilizing Whole Genome Sequencing and Transcription Data to Aid Gene Discovery in Medicago truncatula

Research into the mechanism of symbiotic nitrogen fixation between legumes and rhizobia involves a complex interaction between the organisms, and many genes involved in this remain either uncharacterized or undiscovered. Using forward genetics, mutant plant lines are screened to find new genes without reliance on software-based gene prediction. A large population of Tnt1-mutagenized Medicago truncatula lines is used for this purpose. Herein, the aid of tools like whole genome sequencing (WGS) in this process is explored so that new methods and tools are elucidated. The use of WGS data allows for rapid prediction of all insertions in the genome and has been shown to predict insertion locations that were missed by the TAIL-PCR-based Tnt1 mutant database already in existence. This WGS strategy has been successfully used to find the causal mutations in multiple plant lines. Two WGS strategies are used to analyze insertions in nine sequenced lines and compared with each other and the existing Tnt1 mutant database. It appears that using either WGS method will yield similar results, but the TAIL-PCR-based predictions have much less overlap. Aromaticity, Supramolecular Stacks, and Luminescence Properties of Cyclic Trinuclear Complexes

The dissertation covers three major topics: metal-assisted aromaticity, synthetic approaches to tailor donor-acceptor supramolecular stacks, and photoluminescence properties of cyclic trinuclear complexes (CTCs) of d10 metals. First, multiple theoretical approaches are adapted to discuss in detail the origin of aromaticity of CTCs, putting forward a metal-assisted aromaticity model. Next are the discoveries of donor-acceptor stacked CTC–CTC' complexes from both experimental and computational perspectives, reporting multiple novel crystallography-determined structures and revealing their pertinent intermolecular ground-state charge transfer. The spontaneous binding behavior is also determined by UV-vis and NMR titrations and rationalized as the cooperation of multiple supramolecular interactions, including metallophilicity, electrostatic attraction, and dispersion. The last part includes systematic investigations of photoluminescence properties of halogen-metal-bonded CTCs and sandwich-like cation–π-bonded heptanuclear clusters based on CTCs. The cooperative effects of metal-centered conformation, the heavy-atom and relativistic effects from both the halogen and metal atoms play complementary roles in the phosphorescence process to promote the inter-system crossing and radiative transitions.

Temperature Change and Its Consequences for the Physiology of the Eurythermic Sheepshead Minnow (Cyprinodon variegatus)

The estuarine sheepshead minnow (Cyprinodon variegatus) is the most eurythermic fish species, with a thermal tolerance window between 0.6°C and 45.1°C. However, little is known about the physiological mechanisms that allow this species to survive this temperature range. In order to understand how sheepshead minnow physiology is affected by temperature acclimation and acute changes in temperature, I conducted research on this species using a multi-level approach. I began at the organismal level, and examined the effects of these temperature changes on the sheepshead minnow's metabolic rate and swimming performance. The next chapter investigated the effects of changing temperatures on cardiac function (i.e., tissue/organ specific effects). In the final chapter, I conducted research at the sub-cellular level, and determined how mitochondrial bioenergetics / function is impacted by changing temperatures. This research shows that while sheepshead minnows are able to sustain heart function and mitochondrial respiration over a broad range of temperatures; they also display a plastic temperature response which is associated with the downregulation of standard metabolic rate and cardiac remodeling to maintain force generation. Collectively, these physiological responses may contribute to the sheepshead minnow's ability to maintain physiological and organismal function across a large temperature range.

The Relationship between Racial Colorblindness and the Self-Reported Implementation of Multicultural Teaching Practices among Teachers of Gifted Students

Culturally responsive instruction is recommended as a tool to help mitigate disparities in the recruitment and retention of culturally diverse students in gifted programs. However, the endorsement of colorblind racial ideology has been associated with lower multicultural teaching competency and the adoption of deficit perspectives among teachers. In addition to informing teachers' perceptions of students' abilities and potential, endorsing colorblind racial ideology may make it more difficult for teachers to recognize disparities that adversely impact students from traditionally minoritized and underrepresented groups. The current study examines the relationship between colorblind racial attitudes and multicultural teaching competency among teachers of students who have been identified as gifted and talented. In a large sample of Texas teachers, multiple regression analysis was used to examine the relationship between colorblind racial ideology, multicultural teaching knowledge, and the implementation of culturally responsive teaching practices when teaching gifted and talented students. Results indicate that racial colorblindness, sex, and multicultural teaching knowledge are all significant predictors of the self-reported implementation of culturally responsive instruction, while gifted-specific training and the proportion of students of color had no effects.
