Identification and Characterization of the Pyrimidine Biosynthetic Operon in Streptomyces griseus (open access)

Identification and Characterization of the Pyrimidine Biosynthetic Operon in Streptomyces griseus

To further understand the ATCase/DHOase bifunctional complex formed in Streptomyces, the genes encoding these and other pyrimidine enzymes were identified and characterized. Polymerase chain reaction (PCR) was utilized in this effort. Primers were constructed by selecting conserved regions of pyrimidine genes from known gene and protein sequences of a wide variety of organisms. These sequences were then optimized to Streptomyces codon usage. PCR products were obtained from internal sites within pyrimidine genes and also from primer combinations of different genes. The size, orientation, and partial sequence of the resulting products shows that Streptomyces has a gene organization of pyrR followed by pyrB, pyrC, carA, carB, and pyrF in an operon similar to that found in other Gram-positive bacteria.

Re-examining the Battle of the Bulge : Assessing the Role of Strategic Intelligence and Coalition Warfare Against the 1944 Wehrmacht

The 1944 German Ardennes offensive failed. It was overly ambitious, built on erroneous assumptions, insufficiently supported by logistics, and depended on the weather for success. Yet, the offensive achieved more than it should have given the strength and combat experience of the Allied armies in Europe. Previous attempts to explain the limited success of the German offensive have emphasized the failure of Allied strategic intelligence - Ultra. Intelligence is an accurate, but incomplete explanation for initial German success in the Ardennes. Three conditions allowed the Wehrmacht, approaching its manpower and logistical end, to crush the US First Army. First, coalition warfare so weakened the First Army that it became vulnerable to attack. Second, the Allies failed to develop a unified intelligence network capable of assessing the information that indicated the timing and target of the German attack. Finally, a well-executed German security and deception plan surprised the Allies. The well-executed German offensive manipulated both Allied intelligence and the Anglo-American coalition.

The National Museum of the Pacific War presents an oral interview with Robert E. Galer. Galer was born in Seattle 23 October 1913. In 1935, he graduated from the University of Washington, having earned a commission through ROTC. During flight training at Pensacola, he accepted a regular commission in the Marine Corps. His classmate at Washington was Gregory “Pappy” Boyington, who also trained with him at Quantico. During the ensuing years, Galer became carrier qualified and flew sea planes in the Virgin Islands, had a tour of duty with a fighter squadron in San Diego, and had fighter training in Honolulu. He watched the Japanese attack Pearl Harbor from a friend’s house. Galer went to Guadalcanal in late August 1942, where he flew interdiction missions in the Grumman F4F Wildcat. In fighting for Henderson Field, Galer was shot down three times, was credited with shooting down 11 ½ Japanese planes, and received the Medal of Honor. After receiving the award, he went on speaking tours to sell war bonds. Afterwards, he attended the Army Command and Staff School at Fort Leavenworth. In early 1945, he was assigned as the co-leader of a 25-man team responsible for evaluating a new radar …
