Advanced Dewatering Systems Development
A new fine coal dewatering technology has been developed and tested in the present work. The work was funded by the Solid Fuels and Feedstocks Grand Challenge PRDA. The objective of this program was to 'develop innovative technical approaches to ensure a continued supply of environmentally sound solid fuels for existing and future combustion systems with minimal incremental fuel cost.' Specifically, this solicitation is aimed at developing technologies that can (i) improve the efficiency or economics of the recovery of carbon when beneficiating fine coal from both current production and existing coal slurry impoundments and (ii) assist in the greater utilization of coal fines by improving the handling characteristics of fine coal via dewatering and/or reconstitution. The results of the test work conducted during Phase I of the current project demonstrated that the new dewatering technologies can substantially reduce the moisture from fine coal, while the test work conducted during Phase II successfully demonstrated the commercial viability of this technology. It is believed that availability of such efficient and affordable dewatering technology is essential to meeting the DOE's objectives.

Yucca Mountain licensing support network archive assistant.
This report describes the Licensing Support Network (LSN) Assistant--a set of tools for categorizing e-mail messages and documents, and investigating and correcting existing archives of categorized e-mail messages and documents. The two main tools in the LSN Assistant are the LSN Archive Assistant (LSNAA) tool for recategorizing manually labeled e-mail messages and documents and the LSN Realtime Assistant (LSNRA) tool for categorizing new e-mail messages and documents. This report focuses on the LSNAA tool. There are two main components of the LSNAA tool. The first is the Sandia Categorization Framework, which is responsible for providing categorizations for documents in an archive and storing them in an appropriate Categorization Database. The second is the actual user interface, which primarily interacts with the Categorization Database, providing a way for finding and correcting categorizations errors in the database. A procedure for applying the LSNAA tool and an example use case of the LSNAA tool applied to a set of e-mail messages are provided. Performance results of the categorization model designed for this example use case are presented.

Optical Fiber Sensor Instrumentation for Slagging Coal Gasifiers
Coal gasifier is one of the most promising solutions for clean fossil energy. Refractory thickness monitoring and online real-time temperature measurement is needed for improved reliability and advanced process control for current and future generation power plants. The objective of this program is to design and implement an optical fiber based sensing system that could potentially be used to monitor refractory wall thickness and temperature inside a coal gasifier. For the thickness monitoring, the system should be able to operate at temperatures up to 1000 C. For this temperature range, silica fiber can still work so it is chosen for the sensor design. The measurement is based on a photon counting optical time domain reflectometer. A narrow light pulse is launched into a silica fiber which could be embedded into the gasifier refractory wall, and is partially reflected by the far end of the fiber. The time of flight of the light pulse in the fiber then gives an indication of the position of the fiber end, which is a function of the wall thickness when the fiber is embedded. Results obtained show a measurement accuracy of {+-}2cm in environment of 1000 C with a saw cut fiber end. When …

Transcriptional Response of Desulfovibrio vulgaris Hildenborough to Oxidative Stress Mimicking Environmental Conditions
Sulphate-reducing bacteria are anaerobes readily found in oxic-anoxic interfaces. Multiple defence pathways against oxidative conditions were identified in these organisms and proposed to be differentially expressed under different concentrations of oxygen, contributing to their ability to survive oxic conditions. In this study, Desulfovibrio vulgaris Hildenborough cells were exposed to the highest concentration of oxygen that sulphate-reducing bacteria are likely to encounter in natural habitats, and the global transcriptomic response was determined. 307 genes were responsive, with cellular roles in energy metabolism, protein fate, cell envelope and regulatory functions, including multiple genes encoding heat shock proteins, peptidases and proteins with heat shock promoters. Of the oxygen reducing mechanisms of D. vulgaris only the periplasmic hydrogen-dependent mechanism is up-regulated, involving the [NiFeSe]hydrogenase, formate dehydrogenase(s) and the Hmc membrane complex. The oxidative defence response concentrates on damage repair by metal-free enzymes. These data, together with the down regulation of the Fur operon, which restricts the availability of iron, and the lack of response of the PerR operon, suggest that a major effect of this oxygen stress is the inactivation and/or degradation of multiple metalloproteins present in D. vulgaris as a consequence of oxidative damage to their metal clusters.
