Regulation of Colony-Stimulating Factor-1 Biosynthesis

Recent studies suggest that synthesis of the Colony-stimulating factor (CSF) is a well regulated process. However, the molecular mechanisms of the signal transduction of the various inducers of CSF such as monokines and lymphokines are not well understood. Using Interleukin 1 (IL-1) stimulation of CSF-1 in the MIA PaCa-2 cell line as a model system, the involvement of G-protein has been studied. The IL-1 induction of CSF-1 synthesis can be inhibited by both Pertussis toxin and Cholera toxin, which are known to modify the Gᵢ and Gₛ proteins respectively, thus activating adenylate cyclase to release more cAMP. The toxin inactivation can be prevented by inhibitors of the ADP-ribosylation such as, benzamide and MBAMG. Addition of dibutyryl-cAMP inhibits the IL-1 induced CSF production. Both Theophylline and Forskolin which increase cAMP by inhibiting phosphodiesterase and stimulating adenylate cyclase respectively, also inhibit CSF-1 production. Results from these studies have shown that cAMP level inversely regulates the biosynthesis of CSF-1. Preincubation of MIA PaCa-2 cells with IL-1 and 5'- guanylylimidodiphosphate (GppNHp) prevents the inhibitory effect of pertussis toxin on CSF-1 production. Exploration of Genome Length, Burst Time, and Burst Size of Streptomyces griseus Bacteriophages

Since phages use the host resources to replicate themselves after infection, the different sizes of the phage genome should influence the replication rate. We, therefore, hypothesized that the smaller genomes should burst the cell faster than the larger ones. As well, the shorter genomes would have greater burst sizes because they should replicate faster. Here, we obtained 16 phages of various genome length. All phages were isolated on Streptomyces griseus and available in our phage bank at the University of North Texas. We performed one-step growth studies for the 16 phages, as well as determined the host doubling time from its growth curve. The results show that S. griseus grown in nutrient broth has a doubling time of 5 hours and 22 minutes. This doubling time is used as a guideline for the phage growth studies. Because the filamentous nature of the host caused several difficulties during the experiment, we isolated single cells by sonication and centrifugation. After the cell number was determined by viable cell count, the cells were infected with each type of phage using a multiplicity of infection (MOI) of 0.5. Ultraviolet Radiation Tolerance in High Elevation Copepods from the Rocky Mountains of Colorado, USA

Copepods in high elevation lakes and ponds in Colorado are exposed to significant levels of ultraviolet radiation (UV), necessitating development of UV avoidance behavior and photoprotective physiological adaptations. The copepods are brightly pigmented due to accumulation of astaxanthin, a carotenoid which has photoprotective and antioxidant properties. Astaxanthin interacts with a crustacyanin-like protein, shifting its absorbance from 473 nm (hydrophobic free form, appears red) to 632 nm (protein-bound complex, appears blue). In six sites in Colorado, habitat-specific coloration patterns related to carotenoprotein complex have been observed. The objective of this study was to determine whether pigment accumulation or carotenoprotein expression has a greater effect on resistance to UV exposure. For each site, copepod tolerance to UV was assessed by survivorship during UV exposure trials. Average UV exposure was determined for each habitat. Astaxanthin profiles were generated for copepods in each site. Ability to withstand UV exposure during exposure trials was significantly different between color morphs (p < 0.0001). Red copepods were found to tolerate 2-fold greater levels of UVB than blue or mixed copepods. Effects of Trichinella Soluble Antigens on Macrophage Subpopulations

The immunomodulatory effects of Trichinella spiralis or Trichinella pseudospiralis soluble antigen extracts were examined in an effort to characterize the differences in immune responses seen during these Trichinella infections. The newborn larvae extracts of either parasite exhibited similar potency for stimulating macrophage PGE production; however, the muscle larvae extracts of T. pseudospiralis stimulated greater levels of PGE than did the muscle larvae extracts of T. spiralis. These data clearly indicate that Trichinella antigens possess immunomodulatory capabilities.

This thesis describes the development of an optimized method for culturing dissociated, monolayer neuronal networks from murine frontal cortex and midbrain. It is presented as a guidebook for use by cell culture specialists and laboratory personnel who require updated and complete procedures for use with microelectrode array (MEA) recording technology. Specific cell culture protocols, contamination prevention and control, as well common problems encountered within the cell culture facility, are discussed. This volume offers value and utility to the rapidly expanding fields of MEA recording and neuronal cell culture. Due to increasing interest in determining the mechanisms underlying Parkinson's disease, the newly developed procedures for mesencephalon isolation and culture on MEAs are an important research contribution.

Cowpea, Vigna unguiculata (L.) Walp is an excellent source of protein, vitamins and minerals and a major food crop many parts of Africa. Optimal production levels are hampered by insect pests and diseases. Biotechnological techniques such as tissue culture and genetic engineering can aid in the development of varieties with resistance to insect pests and diseases. The objective of this study was to investigate conditions necessary for the development of a reproducible tissue culture system that can be applied to regenerate transformed cells from culture. The in vitro manipulation of cowpea using Murashige and Skoog (MS) medium, auxins and cytokinins resulted in the formation of callus and rhizogenesis. Calli that were formed were separated into six classes based on color and texture. Yellowish friable callus, yellowish compact, soft yellowish callus and green and white were composed of largely vacuolated cells and were non-regenerative. Friable green callus was the most prevalent callus type and could form of roots in some hormone combinations. Green spots were formed on hard compact green callus. The green spots became nodular, forming root primordia and ultimately giving rise to roots. None of the six calli types gave rise to the formation of shoots. Embryogenic callus was …

Characterization of the Pigment-Protein and Pigment-ester of Xanthomonas Campestris Pv. Juglandis

The objectives of this project were to develop a high performance liquid chromatographic method for separating the pigment esters mixture, to determine the locations of the pigment moiety in the isolated esters using pholosiphases, and to characterize the pigment-protein complex and determine its distribution in other bacteria. Saponification of the two pigment esters 1 and 2 with aqueous KOH yielded two free pigments on TLC plates developed by two solvent systems. The fasters moving of these two free pigments co-chromatographed with the one free pigment produced from each pigment ester by phospholipase A2 treatment. This suggests that the pigment molecule is a methoxy derivative of xanthomonadin and is esterified to the 2-position of the glycerol moiety of each pigment ester. No free pigment was released from phospholipases C and D treatment of the two pigment esters, indicating that pigment is not esterified to the sorbitol or phosphate moiety of pigment esters 1 or 2.

Determination of Dissociation Constants for GABAA Receptor Antagonists using Spontaneously Active Neuronal Networks in vitro

Changes in spontaneous spike activities recorded from murine frontal cortex networks grown on substrate-integrated microelectrodes were used to determine the dissociation constant (KB) of three GABAA antagonists. Neuronal networks were treated with fixed concentrations of GABAA antagonists and titrated with muscimol, a GABAA receptor agonist. Muscimol decreased spike activity in a concentration dependent manner with full efficacy (100% spike inhibition) and a 50% inhibitory concentration (IC50) of 0.14 ± 0.05 µM (mean ± SD, n=6). At 10, 20, 40 and 80 µM bicuculline, the muscimol IC50 values were shifted to 4.3 ± 1.8 µM (n=6), 6.8 ± 1.7 µM (n=6), 19.3 ± 3.54 µM (n=10) and 43.5 µM (n=2), respectively (mean ± SD). Muscimol titration in the presence of 10, 20, 40 µM of gabazine resulted in IC50s values of 20.1 (n=2), 37.17 (n=4), and 120.45 (n=2), respectively. In the presence of 20, 80, and 160 µM of TMPP (trimethylolpropane phosphate) the IC50s were 0.86 (n=2), 3.07 (n=3), 6.67 (n=2) µM, respectively. Increasing concentrations of GABAA antagonists shifted agonist log concentration-response curves to the right with identical efficacies, indicating direct competition for the GABAA receptor. Throughout history floods have been part of the human experience. They are recurring phenomena that form a necessary and enduring feature of all river basin and lowland coastal systems. In an average year, they benefit millions of people who depend on them. In the more developed countries, major floods can be the largest cause of economic losses from natural disasters, and are also a major cause of disaster-related deaths in the less developed countries. Flood disaster mitigation research was conducted to determine how remotely sensed data can effectively be used to produce accurate flood plain maps (FPMs), and to identify/quantify the sources of error associated with such data. Differences were analyzed between flood maps produced by an automated remote sensing analysis tailored to the available satellite remote sensing datasets (rFPM), the 100-year flooded areas "predicted" by the Flood Insurance Rate Maps, and FPMs based on DEM and hydrological data (aFPM). Landuse/landcover was also examined to determine its influence on rFPM errors. These errors were identified and the results were integrated in a GIS to minimize landuse / landcover effects. An Interpretation of Archaic Medical Treatises

Ancient peoples did not distinguish between philosophy, religion, and science. Scientific truth did not exist apart from divine truth. Any new idea, finding, or theory was assimilated into a monolithic mythological structure. This is one of the causes of the underestimation of ancient science: it is always packaged in a myth - the method of preserving information in an oral culture. The mythological medium allowed the preservation and dissemination of hard-won, empirical, scientific knowledge through generations of preliterate peoples. The context for mythological memorization, or simply tradition, needed to be easily and naturally acquired. The ideal context was the anthropomorphic context, the ontogenic context. This is the Grand Allegory - the anthropomorphization of information. Biomyths are essentially biological texts allegorized in esoteric language.

Isolating novel phages using Streptomyces azureus, which produces antibiotic thiostrepton, as a host, and characterizing the genomes may help us to find new tools that could be used to develop antibiotics in addition to contribute to the databases of phages and specifically, Streptomyces phages. Streptomyces phages Alsaber, Omar, Attoomi, Rowa, and ZamZam were isolated using during this study. They were isolated from enriched soil and sequenced by Illumina sequencing method. They were isolated from three different geographical regions. They are siphoviridae phages that create small clear plaques with a diameter of approximately 0.5-1 mm, except for Rowa which has cloudy plaques, and they have varied sizes of their heads and tails. ZamZam was not characterized at this time. The sequencing shows that they are circular genome with 3' sticky overhang and various genomes' sizes with high percentage of GC content with the average of 66%. Alsaber was classified under sub-cluster BD3, while Omar was categorized under sub-cluster BD2. They share the same cluster of Cluster BD. Rowa was placed in Cluster BL and Attoomi is currently a singleton that does not fit into an established cluster. Alsaber yields 76 putative genes with no tRNA, Omar 81 putative genes with 1 …

Investigating the Spatial Relationship between Suicide and Race/Ethnicity: The Case for Alternate Rate Adjustment Techniques in Medical Geography

This work explores potential distortions created by race and ethnicity on the visualization, interpretation, and understanding of the spatial distribution of suicide in the United States. Due to radically different suicide rates among racial/ethnic groups, traditional crude or age-adjusted rates may introduce statistical confounding in both linear and spatial models. Using correlation, choropleth mapping, hot spot analysis, and location-allocation modeling, this work shows how traditional methods of health system planning may unintentionally overlook elevated risk in minority-dominated areas like inner cities, the Texas/Mexico border region, and the Deep South. The final chapter introduces a simulation protocol for examining potential distortions in datasets to identify spatial and non-spatial distortions created by the underlying population composition. Methodologically, this dissertation contributes to the discourse on place context versus population composition. More generally, this research points to potential hazards to creating a more inclusive and equitable healthcare system.

Investigating the Effects of Inhaled Diesel Exhaust Particles on Gut Microbiome, Intestinal Integrity, Systemic Inflammation, and Biomarkers of Cardiovascular Disease in Wildtype Mice

We investigated the hypothesis that exposure to inhaled diesel exhaust PM can alter the gut microbiome and intestinal integrity, thereby promoting systemic inflammatory response and early CVD risk, which are exacerbated by HF diet. Furthermore, we investigated whether the observed exposure and diet-mediated outcomes could be mitigated through probiotic treatment. We performed an exposure study on C57Bl/6 male mice, placed on either a low fat (LF) diet or a high-fat (HF) diet, and exposed via oropharyngeal aspiration to 35 μg diesel exhaust particles (DEP) suspended in 35 μl of sterile saline or sterile saline controls (CON) twice a week for four weeks. A subset of mice on HF diet were dosed with 0.3 g/day (PRO, ~7.5x108 CFU/day) of probiotic Ecologic® Barrier 849 (Winclove Probiotics) in drinking water during the course of the study. For our first aim, we investigated the alterations in the gut microbiome, measured circulating cytokines and lipopolysaccharide (LPS), and measured CVD biomarkers in the heart. Our results revealed that exposure to inhaled DEP results in gut dysbiosis characterized by expansion of the phyla Verrucomicrobia and Proteobacteria and reduction in Actinobacteria, which was exacerbated by HF diet. Physiological Responses of Myriophyllum spicatum to Time Varying Exposures of Diquat, 2,4-D and Copper

The physiological responses of Myriophyllum spicatum to 2,4-D, diquat and copper were quantified using a plant tissue viability assay, and daily measures of dissolved oxygen and pH. Correlations of herbicide tissue residues to physiological response measures were determined and the relationship was used to develop exposure-response models. Diquat and copper had a greater effect on plant tissue viability than was observed for 2,4-D. Diquat produced greater reductions in dissolved oxygen concentrations and pH values than 2,4-D or copper. Copper exposure had the least effect on these parameters. Exposure-response models developed for 2,4-D predicted effective control at plant tissue residues ranging from 4000 to 4700 mg/kg. Aqueous exposure concentrations necessary to produce effective control plant tissue residues ranged from 0.20 to 0.40 mg/L. Exposure-response models developed for diquat predicted effective control at plant tissue residues ranging from 225 to 280 mg/kg. Aqueous exposure concentrations necessary to produce effective control plant tissue residues ranged from 0.113 to 0.169 mg/L. Systematics of Coccidian Parasites (Apicomplexa) from Amphibians and Reptiles in Northcentral Texas

Between February 1986 and October 1988, 863 amphibians and reptiles were collected in northcentral Texas and examined for coccidial parasites. Thirteen percent of amphibians <26% salamanders, 11% frogs and toads) and 28% of reptiles (54% turtles, 25% snakes) harbored 20 previously described and 16 new species of coccidia; overall prevalence of infection was 176/863 < 20%). Sixteen Ambvstoma texanum were infected with Eimeria ambystomae which represents new host and geographic locality records for the coccidium. Forty anurans were found to be passing coccidia, including Pseudacris streckeri. Bufo valliceos and Gastrophryne olivacea. Four new species of coccidia were described from anurans and include Eimeria flexuosa. E. streckeri. Isospora dellcatus and I_. fraaosum. However, oocysts found in B. v.. valliceps were determined experimentally to represent pseudoparasites. Sixty-eight turtles were infected with coccidia, including Chelvdra serpentina, Kinoeternon flavescens. Pseudemvs texana. Terrapeng ornata and Trachemvs scripta eleoans. Fourteen eimerians (5 of which are described as new species) were found in turtles. The new species from turtles include Eimeria cooteri, E. ornata, E. Btvlosa. E. texana and E. trachemvdis. Analysis of Students' Knowledge, Perceptions, and Interest in Engineering Post Teacher Participation in a National Science Foundation (NSF) Research Experience for Teachers (RET) Professional Development

This study examined the impact of the National Science Foundation's Research Experience for Teachers (RET) in engineering at University of North Texas on students after their teachers' participation in the program. Students were evaluated in terms of self-efficacy, knowledge of engineering, perceptions of engineering, and interest in engineering. A 22-item Likert pre/post survey was used for analysis, and participants included 589 students from six high schools, one middle school, and one magnet school. Paired surveys were analyzed to determine if there was a statistically significant difference in attitudes and knowledge after teachers implemented lessons from their time at the RET. Surveys were also analyzed to determine if there was a statistically significant difference in student response based on gender or student school type. Results showed no statistically significant difference in the self-efficacy of students, however there was a statistically significant difference in knowledge, perceptions, and interest in engineering. In addition, there was a statistically significant difference between genders on an isolated question, and seven out of the 22 Likert questions showed a statistically significant difference between student school types.

Evaluation of Diet, Water, and Culture Size for Ceriodaphnia Dubia Laboratory Culturing

Six reagent waters, eleven diets, and two culture sizes were evaluated for culturing C. dubia. Different filtration techniques were used to prepare the reagent waters. The eleven diets were comprised of two algae augmented with eight supplements. Reproduction and growth were assessed to discern differences among C. dubia raised in mass cultures and cultured in individual cups, during which, bacterial population densities, lipid, protein, and carbohydrate concentrations of the diets were measured. Results showed that a glass-distilled, carbon filtered, deionized reagent water and a Selenastrum capricornutum- Cerophyl® diet were optimum for culturing. Mass culturing supported the highest reproduction and growth, while no correlation was found between nutritional measurements and production.

Exploration of Explanatory Variables in the Creation of Linear Regression Models and Logistic Regression Models to Predict the Performance of Preservice Teachers on the Science Portion of the EC-6 TExES Certification Examination

The purpose of this study was to analyze the current and pre-service conditions that can affect student teachers' preparedness to pass the science portion of the EC-6 Texas Examinations for Educator Standards (TExES), one of the mandatory certification exam to become a teacher in Texas. Two types of prediction models were employed in this study: binomial logistic regression and multiple linear regression. The independent variables used in this study were: final grade in BIOL 1082, classification of students, transfer status, taken college biology, taken college chemistry, taken college physics, taken college environmental science, taken college earth science, attending college part-time, number of credits taken during the semester, first-generation college student, relatives with degree in education, and current GPA. The dependent variable of this study was the posttest score on science portion of the EC-6 TExES practice exam. A total of 170 preservice teachers participated this study. This study used students enrolled in BIOL 1082, who volunteered to take a Biology for Educators QualtricsTM survey and the EC-6 TExES practice exam in a pretest (start of semester) and posttest (end of semester) form. Mass Spectrometry-Based Identification of Ceramic-Bound Archaeological Protein Residues: Method Validation, Residue Taphonomy, and Prospects

Despite the variety of successful reports of the preservation, recovery, and identification of archaeological proteins in general, there are few positive reports regarding mass spectrometry-based identification of ceramic-bound proteins. In large part, this shortage is due to the lack of consideration for the unique taphonomic histories of such residues and, in general, methods development. Further, because negative results are rarely published, there is no baseline to which results can be compared. This paper attempts to address these challenges via a multi-pronged approach that uses mass spectrometry and complementary approaches to evaluate ceramic-bound protein preservation in both controlled, actualistic experiments, and in archaeological artifacts. By comparing the results obtained from protein-spiked, experimentally-aged ceramic to those obtained from both faunal and ceramic archaeological materials, an enhanced perspective on protein preservation and subsequent recovery and identification is revealed. This perspective, focusing on taphonomy, reveals why negative results may be the norm for ceramic artifacts when non-targeted methods are employed, and provides insight into how further method development may improve the likelihood of obtaining positive results.

Hemolytic disorders are characterized by hemolysis and are prone to thrombosis. Previously, it has been shown that the RNA released from damaged blood cells activates clotting. However, the nature of RNA released from hemolysis is still elusive. We found that after hemolysis, the red blood cells from both zebrafish and humans release 5.8S rRNA. This RNA activated coagulation in zebrafish and human plasmas. Using both natural and synthetic 5.8S rRNA and its synthetic truncated fragments, we found that the 3'-end 26 nucleotide-long RNA (3'-26 RNA) and its stem-loop secondary structure were necessary and sufficient for clotting activity. Corn trypsin inhibitor (CTI), a coagulation factor XII (FXII) inhibitor blocked 3'-26 RNA-mediated coagulation activation of both zebrafish and human plasma. CTI also inhibited zebrafish coagulation in vivo. 5.8S rRNA monoclonal antibody inhibited both 5.8S rRNA- and 3'-26 RNA-mediated zebrafish coagulation activity. Both 5.8S rRNA and 3'-26 RNA activates normal human plasma but did not activate FXII-deficient human plasma. Understanding Microbial Biodegradation of Environmental Contaminants

The accumulation of industrial contaminants in the natural environments have rapidly become a serious threat for human and animal life. Fortunately, there are microorganisms capable of degrading or transforming environmental contaminants. The present dissertation work aimed to understand the genomic basis of microbial degradation and resistance. The focus was the genomic study of the following bacteria: a) Pseudomonas fluorescens NCIMB 11764, a unique bacterium with specific enzymes that allow cyanide adaptation features. Potential cyanide degradation mechanisms found in this strain included nit1C cluster, and CNO complex. Potential cyanide tolerance genes found included cyanide insensitive oxidases, nitric oxide producing gene, and iron metabolism genes. b) Cupriavidus sp. strain SK-3 and strain SK-4. The genome of both bacteria presented the bph operon for polychlorinated biphenyl (PCB) degradation, but we found differences in the sequences of the genes. Those differences might indicate their preferences for different PCB substrates. c) Arsenic resistant bacterial communities observed in the Atacama Desert. Specific bacteria were found to thrive depending on the arsenic concentration. Examples were Bacteroidetes and Spirochaetes phyla whose proportions increased in the river with high arsenic concentrations. Expression of G-protein Coupled Receptors in Young and Mature Thrombocytes and Knockdown of Gpr18 in Zebrafish

In this study, a novel method based on biotinylated antibodies and streptavidin coated magnetic beads was used to separate the thrombocyte subpopulations from zebrafish whole blood. DiI-C18, a lipophilic dye, labels only young thrombocytes when used at low concentrations. Commercially available biotinylated anti-Cy3 antibody was used to label the chromophore of DiI-C18 on the young thrombocytes and streptavidin coated magnetic beads were added subsequently, to separate young thrombocytes. The remaining blood cells were probed with custom-made biotinylated anti-GPIIb antibody and streptavidin magnetic beads to separate them from other cells. Further, thrombocytes are equivalents of mammalian platelets. Platelets play a crucial role in thrombus formation. The G-protein coupled receptors (GPCRs) present on the platelet surface are involved during platelet activation and aggregation processes. So, thrombocytes were studied for the presence of GPCRs. The GPCR mRNA transcripts expressed in the young and mature thrombocytes were subjected to densitometry analysis and pixel intensities of the bands were compared using one way ANOVA. This analysis did not show significant differences between the young and mature GPCR mRNA transcripts but identified a novel GPCR, GPR18 that was not reported in platelets earlier. The Effect of Natural Gas Well Setback Distance on Drillable Land in the City of Denton, Texas

Municipalities protect human health and environmental resources from impacts of urban natural gas drilling through setback distances; the regulation of distances between well sites and residences, freshwater wells, and other protected uses. Setback distances have increased over time, having the potential to alter the amount and geographical distribution of drillable land within a municipality, thereby having implications for future land use planning and increasing the potential for future incompatible land uses. This study geographically applies a range of setback distances to protected uses and freshwater wells in the city limits of Denton, Texas to investigate the effect on the amount of land remaining for future gas well development and production. Denton lies on the edge of a productive region of the Barnett Shale geological formation, coinciding with a large concentration of drillable land in the southwestern region of the study area. This region will have the greatest potential for impacts to future municipal development and land use planning as a result of future gas well development and higher setback standards. Regulation of Receptors in Neuronal Cilia with Development, Seizures, and Knockouts: Implications for Excitability

Neurons commonly have a primary cilium, which is a non-motile organelle extending from the centrosome into the extracellular space. In most brain regions, neuronal cilia are enriched in either somatostatin receptor type 3 (SstR3) or melanin concentrating hormone receptor type 1 (MCHR1), or both. The present immunohistochemical study provides novel evidence that primary cilia regulate neuronal excitability via G-protein coupled receptors (GPCRs), and that their identity is governed by brain region and by competition, both in adulthood and in postnatal development. The hippocampus, which is particularly vulnerable to seizures, has opposing gradients of SstR3(+) and MCHR1(+) ciliary GPCRs. We hypothesized that there is a competition between these two ciliary GPCRs, which might take place on any level from gene expression to presence in the cilium. We examined whether receptor colocalization occurs transiently in development before ciliary GPCR dominance is established in neurons in the CNS. In postnatal CA1 and CA3, the first GPCR to appear in cilia was the one that will dominate in adults: MCHR1 in CA1 and SstR3 in CA3.
